Objective To investigate miR-183-5p targeting to forkhead box protein O1(FOXO1)and its corresponding effect on the proliferation,migration,invasion,and epithelial-mesenchymal transition(EMT)of non-small cell lung canc...Objective To investigate miR-183-5p targeting to forkhead box protein O1(FOXO1)and its corresponding effect on the proliferation,migration,invasion,and epithelial-mesenchymal transition(EMT)of non-small cell lung cancer(NSCLC)cells.Methods NSCLC tissues and adjacent normal tissues from 60 patients with NSCLC adenocarcinoma were obtained via pathological biopsy or intraoperative resection.Several cell lines were cultured in vitro,including the human normal lung epithelial cell line BEAS-2B and human NSCLC cell lines A549,SPCA-1,PC-9,and 95-D.miR-183-5p and FOXO1 mRNA expression in tissues and cells were detected by qRT-PCR;the corresponding correlations in NSCLC tissues were analyzed using the Pearson test,and the relationship between miR-183-5p expression and clinicopathological parameters was analyzed.The miR-183-5p-mediated regulation of FOXO1 was verified by bioinformatics prediction alongside double luciferase,RNA-binding protein immunoprecipitation(RIP)assay,and pull-down experiments.A549 cells were divided into control,anti-miR-NC,anti-miR-183-5p,miR-NC,miR-183-5p,miR-183-5p+pcDNA3.1,and miR-183-5p+pcDNA3.1-FOXO1 groups.Cell proliferation,invasion,migration,apoptosis,and cell cycle distribution were detected using an MTT assay,clone formation assay,Transwell assay,scratch test,and flow cytometry,respectively.The expression of EMT-related proteins in the cells was analyzed by western blotting.The effect of miR-185-3p silencing on the development of transplanted tumors was detected by analyzing tumor formation in nude mice.Results miR-183-5p expression was significantly higher in NSCLC tissues and cells than in adjacent normal tissues,whereas FOXO1 mRNA expression was significantly down-regulated.There was a significant negative correlation between miR-183-5p and FOXO1 mRNA in NSCLC tissues(P<0.05).Additionally,the expression of miR-183-5p was significantly correlated with tumor size,tumor differentiation,and tumor-node-metastasis stage in patients with NSCLC(P<0.05).miR-183-5p targeted and inhibited FOXO1 expression.Compared to the anti-miR-NC group,the cell proliferation,scratch healing rate,N-cadherin and vimentin protein expression,and the proportion of S phase cells were significantly lower in the anti-miR-183-5p group,whereas the protein expression of E-cadherin andα-catenin and the proportion of G0/G1 phase cells were significantly higher;additionally,the frequency of colony formation and invasion were significantly lower in the anti-miR-183-5p group(P<0.05).Compared to the miR-NC group,the cell proliferation,scratch healing rate,N-cadherin and vimentin protein expression,and the proportion of S phase cells in the miR-183-5p group were significantly higher,whereas the E-cadherin andα-catenin protein expression and the proportion of G0/G1 phase cells were significantly lower;furthermore,the frequency of colony formation and invasion were significantly higher in the miR-183-5p group(P<0.05).Compared with the miR-183-5p+pcDNA3.1 group,the OD value,scratch healing rate,N-cadherin and vimentin protein expression,and the proportion of S phase cells were significantly lower in the miR-183-5p+pcDNA3.1-FOXO1 group,whereas E-cadherin andα-catenin protein expression and the proportion of G0/G1 phase cells were significantly higher;additionally,the frequency of colony formation and invasion was significantly lower in the miR-183-5p+pcDNA3.1-FOXO1 group(P<0.05).Overall,silencing miR-185-3p inhibited the growth of transplanted tumors and promoted FOXO1 expression.Conclusion Overexpression of miR-183-5p can inhibit apoptosis and promote the proliferation,migration,invasion,and EMT,of NSCLC cells by down-regulating FOXO1 expression.展开更多
Recent bathymetric changes in the Changjiang Estuary under the influence of artificial regulation engineerings and basin reservoirs have been analyzed based on the maritime charts since 1997 and recent fieldworks. The...Recent bathymetric changes in the Changjiang Estuary under the influence of artificial regulation engineerings and basin reservoirs have been analyzed based on the maritime charts since 1997 and recent fieldworks. The results indicate a slight erosion of the channels in the upper and middle estuary, continuing deposition and seaward move of the mouth bar crest and intensifying erosion at the nearshore seabed. It is noteworthy that the morphological evolution caused by intensive human activities dominates over the changes from nature process. First, the riverbes are eroded overall in the South Branch (SB), the South Channel (SC) and the upper and middle reaches of the North Channel (NC). The nearshore seabed outside the river mouth is being eroded slightly, which is attributed to the declining sediment supply from the Changjiang Basin due to the construction of the Three Gorges Dam upstream. The sediment above the seabed is very active and coarsened, meanwhile, sand waves are becoming more distinct. Second, a deposition occurs in the North Brach (NB), the mouth of the NC, the mouth bars of the North Passage (NP) and the South Passage (SP) and especially the main channel of the NP, where it shows a massive siltation after the deep waterway project. The reasons for the recent changes are not only the dynamic structure in estuarine mouth bars, but also the supply of sediment resuspension in a local and offshore area. Meanwhile, the severe erosion and siltation in some reaches is related to the construction of estuarine engineerings. It is indicated that the Changjiang Estuary is gradually self-adjusted and adapting to the varying natural factors and intensive human activities. The study on the mechanism of self-regulation of the recent bathymetric changes in the Changjiang Estuary has important and practical significance.展开更多

The effect of hypertension in the elderly is higher than other age groups and it may lead to changes in their quality of life. By the increase of age, the amount of self-care behaviors of patients with hypertension decreased. Therefore, control and self-care behaviors are so important. This study has been conducted to study self-regulation behaviors and Associated Factors in elderly patients with hypertension. This study was a cross sectional, descriptive analytic study, which was conducted using multistage sampling on the elderly 60 years and above with hypertension. The sample size was calculated 365 and the final sample size of 380 people was considered by possible losses. The instruments include of demographic and self-regulatory behaviors questionnaire. The results showed that self-regulation of behavior was 42.3% of the total score. In terms of quality, only 7.4% of the elderly had good behavior, 46.6% of poor performance, and the rest were mediocre performance. Age (p = 0.03), gender (p = 0.02), education (p = 0.001), income level (p = 0.03) and marital status (p = 0.001) significantly associated with behaviors of self-regulation. The results generally indicate unfavorable self-regulation of elderly patients with hypertension. Some variables such as gender, age, marital status, education and income levels were associated with these behaviors. So paying attention to the education of the elderly in care centers is essential.展开更多

Different water-transmitting coating fibers were prepared by coating fiber using coating material with different mass proportions of additives to adhesive. And the coating materials were made from three kinds of inorganic clays as additives respectively and polyvinyl alcohol (PVA) as adhesive. Furthermore, the surface morphology and water-transmitting capacity of coating fiber were studied by SEM, Perkin Elmer Diamond SII thermal multi-analyzer and instrument for quick measurement moisture M30. The experimental results indicate that water-transmitting coating fibers made from three kinds of clays all have water-transmitting capacity. The surface of water-transmitting coating fiber prepared by common clay T is continuous and compact, and the water-transmitting effect is better than coating fibers made from other clays.展开更多

It has been shown that immersion into a virtual competitive game, controlled by physiological responses, causes a wide involvement of the cortices, characterized by a high volume of activation in the mid-temporal, occipital and frontal areas, the cuneus and the precuneus. Objective: The current study aimed to investigate the relationship among personality traits and self regulation among different drug abuse cases. The three categories of drug abuse were based on phases of addiction na...Objective: The current study aimed to investigate the relationship among personality traits and self regulation among different drug abuse cases. The three categories of drug abuse were based on phases of addiction namely current, relapse and remitted cases. Method: The sample consisted of 108 drug abusers taken from different rehabilitation centers of Rawalpindi and Islamabad. The selected drug abusers were at current (n = 40), relapsed (n = 42) and remission phase (n =2 7) of drug addiction;with an age range of 20 to 70 years. The data were collected from private rehabilitation centers of Rawalpindi and Islamabad. In this study, Eysenck Personality Questionnaire Revised EPQR-Short (Eysenck, Eysenck & Barrett, 1985) was used to measure personality traits of the drug abuse cases. Self Regulation Questionnaire (SRQ) developed by Miller and Brown (1991) was used to measure the self regulatory skills. The data were collected from rehabilitation centers. Before collection of data, formal permission was taken from the administration of these centers. Research protocol included an Ethical Proforma explaining rationale of study, ensuring confidentiality of the information to be used for research purpose only and also had personal consent form. In the first phase, pilot study was conducted to check the reliability of translated scale and availability of sample. A sample of 15 drug abusers was selected for pilot study. In the next step (main study);protocol was administered on current, relapsed and remitted cases through individual administration. For remitted cases, the researcher consulted authorities (administration head, and medical doctors) of centers and administered protocol on the days when these participants came for follow-up session. Results: The results of the study indicate strong correlation between self regulation and personality traits (r = 0.27**). Furthermore, most of the participants scored higher on Psychoticism (100%), Introversion (75%) while Neuroticism (58%) trait was found less among drug abuse cases. The study also indicates strong correlation between self regulation and Extraversion-Introversion personality trait (r = 0.20**) while other traits did not have significant correlations with self regulation. Furthermore, the descriptive frequencies reveal that most of the participants scored higher on Psychoticism (100%), Introversion (75%) while Neuroticism (58%) trait was found less among drug abuse cases. Conclusion: From the above mentioned findings, it is concluded that self regulation is associated with personality traits among drug abusers. The comparisons of personality traits and self regulatory skills among the different phases of addiction provide an insight into how these traits interact with self control to regulate impulsive and addictive behaviors.展开更多

The activated carbons were prepared with the temperature rising rates of 5, 10, 15, 20 ℃/min and constant temperatures of 600, 700, 800, 900, 1 000, 1 100, 1 200, 1 300, 1 400, and 1 500 ℃ for 0.5, 1, 2, 3, and 6 h, respectively.And then their structures such as specific surface areas, volumes of pores from 3.7-7.1 nm and average pore diameters were tested. Various humidity conditions were simulated and their self-regulating humidity properties such as absorption/desorption contents, rates and stabilities were evaluated. The results show that the specific surface area, volume of pores from 3.7-7.1 nm and average pore diameter of activated carbon are closely nonlinearly related to the temperature rising rate, temperature and holding time. In general, the activated carbon prepared with the temperature rising rate of 5 ℃/min and kept at 1 100 ℃ for 2 h has the highest absorption/desorption content and rate, the lowest decay factor and then the best stability at all humidity for the fact that it is provided with proper average pore diameter, higher specific surface area and volume of pores from 3.7-7.1 nm. The activated carbons can be prepared with the temperature rising rate of 5 ℃/min and kept at 1000, 900,1 200, 1 300, 1 400, 1 500, 800, 700, and 600 ℃ respectively for 2 h. These resulted properties are attributed to their different average pore diameters and volumes of pores from 3.7-7.1 nm.展开更多

Mitogen-activated protein kinases(MAPKs)are a family of proteins that constitute signaling pathways involved in processes that control gene expression,cell division, cell survival,apoptosis,metabolism,differentiation ...Mitogen-activated protein kinases(MAPKs)are a family of proteins that constitute signaling pathways involved in processes that control gene expression,cell division, cell survival,apoptosis,metabolism,differentiation and motility.The MAPK pathways can be divided into conventional and atypical MAPK pathways.The first group converts a signal into a cellular response through a relay of three consecutive phosphorylation events exerted by MAPK kinase kinases,MAPK kinase,and MAPK.Atypical MAPK pathways are not organized into this three-tiered cascade.MAPK that belongs to both conventional and atypical MAPK pathways can phosphorylate both non-protein kinase substrates and other protein kinases.The latter are referred to as MAPK-activated protein kinases.This review focuses on one such MAPK-activated protein kinase,MAPK-activated protein kinase 5(MK5)or p38-regulated/activated protein kinase(PRAK).This protein is highly conserved throughout the animal kingdom and seems to be the target of both conventional and atypical MAPK pathways.Recent findings on the regulation of the activity and subcellular localization,bona fide interaction partners and physiological roles of MK5/PRAK are discussed.展开更多
BACKGROUND Pancreatic ductal cancer(PDAC)has high malignancy and poor prognosis.Long noncoding RNAs(lncRNAs)are associated with high levels of malignancy,including PDAC.However,the biological and clinical significance...BACKGROUND Pancreatic ductal cancer(PDAC)has high malignancy and poor prognosis.Long noncoding RNAs(lncRNAs)are associated with high levels of malignancy,including PDAC.However,the biological and clinical significance of negative regulator of antiviral response(NRAV)in PDAC is unclear.AIM To study the regulatory role of lncRNA NRAV in PDAC.METHODS GEPIA analyzed lncRNA NRAV and miRNA(miR-299-3p)expression levels in PDAC tissues and measured them in PDAC cells by quantitative measurements in real time.The specific role of NRAV and miR-299-3p in cell proliferation and transfer potential was evaluated by cell formation analysis,Cell Counting Kit-8 and Transwell analysis.The relationship between NRAV and miR-299-3p was studied by predictive bioinformatics,RNA immunoassay,and fluorescence enzyme analysis.In vivo experiments included transplantation of simulated tumor cells under naked mice.RESULTS The expression level of lncRNA NRAV was higher in both tumor tissues and cell lines of PDAC and was negatively associated with the clinical survival of PDAC patients.Functionally,overexpression of NRAV promoted cell proliferation and metastasis of PDAC cells,while knockdown of NRAV reversed these effects.Finally,NRAV was performed as a molecular sponge of miR-299-3p.Moreover,overexpression of miR-299-3p could reverse the promoting effects of NRAV on cell proliferation and metastasis of PDAC cells.CONCLUSION NRAV facilitates progression of PDAC as a molecular sponge of miR-299-3p and may be a potential molecular marker for diagnosis and treatment of PDAC.展开更多

The structure of compound 3 was characterized by ~1H NMR, 13 C NMR, HRMS and single-crystal X-ray diffraction. The compound crystallizes in the monoclinic system, space group P21/n with a = 5.7227(8), b = 27.924(4), c = 6.2282(7) ?, β = 101.892(11)°, V = 973.9(2) ?~3, Z = 4, T = 180.00(10) K, μ(MoKα) = 0.087 mm^(-1), Dc = 1.243 g/cm^3, 3832 reflections measured(3.648≤θ≤26.022°), 1916 unique reflections(Rint = 0.0359, Rsigma = 0.0572) used in all calculations. The final R = 0.0557(I 〉 2σ(I)) and w R = 0.1276(all data). Bioassay showed that 3 displayed excellent activity as plant growth regulator with inducing lateral root formation and enhancing primary root elongation at 0.27 mmol/L(50 ppm) in soybeen(He Feng-50). Good water solubility was found with 50 mg in 1 m L of water. Therefore, application of 3 in agriculture is more environmentally friendly due to cosolvent-free condition, and results in improved abiotic-stress tolerance by affecting the root growth. Human adipose-derived stem cells(hASCs)are a promising cell type for bone tissue regeneration.Circular RNAs(circRNAs)have been shown to play a critical role in regulating various cell differentiation and involve in me...Human adipose-derived stem cells(hASCs)are a promising cell type for bone tissue regeneration.Circular RNAs(circRNAs)have been shown to play a critical role in regulating various cell differentiation and involve in mesenchymal stem cell osteogenesis.However,how circRNAs regulate hASCs in osteogenesis is still unclear.Herein,we found circ_0003204 was significantly downregulated during osteogenic differentiation of hASCs.Knockdown of circ_0003204 by si RNA or overexpression by lentivirus confirmed circ_0003204 could negatively regulate the osteogenic differentiation of hASCs.We performed dual-luciferase reporting assay and rescue experiments to verify circ_0003204 regulated osteogenic differentiation via sponging miR-370-3p.We predicted and confirmed that miR-370-3p had targets in the 3′-UTR of HDAC4 m RNA.The following rescue experiments indicated that circ_0003204 regulated the osteogenic differentiation of hASCs via miR-370-3p/HDAC4 axis.Subsequent in vivo experiments showed the silencing of circ_0003204 increased the bone formation and promoted the expression of osteogenic-related proteins in a mouse bone defect model,while overexpression of circ_0003204 inhibited bone defect repair.Our findings indicated that circ_0003204 might be a promising target to promote the efficacy of hASCs in repairing bone defects.展开更多

The sediment above the seabed is very active and coarsened, meanwhile, sand waves are becoming more distinct. Second, a deposition occurs in the North Brach (NB), the mouth of the NC, the mouth bars of the North Passage (NP) and the South Passage (SP) and especially the main channel of the NP, where it shows a massive siltation after the deep waterway project. The reasons for the recent changes are not only the dynamic structure in estuarine mouth bars, but also the supply of sediment resuspension in a local and offshore area. Meanwhile, the severe erosion and siltation in some reaches is related to the construction of estuarine engineerings. It is indicated that the Changjiang Estuary is gradually self-adjusted and adapting to the varying natural factors and intensive human activities. The study on the mechanism of self-regulation of the recent bathymetric changes in the Changjiang Estuary has important and practical significance.
文摘Hypertension is one of the most important risk factors for heart diseases today. The effect of hypertension in the elderly is higher than other age groups and it may lead to changes in their quality of life. By the increase of age, the amount of self-care behaviors of patients with hypertension decreased. Therefore, control and self-care behaviors are so important. This study has been conducted to study self-regulation behaviors and Associated Factors in elderly patients with hypertension. This study was a cross sectional, descriptive analytic study, which was conducted using multistage sampling on the elderly 60 years and above with hypertension. The sample size was calculated 365 and the final sample size of 380 people was considered by possible losses. The instruments include of demographic and self-regulatory behaviors questionnaire. The results showed that self-regulation of behavior was 42.3% of the total score. In terms of quality, only 7.4% of the elderly had good behavior, 46.6% of poor performance, and the rest were mediocre performance. Age (p = 0.03), gender (p = 0.02), education (p = 0.001), income level (p = 0.03) and marital status (p = 0.001) significantly associated with behaviors of self-regulation. The results generally indicate unfavorable self-regulation of elderly patients with hypertension. Some variables such as gender, age, marital status, education and income levels were associated with these behaviors. So paying attention to the education of the elderly in care centers is essential.

文摘Common clay, Kaolin and Bentonite were used as additives to prepare water-transmitting coating fiber, respectively, and the water-transmitting characteristic of coating fiber was studied. Different water-transmitting coating fibers were prepared by coating fiber using coating material with different mass proportions of additives to adhesive. And the coating materials were made from three kinds of inorganic clays as additives respectively and polyvinyl alcohol (PVA) as adhesive. Furthermore, the surface morphology and water-transmitting capacity of coating fiber were studied by SEM, Perkin Elmer Diamond SII thermal multi-analyzer and instrument for quick measurement moisture M30. The experimental results indicate that water-transmitting coating fibers made from three kinds of clays all have water-transmitting capacity. The surface of water-transmitting coating fiber prepared by common clay T is continuous and compact, and the water-transmitting effect is better than coating fibers made from other clays.
文摘Functional MRI was used to map the brains of subjects on-line during the process of media training for the acquisition and improvement of self-regulation mechanisms. The temporal and spatial dynamics of the new neural network formation were studied in real and simulated (false) biofeedback game, and their qualitative characteristics were discussed. It has been shown that immersion into a virtual competitive game, controlled by physiological responses, causes a wide involvement of the cortices, characterized by a high volume of activation in the mid-temporal, occipital and frontal areas, the cuneus and the precuneus. In both forms of media training, high values of activation volume were identified in the cerebellar structures.

College students, especially freshmen, are facing a large workload and a relatively flexible learning atmosphere compared with their experience in high school. Self-regulation seems to be an important component in acquiring academic achievement for college students. In other words, students who can control their learning processes effectively are characterized as self-regulated learners(Schunk & Zimmerman 1994). Many people believe that everyone could be self-regulated learner,but what differentiates them most is the quality of their self-regulation( as cited in Cohen 2012). Therefore, understanding the role of self-regulation and how to manage students' learning experiences in order to improve their self-regulation ability are significant.

文摘Objective: The current study aimed to investigate the relationship among personality traits and self regulation among different drug abuse cases. The three categories of drug abuse were based on phases of addiction namely current, relapse and remitted cases. Method: The sample consisted of 108 drug abusers taken from different rehabilitation centers of Rawalpindi and Islamabad. The selected drug abusers were at current (n = 40), relapsed (n = 42) and remission phase (n =2 7) of drug addiction;with an age range of 20 to 70 years. The data were collected from private rehabilitation centers of Rawalpindi and Islamabad. In this study, Eysenck Personality Questionnaire Revised EPQR-Short (Eysenck, Eysenck & Barrett, 1985) was used to measure personality traits of the drug abuse cases. Self Regulation Questionnaire (SRQ) developed by Miller and Brown (1991) was used to measure the self regulatory skills. The data were collected from rehabilitation centers. Before collection of data, formal permission was taken from the administration of these centers. Research protocol included an Ethical Proforma explaining rationale of study, ensuring confidentiality of the information to be used for research purpose only and also had personal consent form. In the first phase, pilot study was conducted to check the reliability of translated scale and availability of sample. A sample of 15 drug abusers was selected for pilot study. In the next step (main study);protocol was administered on current, relapsed and remitted cases through individual administration. For remitted cases, the researcher consulted authorities (administration head, and medical doctors) of centers and administered protocol on the days when these participants came for follow-up session. Results: The results of the study indicate strong correlation between self regulation and personality traits (r = 0.27**). Furthermore, most of the participants scored higher on Psychoticism (100%), Introversion (75%) while Neuroticism (58%) trait was found less among drug abuse cases. The study also indicates strong correlation between self regulation and Extraversion-Introversion personality trait (r = 0.20**) while other traits did not have significant correlations with self regulation. Furthermore, the descriptive frequencies reveal that most of the participants scored higher on Psychoticism (100%), Introversion (75%) while Neuroticism (58%) trait was found less among drug abuse cases. Conclusion: From the above mentioned findings, it is concluded that self regulation is associated with personality traits among drug abusers. The comparisons of personality traits and self regulatory skills among the different phases of addiction provide an insight into how these traits interact with self control to regulate impulsive and addictive behaviors.

文摘A self-regulating humidity material with bamboo for the room was prepared. The activated carbons were prepared with the temperature rising rates of 5, 10, 15, 20 ℃/min and constant temperatures of 600, 700, 800, 900, 1 000, 1 100, 1 200, 1 300, 1 400, and 1 500 ℃ for 0.5, 1, 2, 3, and 6 h, respectively.And then their structures such as specific surface areas, volumes of pores from 3.7-7.1 nm and average pore diameters were tested. Various humidity conditions were simulated and their self-regulating humidity properties such as absorption/desorption contents, rates and stabilities were evaluated. The results show that the specific surface area, volume of pores from 3.7-7.1 nm and average pore diameter of activated carbon are closely nonlinearly related to the temperature rising rate, temperature and holding time. In general, the activated carbon prepared with the temperature rising rate of 5 ℃/min and kept at 1 100 ℃ for 2 h has the highest absorption/desorption content and rate, the lowest decay factor and then the best stability at all humidity for the fact that it is provided with proper average pore diameter, higher specific surface area and volume of pores from 3.7-7.1 nm. The activated carbons can be prepared with the temperature rising rate of 5 ℃/min and kept at 1000, 900,1 200, 1 300, 1 400, 1 500, 800, 700, and 600 ℃ respectively for 2 h. These resulted properties are attributed to their different average pore diameters and volumes of pores from 3.7-7.1 nm.

文摘Mitogen-activated protein kinases(MAPKs)are a family of proteins that constitute signaling pathways involved in processes that control gene expression,cell division, cell survival,apoptosis,metabolism,differentiation and motility.The MAPK pathways can be divided into conventional and atypical MAPK pathways.The first group converts a signal into a cellular response through a relay of three consecutive phosphorylation events exerted by MAPK kinase kinases,MAPK kinase,and MAPK.Atypical MAPK pathways are not organized into this three-tiered cascade.MAPK that belongs to both conventional and atypical MAPK pathways can phosphorylate both non-protein kinase substrates and other protein kinases.The latter are referred to as MAPK-activated protein kinases.This review focuses on one such MAPK-activated protein kinase,MAPK-activated protein kinase 5(MK5)or p38-regulated/activated protein kinase(PRAK).This protein is highly conserved throughout the animal kingdom and seems to be the target of both conventional and atypical MAPK pathways.Recent findings on the regulation of the activity and subcellular localization,bona fide interaction partners and physiological roles of MK5/PRAK are discussed.

文摘BACKGROUND Pancreatic ductal cancer(PDAC)has high malignancy and poor prognosis.Long noncoding RNAs(lncRNAs)are associated with high levels of malignancy,including PDAC.However,the biological and clinical significance of negative regulator of antiviral response(NRAV)in PDAC is unclear.AIM To study the regulatory role of lncRNA NRAV in PDAC.METHODS GEPIA analyzed lncRNA NRAV and miRNA(miR-299-3p)expression levels in PDAC tissues and measured them in PDAC cells by quantitative measurements in real time.The specific role of NRAV and miR-299-3p in cell proliferation and transfer potential was evaluated by cell formation analysis,Cell Counting Kit-8 and Transwell analysis.The relationship between NRAV and miR-299-3p was studied by predictive bioinformatics,RNA immunoassay,and fluorescence enzyme analysis.In vivo experiments included transplantation of simulated tumor cells under naked mice.RESULTS The expression level of lncRNA NRAV was higher in both tumor tissues and cell lines of PDAC and was negatively associated with the clinical survival of PDAC patients.Functionally,overexpression of NRAV promoted cell proliferation and metastasis of PDAC cells,while knockdown of NRAV reversed these effects.Finally,NRAV was performed as a molecular sponge of miR-299-3p.Moreover,overexpression of miR-299-3p could reverse the promoting effects of NRAV on cell proliferation and metastasis of PDAC cells.CONCLUSION NRAV facilitates progression of PDAC as a molecular sponge of miR-299-3p and may be a potential molecular marker for diagnosis and treatment of PDAC.

文摘The title compound 1-(3-amino-[1,2,4]triazol-1-yl)-3,3-dimethyl-butan-2-one(3) was synthesized by Hofmann-alkylation reaction of 1-chloro-3,3-dimethyl-butan-2-one(1) and ~1H-[1,2,4]triazol-3-ylamine(2) with equal amount of K_2CO_3 as acid acceptor. The structure of compound 3 was characterized by ~1H NMR, 13 C NMR, HRMS and single-crystal X-ray diffraction. The compound crystallizes in the monoclinic system, space group P21/n with a = 5.7227(8), b = 27.924(4), c = 6.2282(7) ?, β = 101.892(11)°, V = 973.9(2) ?~3, Z = 4, T = 180.00(10) K, μ(MoKα) = 0.087 mm^(-1), Dc = 1.243 g/cm^3, 3832 reflections measured(3.648≤θ≤26.022°), 1916 unique reflections(Rint = 0.0359, Rsigma = 0.0572) used in all calculations. The final R = 0.0557(I 〉 2σ(I)) and w R = 0.1276(all data). Bioassay showed that 3 displayed excellent activity as plant growth regulator with inducing lateral root formation and enhancing primary root elongation at 0.27 mmol/L(50 ppm) in soybeen(He Feng-50). Good water solubility was found with 50 mg in 1 m L of water. Therefore, application of 3 in agriculture is more environmentally friendly due to cosolvent-free condition, and results in improved abiotic-stress tolerance by affecting the root growth. And furthermore, it can be used as a precursor to investigate the function of regulating plant root growth.

文摘Human adipose-derived stem cells(hASCs)are a promising cell type for bone tissue regeneration.Circular RNAs(circRNAs)have been shown to play a critical role in regulating various cell differentiation and involve in mesenchymal stem cell osteogenesis.However,how circRNAs regulate hASCs in osteogenesis is still unclear.Herein,we found circ_0003204 was significantly downregulated during osteogenic differentiation of hASCs.Knockdown of circ_0003204 by si RNA or overexpression by lentivirus confirmed circ_0003204 could negatively regulate the osteogenic differentiation of hASCs.We performed dual-luciferase reporting assay and rescue experiments to verify circ_0003204 regulated osteogenic differentiation via sponging miR-370-3p.We predicted and confirmed that miR-370-3p had targets in the 3′-UTR of HDAC4 m RNA.The following rescue experiments indicated that circ_0003204 regulated the osteogenic differentiation of hASCs via miR-370-3p/HDAC4 axis.Subsequent in vivo experiments showed the silencing of circ_0003204 increased the bone formation and promoted the expression of osteogenic-related proteins in a mouse bone defect model,while overexpression of circ_0003204 inhibited bone defect repair.Our findings indicated that circ_0003204 might be a promising target to promote the efficacy of hASCs in repairing bone defects.

文摘This paper describes the self—adjustment of some tuning-knobs of the generalized predictive controller(GPC).A three feedforward neural network was utilized to on line learn two key tuning-knobs of GPC,and BP algorithm was used for the training of the linking-weights of the neural network.Hence it gets rid of the difficulty of choosing these tuning-knobs manually and provides easier condition for the wide applications of GPC on industrial plants.Simulation results illustrated the effectiveness of the method.
