Effects of Different Seasons on Cutting Propagation of Five Citrus Rootstocks

[Objectives]To explore the cutting propagation mode of citrus rootstock,improve the survival rooting rate of citrus rootstock,and provide theoretical guidance and technical reference for the rooting research of difficult-to-root plants such as citrus.[Methods]Five citrus rootstocks Citrus tangerina Tanaka‘Hongju’,Citrus haniana Hort‘Suanju’,Citrus limonia Osbeck‘Hongningmeng’,Citrus sinensis×Poncirus trifoliata‘Zhicheng’and Poncirus trifoliate(L)Raf.‘Zhike’were used as experimental materials to select the suitable cutting substrate for citrus rootstocks by measuring the physical properties of the substrate.Cutting was carried out in spring,summer,autumn and winter respectively.After cutting,the morphological changes of cuttings were observed regularly,and the callus rate,germination rate and rooting rate of cuttings were recorded.[Results]The best substrate for citrus rootstock cutting was peat soil,vermiculite and fine river sand(2:1:1).The callus of citrus rootstock in different cutting seasons began to appear in 10-22 d,and the callus rate reached 55%-100%.In terms of budding,the budding time was the earliest in summer and autumn,and slightly later in spring and winter;the germination rate of C.limonia Osbeck‘Hongningmeng’was the highest,and the germination rate of C.sinensis×P.trifoliata‘Zhicheng’and P.trifoliate(L)Raf.‘Zhike’was lower;in terms of rooting,C.limonia Osbeck‘Hongningmeng’had the earliest rooting time and the highest rooting rate and could reach 100%in all seasons;the rooting rate of C.tangerina Tanaka‘Hongju’was 50%-80%;the rooting rate of C.haniana Hort‘Suanju’was 60%-80%;C.sinensis×P.trifoliata‘Zhicheng’and P.trifoliate(L)Raf.‘Zhike’showed the earliest rooting time and the highest rooting rate in summer,and the latest rooting time and the lowest rooting rate in winter,which were only 14.5%.Therefore,different citrus rootstock varieties should choose the appropriate cutting time according to their own characteristics.[Conclusions]The results of this study can provide a scientific basis for a large number of cutting propagation of different citrus rootstocks,and have practical guiding significance for large-scale planting. 展开更多

Five Benzylaminopurine (BAP) concentrations were utilized for shoot induction—0.0 mg/L (Control), 0.5 mg/L, 1.0 mg/L, 1.5 mg/L, and 2.0 mg/L and five Indole Buteric Acid (IBA) concentrations—0.0 mg/L(Control), 0.5 mg/L, 1.0 mg/L, 1.5 mg/L, and 2.0 mg/l—were used for the induction of the root. The highest mean amount of shoots (eight) and length of the shoot (3.40 cm) were observed at a concentration of BAP of 0.5 mg/L. Festival also had the highest mean amount of leaves (6) when we used the identical concentration, while RABI-3 and Camarosa did the least well. The IBA of 0.5 mg/L concentration of rooting performed the best across all metrics tested among the five concentrations. The longest (3.3 cm) roots and most roots (7) were likewise obtained from this concentration in Festival. However, RABI-3 and Camarosa performed worse than Festival in the same concentration. Half-strength MS media without IBA concentration showed no response regarding root induction for each of the three cultivars. 展开更多

A study was done to produce a rapid in vitro propagation of three strawberry genotypes and tested in the field under Bangladeshi circumstances. Festival, RABI-3, and Neho strawberry genotypes’ runner tips were cultivated in vitro to induce root induction and multiple shoot proliferation. MS (Murashige and Skoog) media that were basally containing three different concentrations at 1.0, 1.5, and 2.0 of BA (6-benzyl adenine), KIN (6-furfuryl amino purine), or GA<sub>3</sub> (gibberellic acid) at 0.5 mg/L increasing tips of the runner was attained. The culture grew on the medium provided with 1.5 mg/L 6-benzyl adenine and 0.5 mg/L 6-furfuryl amino acid to increase shoot at the best level. Micro-cuttings were rooted on MS media at half strength combined with 0.5 mg/L - 1.5 mg/L IBA (indole butyric acid) or IAA (indole acetic acid). IBA attained 4 - 9 roots and 91% - 96% rooting at 1.0 mg/L. The resulting plantlets grew into hardy plants and took root in the earth. The rooting efficiency index (REI) showed a positive correlation with the rooting rate. In this study we investigated the rooting ability and the growth performance of juvenile single-node leafy stem cuttings of Litsea monopetala (Roxb) Pers. collected from two mature mother trees preserved in the hill forest of Chittagong district, Bangladesh. The rooting ability of cuttings was studied under 0%, 0.1%, 0.2% and 0.4% indole-3-butyric acid (IBA) treatments. Significantly better rooting response (p ≤ 0.05) was observed with 0.1% IBA compared to control (0% IBA). The mean number of roots and the length of the longest root of cuttings in different treatments showed no significant difference (p≤0.05). After transfer into polythene bags from non-mist propagator, rooted cuttings treated with 0%, 0.1% and 0.2% IBA demonstrated the highest (100 ± 0.00%) survival capacity. The mean number of shoots developed in cuttings in the polythene bags in first three weeks varied significantly (p≤0.05) among the treatments. Effects of three fertilizer treatments, viz. T0 (no fertilizer), T1 (10g Urea, 20g TSP, 10g MOP dissolved in 1 L water) and T2 (10g Urea, 20g TSP, 10g MOP dissolved in 2 L water) on initial growth ofstecklings were also measured over a 90-days period. The increment of leaf area of stecklings was significantly higher (p≤0.05) under T0 compared with that under T1 and T2 while the increment of stem length, collar diameter and root biomass varied insignificantly among different fertilizer treatments. The results suggest that rooting juvenile single-node leafy stem cuttings could be an effective mean of regenerating L. monopetala. The application of 0.1% IBA concentration is recommended for rooting of juvenile leafy stem cuttings and application of fertilizer appeared unnecessary for the subsequent growth of stecklings in polythene bags. 展开更多

In this study a reliable protocol was developed for the establishment of commercial in vitro cultures of Tripterygium wilfordii Hook f.. Juvenile shoots from one-year-old elite plants were used as the source of explants. New axillary shoots were obtained after 30 days of culture on a MS medium supplemented with BAP (2.0 mg.L^-1) and NAA (0.1 mg.L^-1). The optimal multiplication medium was a modified MS medium supplemented with BAP (1.0 mg.L^-1) and NAA (0.1 mg.L^-1). This yielded a multiplication rate of 2.4 for each subculture. Slightly more than 92% of shoots rooted when cultured on a modified MS medium containing IBA (0.2 mg.L^-1) and activated charcoal (0.5 mg.L^-1). Activated charcoal promoted both a strong and a high rooting rate during the rooting phase. Plantlets were transferred to pots for a short acclimatization stage in a greenhouse where 95% of the plantlets survived. This highly reproducible procedure can be adopted for large-scale propagation of T. wilfordii. 展开更多

Bambusa vulgaris Schrad ex wendl is a widely cultivated bamboo species in rural Bangladesh for its versatile uses. The vegetative propagation becomes the only viable alternative for this species because B. vulgaris does not set seed after sparse flowering, which makes seedling progenies unavailable. A low-cost propagation trial was conducted to explore the clonal propagation techniques for the species with two types of small branch cuttings, nodal leafy cuttings and tip cuttings. The cuttings, after treating with 0, 0.1%, 0.4%, and 0.8% IBA solutions, were kept in non-mist propagator to let them to root for assessing the rooting ability. The cuttings were rooted in four weeks and were allowed to grow in the polybags for 10 months under nursery condition to assess their steckling capacity. The study reveals that both types of branch cuttings are able to develop roots, shoots, to survive and to form rhizome under the nursery condition. Rooting ability of the cuttings was significantly enhanced by the application of rooting hormone - IBA. The highest rooting percentage in nodal leafy cuttings and the tip cuttings (56.67% and 51.0%, respectively) were observed in 0.8% IBA treatment, followed by 0.4% IBA and the lowest (34.3% and 30.0%, respectively) was in control. The highest number of root developed per cutting (9.77 and 8.33 in nodal leafy cuttings and the tip cuttings, respectively) was also obtained from the cuttings treated with 0.8% IBA solution, followed 0.4% IBA treatment and the lowest (3.1 and 2.1, respectively) was in the cuttings without treatment. However, the length of the longest root varied significantly neither with the cutting types nor the concentrations of IBA solution. Survival percentage of the stecklings in nursery condition was significantly enhanced by IBA. 展开更多

Techniques have been developed for the vegetative propagation of Tetraena mongolica,a rare and endangered species endemic to the western Ordos Plateau in the northcentral portion of China.The purpose of the study was to investigate the rooting characteris-tics of the plant species from the aspects of length and ages of cuttings,external hormone concentration,leaf retention,cutting orientations,and cutting positions.The results obtained from the experiments indicated that the rooting ratio of 2-year or 3-year old cuttings was larger than more than of 4-year old cuttings.The influence of external hormone concentration on rooting was ob-vious;the rooting ratio of cuttings was increased by external hormone treatment,especially when treated by ABT1(ABT No.1 Rooting Powder) 50 mg/L for 3 hours,in which case,the rooting ratio reached 91 percent.Leaf retention influenced rooting sig-nificantly;the rooting rate of cuttings became greater as the leaf retention increased.There was no significant difference in rooting capability between south and north orientations of the ortets.The rooting ratio of the cuttings obtained from the distal portions of shoots was significantly larger than those obtained from the middle and the basal portions of the shoots.On day three after treat-ment,calluses started appearing at the incisions of some cuttings dipped in 500 mg/L IAA(indole acetic acid).Most cuttings were healed or swelled,and 6 percent of cuttings rooted on day 6 after treatment.Higher rooting period occurred 7-10 days after the experiment was set up. 展开更多

The present study investigated the rooting ability and growth performance of juvenile single-node leafy stem cuttings of Holarrhena pubescens, collected from two mature mother trees preserved in the hill forests of Chittagong District, Bangladesh. The rooting ability of cuttings was studied with 0, 0.1%, 0.2% and 0.4% indole-3-butyric acid (IBA) treatments. The better rooting response was observed with 0.1% IBA, but was insignificant compared to other treatments. The mean number of roots and length of the longest root of cuttings in different treatments showed no significant difference. After transfer to the polythene bags from a nonmist propagator, rooted cuttings treated with 0.4% IBA demonstrated the highest (50.00± 0.78%) survival capacity. Regarding initial growth of established rooted cuttings (stecklings) over a 90 d period, the increment of stem length of stecklings under T2 (10 g Urea, 20 g TSP, 10 g MOP dissolved in 2 L water) and leaf area expansion of stecklings under T1 (10 g Urea, 20 g TSP, 10 g MOP dissolved in 1 L water) was significantly higher (p 〈 0.05) compared with that under TO (no fertilizer). The increment of collar diameter and root biomass production varied insignificantly among different fertilizer concentrations, although stecklings under T2 showed the better performance. The results suggest that rooting juvenile single-node leafy stem cuttings can be an effective mean of regenerating H. pubescens. The application of 0.1% IBA or without IBA is recommended for rooting juvenile leafy stem cuttings. Application of fertilizer appears necessary for subsequent growth of stecklings in polythene bags. 展开更多

To solve the problems of microbial contamination and no rooting during the tissue culture of Huperzia serrata, wild seedlings of Huperzia serrata were cultured by indoor hydroponics or soil cultivation methods, then the stem tips were used in tissue culture. The above operation significantly reduced microbial contamination during tissue culture process. Different types and concentrations of hormones were added into the basic medium MS to screen the optimal formula for induced plantlet growth and rooting. It was found that 1/2MSCO was the best medium on which stem, leaves and roots of tissue culture plantlets of Huperzia serrata grew quickly. At the same time, sand planting was a convenient and effective approach for gemmae propagation of Huperzia serrata, the survival rate of seedlings germinated from gemmae was nearly 100%. 展开更多

Vitis aestivalis “Norton/Cynthiana” is a grape native to North America and utilized in the commercial wine industry. The purpose of this research was to determine the appropriate time of year and/or time after last freeze that will yield the most successful propagation rate for Vitis aestivalis. It was hypothesized that the highest rate of propagation would be in early summer. During a year-long study (2015-2016), forty cuttings were taken semi-monthly from a vineyard, treated with 0.1% IBA and placed in a plant growth room set to spring conditions. Daily temperatures were recorded every day as well. In a follow-up study, cuttings were taken weekly from early spring before vine budding until late fall (2017) and treated as previous. After six weeks, cuttings were evaluated for root production. In both studies, rooting success rates were the highest in the month of June at 10.0% - 27.5%, with rates less than 7.5% for all other sampling dates. It was determined that the best time to propagate Vitis aestivalis is in June, 10 - 12 weeks after the last temperature below 0°C or eight to ten weeks after the first budding. 展开更多

Significant differences (P < 0.005) in results are obtained in the stages of initiation, multiplication, and rooting according to their response to the various concentrations of different growth regulators examined, namely BA, IBA, AG</span><sub><span style="font-family:Verdana;">3</span></sub><span style="font-family:Verdana;">. The use of 0.5 mg/l BA and 0.2 mg/l IBA was the most favorable for shoots neoformation. The leafy shoots are propagated in MS medium with BA at a concentration of 1.5 mg/l. The addition of gibberellic acid at 0.2 mg/l in the culture medium allows a good elongation and development of the shoots of the carob. The effect of the age of the plant material used has shown that explants taken from mature carob trees have a low capacity for bud sprouting and shoot proliferation compared to those taken from juvenile trees. Rooting has been successful when the plant material used is taken from young trees on an MS medium containing 2 mg/IBA, with an average number of 3 to 4, roots 1 to 2 cm long, then for the adult material, no rooting was observed. Based on these tests, it appears that micropropagation of the carob from the axillary buds is feasible, but additional work must be done to root this recalcitrant material.</span> 展开更多

The study describes the scope of clonal propagation of guava (Psidium guajava Linn.) by stem cutting collected from mature stockplants. Cuttings were treated with 0, 0.2%, 0.4% and 0.8% IBA solution and rooted in the non-mist propagator. Rooted cuttings were allowed to grow in the polybags filled with soil and cow-dung mixed in the ratio of 3: 1 (by volume) for three months to assess the steckling capacity and initial growth performance. The study reveals that the species is amenable for clonal propagation by mature stem cutting. The highest rooting percentage (60%) was observed in the cuttings treated with 0.4% IBA solution followed by 0.2% IBA and the lowest was in controlled cuttings. The maximum number of primary root (32.7) was developed in the cuttings treated with 0.8% IBA solution followed by 0.4% IBA and the lowest was in the cuttings without IBA treatment. The highest survival percentage (70.9) was observed in the cuttings rooted with 0.4% IBA treatment and the lowest (58.3) was in the cuttings without any treatment. In order to realize the efficient industrial production of <i>Ilex</i> “China Girl”, an orthogonal experiment with 4 factors and 3 levels was designed. Firstly, the optimal orthogonal cutting scheme was selected from 9 treatments. Then through the systematic analysis of the effects of cutting position, substrate, exogenous hormones type and concentration on rooting indexes, such as rooting rate, root number, root length and root effect index, the theoretical optimal scheme was predicted and verified. The results showed that the theoretical optimal scheme (3000 mg/L IBA treatment for 15 s, cutting in mixed matrix with peat soil, perlite and vermiculite ratio of 2:1:1) was the optimal <span>cutting rooting scheme of <i>Ilex</i> “China Girl”. After the treatment of this sc</span>heme, the rooting rate of <i>Ilex</i> “China Girl” reached 100%, the average root number was 51.67 per plant, and the average root length was 6.13 cm. The rooting time was greatly shortened, the rooting rate and rooting effect were greatly improved. In this study, the efficient cutting propagation technology system of <i>Ilex</i> “China Girl” was established, which laid a foundation for the popularization and application of <i>Ilex</i> “China Girl”, and also provided reference for further improving the cutting propagation efficiency of other evergreen holly. To explore the establishment of a tissue culture and rapid propagation system of Tilia amurensis,the effects of basic medium and concentrations and ratios of plant growth regulators on tissue culture and rapid propagation of T.amurensis were studied.The results showed that 1/2 MS medium was the most suitable proliferation medium,and the proliferation coefficient could reach 13.5 after adding 0.05 mg/L 6-BA and 0.03 mg/L IBA;MS medium was the most suitable medium for strong plantlets and rooting,and the best medium for strong plantlets was MS+0.1 mg/L 6-BA+0.1 mg/L IBA+0.03 mg/L GA_(3),with which the average plantlet height reached 5.15 cm;and the best rooting medium was MS+1.0 mg/L6-BA+0.05 mg/L NAA,with which the rooting rate was 93.3%and the number of roots was 5.7 roots. 展开更多

Panax pseudoginseng Wall., a highly medicinal, herbaceous, long-lived plant, grows in the forest litter of shady primary forest. The species is threatened in the natural habitat due to unsustainable harvesting of rhizome for medicine and habitat destruction. The species has very poor adaptive power to the synthetic environment and fails to grow and propagate. Present study was undertaken to develop suitable low cost propagation technique and produce clonal planting materials through rhizome splitting and root cutting. Rhizomes and roots were cut into segments and sowed in the raised soil bed prepared by mixing decayed wood powder, sand and top black soil at 1:1:3 ratios in a shaded poly house. Of the sowed rhizome segments ~55% segments remained recalcitrant to morphogenetic response in the first year of the study and the response improved in the subsequent years. In the third year ~51% rhizome segments responded positively. The horizontal root cuts mostly remained morphogenetically dormant but inclined root cuts exhibited better response. The morphogenetic response from the roots was comparatively very poor compare to rhizome segments. The plantlets formed both rhizome segments and root cuttings were maintained for three years in the bed/pots followed by transferring in the cultivated plots. 展开更多

Propagation models are the foundation for radio planning in mobile networks. They are widely used during feasibility studies and initial network deployment, or during network extensions, particularly in new cities. They can be used to calculate the power of the signal received by a mobile terminal, evaluate the coverage radius, and calculate the number of cells required to cover a given area. This paper takes into account the standard K factors model and then uses the Ion motion optimization (IMO) algorithm to set up a propagation model adapted to the physical environment of each of the Cameroonian cities of Yaoundé and Bertoua for different frequencies and technologies. Drive tests were made on the CDMA network in the city of Yaoundé on one hand and on an LTE TDD network in the city of Bertoua on the other hand. IMO is used as the optimization algorithm to deduct a propagation model which fits the environment of the two considered towns. The calculation of the root-mean-square error (RMSE) between the actual data from the drive tests and the prediction data from the implemented model allows the validation of the obtained results. A comparative study made between the RMSE value obtained by the new model and those obtained by the Okumura-Hata and K factors standard models, allowed us to conclude that the new model obtained in each of these two cities is better and more representative of our local environment than the Okumura-Hata currently implemented. The implementation shows that IMO can perform well and solve this kind of optimization problem;the newly obtained models can be used for radio planning in the cities of Yaounde and Bertoua in Cameroon. 展开更多

【目的】研究灵武长枣根蘖繁殖与嫁接繁殖果实品质差异的内在机理,为提高其果实品质提供基础数据。【方法】以灵武长枣根蘖繁殖与嫁接繁殖白熟期(NB)、着色期(NZ)、成熟期(NC)和嫁接繁殖白熟期(JB)、着色期(JZ)、成熟期(JC)的果实为试... 【目的】研究灵武长枣根蘖繁殖与嫁接繁殖果实品质差异的内在机理,为提高其果实品质提供基础数据。【方法】以灵武长枣根蘖繁殖与嫁接繁殖白熟期(NB)、着色期(NZ)、成熟期(NC)和嫁接繁殖白熟期(JB)、着色期(JZ)、成熟期(JC)的果实为试验材料,采用高通量测序技术进行果实转录组测序,筛选与根蘖和嫁接繁殖的灵武长枣果实品质差异相关的基因,初步探究根蘖和嫁接繁殖灵武长枣果实不同时期基因表达差异情况。【结果】18个灵武长枣样品经过转录组测序,得到质控数据138.26 GB,将其比对到参考基因组冬枣上,18个样品的Cleans reads与参考基因组比对效率在93.77%以上。主成分分析结果显示,3次生物学重复样品聚集程度较高且组间存在分离现象,说明其基因表达量存在差异。差异分析结果表明,灵武长枣果实发育各时期组间分析对比中,果实白熟期(JB-vs-NB)、着色期(JZ-vs-NZ)和成熟期(JC-vs-NC)各组间分别筛选出65、4 509和1 534个差异基因。GO功能富集到生物过程、分子功能和细胞组分三大类中的50个亚类中,KEGG通路注释将差异基因显著富集在次生代谢物生物合成、代谢途径、氨基酸生物合成等通路中,成熟期差异基因富集在糖酸相关通路的显著性较高。【结论】在淀粉和蔗糖代谢、半乳糖代谢、氨基糖和核苷酸糖代谢,乙醛酸和二羧酸代谢、丁酸代谢5条与果实可溶性糖和总酸含量相关的代谢途径中筛选出了ncbi_107424266、ncbi_107414286、ncbi_107425230、ncbi_107429837等31个差异基因,分别在果实发育不同时期的糖酸代谢途径中差异表达,调控各时期果实中可溶性糖及总酸含量。 展开更多
