Among the various biological and physical analyses per- formed for full study on this biopharmaceutic, the determination of the concentration preparations throughout manufacturing and subsequent handling in hospital is particularly relevant. In the present work, the study and validation of a method for quantifying intact CTX by reverse-phase high-perfor- mance liquid chromatography with diode array detection ((RP)HPLC/DAD) is presented. With that end, we checked the performance of a chromatographic method for quantifying CTX and conducted a study to validate the method as stability-indicating in accordance with the International Conference on Harmo- nization guidelines (ICH) for biotechnological drugs; therefore, we evaluated linearity, accuracy, preci- sion, detection and quantification limits, robustness and system suitability. The specificity of the method and the robustness of the mAb formulation against external stress factors were estimated by compre- hensive chromatographic analysis by subjecting CTX to several informative stress conditions. As de- monstrated, the method is rapid, accurate, and reproducible for CTX quantification. It was also suc- cessfully used to quantify CTX in a long-term stability study performed under hospital conditions.展开更多

Thirty bariatric patients were interviewed before bariatric surgery and at a 12-month follow-up, and fulfilled self-report measures about health problems, health complaints and coping strategies before surgery, at 6-and 12-month follow-ups. Before surgery, failure cases differ from success on the conceptualization of obesity, However, there are no other differences between groups. At 6-and 12-month follow-ups, failure cases had the highest BMI, health problems and complaints and less % EWL than success cases. One year after the surgery, one in each three persons did not lose the expected weight, i.e., are failure cases. Before surgery, there are no differences between success and failure cases in the report of health problems, health complaints and coping strategies, but they have different conceptualizations of their obesity and treatment. One year after the surgery, success cases understood bariatric surgery as an important moment in their lives related to their expected results, whereas failures valued unexpected dimensions and still waiting for a miracle surgery without their personal commitment. Accordingly, it is necessary to consider lifestyle changes in the obesity treatment process.展开更多

The reverse dry reforming reaction (RDR, 2H2 + 2CO←→CH4 + CO2), combining CO methanation with water-gas-shift reaction, can significantly decrease the H2/CO molar ratio to 1 for SNG production. A detailed thermodynamic analysis of RDR reaction was carried out based on the Gibbs free energy minimization method. The effect of temperature, pressure. H2/CO ratio and the addition of H2O, CH4, CO2, O2 and C2H4 into the feed gas on CO conversion, CH4 and CO2 selectivity, as well as CH4 and carbon yield, are discussed. Experimental results obtained on homemade impregnated Ni/Al2O3 catalyst are compared with the calculations. The results demonstrate that low temperature (200-500 °C), high pressure (1-5 MPa) and high H2/CO ratio (at least 1) promote CO conversion and CH4 selectivity and decrease carbon yield. Steam and CO2 in the feed gas decrease the CH4 selectivity and carb on yield, and enhance the CO2 con tent. Extra CH4 elevates the CH4 content in the products, but leads to more carbon formation at high temperatures. O2 significantly decreases the CH4 selectivity and C2H4 results in the generation of carbon.展开更多

A reverse design and simulation software was compiled based on Visual Basic 6.0 to obtain the mechanism parameters, including the pitch curves and tooth profiles of the non-circular gears. A test-bed of this mechanism was developed according to these parameters. The kinematic performance of this weft insertion mechanism was tested by high-speed video tape recorder with its corresponding video analytical software Blaster's MAS. And the test results were identical with the theoretical calculation. Compared with the conjugate cam weft insertion mechanism which is applied on TT96 rapier loom, under the condition of same weft insertion rate, the variation of acceleration of this mechanism is smaller. This novel weft insertion mechanism can meet the requirements of weft insertion on rapier looms with smooth and steady motion.展开更多

In this study, we aimed to investigate the changes oftitin and a-smooth muscle actin (α-SMA) after long time of AF reversal. Methods Twenty-four goats were randomized into four groups: (1) sinus rhythm (SR), (2) 3 months AF (3-too AF), (3) 3 months SR after 3 months AF (3-mo post AF), (4) 6 months SR after 3-mo AF (6-mo post AF), with 6 in each group. By pacing on the anterior bottom of left atria appendage (LAA), we established a goat model of chronic AF. Atria effective refractory period (AERP) was measured with electrophysiological methods. Ultra-structure was studied with echocardiography, light and electron microscopy. Titin and α-SMA protein expressions were determined by Western blot. Results The animals underwent high rate pacing on LAA for a mean of 42.23± 21.70 days before presenting AF. Electrophysiological analysis revealed that AERP completely resumed in 3-mo post AF goats. Echocardiography displayed that the size of left atrium resumed almost in 6-too post AF goats (P〈 0.01). Pathological and electron microscopic examination revealed the disorder of myofibrils, augmentation of intercellular space, myolysis, accumulation of glycogen, and numerous bigger mitochondria among atrial cardiomyocytes in 3-mo AF goats. They recovered mostly in 6-mo post AF goats. Western blot showed that the band density oftitin significantly reduced in 3-mo AF goats compared to SR ones [1826 ± 319 vs 5012±854, P 〈 0.01]. In 3- and 6-mo post AF goats, titin increased gradually and it reversed completely in 6-mo post AF goats (3841 ± 601 and 4523 ±833 respectively, P 〈 0.01). Conversely, the band density ofa-SMAwas significantly higher in 3-mo AF goats (5324 ± 948) than in SR ones (1619 ±271, P 〈 0.01). In 3- and 6-mo post-AF goats, α-SMA decreased gradually, and it recovered mostly in 6- mo post AF goats (4437 ± 792 and 2205 ± 540 respectively, P〈 0.01,). Conclusions These data indicate that the reversal of dedifferentiation of atrial cardiomyocyts is a very slow process, and it is definitely essential for normal cardiac function .展开更多
