In order to explore the mutagenicity and mutagenic process of GMA, plasmid pBR322 was used for in vitro binding, mutant screening, and restriction enzyme mapping. The binding between GMA and DNA in vitro has been verified by means of a spectrophotometric method. When pBR322 and GMAbound pBR322 were used to transform Eschenchia coli HB101, the following results were obtained: (1) The transformation efficiency of GMA-bound pBR322 was much lower than that of pBR322 alone. (2) GMA-bound pBR322 induced phenotype changes in competent cells (i.e., tetracycline-resistance inactivation or ampicillin-resistance inactivation). There were two mutants of pBR322, Ap~RTc~S and Ap~STc~R, in the transformants and a deductive mutant Ap~STc~S in the nontranstormants. (3) All of the selected mutants were stable and heritable. (4) When restriction enzyme maps were used to analyze the mutant Ap~RTc~S, four of seven maps were changed. some sites were shifted to other resistant gene regions, for example, sites of Bgll, EcoRl, Ilindlll. Hinclll, etc., and there was a new recognition site for Hindi (252). We did not observe any DNA fragment insertion or deletion on any maps. Our results suggest that when GMA is covalently linked to the plasmid DNA, it gives rise to a premutagenic lesion of DNA that is converted in vivo into a point mutation. Restriction maps in the regions of rDNA spacers were compared between giant panda( Ailuropoda melanoleuca ), lesser panda( Ailurus fulgens ), Asiatic black bear( Selenarctos thibetanus ), sun bear( Helarctos malayanus ), raccoon( Procyon lotor ) and lynx( Felis lynx ). Phylogenetic trees for these species were constructed using maximum likelihood and parsimony method. The results show that in respect to rDNA RFLPs, the giant panda is more closely related to bear than to lesser panda; while the lesser panda is slightly related to the raccoon.展开更多

Sequences from chimpanzee, orangutan and human were also amplified, but they were shorter than predicted. We were unable to amplify sequences from spider monkey, salmon, several rodents, bandicoot, sheep and pig . Analysis of the restriction map revealed several conserved RE sites in the amplified sequences of the primates. The results support the view that monkey has more close relationship with human than any other species at molecular level.展开更多
The present study deals with the monitoring for a real implementation of management policies in marine environments and the potential conflicts between professional and recreational fishery in the coastal areas of the...The present study deals with the monitoring for a real implementation of management policies in marine environments and the potential conflicts between professional and recreational fishery in the coastal areas of the Mediterranean Sea.A comprehensive database of fishing effort and fisheries infringements from professional small-scale and recreational fisheries was screened to identify hotspots areas of fishing pressure in the coastal zones of Ionian Sea.Mapping points showed that the number of the recorded infringements conducted both by professional and recreational fishers are too low(1 and 6 recorded infringements per 104 km2 of vessel days per year)and that fishing effort,and subsequently the recorded infringements,are not evenly distributed but concentrated on specific fishing grounds.These revealed high-risk areas prone to illegal fishing activities and are implying problems in the implementation of the fisheries regulations rather than a low delinquency of the fishers to comply with the rules.Findings represents a step forward in applying tracking technology to the surveillance of small-scale fishery and are crucial towards the specification of the critical zones for setting an efficient control system.展开更多

Methods A renin 2 gene restriction map was constructed by endonuclease digest ion and Southern blotting hybridization. Transgenic rats were produced via micro injection method.Results The 24 kb fragments containing mouse full length ren 2 and it s flanking sequence were cleaved by single enzymes (EcoRⅠ, KpnⅠ and BamHⅠ) an d combined enzymes (EcoRⅠ/KpnⅠ, KpnⅠ/BamHⅠ and BamHⅠ/EcoRⅠ), respectively. The digests were electrophoresed in 0.8% agarose plates and transferred onto NC membranes. Radioactive 735 bp and 1400 bp probes obtained from half and full l ength renin 1 cDNA were used in southern blotting hybridization. According to t he electrophoresis and hybridization patterns, a ren 2 restriction map was cons tructed. 1603 fertilized rat ova after injection with purified 24 kb renin 2 ge ne were implanted into the oviducts of 81 pseudopregnant recipients in about 2 0 ova per female rat. 306 progenies were obtained from 50 foster mothers (averag e of pregnancies was 56.6%). 248 survived pups were identified by PCR analys is and Southern hybridization, and eight positive rats were found to be the transgenic rats (founder, F). All of them carried long fragments (24 kb) of renin 2 gene with normal blood pressure. Preliminary breeding and screening were carried out in the founder. Total survival pups (17.8%) and overall efficiencies (1%) were h arvested as the same as those reported in the literatures. A systemic observatio n and the problems occurred during production of transgenic rats were also descr ibed besides the technique procedure used in this study.Conclusions Mapping of full length murine ren 2 can be used in invest igation of the structure and function of the gene. The results denoted that the ren 2 tran sgenic rats were successfully established in this study and the technique used i n the production of transgenic rats was proved to be valid in leading to wide s pread application of transgenic technique to many other related researches.展开更多
