The influence of temperature-dependent properties on thermal stresses response and optimum design of newly developed ceramic-metal functionally graded materials under cyclic thermal loaning and high temperature gradient environment is studied. The thermal conductivity of material is considered to be dependent on the temperature. In this paper, the thermal stresses response of the material is calculated rising a nonlinear finite element method. Emphasis is placed on the influence of temperature-dependent properties on the thermal stresses response characteristics, the thermal stresses relaxation property and the thermal stresses history under the different graded compositional distributions and different heat flux magnitudes. Through tile analysis. it is suggested that the influence of temperature-dependent properties can not be neglected In the thermal stresses response analysis and the optimum design process of the material must be based on the temperature-dependent thermo-elastic-plastic theory. 展开更多

White Hypsizygus marmoreus is a popular edible mushroom. Its mycelium is easy to be contaminated by Penicillium, which leads to a decrease in its quality and yield.Penicillium could compete for limited space and nutrients through rapid growth and produce a variety of harmful gases, such as benzene, aldehydes, phenols, etc.,to inhibit the growth of H.marmoreus mycelium.A series of changes occurred in H.marmoreus proteome after contamination when detected by the label-free tandem mass spectrometry(MS/MS)technique.Some proteins with up-regulated expression worked together to participate in some processes, such as the non-toxic transformation of harmful gases, glutathione metabolism, histone modification, nucleotide excision repair, clearing misfolded proteins, and synthesizing glutamine, which were mainly used in response to biological stress. The proteins with down-regulated expression are mainly related to the processes of ribosome function, protein processing, spliceosome, carbon metabolism, glycolysis, and gluconeogenesis. The reduction in the function of these proteins affected the production of the cell components, which might be an adjustment to adapt to growth retardation. This study further enhanced the understanding of the biological stress response and the growth restriction adaptation mechanisms in edible fungi. It also provided a theoretical basis for protein function exploration and edible mushroom food safety research. 展开更多

The molecular response of wheat(Triticum aestivum L.,cv.Yangmai 13) seedlings to heavy metal(Cd,Hg) and 1,2,4-trichlorobenzene(TCB) stresses were examined by two-dimensional gel electrophoresis, image analysis, and peptide mass fingerprinting. The results showed inhibitions of root and shoot growth by Cd,Hg,and TCB.These stresses led to water deficit and lipid phosphorylation in the seedling which also promoted protein phophorylation in the leaves.Hg stress inhibited protein synthesis while Cd and TCB stresses induced or up-regulated more proteins in the leaves. Most of these induced proteins played important roles in the biochemical reactions involved in tolerance of wheat to Cd and TCB stresses. The primary functions of Cd-and TCB-induced proteins included methionine metabolism,Rubisco modification, protein phosphorylation regulation, protein configuration protection,H+ transmembrane transportation and also the synthesis of ethylene, defense substances and cell wall compounds. 展开更多

However, species may exhibit stress response variation depending on their specific environmental and selection pressures. Blind cavefish dwell in cave environments, which differ markedly in stressors and resource availability compared to surface aquatic environments. However, whether blind cavefish exhibit differences in stress response as an adaptation to their cave environments remains unclear. Here, we investigated differences in stress response in six closely related Triplophysa species, including three blind cavefish (T. longibarbata, T. jiarongensis, and T. rosa) and three normal-sighted river fish (T. nasobarbatula, T. dongsaiensis, and T. bleekeri). Results showed that blind cavefish exhibited a range of distinct behavioral responses compared to sighted river fish, including greater levels of activity, shorter duration of freezing, absence of erratic movements or thrashing behavior, and opposite behavioral trends over time. Furthermore, the cavefish species demonstrated attenuated increases in metabolic rate in response to stressors related to novel environments. Cave-dwelling T. rosa also exhibited lower basal hypothalamic-pituitary-inter-renal (HPI) axis-related gene expression levels and stress hormone concentrations compared to river-dwelling T. bleekeri. These results suggest that blind cavefish may have lost their behavioral stress response, potentially mediated by a reduction in basal activity of the HPI axis, thus enabling the conservation of energy by reducing unnecessary expenditure in energy-limited caves. 展开更多

Plant male reproduction is a fine-tuned developmental process that is susceptible to stressful environments and influences crop grain yields.Phytohormone signaling functions in control of plant normal growth and development as well as in response to external stresses, but the interaction or crosstalk among phytohormone signaling, stress response, and male reproduction in plants remains poorly understood.Cross-species comparison among 514 stress-response transcriptomic libraries revealed that ms33-6038,a genic male sterile mutant deficient in the Zm Ms33/Zm GPAT6 gene, displayed an excessive drought stress-like transcriptional reprogramming in anthers triggered mainly by disturbed jasmonic acid(JA)homeostasis. An increased level of JA appeared in Zm Ms33-deficient anthers at both meiotic and postmeiotic stages and activated genes involved in JA biosynthesis and signaling as well as genes functioning in JA-mediated drought response.Excessive accumulation of JA elevated expression level of a gene encoding a WRKY transcription factor that activated the Zm Ms33 promoter. These findings reveal a feedback loop of Zm Ms33-JA-WRKY-Zm Ms33 in controlling male sterility and JA-mediated stress response in maize, shedding light on the crosstalk of stress response and male sterility mediated by phytohormone homeostasis and signaling. 展开更多

To investigate the stress response characteristics and shear stress transfer mechanism of BFRP(basalt fiber reinforced plastics)anchors under rainfall conditions and to explore the reinforcement effect of BFRP anchors,a comparative indoor physical model test was conducted in this study using loess mudstone slope as a typical case, and multi-attribute response data, such as slope displacement,BFRP anchor strain and axial force, were obtained.Based on the variation law of slope displacement,it can be concluded that the deformation displacement of the slope on the side reinforced by BFRP anchors is smaller than that of slopes reinforced with steel anchors;the bearing capacity of BFRP anchor is given priority in time, and the anchoring effect of BFRP anchor at the top of the slope is given priority, and the axial force value of anchor is characterized by a surface amplification effect in space;the axial force is higher around the anchor head and tends to decay as a negative exponential function as the anchor extends in the direction of the interior of the slope;the shear stress of BFRP anchor and anchorage body are unevenly distributed along the axial direction, with the maximum value occurring in the free section near the slope surface and decreases as the BFRP anchor extends towards the interior of the slope. These results can provide a theoretical basis for the optimal design of BFRP anchors. 展开更多

Although tocopherols are essential for rice development, the molecular details by which their absence affects development remain to be determined. To study how tocopherols function during rice development,we performed a transcriptome deep sequencing(RNA-seq)analysis of the rice cultivar Nipponbare(Nip)and the tocopherol-deficient mutant small grain and dwarf 1-2(sgd1-2).We identified 563 differentially expressed genes that were enriched in Gene Ontology categories associated with metabolism, stress, cellular responses, and transcriptional regulation. We determined that the total fatty acid composition of Nip and sgd1-2 was comparable, although cell membrane penetrability in sgd1-2 was significantly higher than in Nip under optimal growth conditions, indicating that tocopherol deficiency induces cell membrane damage. The expression levels of dehydration-responsive element binding 1(DREB1)genes and free proline content in sgd1-2 were also higher than those in Nip.We also showed that the DELLA protein SLENDER RICE1(SLR1)accumulated in sgd1-2,resulting in significant changes in the global transcriptome. Our study confirms that the lack of tocopherol accumulation in rice induced ectopic stress responses and limited growth by enhancing SLR1 abundance through increasing SLR1 transcript levels. These results provide new insights into tocopherol during rice development. 展开更多

Over the past century,age-related diseases, such as cancer,type-2 diabetes, obesity, and mental illness, have shown a significant increase, negatively impacting overall quality of life.Studies on aged animal models have unveiled a progressive discoordination at multiple regulatory levels, including transcriptional, translational, and post-translational processes, resulting from cellular stress and circadian derangements. The circadian clock emerges as a key regulator, sustaining physiological homeostasis and promoting healthy aging through timely molecular coordination of pivotal cellular processes, such as stem-cell function, cellular stress responses, and inter-tissue communication, which become disrupted during aging.Given the crucial role of hypothalamic circuits in regulating organismal physiology, metabolic control, sleep homeostasis, and circadian rhythms, and their dependence on these processes, strategies aimed at enhancing hypothalamic and circadian function, including pharmacological and non-pharmacological approaches, offer systemic benefits for healthy aging.Intranasal brain-directed drug administration represents a promising avenue for effectively targeting specific brain regions, like the hypothalamus, while reducing side effects associated with systemic drug delivery, thereby presenting new therapeutic possibilities for diverse age-related conditions. 展开更多

BACKGROUND Pulmonary fibrosis(PF)is a chronic interstitial lung disease characterized by fibroblast proliferation and extracellular matrix formation, causing structural damage and lung failure.Stem cell therapy and mesenchymal stem cells-extracellular vesicles(MSC-EVs)offer new hope for PF treatment.AIM To investigate the therapeutic potential of MSC-EVs in alleviating fibrosis, oxidative stress, and immune inflammation in A549 cells and bleomycin(BLM)-induced mouse model.METHODS The effect of MSC-EVs on A549 cells was assessed by fibrosis markers[collagen I andα-smooth muscle actin(α-SMA),oxidative stress regulators[nuclear factor E2-related factor 2(Nrf2)and heme oxygenase-1(HO-1),and inflammatory regu-lators[nuclear factor-kappaB(NF-κB)p65,interleukin(IL)-1β,and IL-2].Similarly,they were assessed in the lungs of mice where PF was induced by BLM after MSC-EV transfection.MSC-EVs ion PF mice were detected by pathological staining and western blot.Single-cell RNA sequencing was performed to investigate the effects of the MSC-EVs on gene expression profiles of macrophages after modeling in mice.RESULTS Transforming growth factor(TGF)-β1 enhanced fibrosis in A549 cells, significantly increasing collagen I andα-SMA levels.Notably, treatment with MSC-EVs demonstrated a remarkable alleviation of these effects.Similarly, the expression of oxidative stress regulators, such as Nrf2 and HO-1,along with inflammatory regulators, including NF-κB p65 and IL-1β,were mitigated by MSC-EV treatment.Furthermore,in a parallel manner,MSC-EVs exhibited a downregulatory impact on collagen deposition, oxidative stress injuries, and inflammatory-related cytokines in the lungs of mice with PF.Additionally, the mRNA sequencing results suggested that BLM may induce PF in mice by upregulating pulmonary collagen fiber deposition and triggering an immune inflammatory response. The findings collectively highlight the potential therapeutic efficacy of MSC-EVs in ameliorating fibrotic processes, oxidative stress, and inflammatory responses associated with PF.CONCLUSION MSC-EVs could ameliorate fibrosis in vitro and in vivo by downregulating collagen deposition, oxidative stress, and immune-inflammatory responses. 展开更多

We examined the possible multiple defects induced by acute and prolonged exposure to high levels of manganese(Mn) solution by monitoring the endpoints of lifespan, development, reproduction, and stress response. Our data suggest that acute exposure(6 h) to Mn did not cause severe defects of life span, development, and reproduction, similarly,no significant defect could be found in animals exposed to a low concentration of Mn(2.5 μmol/L) for 48 h.In contrast, prolonged exposure(48 h) to high Mn concentrations(75 and 200 μmol/L) resulted in significant defects of life span, development, and reproduction,as well as the increase of the percentage of population with hsp-16.2::gfp expression indicating the obvious induction of stress responses in exposed animals. Moreover, prolonged exposure(48 h) to high concentrations(75 and 200 μmol/L) of Mn decreased the expression levels of antioxidant genes of sod-1,sod-2,sod-3,and sod-4 compared to control. Therefore, prolonged exposure to high concentrations of Mn will induce the severe defects of life span, development, and reproduction in nematodes possibly by affecting the stress response and expression of antioxidant genes in Caenorhabditis elegans. 展开更多

A proteomic approach including two-dimensional electrophoresis and mass spectrometric (MALDI-TOF MS) analyses was used to investigate the responses to cadmium (Cd) stress in seedlings of rice (Oryza sativa L.) varieties Shanyou 63 and Aizaizhan. Cd stress significantly inhibited root and shoot growth, and affected the global proteome in rice roots and leaves, which induced or upregulated the expression of corresponding proteins in rice roots and leaves when rice seedlings were exposed to 0.1 or 1.0 mmol/L Cd. The Cd-induced proteins are involved in chelation and compartmentation of Cd, elimination of active oxygen free radicals, detoxification of toxic substances, degradation of denatured proteins or inactivated enzymes, regulation of physiologic metabolism and induction of pathogenesis-related proteins. Comparing the Cd-induced proteins between the two vadeties, the β-glucosidase and pathogenesis-related protein family 10 proteins were more drastically induced by Cd stress in roots and leaves of Aizaizhan, and the UDP-glucose protein transglucosylase and translational elongation factor Tu were induced by 0.1 mmol/L Cd stress in roots of Shanyou 63. This may be one of the important mechanisms for higher tolerance to Cd stress in Shanyou 63 than in Aizaizhan. 展开更多

BACKGROUND Thoracic surgery for radical resection of lung tumor requires deep anesthesia which can lead to an adverse inflammatory response, loss of hemodynamic stability, and decreased immune function.Herein,we evaluated the feasibility and benefits of ultrasound-guided paravertebral nerve block anesthesia,in combination with general anesthesia, for thoracic surgery for lung cancer. The block was performed by diffusion of anesthetic drugs along the paravertebral space to achieve unilateral multi-segment intercostal nerve and dorsal branch nerve block.AIM To evaluate the application of ultrasound-guided paravertebral nerve block anesthesia for lung cancer surgery to inform practice.METHODS The analysis was based on 140 patients who underwent thoracic surgery for lung cancer at our hospital between January 2018 and May 2020.Patients were randomly allocated to the peripheral+general anesthesia(observation)group(n=74)or to the general anesthesia(control)group(n=66).Patients in the observation group received ultrasound-guided paravertebral nerve block anesthesia combined with general anesthesia, with those in the control group receiving an epidural block combined with general anesthesia.Measured outcomes included the operative and anesthesia times,as well as the mean arterial pressure(MAP),heart rate(HR),and blood oxygen saturation(SpO;)measured before surgery,15 min after anesthesia(T1),after intubation,5 min after skin incision, and before extubation(T4).RESULTS The dose of intra-operative use of remifentanil and propofol and the postoperative use of sufentanil was lower in the observation group(1.48±0.43 mg,760.50±92.28 mg,and 72.50±16.62 mg,respectively)than control group(P<0.05).At the four time points of measurement(T1 through T4),MAP and HR values were higher in the observation than control group(MAP,90.20±9.15 mmHg,85.50±7.22 mmHg,88.59±8.15 mmHg,and 90.02±10.02 mmHg,respectively;and HR,72.39±8.22 beats/min,69.03±9.03 beats/min,70.12±8.11 beats/min,and 71.24±9.01 beats/min,respectively;P<0.05).There was no difference in SpO;between the two groups(P>0.05).Postoperative levels of epinephrine, norepinephrine, and dopamine used were significantly lower in the observation than control group(210.20±40.41 pg/mL,230.30±65.58 pg/mL,and 54.49±13.32 pg/mL,respectively;P<0.05).Similarly,the postoperative tumor necrosis factor-αand interleukin-6 levels were lower in the observation(2.43±0.44 pg/mL and 170.03±35.54 pg/mL,respectively)than control group(P<0.05).There was no significant difference in the incidence of adverse reactions between the two groups(P>0.05).CONCLUSION Ultrasound-guided paravertebral nerve block anesthesia improved the stress and hemodynamic response in patients undergoing thoracic surgery for lung cancer, with no increase in the rate of adverse events. 展开更多

Objective To investigate how acetamiprid, a new insecticide, affects the activity of superoxide dismutase (SOD), catalase (CAT), and ATPase and the SOD isozyme patterns in two G-bacteria, E. coli K12 anti Pse.FH2, and one G^+ bactemm, B. subtilis. Methods The SOD, CAT, and ATPase specific activities of cell lysates were determined spectrophotometrically at 550 nm, 240 nm, and 660 nm, respectively, with kits A001, A016, and A007. SOD isozyme patterns were detected by native PAGE analysis. Results SOD and CAT activities in the tested bacteria increased significantly in a concentration-dependent manner after different concentrations of acetamiprid were applied. The activity of SOD in B. subtilis and Pse.FH2 was stimulated and reached the highest level after treatment with 100 mg/L acetamiprid for 0.5 h. For Pse.FH2, there was another stimulation of SOD activity after acetamiprid application for about 8.0 h and the second stimulation was stronger than the first. The stimulation by acetamiprid showed a relative lag for E. coli K12. Acetamiprid seemed to exhibit a similar effect on CAT activity of the two G bacteria and had an evident influence on ATPase activity in the three bacteria within a relatively short period. Only one SOD isozyme was detectable in Pse.FH2 and B. subtilis, while different isozyme compositions in E. coli could be detected by native PAGE analysis. Conclusion Acetamiprid causes a certain oxidative stress on the three bacteria which may not only elevate SOD and CAT activities but also generate new SOD isozymes to antagonize oxidative stress. However, this oxidative stress lasts for a relatively short time and does not cause a long-term damage. 展开更多

The homeodomain-leucine zipper(HD-Zip)family has been shown to perform amultitude of functions during plant development and stress responses;however, the familymembers and functions have not been identified in apple(Malus×domestica). In this study,83 HD-Zips(MdHDZs)were identified in the apple genome. They were assembled into four subgroups according to the classification in Arabidopsis, where MdHDZs in the same subgroup had similar gene structures and conserved protein motifs.Putative cis-element analysis of MdHDZs promoter regions uncovered numerous elements related to the response of stress and plant hormones. In addition, twelve transcripts of the MdHDZs showed different expression patterns under salt, drought, low temperature and ABA stresses by quantitative reverse transcription-PCR(qRT-PCR)assay.To further explore the function of MdHDZs in apple,MdHDZ3 was selected to verify its function under salt, low temperature and ABA stresses;and genetic transformation was used to obtain MdHDZ3 transgenic apple calli. The results demonstrated that MdHDZ3 increased sensitivity to salt, low temperature and abscisic acid in apple calli, suggesting that MdHDZ3 plays an important role in response to stresses.Subcellular localization and three-dimensional structural analysis revealed that MdHDZ3 was a nuclear-localized protein.Taken together, these findings provide potential information for further identification of HD-Zip proteins in apple. 展开更多
