Single-arm trials for domestic oncology drug approvals in China

The urgent need for effective cancer treatments,particularly for advanced and relapsed cases in which standard therapies are inadequate,has spurred the development of innovative therapeutic drugs1,2.Among the strategies to expedite drug development,the use of single-arm trials(SATs)is emerging as a promising avenue with substantial potential to shorten drug approval timelines and accelerate market entry.

Research and Enlightenment of Accelerated Approval Pathway in China

Objective To provide suggestions to improve the system of accelerated review and approval in China and to speed up the development and marketing of innovative drugs.Methods A comparative analysis was made to study the accelerated review,conditional approval and breakthrough therapy between China and the United States.Combined with the implementation of accelerated pathway in China,some suggestions were put forward.Results and Conclusion The implementation of accelerated drug review and approval system can significantly speed up the process of new drug development,which will improve the accessibility of innovative drugs.However,there are some problems in the review procedures and the implementation of supportive documents in China.In view of the above problems and combined with the experiences of US FDA,we should strengthen the construction of institutional system and reviewer team,and improve the efficiency of drug research and development.

Rapid evolution of T2/S-RNase genes in Fragaria linked to multiple transitions from self-incompatibility to self-compatibility

The T2/RNase gene family is widespread in eukaryotes,and particular members of this family play critical roles in the gametophytic self-incompatibility(GSI) system in plants.Wild diploid strawberry(Fragaria)species have diversified their sexual systems via self-incompatible and self-compatible traits,yet how these traits evolved in Fragaria remains elusive.By integrating the published and de novo assembled genomes and the newly generated RNA-seq data,members of the RNase T2 gene family were systematically identified in six Fragaria species,including three self-incompatible species(Fragaria nipponica,Fragaria nubicola,and Fragaria viridis) and three self-compatible species(Fragaria nilgerrensis,Fragaria vesca,and Fragaria iinumae).In total,115 RNase T2 genes were identified in the six Fragaria genomes and can be classified into three classes(Ⅰ-Ⅲ) according to phylogenetic analysis.The identified RNase T2 genes could be divided into 22 homologous gene sets according to amino acid sequence similarity and phylogenetic and syntenic relationships.We found that extensive gene loss and pseudogenization coupled with small-scale duplications mainly accounted for variations in the RNase T2 gene numbers in Fragaria.Multiple copies of homologous genes were mainly generated from tandem and segmental duplication events.Furthermore,we newly identified five S-RNase genes in three self-incompatible Fragaria genomes,including two in F.nipponica,two in F.viridis,and one in F.nubicola,which fit for typical features of a pistil determinant,including highly pistil-specific expression,highly polymorphic proteins and alkaline isoelectric point(pI),while no S-RNase genes were found in all three selfcompatible Fragaria species.Surprisingly,these T2/S-RNase genes contain at least one large intron(>10 kb).This study revealed that the rapid evolution of T2/S-RNase genes within the Fragaria genus could be associated with its sexual mode,and repeated evolution of the self-compatible traits in Fragaria was convergent via losses of S-RNase.

Testing and Approval Procedures for New Slab Track Systems in Europe

This paper describes the required testing and approval procedures of new track systems operated in Europe. To begin with, test methods and performance specifications according to European Standard （EN） are outlined. These include the repeated loading test, the determination of the static and dynamic stiffness of rail pads, clamping force and longitudinal rail resistance. The fact that labor tests are unable to simulate all the conditions in situ shows that these labor tests are not sufficient for the evaluation of the long term behaviour of a new track system： a test track of sufficient length must be constructed and exposed to traffic loads. In Europe to be accredited as a new system, a new slab track system must have a trial time of more than two years, during which the features of the whole system can be recognized. In the second part of this paper, the experience of the Institute of Road, Railway and Airfield Construction of TUM concerning the measuring methods of slab track systems carrying traffic are outlined. Also the approval procedure of the new slab tracks in Germany is discussed.

Effects of China's Administrative Approval Reforms on Transaction Cost and Economic Growth

Chinas economic growth miracle despite complicated administrative approval formalities prompted us to raise the following question: Is it true that reforming administrative approval will promote economic growth? Based on our analysis on market entry, we discovered that reforming administrative approval will spur economic growth by reducing transaction cost for firms. Administrative approval may suppress social cost and propel Chinas economic growth; Chinas gradualist approval reforms may indeed propel economic growth by reducing transaction cost for firms. With the data of prefecture-level cities during 2000-2013 and data of companies listed on the SME board during 2010-2014, we tested the effects of approval reforms on economic growth and on transaction cost, and employed instrumental variable and PSMfor the treatment of the endogeneity problem-all these tests led to robust and consistent results. Moreover, we discovered that difference in government policy implementation in the approval process is the root cause of corruption and rent seeking.

Region coverage control for multiple stratospheric airships with combined self-/event-triggered mechanism

The region coverage control problem of multiple stratospheric airships system is firstly addressed in this paper.Towards it,we propose a two-layer control framework with the artificial potential field(APF)-based region coverage control law and the adaptive tracking control law.The APF-based region coverage control law ensures the coverage task is achieved until every single stratospheric airship ends up performing station keeping where near the respective global minimum point,in which an innovative solution to the local minimum problem is put forward.The adaptive tracking control law is designed to realize motion control using tracking the desired velocity and angular velocity given by coverage control law,with the consideration of several practical control problems as unknown individual differences and external disturbances.To save resources,the combined self-/event-triggered mechanism designed therein significantly reduces the times of state information transmission and control law calculation.The effectiveness of the proposed control framework is verified through simulations.

Investigation on the Benefits and Satisfaction Degree of the Electronic Official Document On-line Submission and Approval System-A Case Study of a Medical Center in Southern Taiwan

Purpose:The aim of the study was to investigate the actual benefits of the electronic official document online submission and approval system and the satisfaction of hospital staff in a medical center in southern Taiwan,and to find out whether there are any differences between medical institutions and general government personnel.Methods:A cross-sectional study was conducted to investigate satisfactory outcome with questionnaires.The subjects were administrators,healthcare professionals and medical personnel of a medical center in the southern part of Taiwan who had signed electronic documents online.A total of 395 questionnaires were sent out,147 of which were valid,and the rate of collecting data survey was 37%.We analyzed with SPSS version 20.Results:The official document approval system was mainly used by administrative units and contractors,accounting for more than 50%of users.Besides,the frequency of use was at least more than once a week.As for the user’s perception of operating system,most people thought that it is easier to choose the format of official document and to set up the duty agent on leave,but in the part of the signing and approval process setting or modifying,it was considered more difficult,accounting for 38.1%.In terms of perceptual usefulness,the average value was 3.81,which showed that the user agreed that the system has met the needs of daily official documents.When some users of service area encountered problems with their use,the clerical staff were able to provide services immediately and have the professional ability to resolve problems,in order to agree to the majority,accounting for 53.7%.In addition,nearly 60%of users rated the official document system positively,with an average of 3.84 satisfaction,which was higher than the certified value of 3,conforming to the standards for satisfaction with the use of official documents.In addition,the role authorization,perceptual easy-to-use and service area were significant(p<0.05).The perceptual usefulness of Subordinate units was also significant(p=0.016).The frequency of use,perceptual easy-to-use,perceptual usefulness,service area and satisfaction were significantly different(p<0.05).The correlation coefficient between perceptual usefulness and user satisfaction was 0.833,indicating that there was a high correlation.The daily usage frequency of contractors was higher than supervisors.However,supervisors had the highest frequency of use every quarter(p=0.135).There was no significant difference between contractors and supervisors in the frequency of use.Conclusion:It is suggested that education and training on the operation of the electronic official document on-line submission and approval system should be conducted,which can enhance the education and training of supervisors and medical personnel.Continually,invite supervisors and medical personnel to provide advices on the official document system as a reference for future improvements of the system.

Japanese Social Mentality and the Change in the Approval Rates of the Abe Administration

From June to July of 2017, the approval rates of the Abe administration fell sharply, in stark contrast to the long-term stability since2012. The stability of Abe's approval rates originated from the people's negative support for him and their opposition to the Security Laws and other important events as well as the scandals under his administration. The opposition and negative support originated from the people's "acquired helplessness","Abenomics", the sharp drop in the opportunity costs of voting and other conditions. The sudden fall of Abe's approval rates was not due to the change in the aforementioned conditions, rather because of the collapse of his image including personality owing to Abe's perceived arrogance and his estrangement with the people. In the context of the temporary stabilization of approval rates, Abe dissolved the House of Representatives ahead of schedule and held a general election, taking full advantage of the internal strife between the opposition parties, to achieve his goal of ruling until the 2020 Tokyo Olympics. His victory in the recent election does not accurately demonstrate that Abe is completely immune to negative impact, thus he will to use the Korean Peninsula crisis and other issues to demonstrate his ability of safeguarding Japan's security, seek reconstruction of his image, and further enhance his approval rate to stabilize his administration.
