Background: Exposure to solar UV rays has deleterious effects on skin appearance through physiological and structural alterations that eventually lead to skin photo-damage. Aims: To test the photo-protective effect of a novel preparation, Dead Sea Osmoter Concentrate (DSOC), containing high concentration of Dead Sea water. Methods: Human skin organ culture was used as a model to assess the biological effects of UVB irradiation and the protective effect of topical application of DSOC preparation. Skin pieces were analyzed for mitochondrial activity by Alamar blue (resazurin) assay, for apoptosis by caspase 3 assay, for detoxification by proteasome 20S sub-unit activity and for skin hydration by aquaporin 3 (AQP3) membrane channels expression. Human subjects were tested to evaluate the effect of DSOC Serum, a topical formula for facial skin, on skin hydration by a corneometer. Results: UVB irradiation induced cell apoptosis in the epidermis of skin organ cultures and decreased their proteasome activity and AQP3 expression. Topical application of DSOC significantly attenuated all these effects. In human subjects, an elevation in the intense skin moisture, which was higher relatively to subjects’ regular moisturizer, was observed. Moreover, DSOC Serum boosted the hydration effect together with regular moisturizer. Conclusions: The results clearly demonstrate that DSOC preparation can significantly improve the skin capability in coping with UVB rays stress in different cellular functions such as anti-apoptotic properties as well as detoxification and hydration recovery that can attenuate biological effects of skin photo-damage. Topical application of DSOC Serum had contributed to skin appearance by a strong hydration impact, also as a booster. 展开更多

Protection from Near-Infrared to Prevent Skin Damage

Over half of the solar energy consists of near-infrared, and in addition to natural near-infrared, humans are increasingly exposed to artificial near-infrared from electrical appliances. Thus, we are exposed to tremendous amounts of near-infrared. Despite the wide prevalence of a variety of ultraviolet blocking materials, the necessity to protect against near-infrared has not been well recognized. To clarify the necessity to protect against near-infrared, we assessed cell viability of human fibroblast cells after water-filtered broad-spectrum near-infrared (1100 - 1800 nm together with a water-filter that excludes wavelengths 1400 - 1500 nm) treatment using 2 sets of transparent polycarbonate plates, one to block ultraviolet and the other to block both ultraviolet and near-infrared. The cell viability was significantly decreased after 10 rounds of near-infrared irradiation at 20 J/cm2 in near-infrared treated cells without a protective polycarbonate plate and near-infrared treated cells using the polycarbonate plate to block only ultraviolet. Assuming that the cell viability of the non-irradiated control to be 100, the cell viability of the near-infrared treated cells without any protection was 0.2. The cell viability of the near-infrared treated cells with the polycarbonate plate to block only ultraviolet was 0.3, whereas both ultraviolet and near-infrared protected cells retained a viability of 85.1. The results of this study indicate that protection from not only ultraviolet but also near-infrared should be considered to prevent skin damage. 展开更多

Thermal protective clothing has been recognized as the primary shielding against emergency fire hazard and inflammable gas leakage. Therefore,the thermal response of human covered with thermal protective clothing under high temperature is the key work to investigate the thermal insulation of thermal protective clothing. A coupling model composed of thermal protective clothing,air gap and human skin is established and the temperature of the micro-system is numerically solved via the finite element method( FEM).Especially,the heat transfer of air gap located between clothing and human skin considering conduction and radiation is established while the human skin layers involve the effect of blood perfusion. Then the effect of thermophysical properties( thermal conductivity and volumetric capacity) of fabric and thickness of fabric and air on the thermal response of the micro-system is elucidated and compared.The results indicate that the volumetric heat capacity of fabric is the key parameter to affect the thermal shielding performance of thermal protective clothing,and the thicker fabric thickness and air gap thickness can improve the thermal protective properties of the micro-system. 展开更多

Skin cancer rates have risen over the past decades,making it imperative that adults understand the need for protection from sun exposure.Though some risk factors have been identified as predictive for skin cancers,there is a lack of synthesized information about factors that influence adults in their decisions to engage in sun protective behaviors.The purpose of this paper is to present the current state of the science on influential factors for sun protective behaviors in the general adult population.A rigorous literature search inclusive of a generally White,Caucasian,and non-Hispanic adult population was performed,and screening yielded 18 quantitative studies for inclusion in this review.Findings indicate that modifiable and non-modifiable factors are interdependent and play a role in sun protective behaviors.This study resulted in a proposed conceptual model for affecting behavioral change in sun protection including the following factors:personal characteristics,cognitive factors,family dynamics,and social/peer group influences.These factors are introduced to propose tailored nursing interventions that would change current sun protective behavior practice.Key implications for nursing research and practice focus on feasibility of annual skin cancer screening facilitated by advanced practice nurses,incorporating the identified influential factors to reduce skin cancer risk and unnecessary sun exposure. 展开更多

Background:Nursing staff are at much greater risk of infection(Ebola virus diseases and severe acute respiratory syndrome)due to their exposure to highly infectious bodily fluids and droplet nuclei and the need for personal protective equipment(PPE)to reduce the transmission risk.Aim:The present study was conducted to estimate the prevalence of skin injuries and their types due to PPE usage among nursing staff in tribal India.Materials and Methods:This descriptive cross-sectional study was conducted in the dedicated coronavirus disease-2019(COVID-19)hospital for a period of 4 months among 144 nursing staff wearing Grade 2 and 3 PPE kits.Study subjects were approached through social networking websites and survey questionnaires(Google forms)according to relevant guides,and research literature was used to collect the details regarding baseline,duty,and skin injury characterization.Chi-square test was used to find the association between skin injury and baseline or duty characteristics,and the association was significant at a P<0.05.Results:It was observed that 54.7%of nursing staff were working for 6 or more hours and 16.5%of subjects were wearing the PPE kit for 5 or more hours per day.Skin injury was reported by 86.3%of the subjects after using PPE.Skin injuries among nurses were statistically significant with their current place of stay and daily duty hours(P<0.05).Conclusion:In this study,it was found that 86.3%of subjects had suffered from skin injuries by using PPE while caring for COVID-19 patients during duty hours,so an effective preventive measure should be adopted. 展开更多

New Insights in the Skin Protective Activity of a Dexpanthenol Containing Formula (BEPANTHEN<sup>®</sup>) in a Diaper Rash-Like Model

<strong>Background:</strong> Dexpanthenol containing formula (BEPANTHEN<sup>®</sup>), formulated as a water in oil preparation, is currently widely marketed as a diaper care product aiming... <strong>Background:</strong> Dexpanthenol containing formula (BEPANTHEN<sup>®</sup>), formulated as a water in oil preparation, is currently widely marketed as a diaper care product aiming to protect baby’s buttocks and repair diaper dermatitis. Dexpanthenol is a well-known moisturizer with barrier-improving properties and the oily phase of the water in oil preparation forms a lipophilic film on the skin surface that isolates the skin from irritants (feces and urine). Prolonged contact with irritants triggers a local inflammation cascade responsible for the cutaneous erythema. To further investigate the protective properties of skin barrier preparations, we took advantage of an <i>ex vivo</i> model of healthy human skin discs especially designed to evaluate protective and/or repairing effects of topical preparations recommended for baby’s buttocks through the measurement of interleukin-1 alpha release (a cytokine considered as the <em>Primum movens</em> of the skin inflammatory reaction), following the application of different irritants. <strong>Methods: </strong>Healthy human skin discs have been incubated in the absence (control) or in the presence of two irritants,<em> i.e.</em> a “urine like + urease” preparation and sodium dodecyl sulfate, and in the presence of three ointments, one containing dexpanthenol, but not the other two. At the end of the incubation period, interleukin-1 alpha (IL-1<em>α</em>) was quantified in the explants culture media.<strong> Results: </strong>“Urine like + urease” preparation (ULU) and sodium dodecyl sulfate (SDS) both increased IL-1<em>α</em> production of skin explants by 181.1% (p < 0.001) and 88.3% (p < 0.001), respectively. The dexpanthenol containing formula significantly inhibited the ULU- and the SDS-induced IL-1<em>α</em> release by 67.42% (p < 0.001) and 46.55% (p < 0.001), respectively. Under the same experimental conditions, one of the formulas without dexpanthenol significantly inhibited the ULU-induced IL-1<em>α</em> release by 45.94% (p < 0.01) but not the SDS-induced one, and the other tested formulation displayed no significant effect on the IL-1<em>α</em> production regardless of the irritant applied. Moreover, the effect of the dexpanthenol containing formula on the ULU-induced IL-1<em>α</em> release was significantly higher than the effect of the other formula;a difference of 19.6 % (p < 0.05) was observed.<strong> Conclusion: </strong>Dexpanthenol containing formula (BEPANTHEN<sup>®</sup>) provides good protection of baby’s buttocks against irritants. Its protective effect seems to be superior compared with other products, which did not contain this ingredient. Moreover, the results obtained in the present study suggest that dexpanthenol displays <i>per se</i> a real IL-1<em>α</em> production inhibitory effect. This work, however, consists of preliminary studies and additional investigations involving more formulas and end-points such as the quantification of other pro- or anti-inflammatory cytokines and/or resolvins for example, are needed to better understand the cutaneous protective effect of dexpanthenol. 展开更多

Background:Collagen peptides(CP),including tripeptides and elastin peptides(EP),are known for their in vitro and in vivo anti-skin aging effects.Despite positive results in animal models,the combination effects of CP and EP and the bioavailability of CP in human studies,particularly in young and middle-aged women,remain underexplored.Objective:To evaluate the effects of an orally administered collagen drink combining CP and EP on the skin health of young and middle-aged women.Materials and Methods:A single-center,randomized,double-blind,parallel-controlled trial was conducted,utilizing the WONDERLABR fish collagen tripeptide beverage.Participants consumed the drink over an 8-week period.Results:Compared to the placebo group,the collagen drink group showed significant improvements in skin hydration(39.19%increase),transepidermal water loss(33.45%decrease),skin elasticity(25.37%increase),dermal collagen content(21.64%increase),pore size(7.94%decrease),wrinkle length(18.09%decrease),skin smoothness(2.85%improvement),and skin roughness(15.32%decrease).Overall pore volume decreased by 60%,and visual assessments indicated a decrease in skin luminosity by 15.20%and smoothness index by 22.55%.Mass spectrometry demonstrated a significant increase in collagen efficacy components,including blood pH and GPH levels(P<0.05).Conclusion:The study confirmed the combination nourishing and anti-skin aging effects of EP and CP on the skin of young and middle-aged women,demonstrating significant improvements in various skin parameters and good bioavailability of collagen peptides. 展开更多

Development of a Reproducible Rating System for Sun Protective Clothing That Incorporates Body Surface Coverage

Skin cancer is the most common form of cancer in humans, and typically affects sun-exposed parts of the body. Sunny Australia and New Zealand have the highest incidence of skin cancer globally. Clothing provides a protective barrier that reduces the amount of ultraviolet radiation (UVR) reaching the skin. Australia pioneered the development of a relative ranking of the sun-protective capabilities of clothing based on the transmission of UVR through fabric. Standardized Ultraviolet Protection Factor (UPF) measurement procedures and associated labeling specifications are documented in the Australian and New Zealand Standard, AS/NZS 4399:1996. The standard was intended to enable consumers to make informed choices. Since its introduction, this standard has been adopted almost universally by the textile industry, and is still in use almost two decades on, with plans to revise it only commencing recently. However, AS/NZS 4399:1996 does not consider garment design, particularly in relation to body surface coverage. Although swim-shirts have grown in popularity in Australia since the late 1990s, particularly among children, clothing remains under-utilized as a form of sun-protection in contemporary society. Skin cancer prevention campaigns should emphasize the sun-protective benefits of clothing and collaboration with the fashion industry is urgently needed to improve the aesthetic appeal, comfort, durability and affordability of sun-protective clothing to increase its popularity in skin cancer prone populations. In light of recent evidence showing that high UPF clothing which covers more of the body surface reduces pigmented mole development in children (major risk factor for melanoma), the rating system for sun-protective clothing should incorporate body surface covered as well as the UPF of the fabric. We discuss progress towards developing a protocol for measuring the extent of coverage of sun-protective garments. Once fully evaluated and refined, the testing protocol developed from this research may influence future revisions of international standards for evaluating and classifying sun-protective clothing. 展开更多

鲟鱼皮GPC的制备及其对PC12细胞的保护作用

目的:对比热解和酶解法所得明胶-肽复合物(GPC)的制备效率和活性。方法:以鲟鱼皮为原料,以GPC得率为指标,优化鲟鱼皮预处理条件及热解法制备GPC的条件。以热解法所得最大分子质量的GPC为底物,以三氯乙酸可溶性寡肽(TCA-SP)为指标,研究... 目的:对比热解和酶解法所得明胶-肽复合物(GPC)的制备效率和活性。方法:以鲟鱼皮为原料,以GPC得率为指标,优化鲟鱼皮预处理条件及热解法制备GPC的条件。以热解法所得最大分子质量的GPC为底物,以三氯乙酸可溶性寡肽(TCA-SP)为指标,研究几种蛋白酶对该底物的水解效率。比较TCA-SP得率最高的热解法和酶解法制备GPC对正常和D-半乳糖致氧化损伤PC12细胞活力的影响。结果:碱处理对鲟鱼皮GPC提取无明显贡献,酸处理对GPC提取率影响较大,最适条件为0.25 mol/L HCl处理鲟鱼皮6 h。高温热解法制备GPC的最适条件为100℃提取2 h,此时GPC和TCA-SP得率最高,分别为34.72%,31.22%。低温热解法以60℃提取1 h所得GPC的黏度最大,TCA-SP最低,提示该产物分子质量最大,结构最完整。以GPC-60℃-1 h为底物,分别用几种蛋白酶单独水解,其中中性蛋白酶水解0.5 h所得GPC中的TCA-SP含量最高(28.74%)。GPC-100℃-2 h和GPC-N-0.5 h底物在质量浓度为50~1000μg/mL范围,对正常PC12细胞活力均无影响。在质量浓度为12.5~200μg/mL范围对D-半乳糖所致PC12细胞氧化损伤均有一定的保护作用,且两者间无明显差异。结论:高温热解法和中性蛋白酶水解法所得GPC中的TCA-SP含量及对PC12的保护作用相近。 展开更多

低温环境手部皮肤温度与心理参数的响应规律

为了减小低温环境中人员手部冻伤风险并提高工作效率,通过手部冷应激试验来揭示局部冷暴露下手部皮肤温度与心理参数的响应规律。首先,招募了12名男性受试者参与试验;然后,利用人工气候室模拟低温环境,受试者穿着防护服暴露于16℃环境中... 为了减小低温环境中人员手部冻伤风险并提高工作效率,通过手部冷应激试验来揭示局部冷暴露下手部皮肤温度与心理参数的响应规律。首先,招募了12名男性受试者参与试验;然后,利用人工气候室模拟低温环境,受试者穿着防护服暴露于16℃环境中,手部暴露于16℃(对照组)和-10℃(试验组)2种工况;最后,在试验期间连续测量手掌、手背、大拇指、食指、中指、无名指、小拇指7个部位的皮肤温度,以及心理参数包括手部热感觉(TSV)和手部疼痛感(PSV)。结果表明:随着局部冷暴露时间的增加,手部皮肤温度会呈现先急剧降低,后趋于稳定的变化趋势;试验组的大拇指与手背的温差最大;手部皮肤温度与TSV呈线性关系(R^(2)=0.86),手部皮肤温度为30~35℃时手部处于舒适状态,25℃左右时感觉凉爽,10~15℃时感觉很冷;手部皮肤温度与PSV呈非线性关系(R^(2)=0.98),相较于TSV滞后,疼痛感发生于手部皮肤温度为15~20℃之间,之后手部皮肤温度呈指数曲线急剧下降,当手部皮肤温度为12℃左右时,疼痛感无法忍受。应注重低温环境下的手部防护,裸手处于-10℃的环境时最大冷暴露时间不应超过10 min,既要保证手部的舒适性,也要保证作业时的灵活度。研究成果可为降低冻伤发生概率、设计保暖服装、研发个体防护装备等提供基础数据与技术支撑。 展开更多

鮟鱇鱼皮活性肽的分离纯化及其对HK-2细胞损伤保护的作用

【目的】为探讨鮟鱇(Lophius litulon)鱼皮活性肽(Monkfish skin peptides,MSP)对棕榈酸(Palmitic acid,PA)诱导人肾皮质近曲小管上皮细胞(HK-2细胞)损伤保护的作用。【方法】以新鲜鮟鱇鱼皮为材料,选取加酶量、提取时间、pH、温度和料... 【目的】为探讨鮟鱇(Lophius litulon)鱼皮活性肽(Monkfish skin peptides,MSP)对棕榈酸(Palmitic acid,PA)诱导人肾皮质近曲小管上皮细胞(HK-2细胞)损伤保护的作用。【方法】以新鲜鮟鱇鱼皮为材料,选取加酶量、提取时间、pH、温度和料液比为因子,以1,1-二苯基-2-三硝基苯肼自由基(DPPH·)清除率为响应值,分别进行单因素实验和响应面实验,优化鮟鱇鱼皮肽的提取工艺;对得到的鮟鱇鱼皮肽经过超滤、葡聚糖凝胶过滤层析和全自动高压制备分离纯化得到MSP;利用液相二级质谱和多肽从头(de novo)测序技术对其进行结构分析,筛选抗氧化活性较强的肽段;通过构建PA诱导的HK-2细胞损伤模型,测定MSP对HK-2细胞中氧化应激指标、脂质代谢指标和炎症因子水平的影响。【结果】确定酸性蛋白酶与胃蛋白酶质量比2∶1的复配比例,加酶量3 800 U/g、pH 3、酶解时间4 h、温度50℃和料液比1 g∶10 mL为鮟鱇鱼皮活性肽的最佳酶解条件,此条件下制备的鮟鱇鱼皮肽对DPPH·的清除率为78.84%;经超滤,酶解多肽中<3 ku组分抗氧化活性最强,收集该组分进行凝胶色谱分离后,得到F1、F2和F3共3个峰,其中F1组分DPPH·清除率最高。F1组分经高效液相色谱法(HPLC)分离纯化后得到6个吸收峰,其中H2组分吸收峰强度最高,经氨基酸序列鉴定,从中筛选出3种寡肽,分别为Phe-Glu-Ala-Thr-Ser-Gln-Glu-Leu(FEATSQEL,FL-8)、Leu-Val-Ser-Cys-Ser-Ser-Leu(LVSCSSL,LL-7)和Pro-Leu-Glu-Asn-Tyr(PLENY,PY-5)。3条多肽在HK-2细胞脂质损伤模型中的修复作用各不相同:FL-8和PY-5能提高HK-2细胞模型的抗氧化能力,使细胞内谷胱甘肽过氧化物酶(GSH-Px)活性和总抗氧化能力(T-AOC)显著升高,降低细胞中活性氧的荧光强度,并显著降低丙二醛(MDA)水平,而LL-7能降低GSH-Px活性与T-AOC能力,能显著降低MDA的含量;在调节细胞的脂质代谢方面,FL-8与PY-5分别在不同程度上降低总胆固醇、甘油三酯和低密度脂蛋白-胆固醇含量,并显著增加高密度脂蛋白-胆固醇含量,而LL-7则会加剧细胞内的脂质蓄积;FL-8与PY-5显著降低炎症因子白细胞介素-18(IL-18)的水平,而LL-7则相反(α=0.05)。【结论】制备的MSP对PA诱导的HK-2具有保护作用,可为鮟鱇鱼皮活性肽的利用和缓解脂质肾毒性产品的开发提供理论基础和实验依据。 展开更多

为探究薰衣草精油(Lavender essential oil,LEO)对皮肤屏障紫外损伤的保护作用及相关机制,采用气相色谱-质谱联用(Gas chromatography-mass spectrometry,GC-MS)法对LEO的成分进行鉴定分析,并采用3D表皮皮肤模型,以600 mJ/cm^(2)剂量的... 为探究薰衣草精油(Lavender essential oil,LEO)对皮肤屏障紫外损伤的保护作用及相关机制,采用气相色谱-质谱联用(Gas chromatography-mass spectrometry,GC-MS)法对LEO的成分进行鉴定分析,并采用3D表皮皮肤模型,以600 mJ/cm^(2)剂量的中波紫外线(Ultraviolet B,UVB)进行辐照,建立皮肤屏障紫外损伤模型;实验设置未给药+UVB未照射组、未给药+UVB照射组、维生素E/地塞米松+UVB照射组及LEO+UVB照射组,通过苏木精-伊红染色法、酶联免疫吸附测定法、免疫组化法和免疫荧光法等探究不同组别对氧化应激、炎症反应、屏障结构相关蛋白以及CD44受体的影响。结果显示,LEO经GC-MS检测分析后,共鉴定出51种化学成分,主要成分为乙酸芳樟酯、芳樟醇、乙酸薰衣草酯等。UVB会造成皮肤屏障损伤,表现为ROS水平升高,SOD酶活性降低,炎症因子表达增多,屏障相关蛋白及CD44受体表达减少。LEO对UVB诱导的皮肤屏障损伤具有一定的保护效果,主要通过抑制UVB诱导的白介素1α、肿瘤坏死因子α和前列腺素E2表达量增加及ROS水平上升(P<0.05),提升受损皮肤模型中4种屏障相关蛋白和CD44受体蛋白表达量及SOD酶活力来实现(P<0.01)。以上结果表明LEO在减少皮肤屏障紫外损伤方面具有较大的应用前景,为其在护肤品中的应用提供一定的理论基础。 展开更多
