The structure was divided into two substructures whereby a physical model of the bottom three stories was tested in the laboratory and the upper 37 stories were simulated numerically using ABAQUS. An overlapping domain method was employed for the bottom three stories to ensure the validity of the boundary conditions of the superstructure. Mixed control was adopted in the test. Displacement control was used to apply the horizontal displacement, while two controlled force actuators were applied to simulate the overturning moment, which is very large and cannot be ignored in the substructure hybrid test of high-rise buildings. A series of tests with earthquake sources of sequentially increasing intensities were carried out. The test results indicate that the proposed hybrid test method is a solution to reproduce the seismic response of high-rise concrete shear wall buildings. The seismic performance of the tested precast high-rise building satisfies the requirements of the Chinese seismic design code.展开更多

The air pressur...To investigate the natural frequencies and towing behaviors of a 3-bucket foundation platform at different drafts, the decay and towing experiments were carried out in a towing tank on a scale of 1:20. The air pressure inside the bucket foundations, the water pressure at the bottom of the bucket foundations, the acceleration of the platform and the towing force were determined in the test process. The time-history curves of the measured parameters were obtained, and the frequency responses of the parameters at different drafts were analyzed by means of fast Fourier transform(FFT). The results showed that the platform natural frequency of heave decreased slightly with the rise of draft. The natural frequencies of roll and pitch are much lower than that of heave, and they increased slightly with the increase of draft. When towing in the following sea, the maximum acceleration of surge, sway and heave has downward trends with the increase of draft, but the change range decreased gradually with the increase of draft. When the draft is 5.0 m(the ratio of draft to bucket height is 0.56), the towing dynamic responses achieve the maximum, which is not conducive to the towing of the platform. When the draft is 6.0 m(the ratio of draft to bucket height is 0.67), the towing dynamic responses are the most stable.展开更多

Usually, ki...Kissing bonds are defects in the adhesive bonds with intimate contact of touching surface but considerably lowered shear strength. Their detection specifically in the aerospace area is so not satisfactory. Usually, kissing bonds are inconspicuous in ultrasonic C-scans. However, the determination of attributes in the time domain and the frequency domain of an ultrasound signal provides the opportunity to derive a pattern for bonded area. Deviations from the pattern found in inconspicuous bonding areas indicate kissing bonds. The survey described here deals with the manufacturing of adhesively joint samples that purposefully include kissing bonds, as well as potential solutions for detecting them through ultrasonic testing combined with pattern recognition. The properties of the epoxy-based adhesive were varied by changing the mixing ratios between resin and hardener. Samples with a mixing ratio far apart from the manufacturer’s recommendation with an inconspicuous appearance in a C-scan, but low shear strength values were taken for further evaluation. After a definition and learning phase, a 100 percent hit rate to separate good bondings from kissing bonds could be derived in a blind test. The discriminating feature found is due to the frequency shift between good and kissing bonds as well as the relative amplitude of the second peak.展开更多
This study describes the development of novel protocols extending the real-time ultrasonic reflectometry(UTDR) for the detection of membrane fouling in hollow fiber module during ultrafiltration(UF) of oily water trea...This study describes the development of novel protocols extending the real-time ultrasonic reflectometry(UTDR) for the detection of membrane fouling in hollow fiber module during ultrafiltration(UF) of oily water treatment. A specially designed acoustic sensor with a frequency of 2.5 MHz was used. The hollow fiber membranes used were polysulphone(PSf) UF membranes with MWCO 40 kDa. The wastewaters with three different oily concentrations of 100, 500 and 1 000 mg/L were investigated. Diesel oil was utilized as the primary foulant. The results show that the permeate flux declines with operation time and its value becomes lower with the increase of the oily concentration in wastewater. It is found that ultrasonic measurement can detect the fouling and cleaning processes. A new signal analysis protocol-ultrasonic reflected energy was developed. Ultrasonic reflected energy obtained indicates the deposition of oily layer as a function of operation time and its removal after cleaning. The overall flux decline is reasonably correlated with the changes in ultrasonic reflected energy. This research provides the evidence that the ultrasonic reflectometry technique is capable of monitoring membrane fouling and cleaning in hollow fiber modules.展开更多

Having been separately transferred into the hosts E. coli BL21 and E. coli JM109, these two strains can express fusion proteins: His-tagged PDZ(PDZ domain) and maltose binding protein-BAR( MBP-BAR domain) respectively, as confirmed by both SDS-PAGE and Wostem blotting. The interaction between these two domains is dose-dependence, as identified by a pull-down test. Moreover, it has been shown from the ELISA analysis that the actual amount of PDZ bound to MBP-BAR-amylose beads reaches ( 16 ± 0. 5)%, as calculated by the molar ratio of PDZ to MBP-BAR. In addition, the interaction between BAR(bait) and PDZ(prey) in vivo was also examined with a yeast two-hybrid system.展开更多
