Effects of Transgenic DREB Toybean Dongnong50 on Diversity of Soil Nitrogen-fixing Bacteria

Drought is a bottleneck for worldwide soybean production which is getting more serious as the climate continues to worsen. Dehydration responsive element binding(DREB) is a kind of transcription factor that regulates the expression of stress tolerance-related genes in response to drought, high salinity and cold stress in plant. Soybean with DREB gene possesses the drought resisting capability which is helpful to increase the yield. However, the potential risk of genetically modified plants(GMPs) on soil microbial community is still in debate. In order to understand the effects of transgenic DREB soybean on the nitrogen-fixing bacteria, the diversity of nif H gene in pot experiments planted transgenic soybean and near-isogenic nontransgenic soybean under normal water condition and drought stress condition was analyzed by PCR-DGGE and sequence analysis. The results showed that transgenic soybean under normal water condition decrease the diversity of the nitrogen-fixing bacteria in the seeding stage and flowering stage, but had no notable effect in other stages. Under drought stress, transgenic soybean reduced the diversity of the nitrogen-fixing bacteria in the flowering stage, but had no notable effects on other stages. Phylogenic analysis revealed that g7, g13, g15 and g19 had a close relationship with Alphaproteobacteria, g12 had a close relationship with Azonexus, others were related to Betaproteobacteria and Burkholderia. 展开更多

Bacterial populations isolated from treated soil,artificially contaminated with lead(Pb)and cadmium(Cd)and undergoing a phytoremediation process were studied to determine their potential application in soil remediation.The physicochemical parameters evaluated in the soil varied significantly.Ten bacterial strains were selected from each polluted soil to test tolerance and growth in contaminated media.The concentrations of heavy metals tested were 1,000 ppm for lead and 850 ppm for cadmium.These strains were morphologically identified through Gram staining.Four strains showing the most significant growth in both contaminants were then selected to verify their tolerance to different concentrations of heavy metals.The results demonstrated that the selected bacteria have high tolerance to Pb,resisting inhibition up to 2,000 ppm.In contrast,strains exposed to cadmium tended to slow their growth as the concentration increased. 展开更多

The salt-resistant nitrogen-fixing cyanobacteria 888 was experimentally applied to the reclamation of saline and alkali soil in Songnen Plain in China. The pH, electrical conductivity (EC) and sodium adsorption ratio (SAR) of different saline soils were studied and compared. Results show that different saline soils exhibit various physico-chemical properties. Saline-sodic soils in Songnen Plain are ameliorated by using nitrogen-fixing blue-green algae 888 in the experiment. It is indicated that cyanobacteria 888 can grow in saline and alkaline soils, and the conditions favorable for its growth are soil moisture of 50% and dry algae inoculation at 0.03 mg/cm2. The main actions of nitrogen-fixing cyanobacteria are keeping the adsorbability of rubber sheath for sodium, increasing the organic matter content of the soils and decreasing the pH and the degree of salinity in the soils. But the arid climate and soil depth are the main factors that limit the restoration of saline and alkaline soils. 展开更多

Effects of N,P and K Fertilizers on Growth of Clover Nitrogen-fixing Rhizobia and Soil Fertility after Plantation

In order to develop organic rice and increase paddy soil fertility by cloverorganic rice rotation, the effects of N, P and K fertilizers on growth of clover nitrogen-fixing rhizobia and soil fertility after plantation were investigated, thereby providing certain reference for reasonable cultivation of clover and improvement of soil fertility. A two-year experiment was conducted from 2012 to 2013. The clover was cultivated after rice every year, and different levels of N, P and K fertilizers were applied before winter. In the treatment group, no fertilizer was applied. The effects of different fertilizers and different application amounts on clover yield, nitrogen-fixing rhizobia quantity, nitrogen-fixing rhizobia weight and soil fertility after plantation were analyzed. The results showed that the application of N, P and K fertilizers in the early stage had significantly effect on the growth of clover. When the application amount of N fertilizer was 75 kg/hm^2(N 46%), the clover yield, nitrogen-fixing rhizobia quantity and nitrogen-fixing rhizobia weight were highest. The soil nitrogen content after plantation increased with the increase of the application amount of N fertilizer, while the P and K content decreased and then increased with the increased application amounts. Soil available P content increased with the increased application amounts of N and P fertilizers, but it did not change significantly with the increased application amount of K fertilizer. Soil available K content increased first and then decreased with the increased application amounts of N and P fertilizers.When the application amounts of N and P fertilizers were 150(N 46%) and 300(P_2O_5 12%) kg/hm^2, soil available K content reached the maximum. Soil organic matter content increased with the increased application amounts of N, P and K fertilizers. Therefore, in the cultivation of clover, appropriate application of N, P and K fertilizers in the early stage can improve clover yield and soil fertility. 展开更多

The yield of winter wheat is hindered by drought and low temperature in the Loess Plateau of China.Two common mulching methods to conserve soil moisture,ridge furrows with plastic film mulching (RP) and flat soil surfaces with plastic film mulching (FP) are helpful for wheat production.Our previous study indicated that FP could improve wheat yield more effectively than RP,but the reason remains unclear.The effect of mulching method on functional bacteria also needs to be further studied.In this study,winter wheat was employed to evaluate the impacts of mulching method on soil temperature,moisture content,microorganisms and grain yield.The results showed that FP had a warming effect when the soil temperature was low and a cooling effect when the temperature was too high.However,the ability to regulate soil temperature in the RP method was unstable and varied with year.The lowest negative accumulated soil temperature was found in the FP treatment,which was 20–89 and 43–99%lower than that of the RP and flat sowing with non-film mulching control (NP) treatments,respectively.Deep soil moisture was better transferred to topsoil for wheat growth in the FP and RP treatments than the NP treatment,which made the topsoil moisture in the two treatments (especially FP) more sufficient than that in the NP treatment during the early growing stage of wheat.However,due to the limited water resources in the study area,there was almost no difference between treatments in topsoil water storage during the later stage.The wheat yield in the FP treatment was significantly higher,by 12–16and 23–56%,respectively,than in the RP and NP treatments.Significant positive correlations were observed among the negative accumulated soil temperature,spike number and wheat yield.The Chao1 and Shannon indices in the RP treatment were 17 and 3.9%higher than those in the NP treatment,respectively.However,according to network relationship analysis,the interspecific relationships of bacteria were weakened in the RP treatment.Phosphorus solubilizing,ammonification and nitrification bacteria were more active in the RP than in the FP treatment,and microbes with nitrate reduction ability and plant pathogens were inhibited in the RP treatment,which improved nutrient availability and habitat for wheat. 展开更多

Environmental DNA was extracted... Soil contains a great diversity of microorganisms, among which are bacteria. This study aimed to explore bacterial diversity in soil samples in Brazzaville in the Republic of the Congo. Environmental DNA was extracted. The illumina MiSeq sequencing was held and the diversity indices have been computed. Illumina MiSeq sequencing revealed 21 Phyla, four of which were abundant: Proteobacteria, Acidobacteria, Actinobacteria and Bacteroidetes. Soil microbial communities in the studied samples were phylogenetically diverse but with a stable community structure. 17 classes are represented with relative abundances of Rihzobiales, Bacillales, Actinomycetales and Acidobacteriales. 40 families, the Alphaproteobacteria, the Bacilli and the 12 Actinobacteria. 83 orders among which the Rhizobiales are the most abundant followed by Bacillales and the least abundant followed by the Flavobacteriaceae. Of the 28 genera listed, the Bradyrhizobium is the most dominant in Mw3 and Mw4. 25 listed species, Bradyrhizobium, Bacillus, Actinoplanes, and Candidatu coribacter Acidobacterium are the most abundant species. The Shannon indices of Mw3 and Mw4 are equal, the H’max of Mw4 is greater than the H’max of Mw3. The Simpson index of Mw4 is equal to the Simpson index of Mw3, and the Pielou index (J) of Mw4 is less than the R of Mw3, but very close. This study opens interesting perspectives on the knowledge and exploitation of telluric bacteria in several areas of life. 展开更多

A two-year field experiment conducted under dryland conditions in semi-humid and drought-prone regions of China aimed to assess the effect of ammonia-oxidizing bacterial on maize water use efficiency and yield.A heterotrophic ammonia-oxidizing bacteria(HAOB)strain S2_8_1 was used.Six treatments were applied:(1)no irrigation+HAOB strain(DI),(2)no irrigation+blank culture medium(DM),(3)no irrigation control(DCK),(4)irrigation+HAOB(WI),(5)irrigation+blank culture medium(WM),and(6)irrigation control(WCK).Results revealed that HAOB treatment increased maize growth,yield,and water use efficiency over controls,regardless of whether the year was wet or dry.This improvement was attributed to the accelerated nitrification in the rhizosphere soil due to HAOB inoculation,which subsequently led to increased levels of leaf cytokinins.Overall,these findings suggest that HAOB inoculation holds promise as a strategy to boost water use efficiency and maize productivity in dryland agriculture. 展开更多

Phthalic acid is a main pollutant, which is also an important reason for the continuous cropping effect of tobacco. In order to degrade the phthalic acid accumulated in the environment and relieve the obstacle effect of tobacco continuous cropping caused by the accumulation of phthalic acid in the soil. In this study, phthalate degrading bacteria B3 is screened from continuous cropping tobacco soil. The results of biochemical identification and 16sDNA comparison show that the homology between degrading bacterium B3 and Enterobacter sp. is 99%. At the same time, the growth of Enterobacter hormaechei subsp. B3 and the degradation of phthalic acid under different environmental conditions are studied. The results show that the environment with a temperature of 30˚C, PH of 7, and inoculation amount of not less than 1.2%, which is the optimal growth conditions for Enterobacter sp. B3. In an environment with a concentration of phthalic acid not exceeding 500 mg/L, Enterobacter sp. B3 has a better effect on phthalic acid degradation, and the degradation rate can reach 77% in 7 d. The results of indoor potting experiments on tobacco show that the degradation rate of phthalic acid by Enterobacter B3 in the soil is about 45%, which can reduce the inhibitory effect of phthalic acid on the growth of tobacco seedlings. This study enriches the microbial resources for degrading phthalic acid and provides a theoretical basis for alleviating tobacco continuous cropping obstacles. 展开更多

Peatlands are unique and complex natural ecosystems that are part of the most important carbon reservoirs on our planet, home to a diversity of microorganisms responsible for fermentation, humification or peat. The aim is to understand chemical and biological indicators of peatland soils. This work aims to determine the physicochemical and bacteriological profile and lipolytic activity of soil bacteria in Bilanko peatlands. The bacterial profile with the production of lipases is carried out by classical microbiology techniques. The results show that the soils are moderately acidic with temperatures of 27.8˚C ± 0.01˚C for Bilanko and 27.1˚C ± 0.57˚C for Ngamakala. The electroconductivity (EC) varies from (9.52 ± 0.002) μS/cm to (39.01 ± 1.4) μS/cm with low turbidity of (2.04 ± 0.66) mg/L to (31.02 ± 0.84) mg/L and low ion concentrations with, however, a richness in phenolic compounds for Bilanko compared to Ngamakala. FMAT diversity ranged from (1.71 ± 0.88)∙104 UFC/g to (2.92 ± 0.07)∙105 UFC/g for Bilanko and (1.30 ± 0.73)∙104 UFC/g to (2.89 ± 0.06)∙104 UFC/g for Ngamakala. Bacillus loads ranged from (5.20 ± 1.40)∙103 CFU/g to (1.22 ± 0.13)∙104 CFU/g and from (1.11 ± 0.13)∙104 CFU/g to (9.20 ± 2.05)∙103 CFU/g;enterobacteria loads from (1.40 ± 0.76)∙103 CFU/g to (8.80 ± 1.73)∙103 CFU/g and from (1.01 ± 0.02)∙103 CFU/g to (9.20 ± 2.05)∙103 CFU/g;in Pseudomonas from 0 to (2.30 ± 0.53)∙102 CFU/g and from 0 to (8.90 ± 2.35)∙102 CFU/g for Bilanko and Ngamakala respectively. These results reveal a variation in bacterial similarity and distribution in the Bilanko and Ngamakala peat bogs. 展开更多

A pot experiment was conducted to examine the effects of a phosphate solubilizing bacterium(PSB),Enterobacteria sp.EnHy-401,on the availability of insoluble accumulative phosphorus(P)and growth of wheat(Triticum Ningmai No.13)plants in sterile saline soil.Our results showed that the strain EnHy-401 had the ability to activate the insoluble accumulated phosphorus in saline soil and enhanced nutrient uptake efficiency by wheat plants,then conferred resistance in wheat plants to salt stress and resulted in a significant growth increase.In saline soil inoculated with Enterobacteria sp.EnHy-401,available phosphorus and exchangeable calcium was increased from 6.4 mg/kg and 1 162 mg/kg to 10.3 mg/kg and 1 214 mg/kg,respectively.Wheat seedling grown in soil inoculated with the EnHy-401 strain increased shoot weight by 28.1% and root weight by 14.6% when compared to the control.P,Ca,K and Mg contents in shoots increased 34.4%,36.3%,31.5%,and 6.3% compared to the control,respectively.the fact that the increases in available P,biomass P,and Ca2+ concentration in saline soil treated with PSB Enterobacter sp.EnHy-401 inocula,and high relativity between the P,Ca,K,and Mg content in wheat tissue and dry matter indicated that PSB Enterobacter sp.EnHy-401 suppressed the adverse effect of salinity stress in plants through nutrient(P and Ca)supply and nutrient(K and Mg)uptake enhancement.The phosphate solubilizing activity of Enterobacteria sp.EnHy-401 and the amelioration of salt stress on wheat plants by the strain varied with the salinity levels and content of organic matter in the saline soil. 展开更多

Black soil (Mollisol) is one of the main soil types in northeastern China. Biolog and polymerase chain reactiondenaturing gradient gel electrophoresis (PCR-DGGE) methods were used to examine the influence of various fertilizer combinations on the structure and function of the bacterial community in a black soil collected from Harbin, Heilongjiang Province. Biolog results showed that substrate richness and catabolic diversity of the soil bacterial community were the greatest in the chemical fertilizer and chemical fertilizer+manure treatments. The metabolic ability of the bacterial community in the manure treatment was similar to the control. DGGE fingerprinting indicated similarity in the distribution of most 16S rDNA bands among all treatments, suggesting that microorganisms with those bands were stable and not influenced by fertilization. However, chemical fertilizer increased the diversity of soil bacterial community. Principal com- ponent analysis of Biolog and DGGE data revealed that the structure and function of the bacterial community were similar in the control and manure treatments, suggesting that the application of manure increased the soil microbial population, but had no effect on the bacterial community structure. Catabolic function was similar in the chemical fertilizer and chemical fertilizer+manure treatments, but the composition structure of the soil microbes differed between them. The use of chemical fertilizers could result in a decline in the catabolic activity of fast-growing or eutrophic bacteria. 展开更多

The effect of forests clear felling and associated burning on the population of soil nitrogen transforming bacteria and actinomycetes are reported at three pair sites of Chittagong University campus, Bangla- desh in monsoon tropical climate. Clear felled area or burnt site and 15-21 year mixed plantation of native and exotic species, situated side by side on low hill having Typic Dystrochrepts soil was represented at each pair site. At all the three pair sites, clear felled area or burnt site showed very significantly (p~_0.001) lower population of actinomycetes, Rhizobium, Nitrosomonas, Nitrobacter and ammonifying as well as deni- trifying bacteria compared to their adjacent mixed plantation. From en- vironmental consideration, this finding has implication in managing natural ecosystem. 展开更多

In this study we evaluated the bacterial diversity in a soil sample from a site next to a chemical industrial factory previously contaminated with heavy metals. Analysis of 16S rDNA sequences amplified from DNA directly extracted from the soil revealed 17 different bacterial types (genera and/or species). They included Polyangium spp., Sphingomonas spp., Variovorax spp., Hafina spp., Clostridia, Acidobacteria, the enterics and some uncultured strains. Microbes able to tolerate high concentrations of cadmium (500μmol/L and above) were also isolated from the soil. These isolates included strains of Acinetobacter (strain CD06), Enterobacter sp. (strains CD01, CD03, CD04 and CD08) (similar strains also identified in culture-independent approach) and a strain of Stenotrophomonas sp. The results indicated that the species identified from direct analysis of 16S rDNA of the soil can be quite different from those strains obtained from enrichment cultures and the microbial activities for heavy metal resistance might be more appropriately addressed by the actual isolates. 展开更多

An ancient irrigated paddy soil from the Neolithic age was excavated at Chuodunshan Site in the Yangtze River Delta, close to Suzhou, China. The soil organic matter (SOM) content in the prehistoric rice soil is comparable to the average SOM content of present rice soils in this region, but it is about 5 times higher than that in the parent materials. As possible biomarkers to indicate the presence of the prehistoric paddy soil, the bacterial communities were investigated using the techniques of aerobic and anaerobic oligotrophic bacteria enumeration, Biolog analysis, and polymerase chain reaction-denaturing gradient gel electrophoresis (PCR-DGGE). The results showed that in the buried soil layers, the prehistoric paddy soil had the largest number of aerobic and anaerobic oligotrophic bacteria, up to 6.12 and 5.86 log cfu g-1 dry soil, respectively. The prehistoric paddy soil displayed better carbon utilization potential and higher functional diversity compared to the parent materials and a prehistoric loess layer. The Shannon index and richness based on DGGE profiles of bacterial 16S rRNA genes were higher in prehistoric paddy soil than those in the prehistoric loess soil. It might be concluded that the prehistoric irrigated rice cultivation accumulated the SOM in plowed soil layer, and thus increased soil bacterial populations, metabolic activity, functional diversity and genetic diversity. Bacterial communities might be considered as the sensitive indicators of the presence of the prehistoric paddy soil in China's Yangtze River Delta. 展开更多

Adding phosphate- based fertilizers to increase agricultural yields is a widely used practice;however, the bio- availability of P remains low due to chemical transformations of P into insoluble forms. Thus, phosphate solubilizing bacteria (PSB) play an important role in reducing P deficiency in soil. The goal of this study was to assess biotechnological potential of phosphate-solubilizing bacterial strains. In this study, phosphate solubilizing microorganisms (PSM) were isolated from different soil samples of Southern regions of Kazakhstan. The biological activity of PSM was studied based on their effect on the growth of wheat seeds. The different taxonomic genera of these PSM were identified: Arthrobacter spp., Aureobacterium spp., Azotobacter spp., Bacterium spp., Baccillus spp. Finally, phosphate- solubilizing activity of isolated strains of PSM was assessed. 展开更多

In addition to bacteria,the contribution of fungi to nitrous oxide(N_(2)O)production has been recognized but the responses of these two broad and unrelated groups of microorganisms to global environmental changes,atmospheric nitrogen(N)deposition,and precipitation in terms of N_(2)O production are unclear.We studied how these two microbial-mediated N_(2)O production pathways responded to soil moisture conditions and to N addition in an N-limited temperate forest.Soils from a long-term N addition experiment in Changbai Mountain,northeastern China were incubated.Varied concentrations of cycloheximide and streptomycin,both inhibitors of fungal and bacterial activity,were used to determine the contributions of both to N_(2)O production in 66%,98%and 130%water-filled pore spaces(WFPS).The results showed that N_(2)O production decreased significantly with increasing cycloheximide concentration whereas streptomycin was only inhibiting N_(2)O emissions at 98%and 130%WFPS.The bacterial pathway of N_(2)O production in N-addition(Nadd)soil was significantly more dominant than that in untreated(Namb)soil.The difference in the fungal pathway of N_(2)O production between the soil with nitrogen addition and the untreated soil was not significant.Net N_(2)O emissions increased with increasing soil moisture,especially at 130%WFPS,a completely flooded condition.Bacteria dominated carbon dioxide(CO_(2))and N_(2)O emissions in Nadd soil and at 130%WFPS regardless of N status,while fungi dominated CO_(2)and N_(2)O emissions in soil without N addition at 66%and 98%WFPS.The results suggest that flooded soil is an important source of N_(2)O emissions and that bacteria might be better adapted to compete in fertile soils under anoxic conditions. 展开更多

The utilization of cattle and poultry manure as organic fertilizer improves soil productivity, but arsenic contaminated poultry dung may interfere in soil metabolism and soil fertility. The study was conducted to assess the effects of poultry dung as well as arsenic contamination on soil properties in 1%, 3% and 5% poultry dung amended soil and 1, 5 and 10 ppm sodium arsenite contaminated soil. pH and conductivity were found to be increasing with increase in poultry dung in soil. Other chemical parameters like nitrate, phosphate and organic carbon were found higher in poultry dung amended soil than that of arsenic contaminated soil. Soil bacteria, CO2 evolution and enzymatic activities like amylase, invertase and dehydrogenase were also found higher in poultry dung amended soil suggesting the effectiveness of poultry dung in enhancing soil productivity, even if it was contaminated by As through feed additive. 展开更多
