BACKGROUND: In a multidisciplinary conference patients with advanced non-resectable hepatocellular carcinoma (HCC) were stratified according to their clinical status and tumor extent to different regional modalities o...BACKGROUND: In a multidisciplinary conference patients with advanced non-resectable hepatocellular carcinoma (HCC) were stratified according to their clinical status and tumor extent to different regional modalities or to best supportive care. The present study evaluated all patients who were stratified to repeated transarterial chemoembolization (TACE) from 1999 until 2003 in terms of tumor response, toxicity, and survival. A moderate embolizing approach was chosen using a combination of degradable starch microspheres (DSM) and iodized oil (Lipiodol) in order to combine anti-tumoral efficiency and low toxicity. METHODS: Fourty-seven patients were followed up prospectively. TACE treatment consisted of cisplatin (50 mg/m2), doxorubicin (50 mg/m2), 450-900 mg DSM, and 5-30 ml Lipiodol. DSM and Lipiodol were administered according to tumor vascularization. Patient characteristics,toxicity, and complications were outlined. In multivariate regression analyses of pre-treatment variables from a prospective database, predictors for tumor response and survival after TACE were determined. RESULTS: 112 TACE courses were performed (2.4±1.5 courses per patient). Mean maximum tumor size was 75 (± 43) mm, in 68% there was bilobar disease. Best response to TACE treatment was: progressive disease (PD) 9%, stable disease (SD) 55%, partial remission (PR) 36%, and complete remission (CR) 0%. Multivariate regression analyses identified tumor size ≤75 mm, tumor number ≤5, and tumor hypervascularization as predictors for PR. The overall 1-, 2-, and 3-year-survival rates were 75%, 59%, and 41%, respectively, and the median survival was 26 months. Low α-fetoprotein levels (<400 ng/ml) (Odds ratio=3.3) and PR as best response to TACE (Odds ratio=6.7) were significantly associated with long term survival (>30 months, R2=36%). Grade 3 toxicity occurred in 7.1% (n=8), and grade 4 toxicity in 3.6% (n=4) of all courses in terms of reversible leukopenia and thrombocytopenia. The incidence of major complications was 5.4% (n=6). All complications were managed conservatively. The mortality within 6 weeks after TACE was 2.1% (one patient). CONCLUSIONS: DSM and Lipiodol were combined successfully in the palliative TACE treatment of advanced HCC resulting in high rates of tumor response and survival at limited toxicity. Favourable tumor response was associated with tumor extent and vascularization. TACE using DSM and Lipiodol can be considered a suitable palliative measure in patients who might not tolerate long acting embolizing agents.展开更多
Background:Starch is an important substance that supplies energy to ruminants.To provide sufficient energy for high-yielding dairy ruminants,they are typically fed starch-enriched diets.However,starch-enriched diets h...Background:Starch is an important substance that supplies energy to ruminants.To provide sufficient energy for high-yielding dairy ruminants,they are typically fed starch-enriched diets.However,starch-enriched diets have been proven to increase the risk of milk fat depression(MFD)in dairy cows.The starch present in ruminant diets could be divided into rumen-degradable starch(RDS)and rumen escaped starch(RES)according to their different degradation sites(rumen or intestine).Goats and cows have different sensitivities to MFD.Data regarding the potential roles of RDS in milk fat synthesis in the mammary tissue of dairy goats and in regulating the occurrence of MFD are limited.Results:Eighteen Guanzhong dairy goats(day in milk=185±12 d)with similar parity,weight,and milk yield were selected and randomly assigned to one of three groups(n=6),which were fed an LRDS diet(Low RDS=20.52%),MRDS diet(Medium RDS=22.15%),or HRDS diet(High RDS=24.88%)for 5 weeks.Compared with that of the LRDS group,the milk fat contents in the MRDS and HRDS groups significantly decreased.The yields of short-,mediumand long-chain fatty acids decreased in the HRDS group.Furthermore,increased RDS significantly decreased ruminal B.fibrisolvens and Pseudobutyrivibrio abundances and increased the trans-10,cis-12 conjugated linoleic acid(CLA)and trans-10 C18:1 contents in the rumen fluid.A multiomics study revealed that the HRDS diet affected mammary lipid metabolism down-regulation of ACSS2,MVD,AGPS,SCD5,FADS2,CERCAM,SC5D,HSD17B7,HSD17B12,ATM,TP53RK,GDF1 and LOC102177400.Remarkably,the significant decrease of INSIG1,whose expression was depressed by trans-10,cis-12 CLA,could reduce the activity of SREBP and,consequently,downregulate the downstream gene expression of SREBF1.Conclusions:HRDS-induced goat MFD resulted from the downregulation of genes involved in lipogenesis,particularly,INSIG1.Specifically,even though the total starch content and the concentrate-to-fiber ratio were the same as those of the high-RDS diet,the low and medium RDS diets did not cause MFD in lactating goats.展开更多

METHODS: DSM were injected into the proper hepatic artery through a sila...AIM: To observe the dynamic changes of liver microcirculation in vivo after arterial embolization with degradable starch microspheres (DSM). METHODS: DSM were injected into the proper hepatic artery through a silastic tube inserted retrogradely in gastroduodenal artery (GDA) of SD rats. Fluorescent microscopy was used to evaluate the dynamic changes of blood flow through the terminal portal venules (TPVs), sinusoids and terminal hepatic venules (THVs). The movements of DSM debris were also recorded. Six hours after injection of DSM, percentages of THVs with completely stagnant blood flow were recorded. RESULTS: Two phases of blood flow change were recorded. In phase one: after intra-arterial injection of DSM, slow or stagnant blood flow was immediately recorded in TPVs, sinusoids and THVs. This change was reversible, and blood flow resumed completely. In phase two: after phase one, blood flow in TPVs changed again and three patterns of blood flow were recorded. Six hours after DSM injection, 36.9% ± 9.2% of THVs were found with completely stagnant blood flow. CONCLUSION: DSM can stop the microcirculatory blood flow in some areas of liver parenchyma. Liver parenchyma supplied by arteries with larger A-P shunt is considered at a higher risk of total microcirculatory blood stagnation after injection of DSM through hepatic artery.展开更多
Since starch digestion in the small intestine provides more energy than digestion in the rumen of ru-minants,reducing dietary rumen degradable starch(RDS)content is beneficial for improving energy utilization of starc...Since starch digestion in the small intestine provides more energy than digestion in the rumen of ru-minants,reducing dietary rumen degradable starch(RDS)content is beneficial for improving energy utilization of starch in ruminants.The present study tested whether the reduction of rumen degradable starch by restricting dietary corn processing for growing goats could improve growth performance,and further investigated the possible underlying mechanism.In this study,twenty-four 12-wk-old goats were selected and randomly allocated to receive either a high RDS diet(HRDS,crushed corn-based concen-trate,the mean of particle sizes of corn grain=1.64 mm,n=12)or a low RDS diet(LRDS,non-processed corn-based concentrate,the mean of particle sizes of corn grain>8 mm,n=12).Growth performance,carcass traits,plasma biochemical indices,gene expression of glucose and amino acid transporters,and protein expression of the AMPK-mTOR pathway were measured.Compared to the HRDS,LRDS tended to increase the average daily gain(ADG,P=0.054)and decreased the feed-to-gain ratio(F/G,P<0.05).Furthermore,LRDS increased the net lean tissue rate(P<0.01),protein content(P<0.05)and total free amino acids(P<0.05)in the biceps femoris(BF)muscle of goats.LRDS increased the glucose concen-tration(P<0.01),but reduced total amino acid concentration(P<0.05)and tended to reduce blood urea nitrogen(BUN)concentration(P=0.062)in plasma of goats.The mRNA expression of insulin receptors(INSR),glucose transporter 4(GLUT4),L-type amino acid transporter 1(LAT1)and 4F2 heavy chain(4F2hc)in BF muscle,and sodium-glucose cotransporters 1(SGLT1)and glucose transporter 2(GLUT2)in the small intestine were significantly increased(P<0.05)in LRDS goats.LRDS also led to marked activation of p70-S6 kinase(S6K)(P<0.05),but lower activation of AMP-activated protein kinase(AMPK)(P<0.05)and eukaryotic initiation factor 2a(P<0.01).Our findings suggested that reducing the content of dietary RDS enhanced postruminal starch digestion and increased plasma glucose,thereby improving amino acid utilization and promoting protein synthesis in the skeletal muscle of goats via the AMPK-mTOR pathway.These changes may contribute to improvement in growth performance and carcass traits in LRDS goats.展开更多

In the glucose-fed chemostat, the aceticlastic methanogen Methanosaeta spp. and hydrogenotrophic methanogen Methanoculleus spp. predominated at low dilution rates, whereas Methanosaeta spp. and the hydrogenotrophic Methanobacterium spp. predominated together when dilution rates were greater than 0.1 d 1. Bacteria affiliated with the phyla Bacteroidetes, Spiro- chaetes, and Actinobacteria predominated at dilution rates of 0.05, 0.1, and 0.15 d-l, respectively, while Firmicutes predominated at higher dilution rates (0.2 and 0.25 d-l). In the starch-fed chemostat, the aceticlastic and hydrogeno- trophic methanogens coexisted at all dilution rates. Although bacteria belonging to only two phyla were mainly responsible for starch degradation (Spirochaetes at the dilution rate of 0.08 d-1 and Firmicutes at other dilution rates), different bacterial genera were identified at different dilution rates. With the exception of Archaea in the glucose-fed chemostat, the band patterns revealed by denaturing gradient gel electrophoresis (DGGE) of the microbial communities in the two chemostats displayed marked changes during long-term operation at a constant dilution rate. The ethylene insensitive 3/ethylene insensitive3-like(EIN3/EIL)plays an indispensable role in fruit ripening.However,the regulatory mechanism that links post-translational modification of EIN3/EIL to fruit ripening is...The ethylene insensitive 3/ethylene insensitive3-like(EIN3/EIL)plays an indispensable role in fruit ripening.However,the regulatory mechanism that links post-translational modification of EIN3/EIL to fruit ripening is largely unknown.Here,we studied the expression of 13 MaE IL genes during banana fruit ripening,among which MaE IL9 displayed higher enhancement particularly in the ripening stage.Consistent with its transcript pattern,abundance of MaE IL9 protein gradually increased during the ripening process,with maximal enhancement in the ripening.DNA affinity purification(DAP)-seq analysis revealed that MaE IL9 directly targets a subset of genes related to fruit ripening,such as the starch hydrolytic genes MaA MY3D and MaB AM1.Stably overexpressing MaE IL9 in tomato fruit hastened fruit ripening,whereas transiently silencing this gene in banana fruit retarded the ripening process,supporting a positive role of MaEIL9 in fruit ripening.Moreover,oxidation of methionines(Met-129,Met-130,and Met-282)in MaEIL9 resulted in the loss of its DNA-binding capacity and transcriptional activation activity.Importantly,we identified MaEIL9 as a potential substrate protein of methionine sulfoxide reductase A MaMsrA4,and oxidation of Met-129,Met-130,and Met-282in MaEIL9 could be restored by MaMsrA4.Collectively,our findings reveal a novel regulatory network controlling banana fruit ripening,which involves MaMsrA4-mediated redox regulation of the ethylene signaling component MaEIL9.展开更多
