The neuronal differentiation of mesenchymal stem cells offers a new strategy for the treatment of neurological disorders.Thus,there is a need to identify a noninvasive and sensitive in vivo imaging approach for real-time monitoring of transplanted stem cells.Our previous study confirmed that magnetic resonance imaging,with a focus on the ferritin heavy chain 1 reporter gene,could track the proliferation and differentiation of bone marrow mesenchymal stem cells that had been transduced with lentivirus carrying the ferritin heavy chain 1 reporter gene.However,we could not determine whether or when bone marrow mesenchymal stem cells had undergone neuronal differentiation based on changes in the magnetic resonance imaging signal.To solve this problem,we identified a neuron-specific enolase that can be differentially expressed before and after neuronal differentiation in stem cells.In this study,we successfully constructed a lentivirus carrying the neuron-specific enolase promoter and expressing the ferritin heavy chain 1 reporter gene;we used this lentivirus to transduce bone marrow mesenchymal stem cells.Cellular and animal studies showed that the neuron-specific enolase promoter effectively drove the expression of ferritin heavy chain 1 after neuronal differentiation of bone marrow mesenchymal stem cells;this led to intracellular accumulation of iron and corresponding changes in the magnetic resonance imaging signal.In summary,we established an innovative magnetic resonance imaging approach focused on the induction of reporter gene expression by a neuron-specific promoter.This imaging method can be used to noninvasively and sensitively detect neuronal differentiation in stem cells,which may be useful in stem cell-based therapies. 展开更多

Adaptive proportional integral differential control based on radial basis function neural network identification of a two-degree-of-freedom closed-chain robot

Accurate trajectory control of a robot is essential in practical-use. This paper presents an adaptive proportional integral differential (PID) control algorithm based on radial basis function (RBF) neural network for trajectory tracking of a two-degree-of-freedom (2-DOF) closed-chain robot. In this scheme, an RBF neural network is used to approximate the unknown nonlinear dynamics of the robot, at the same time, the PID parameters can be adjusted online and the high precision can be obtained. Simulation results show that the control algorithm accurately tracks a 2-DOF closed-chain robot trajectories. The results also indicate that the system robustness and tracking performance are superior to the classic PID method. 展开更多

Identification of differentially expressed genes in dorsal root ganglion in early diabetic rats

Objective To screen and identify differentially expressed genes in the dorsal root ganglion (DRG) in early experimental diabetic rats. Methods Diabetic model rats were induced by single intraperitoneal injection of streptozotocin (STZ). At the second week after STZ injection, the sensory nerve conduction velocities (SNCV) of sciatic nerve were measured as an indicator of neuropathy. The technique of silver-staining mRNA differential display polymerase chain reaction (DD-PCR) was used to detect the levels of differentially expressed genes in rat DRG. The cDNA fragments that displayed differentially were identified by reverse-hybridization, cloned and sequenced subsequently, and then confirmed by Northern blot. Results The SNCV in the diabetic model group [n = 9, (45.25±10.38) m/s] reduced obviously compared with the control group [n = 8, (60.10± 11.92) m/s] (P 〈 0.05). Seven distinct cDNA clones, one was up-regulated gene and the others were downregulated ones, were isolated by silver-staining mRNA differential display method and confirmed by Northern blot. According to the results of sequence alignment with GenBank data, majority of the clones had no significant sequence similarity to previously reported genes except only one that showed high homology to 6-pyruvoyl-tetrahydropterin synthase mRNA (accession No., BC059140), which had not been reported to relate to diabetic neuropathy. Conclusion These differentially expressed genes in the diabetic DRG may contribute to the pathogenesis of diabetic peripheral neuropathy. 展开更多

The electron transport of linear atomic chain trodes was investigated by using the density Green's function method. We have calculated of MgB2 sandwiched between Au(100) elecfunctional theory with the non-equilibrium the corresponding cohesion energy and conductance of junctions in different distance. It is found that, at the equilibrium position, the Au-B bond-length is 1.90 A, the B-Mg bond-length is 2.22 A, and the equilibrium conductance is 0.51G0 (Go=2e^2/h). The transport channel is almost formed by the π antibonding orbitals, which was made up of the Px and Py orbital electrons of B and Mg atoms. In the voltage range of -1.5 to 1.5 V, the junctions show the metallic behaviors. When the voltage is larger than 1.5 V, the current decreases gradually and then negative differential resistance appears almost symmetrically on both positive and negative bias. 展开更多

AIM: To investigate SBA2 expression in CRC cell lines and surgical specimens of CRC and autologous healthy mucosa. METHODS: Reverse transcription-polymerase chain reaction (RT-PCR) was used for relative quantification of SBA2 mRNA levels in 4 human CRC cell lines with different grades of differentiation and 30 clinical samples. Normalization of the results was achieved by simultaneous amplification of beta-actin as an internal control. RESULTS: In the exponential range of amplification, fairly good linearity demonstrated identical amplification efficiency for SBA2 and beta-actin (82%). Markedly lower levels of SBA2 mRNA were detectable in tumors, as compared with the coupled normal counterparts P【0.01). SBA2 expression was significantly (0.01】P 【 0.05) correlated with the grade of differentiation in CRC, with relatively higher levels in well-differentiated samples and lower in poorly-differentiated cases. Of the 9 cases with lymph nodes affected, 78% (7/9) had reduced SBA2 mRNA expression in contrast to 24% (5/21) in non-metastasis samples 0.01】P【0.05). CONCLUSION: SBA2 gene might be a promising novel biomarker of cell differentiation in colorectal cancer and its biological features need further studies. 展开更多

Hypobaric hypoxia can cause severe brain damage and mitochondrial dysfunction, and is involved in hypoxic brain injury. However, little is currently known about the mechanisms responsible for mi- tochondrial dysfunction in hypobaric hypoxic brain damage. In this study, a rat model of hypobaric hypoxic brain injury was established to investigate the molecular mechanisms associated with mi- tochondrial dysfunction. As revealed by two-dimensional electrophoresis analysis, 16, 21, and 36 differential protein spots in cerebral mitochondria were observed at 6, 12, and 24 hours post-hypobaric hypoxia, respectively. Furthermore, ten protein spots selected from each hypobaric hypoxia subgroup were similarly regulated and were identified by mass spectrometry. These de- tected proteins included dihydropyrimidinase-related protein 2, creatine kinase B-type, is- ovaleryI-CoA dehydrogenase, elongation factor Ts, ATP synthase beta-subunit, 3-mercaptopyruvate sulfurtransferase, electron transfer flavoprotein alpha-subunit, Chain A of 2-enoyI-CoA hydratase, NADH dehydrogenase iron-sulfur protein 8 and tropomyosin beta chain. These ten proteins are all involved in the electron transport chain and the function of ATP synthase. Our findings indicate that hypobaric hypoxia can induce the differential expression of several cerebral mitochondrial proteins, which are involved in the regulation of mitochondrial energy production. 展开更多

Methods: Total RNA from ventral prostates of control and flutamide treated rats were isolated. Differentially expressed transcripts were identified using differential display reverse transcriptase polymerase chain reaction. The effect of castration on the expression of flutamideregulated transcripts was studied. Results: We have identified β2-microglobulin, cytoplasmic FMR1 interacting protein 2 and pumilio 1 as flutamide induced and spermine binding protein and ribophorin Ⅱ as flutamide repressed targets in the rat ventral prostate. Although flutamide treatment caused an induction of pumilio 1 rnRNA, castration had no effect. Conclusion: Castration and flutamide treatments exert differential effects on gene expression. Flutamide might also have direct AR independent effects, which might have implications in the emergence of androgen independent prostate cancer and the failure of flutamide therapy. 展开更多

差分隐私增强的大米区块链品控模型

[目的/意义]针对传统大米品质监管追溯系统中存在的品控数据链机制不够完善、品控信息可追溯程度不足、数据上链效率低及隐私信息泄露等问题,提出一种差分隐私增强的大米区块链品控模型。[方法]首先,结合大米全产业链,设计数据传输流程,涵盖种植、收购、加工、仓储和销售等各环节,有效保证品控数据链的连续性;其次,为解决上链数据量大、上链效率低问题,将大米全产业链各环节关键品控数据存储于星际文件系统(InterPlanetary File System, IPFS),然后将存储完成后返回的哈希值上链;最后,为提高品控模型信息可追溯程度,将种植环节关键品控数据中涉及隐私的部分信息通过差分隐私(Differential Privacy)处理后展示给用户,模糊化个体数据,以提高品控信息可信度,同时也保护了农户种植隐私。基于该品控模型,设计了差分隐私增强的大米区块链品控系统,并在相关大米企业实际运行。[结果与讨论]经测试,差分隐私增强的大米区块链品控系统全产业链单环节数据完成存储平均耗时1.125 s,信息追溯查询平均耗时0.691 s。与传统大米品质监管追溯系统相比,单环节数据存储时间缩短6.64%,信息追溯查询时间缩短16.44%。[结论]研究提出的模型不仅提高了品控数据连续性和信息可追溯程度,同时保护了农户的隐私,还在一定程度上提升了品控数据存储及信息追溯查询的效率,可为大米品质监管与信息追溯系统的设计和改进提供参考。 展开更多

考虑低碳补贴和技术延迟的供应链效应研究

为促进我国低碳经济的发展,政府对低碳产业采取补贴政策。考虑到技术普遍存在延迟效应,为避免其对政府低碳补贴、供应链各成员利润和社会福利带来的动态影响,选择一个零售商和一个制造商组成的二级供应链,并构建了其与政府之间关于延迟... 为促进我国低碳经济的发展,政府对低碳产业采取补贴政策。考虑到技术普遍存在延迟效应,为避免其对政府低碳补贴、供应链各成员利润和社会福利带来的动态影响,选择一个零售商和一个制造商组成的二级供应链,并构建了其与政府之间关于延迟效应的微分博弈模型。本文分别统计了长视策略与短视策略两种情况分别在分散型决策和集中型政策下的最优策略、碳减排量和企业信誉演化路径,并将结果对比分析后经研究认为:政府的低碳补贴比率和技术延迟时间呈负相关,对企业的经营发展起到了促进作用。而技术延迟时间过长则会使得制造商选择短视政策,从而不利于企业利润和社会福利的发展。集中型决策下的减排量和社会福利总是最优的,但延迟时间在一定范围内供应链利润才是最优的,一旦超过这个范围分散型决策下的供应链利润高。 展开更多

考虑决策主体的有限理性行为,构建零售商双渠道差异定价的跨渠道退货价格博弈模型,从系统复杂特性的角度分析供应链系统的局部渐近稳定性和演化行为。根据朱利判据给出系统的稳定域,借助数值仿真刻画系统的动力学行为特征,探究系统状态... 考虑决策主体的有限理性行为,构建零售商双渠道差异定价的跨渠道退货价格博弈模型,从系统复杂特性的角度分析供应链系统的局部渐近稳定性和演化行为。根据朱利判据给出系统的稳定域,借助数值仿真刻画系统的动力学行为特征,探究系统状态对价格决策和企业利润的影响,并对失稳系统进行有效控制。研究结果表明,价格调整速度过快容易导致供应链系统经倍周期分岔而陷入混沌状态,存在价格调整的稳定域以确保系统稳定;作为博弈主导者的制造商调整批发价格会影响整个系统的稳定性,而零售商的价格调整仅影响自身稳定;混沌的系统对制造商和零售商都不利,倍周期分岔的系统对制造商有利而对零售商有害,稳定的系统对两者都有利;延迟反馈控制法能有效控制混沌系统;消费者满意度对价格的影响受制于供应链系统状态的制约。 展开更多
