Induced neural stem cells regulate microglial activation through Akt-mediated upregulation of CXCR4 and Crry in a mouse model of closed head injury

Microglial activation that occurs rapidly after closed head injury may play important and complex roles in neuroinflammation-associated neuronal damage and repair.We previously reported that induced neural stem cells can modulate the behavior of activated microglia via CXCL12/CXCR4 signaling,influencing their activation such that they can promote neurological recovery.However,the mechanism of CXCR4 upregulation in induced neural stem cells remains unclear.In this study,we found that nuclear factor-κB activation induced by closed head injury mouse serum in microglia promoted CXCL12 and tumor necrosis factor-αexpression but suppressed insulin-like growth factor-1 expression.However,recombinant complement receptor 2-conjugated Crry(CR2-Crry)reduced the effects of closed head injury mouse serum-induced nuclear factor-κB activation in microglia and the levels of activated microglia,CXCL12,and tumor necrosis factor-α.Additionally,we observed that,in response to stimulation(including stimulation by CXCL12 secreted by activated microglia),CXCR4 and Crry levels can be upregulated in induced neural stem cells via the interplay among CXCL12/CXCR4,Crry,and Akt signaling to modulate microglial activation.In agreement with these in vitro experimental results,we found that Akt activation enhanced the immunoregulatory effects of induced neural stem cell grafts on microglial activation,leading to the promotion of neurological recovery via insulin-like growth factor-1 secretion and the neuroprotective effects of induced neural stem cell grafts through CXCR4 and Crry upregulation in the injured cortices of closed head injury mice.Notably,these beneficial effects of Akt activation in induced neural stem cells were positively correlated with the therapeutic effects of induced neural stem cells on neuronal injury,cerebral edema,and neurological disorders post–closed head injury.In conclusion,our findings reveal that Akt activation may enhance the immunoregulatory effects of induced neural stem cells on microglial activation via upregulation of CXCR4 and Crry,thereby promoting induced neural stem cell–mediated improvement of neuronal injury,cerebral edema,and neurological disorders following closed head injury. 展开更多

Stem cell-based therapies have been proposed as a potential treatment for neural regeneration following closed head injury.We previously reported that induced neural stem cells exert beneficial effects on neural regeneration via cell replacement.However,the neural regeneration efficiency of induced neural stem cells remains limited.In this study,we explored differentially expressed genes and long non-coding RNAs to clarify the mechanism underlying the neurogenesis of induced neural stem cells.We found that H19 was the most downregulated neurogenesis-associated lnc RNA in induced neural stem cells compared with induced pluripotent stem cells.Additionally,we demonstrated that H19 levels in induced neural stem cells were markedly lower than those in induced pluripotent stem cells and were substantially higher than those in induced neural stem cell-derived neurons.We predicted the target genes of H19 and discovered that H19 directly interacts with mi R-325-3p,which directly interacts with Ctbp2 in induced pluripotent stem cells and induced neural stem cells.Silencing H19 or Ctbp2 impaired induced neural stem cell proliferation,and mi R-325-3p suppression restored the effect of H19 inhibition but not the effect of Ctbp2 inhibition.Furthermore,H19 silencing substantially promoted the neural differentiation of induced neural stem cells and did not induce apoptosis of induced neural stem cells.Notably,silencing H19 in induced neural stem cell grafts markedly accelerated the neurological recovery of closed head injury mice.Our results reveal that H19 regulates the neurogenesis of induced neural stem cells.H19 inhibition may promote the neural differentiation of induced neural stem cells,which is closely associated with neurological recovery following closed head injury. 展开更多

The method of numerical simulation was applied to investigate gas preheating and start-up process in a drained aluminum reduction cell. The transient temperature and velocity fields of a 156 kA drained aluminum reduction cell were numerically calculated. The results show that the method of gas preheating and bake-out can basically meet the technical requirements of the start-up process for the drained cell. If the same distributing scheme of gas nozzle as that in the general cells is used, there are problems of great temperature gradients and low temperature zone at the top of cathode near the side of nozzles. In order to promote the effect of gas preheating and baking the drained cell, the jetting angle of end nozzle is adjusted and the temperature distribution in the drained cell is obviously improved. By means of simulating the temperature field in the case that jetting angle varies from 0? to 30?, it is concluded that better temperature distribution can be obtained if the jetting angle of end nozzle is approximately 15?. 展开更多

AIM: To observe the inhibition of hepatitis B virus (HBV) replication and expression in HepG2.2.15 cells by combination of small interfering RNAs (siRNAs). METHODS: Recombinant plasmid psiI-HBV was constructed and transfected into HepG2.2.15 cells. At 48 h, 72 h and 96 h after transfection, culture media were collected and cells were harvested for HBV replication assay. HBsAg and HBeAg in the cell culture medium were detected by enzyme-linked immunoadsorbent assay (ELISA). Intracellular viral DNA and covalently closed circular DNA (cccDNA) were quantified by real-time polymerase chain reaction (PCR). HBV viral mRNA was reverse transcribed and quantified by reverse-transcript PCR (RT-PCR). RESULTS: siRNAs showed marked anti-HBV effects. siRNAs could specifically inhibit the expression of HBsAg and the replication of HBV DNA in a dosedependent manner. Furthermore, combination of siRNAs, compared with individual use of each siRNA, exerted a stronger inhibition on antigen expression and viral replication. More importantlycombination of siRNAs significantly suppressed HBV cccDNA amplification. CONCLUSION: Combination of siRNAs mediates a stronger inhibition on viral replication and antigenexpression in HepG2.2.15 cells, especially on cccDNA amplification. 展开更多

The quasi-static compressive mechanical behavior and deformation mechanism of closed-cell magnesium foams were studied, and the ef- fects of the density of magnesium foams on the compressive and energy absorption properties were also discussed. The results show that the compressive process of closed-cell magnesium foams is characterized by three deformation stages: linear elastic stage, collapsing stage and densification stage. At the linear elastic stage, the peak compressive strength (t70) and Young's modulus (E0) increase as the density increases Magnesium foams can absorb energy at the collapsing stage. In a certain strain range, the energy absorption capacity also increases as the density of magnesium foams increases. 展开更多

The closed-cell aluminum foams (specimenρ=0.31 g/cm3, diameter of 100 mm, and thickness of 20 mm for sound absorption testing; specimenρ=0.51 g/cm3, length of 1 240 mm, width of 1 100 mm, and thickness of 30 mm for sound insulation testing) were prepared by the method of molten body transitional foaming process. Its sound absorption property under frequency of 160-2 000 Hz and the sound insulation property under frequency of 100-4 000 Hz were tested. The sound absorption results show that the sound absorption property is much better under middle frequencies than that under low and high frequencies. The sound absorption coefficient climbs when frequency increases from 160 Hz to 800 Hz and then drops when frequency is increased from 800 Hz to 2 000 Hz. The function of the sound absorption mainly depends on the Helmholtz resonator, the microphone as well as cracks of closed-cell aluminum foam. The sound insulation experiments show that the sound reduction index (R) is small under low frequencies, and large under high frequencies; the weighted sound reduction index (Rw) and the highest sound reduction index (R) can reach around 30.8 dB and 43 dB, respectively. 展开更多

The vesicant problem during the process of preparing closed-cell aluminum foam by molten body transitional foaming process was discussed and the effect of granularity and addition of TiH2on porosity of closed-cell aluminum foam was investigated. The static compressive behavior of closed-cell aluminum foam and the influence of porosity on static compressive property of closed-cell aluminum foam were researched as well. The results show that with increasing granularity of TiH2,the porosity of closed-cell aluminum foam firstly increases and then decreases gradually, the granularity should be controlled in the range of 38-74μm which can result in higher porosity. The porosity of closed-cell aluminum foam increases with the increasing addition of TiH2, and the addition of TiH2 should be controlled from 1.5% to 2.5% which can result in homogeneous cell and moderate strength of closed-cell aluminum foam. The compressive process of closed-cell aluminum foam obviously displays linear elastic phase, plastic collapse phase, and densification phase, and the compressive strength grows with decreasing porosity. 展开更多

Closed-cell aluminum foam was shot peened at different processing time (0 s, 5 s, 10 s, and 20 s), the intensity was the 0.12 mmA. The X-ray diffraction results showed that the reflections became weakened obviously with the shot peened time increased. Combined with Popa model and lognormal distribute model, the surface microstructure of closed-cell aluminum foam was inves-tigated by using the Rietveld whole pattern fitting analysis method. The results revealed that domain size and microstrain fluctuated along different reflection directions after shot peened, which attributed to the random and anisotropic deformation direction. With the shot peened processing time prolonged, a decrease in domain size and an increase in microstrain were also observed. Moreover, the corrosion behavior of closed-cell aluminum foam was studied by weight-loss test. The results indicated that corrosion properties of specimen subjected to shot peened processing was better than the unpeened specimens. 展开更多

目的探究小切口切除术联合持续负压封闭引流对浆细胞性乳腺炎(PCM)患者疗效、乳房毁损率及换药次数的影响。方法选取2016年6月至2021年6月西安交通大学第二附属医院收治的PCM女性患者163例作为研究对象,随机将其分为试验组(75例)和对照... 目的探究小切口切除术联合持续负压封闭引流对浆细胞性乳腺炎(PCM)患者疗效、乳房毁损率及换药次数的影响。方法选取2016年6月至2021年6月西安交通大学第二附属医院收治的PCM女性患者163例作为研究对象,随机将其分为试验组(75例)和对照组(88例)。对照组患者采用持续负压封闭引流进行治疗,试验组在对照组基础上联合使用小切口切除术,对比两组患者的临床疗效、乳房完整性、手术指标、不良反应及复发率情况。结果对照组总有效率(86.36%)低于试验组(97.33%)(P<0.05);乳房完整性比较,试验组优秀的占52.00%,差的占1.33%;对照组优秀的占31.82%,差的占11.36%,试验组乳房完整性优于对照组(P<0.05);试验组手术时间、切口长度和愈合时间均优于对照组(P<0.05);试验组不良反应发生率(4.00%)低于对照组(6.82%),差异无统计学意义(P>0.05);试验组复发率(2.67%)低于对照组(14.77%)(P<0.05)。结论小切口切除术联合持续负压封闭引流能提高PCM患者的临床疗效,降低乳房毁损率。 展开更多

本文报道1例急性粒-单核细胞白血病(急性髓系白血病M4型)(CBFB基因阳性)患者,经亲缘半相合造血干细胞移植后并发下肢微血栓形成,并总结其诊治经验。该患者经常规化疗后获完全缓解,但又于2年后复发,遂予行亲缘半相合造血干细胞移植。在... 本文报道1例急性粒-单核细胞白血病(急性髓系白血病M4型)(CBFB基因阳性)患者,经亲缘半相合造血干细胞移植后并发下肢微血栓形成,并总结其诊治经验。该患者经常规化疗后获完全缓解,但又于2年后复发,遂予行亲缘半相合造血干细胞移植。在患者接受移植后的常规治疗期间,动态观察其凝血相关指标[包括D-二聚体(D-dimer, D-D)、抗凝血酶Ⅲ(antithrombinⅢ,AT-Ⅲ)、血管性血友病因子(von Willebrand factor, vWF)、纤维蛋白原降解产物(fibrinogen degradation products, FDP)、纤维蛋白单体(fibrin monomer, FM)]。在移植后100 d左右,患者出现黄疸及转氨酶升高,采用经右下肢股静脉置管行血浆置换治疗3次。在去除静脉置管1周后,患者出现左踝部指凹性水肿,血管B超检查提示其左下肢腓肠肌浅静脉丛有微血栓形成;凝血指标检测显示,D-D、vWF、FDP、FM等指标升高,AT-Ⅲ水平降低。予口服拜瑞妥5 mg/d抗栓治疗2周后,患者上述凝血指标渐趋正常,左下肢肌间静脉丛血栓和肢肿消失。在造血干细胞移植后,除密切检查血管B超,动态监测D-D、AT-Ⅲ、vWF、FDP、FM等凝血指标可预测移植相关微血栓和高凝状态,并为临床开展有效抗的凝治疗提供实验依据。 展开更多

机翼梁模型对于颤振预测和飞行载荷识别十分重要。采用MATLAB+Hypermesh联合建模方法建立细节模型,基于闭室原理建立梁单元机翼模型(简称梁模型),对梁模型进行了模态试验,并采用秃鹫算法对梁模型进行修正。比较了细节模型与修正后梁模... 机翼梁模型对于颤振预测和飞行载荷识别十分重要。采用MATLAB+Hypermesh联合建模方法建立细节模型,基于闭室原理建立梁单元机翼模型(简称梁模型),对梁模型进行了模态试验,并采用秃鹫算法对梁模型进行修正。比较了细节模型与修正后梁模型的频率误差与MAC值,结果显示,修正后的梁模型能够达到和细节模型相当的精度,梁模型的频率误差在5%以内,MAC值对角元素大于0.75。基于正态分布的不确定性反向传递研究结果表明,细节模型保留的局部信息较多,用于不确定性研究结果更加准确,未经修正的梁模型精度过低,不能用于不确定性研究,而修正后的梁模型也能满足不确定性研究的精度要求。 展开更多

车用石墨板燃料电池低温冷启动能力弱,是影响燃料电池技术在北方寒冷地区大规模推广的重要瓶颈。饥饿自升温是一种常见的低温冷启动策略,其基本原理是通过降低反应物供应速率来增加过电势,短时间在电池内部产生大量热量从而实现快速升... 车用石墨板燃料电池低温冷启动能力弱,是影响燃料电池技术在北方寒冷地区大规模推广的重要瓶颈。饥饿自升温是一种常见的低温冷启动策略,其基本原理是通过降低反应物供应速率来增加过电势,短时间在电池内部产生大量热量从而实现快速升温。该方法原理简单,但对电堆单体初始含水量一致性要求高、且易出现单片反极和尾排氢浓度超标的情况,影响系统安全性和电堆耐久性。针对上述问题,课题组研制了单体多通道交流阻抗在线测试装置,提出了面向单片阻抗一致性的电堆优化吹扫策略,建立了基于定电压变流量泵氢控制的低温冷启动方法,实现了低温启动瞬态过程的高产热、高安全、高动态的电压、电流、进堆/旁通空气流量的多目标多参数的耦合协调控制。台架实验结果表明,采用优化吹扫策略后,单片阻抗间最大差值由0.7降低至0.2 mΩ以下;燃料电池发动机系统可实现124 s内-40℃下快速启动,且重复性好。相关技术在北京冬奥会燃料电池示范中获得了应用,验证了其有效性。 展开更多

固体氧化物燃料电池(Solid Oxide Fuel Cell,SOFC)是一种能量转换装置,可以通过电化学反应将燃料中的化学能直接转换为电能。目前,SOFC大多被视为固定式发电技术,然而因其具有燃料灵活性、高效率和高能量密度等特点,在移动式发电领域如... 固体氧化物燃料电池(Solid Oxide Fuel Cell,SOFC)是一种能量转换装置,可以通过电化学反应将燃料中的化学能直接转换为电能。目前,SOFC大多被视为固定式发电技术,然而因其具有燃料灵活性、高效率和高能量密度等特点,在移动式发电领域如辅助动力、无人机动力、远程电源等也具有广阔应用前景。与固定式SOFC应用相比,移动式SOFC研究进展起步相对较晚,技术较不完善。移动式SOFC系统通常包括电堆、重整器、供气装置、尾燃器、换热器和储能组件等核心组件。开发SOFC移动系统需要考虑系统的鲁棒性、易用性以及发电能力等,而移动式SOFC系统部件相对较复杂,这些部件的耦合匹配特性又会显著影响系统的紧凑性、启动特性及体积/质量功率密度。从热特性、启动策略及高功率密度混合动力系统集成等方面对国内外移动式SOFC系统集成技术的研究进展进行综述。最后,基于对现有研究成果的总结提出了未来移动式SOFC系统集成中存在的挑战,及在材料与工艺的改进、新型启动策略和混合系统设计与优化等方面的发展方向。 展开更多
