目的探讨Sprouty2蛋白与人乳腺癌MCF-7细胞增殖与迁徙的关系。方法通过siRNA技术干扰MCF-7细胞Sprouty2基因的表达,通过Real time PCR、Western blotting以及细胞免疫荧光检测Sprouty2基因的干扰效果,划痕实验以及Transwell实验观察细...目的探讨Sprouty2蛋白与人乳腺癌MCF-7细胞增殖与迁徙的关系。方法通过siRNA技术干扰MCF-7细胞Sprouty2基因的表达,通过Real time PCR、Western blotting以及细胞免疫荧光检测Sprouty2基因的干扰效果,划痕实验以及Transwell实验观察细胞迁移和侵袭的能力,Western blotting检测MMP-2、MMP-9和MMP-13的表达。结果通过siRNA干扰,有效地干扰了Sprouty2基因的表达,建立了Sprouty2基因低表达的细胞株;Sprouty2基因沉默后,细胞的迁移和侵袭能力明显增强;沉默Sprouty2基因的MCF-7细胞中MMP-2、MMP-9和MMP-13蛋白较对照组均升高。结论 Sprouty2基因下调后,MCF-7细胞的迁移和侵袭能力明显提高,可能与MMP-2、MMP-9和MMP-13蛋白有关。展开更多
