An efficient method for constructing a random insertional mutant library for forward genetics in Nannochloropsis oceanica

Insertional mutation,phenotypic evaluation,and mutated gene cloning are widely used to clone genes from scratch.Exogenous genes can be integrated into the genome during non-homologous end joining(NHEJ)of the double-strand breaks of DNA,causing insertional mutation.The random insertional mutant library constructed using this method has become a method of forward genetics for gene cloning.However,the establishment of a random insertional mutant library requires a high transformation efficiency of exogenous genes.Many microalgal species show a low transformation efficiency,making constructing random insertional mutant libraries difficult.In this study,we established a highly efficient transformation method for constructing a random insertional mutant library of Nannochloropsis oceanica,and tentatively tried to isolate its genes to prove the feasibility of the method.A gene that may control the growth rate and cell size was identified.This method will facilitate the genetic studies of N.oceanica,which should also be a reference for other microalgal species.

The Book Publishing and Analysis of Modern Peking University Library

The paper is based on literature search,introducing and analyzing the book publishing situation of modern Peking University Library from the perspective of the publisher being Peking University Library and its predecessor.It is pointed out that the majority of the book printing methods published by the modern Peking University Library are lead printing,and among the types of Chinese library classification,catalogue of library collections is the most common.

Symbols in The Magic Barrel

The article takes the famous modern American Jewish writer Bernard Malamud’s novel The Magic Barrel as the object of study,and uses symbolism to interpret it,analyzing it one by one in terms of the magic barrel,the colors,the seasons and the windows,respectively,so as to reveal the process of getting rid of the childishness within the novel’s male protagonist,Leo Finkel,who is maturing,as well as the novel’s Jewish theme of searching for the soul of the self.

Analysis of the Books Issued by Yadong Library, Published by Peking University Publishing Department: Taking Several Bibliography Advertisements Carried in Newspapers and Magazines From 1919-1922 as Examples

Based on several bibliographic advertisements carried in newspapers and magazines from 1919 to 1922 for books published by Peking University Publishing Department,and issued by Yadong Library,the paper analyzes the characteristics of the books published by Peking University Publishing Department during this period,and points out that Yadong Library entered its golden period of development due to move from the alley to the main road and obtain the management rights of the books published by Peking University Publishing Department.

高校图书馆员数据素养提升策略——来自Library carpentry课程项目的经验

通过网络调查和文献调研分析了图书馆员的数据素养提升的Library carpentry课程项目,从发起背景、课程团队、经费支持、课程目标、课程内容等方面调查了Library carpentry的管理和运行实践,从项目组织、课程和教学方面总结出Library carpentry项目的特点。根据Library carpentry的成功经验,从开拓图书馆服务视野、打造实用化课程体系、推动教育可持续发展,搭建协同教育平台等角度思考对我国图书馆员数字素养教育的启示。

Linker Library:基于几何特征的骨架跃迁片段库

骨架跃迁是目前应用最广泛的药物设计策略之一,但是现有骨架跃迁方法产生的化合物大多为已报道的先导化合物的衍生物或类似物,化学结构缺乏新颖性。针对现有骨架跃迁方法的局限性,提出了一种保持药效团与骨架之间的相对距离和角度不变的骨架跃迁策略,构建一个包含骨架几何特征的linker片段库:Linker Library。该片段库可以根据骨架中心到连接点之间距离和角度,推荐结构新颖且可保持官能团相对位置的片段,有助于指导化合物的骨架跃迁并加速药物发现进程。

一类新的MDS symbol-pair码

为了对抗符号对(symbol-pair)读取信道中的符号对错误,符号对码应运而生。与经典纠错码类似,极小对距离越大,符号对码的纠错能力越强,因此,构造具有较大极小符号对距离的符号对码至关重要。根据多项式重根的判别方法,通过分析有限域F p上线性方程组的解,得到了一类最大距离可分的符号对(MDS symbol-pair)码,其长度为4 p,极小符号对距离为10。结果表明,在同样码长的情况下,新构造的MDS符号对码具有较大的极小符号对距离。

Small Amplicons Mutation Library for Vaccine Screening by Error-Prone Polymerase Chain Reaction

Library construction is a common method used to screen target genes in molecular biology.Most library constructions are not suitable for a small DNA library(<100 base pair(bp))and low RNA library output.To maximize the library’s complexity,error-prone polymerase chain reaction(PCR)was used to increase the base mutation rate.After introducing the DNA fragments into the competent cell,the library complexity could reach 109.Library mutation rate increased exponentially with the dilution and amplification of error-prone PCR.The error-prone PCR conditions were optimized including deoxyribonucleotide triphosphate(dNTP)concentration,Mn^(2+)concentration,Mg^(2+)concentration,PCR cycle number,and primer length.Then,a RNA library with high complexity can be obtained by in vitro transcription to meet most molecular biological screening requirements,and can also be used for mRNA vaccine screening.

A robust symbol timing strategy for cellular systems

In cellular systems,establishing the initial symbol timing of potential preambles is the first step of a cell search.The envelope fluctuation of the downlink signal hinders the successful timing of conventional symbol timing methods.To solve this problem,a hybrid timing strategy is proposed with two novel detectors,namely the normalized replica-based detector and normalized differential detector.The strategy first detects all potential preambles via the normalized replica-based detector and then employs the normalized differential detector to verify the target preamble,which comes from the target cell and has the highest power.The strategy is unaffected by envelope fluctuation and has computational complexity comparable to that of conventional methods.Simu-lations and real-data tests show that the hybrid timing strategy is robust and practical for initial symbol timing.

Dock-able linear and homodetic di, tri, tetra and pentapeptide library from canonical amino acids: SARS-CoV-2 Mpro as a case study

Peptide-based therapeutics are increasingly pushing to the forefront of biomedicine with their promise of high specificity and low toxicity.Although noncanonical residues can always be used,employing only the natural 20 residues restricts the chemical space to a finite dimension allowing for comprehensive in silico screening.Towards this goal,the dataset comprising all possible di-,tri-,and tetra-peptide combinations of the canonical residues has been previously reported.However,with increasing computational power,the comprehensive set of pentapeptides is now also feasible for screening as the comprehensive set of cyclic peptides comprising four or five residues.Here,we provide both the complete and prefiltered libraries of all di-,tri-,tetra-,and penta-peptide sequences from 20 canonical amino acids and their homodetic(N-to-C-terminal)cyclic homologues.The FASTA,simplified molecular-input line-entry system(SMILES),and structure-data file(SDF)-three dimension(3D)libraries can be readily used for screening against protein targets.We also provide a simple method and tool for conducting identity-based filtering.Access to this dataset will accelerate small peptide screening workflows and encourage their use in drug discovery campaigns.As a case study,the developed library was screened against severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)main protease to identify potential small peptide inhibitors.

Identification and epitope mapping of anti-p72 single-chain antibody against African swine fever virus based on phage display antibody library

African swine fever virus(ASFV)is a lethal pathogen that causes severe threats to the global swine industry and it has already had catastrophic socio-economic effects.To date,no licensed prophylactic vaccine exists.Limited knowledge exists about the major immunogens of ASFV and the epitope mapping of the key antigens.As such,there is a considerable requirement to understand the functional monoclonal antibodies(mAbs)and the epitope mapping may be of utmost importance in our understanding of immune responses and designing improved vaccines,therapeutics,and diagnostics.In this study,we generated an ASFV antibody phage-display library from ASFV convalescent swine PBMCs,further screened a specific ASFV major capsid protein(p72)single-chain antibody and fused with an IgG Fc fragment(scFv-83-Fc),which is a specific recognition antibody against ASFV Pig/HLJ/2018 strain.Using the scFv-83-Fc mAb,we selected a conserved epitope peptide(221MTGYKH226)of p72 retrieved from a phage-displayed random peptide library.Moreover,flow cytometry and cell uptake experiments demonstrated that the epitope peptide can significantly promote BMDCs maturation in vitro and could be effectively uptaken by DCs,which indicated its potential application in vaccine and diagnostic reagent development.Overall,this study provided a valuable platform for identifying targets for ASFV vaccine development,as well as to facilitate the optimization design of subunit vaccine and diagnostic reagents.

Training-based symbol detection with temporal convolutional neural network in single-polarized optical communication system

In order to reduce the physical impairment caused by signal distortion,in this paper,we investigate symbol detection with Deep Learning(DL)methods to improve bit-error performance in the optical communication system.Many DL-based methods have been applied to such systems to improve bit-error performance.Referring to the speech-to-text method of automatic speech recognition,this paper proposes a signal-to-symbol method based on DL and designs a receiver for symbol detection on single-polarized optical communications modes.To realize this detection method,we propose a non-causal temporal convolutional network-assisted receiver to detect symbols directly from the baseband signal,which specifically integrates most modules of the receiver.Meanwhile,we adopt three training approaches for different signal-to-noise ratios.We also apply a parametric rectified linear unit to enhance the noise robustness of the proposed network.According to the simulation experiments,the biterror-rate performance of the proposed method is close to or even superior to that of the conventional receiver and better than the recurrent neural network-based receiver.

Research on the Value Positioning of University Library Based on Customer Delivered Value

Construction of Smart Library Based on the Agricultural Information from Metaverse Horizon

Metaverse is a virtual world,but it is possible and even becomes a trend that metaverse interacts with the real world along with the sci-tech and social development.Smart library is a main direction of modern library development,from informatization to digitization and then intelligent-based,modernization of library has been improved qualitatively.Emergence of metaverse brings a brand-new horizon to the generation and application of agricultural information,this paper made primary explorations the requirements and action mechanism of agricultural information on the construction of smart library within the external constraint of metaverse.

Application of Blank Symbols in Japanese Architecture and Indoor Designs

A symbol is an expression of meaning,while blank symbols express special meanings.By focusing on the application of blank symbols in Japanese architecture and indoor designs,we analyzed the aesthetic principles in Japanese architecture and indoor designs from the perspective of semiotics,such as“Kongji,”“Emptiness,”and“Dying out,”and their minimalist and pure design concepts.Traditional Chinese culture was also further explored,especially the profound influence of the“Chan sect”and the philosophy of“unity of heaven and mankind”on Japanese architecture and designs.This study aims to facilitate the coexistence and mutual appreciation of Chinese and Japanese architectural designs.

Crack Growth Rate Model Derived from Domain Knowledge-Guided Symbolic Regression

Machine learning(ML)has powerful nonlinear processing and multivariate learning capabilities,so it has been widely utilised in the fatigue field.However,most ML methods are inexplicable black-box models that are difficult to apply in engineering practice.Symbolic regression(SR)is an interpretable machine learning method for determining the optimal fitting equation for datasets.In this study,domain knowledge-guided SR was used to determine a new fatigue crack growth(FCG)rate model.Three terms of the variable subtree ofΔK,R-ratio,andΔK_(th)were obtained by analysing eight traditional semi-empirical FCG rate models.Based on the FCG rate test data from other literature,the SR model was constructed using Al-7055-T7511.It was subsequently extended to other alloys(Ti-10V-2Fe-3Al,Ti-6Al-4V,Cr-Mo-V,LC9cs,Al-6013-T651,and Al-2324-T3)using multiple linear regression.Compared with the three semi-empirical FCG rate models,the SR model yielded higher prediction accuracy.This result demonstrates the potential of domain knowledge-guided SR for building the FCG rate model.

Behavioral Intention to Continue Using a Library Mobile App

To meet the needs of today’s library users,institutions are developing library mobile apps(LMAs),as their libraries are increasingly intelligent and rely on deep learning.This paper explores the influencing factors and differences in the perception of LMAs at different time points after a user has downloaded an LMA.A research model was constructed based on the technology acceptance model.A questionnaire was designed and distributed twice to LMA users with an interval of three months to collect dynamic data.The analysis was based on structural equation modeling.The empirical results show that the perceived ease of use,the perceived usefulness,the social influence,and the facilitating conditions affected the users’behavioral intention,but their impacts were different at different times.As the usage time increases,the technology acceptance model is still universal for understanding the user perception of LMA.In addition,two extended variables(social impact and convenience)also affect the user’s behavior intention.User behavior is dynamic and changed over time.This study is important both theoretically and practically,as the results could be used to improve the service quality of LMAs and reduce the loss rate of users.Its findings may help the designers and developers of LMAs to optimize them from the perspective of a user and improve the service experience by providing a deeper understanding of the adoption behavior of information systems by LMA users.

Analysis of the Influence of Clothing on Characterization from Symbolism: Taking The Scarlet Letter, Daisy Miller, and Dry September as Examples

In the novels, the clothing worn by the characters are symbols made and used by the authors to express the characters’ personality and deepen the theme, and grasping their symbolism helps to enter into the interior of the novels and understand their essence. This paper will analyze and interpret the clothing of typical female characters in The Scarlet Letter, Daisy Miller and Dry September from the perspective of symbolism, so as to illustrate how the symbolism of clothing, as a kind of symbols, shapes the image of the main characters.
