Parkinson’s disease is typically characterized by the progressive loss of dopaminergic neurons in the substantia nigra pars compacta.Many studies have been performed based on the supplementation of lost dopaminergic ...Parkinson’s disease is typically characterized by the progressive loss of dopaminergic neurons in the substantia nigra pars compacta.Many studies have been performed based on the supplementation of lost dopaminergic neurons to treat Parkinson’s disease.The initial strategy for cell replacement therapy used human fetal ventral midbrain and human embryonic stem cells to treat Parkinson’s disease,which could substantially alleviate the symptoms of Parkinson’s disease in clinical practice.However,ethical issues and tumor formation were limitations of its clinical application.Induced pluripotent stem cells can be acquired without sacrificing human embryos,which eliminates the huge ethical barriers of human stem cell therapy.Another widely considered neuronal regeneration strategy is to directly reprogram fibroblasts and astrocytes into neurons,without the need for intermediate proliferation states,thus avoiding issues of immune rejection and tumor formation.Both induced pluripotent stem cells and direct reprogramming of lineage cells have shown promising results in the treatment of Parkinson’s disease.However,there are also ethical concerns and the risk of tumor formation that need to be addressed.This review highlights the current application status of cell reprogramming in the treatment of Parkinson’s disease,focusing on the use of induced pluripotent stem cells in cell replacement therapy,including preclinical animal models and progress in clinical research.The review also discusses the advancements in direct reprogramming of lineage cells in the treatment of Parkinson’s disease,as well as the controversy surrounding in vivo reprogramming.These findings suggest that cell reprogramming may hold great promise as a potential strategy for treating Parkinson’s disease.展开更多

The bacterium was strictly anaerobic; it does not endure acid and its maximum surviving temperature was 70℃. Many short chain organic compounds can be utilized as electron donors, which were acetate, formate, lactate, propionate, pyruvate, butyrate, succinate, malate, fumarate, valerate, caproate, heptanoate, octanoate, decanoate, tridecanoate, pentadecanoate, palmitate, heptadecanoate or ethanol, while sulfate and sulfite were used as electron acceptors. The following substrates were not utilized: benzoate undecanoate, dodecanoate, tetradecane, propanol, butanol, H2+CO2 (80/20%; v/v) and acetate (1mM) + H2. When lactate was used as electron donors, sulfite and thiosulfate, but not sulfer and nitrate, can be used as electron acceptors. Strain CW-03 was motile, curved rod, Gram-positive, pole flagellum and spore-forming. On the basis of 16S rRNA sequence alignment (accession numbers: AY703032), CW-03 should be included in the genus Desulfotomaculum with BlAST analysis on line. However, some of its physiology and multiple sequence alignments were different from other members of this genus. Therefore, CW-03 should be recognized as a new species, for which we propose the name Desulfotomaculum chinamiddle (Bacteria, Firmicutes, Clostridia, Clostridiales, Peptococcaceae).展开更多

It allows collapsing the tree branches at different taxo...We describe an interactive viewer for the All-Species Living Tree(LVTree). The viewer incorporates treeing and lineage information from the ARB-SILVA website. It allows collapsing the tree branches at different taxonomic ranks and expanding the collapsed branches as well, keeping the overall topology of the tree unchanged. It also enables the user to observe the consequence of trial lineage modifications by re-collapsing the tree. The system reports taxon statistics at all ranks automatically after each collapsing and re-collapsing. These features greatly facilitate the comparison of the 16 S rRNA sequence phylogeny with prokaryotic taxonomy in a taxon by taxon manner.In view of the fact that the present prokaryotic systematics is largely based on 16 S rRNA sequence analysis, the current viewer may help reveal discrepancies between phylogeny and taxonomy. As an application, we show that in the latest release of LVTree, based on 11,939 rRNA sequences, as few as 24 lineage modifications are enough to bring all but two phyla(Proteobacteria and Firmicutes) to monophyletic clusters.展开更多
The Patriarch’s Hall Anthology(Zutang Ji祖堂集),is a work written in the style of“Lamp Records(灯录)”and maintains a deep connection with Xuefeng’s lineage(雪峰系)of Shitou Sect(石头宗).It therefore has obvious se...The Patriarch’s Hall Anthology(Zutang Ji祖堂集),is a work written in the style of“Lamp Records(灯录)”and maintains a deep connection with Xuefeng’s lineage(雪峰系)of Shitou Sect(石头宗).It therefore has obvious sectarian tendencies.In terms of the Dharma inheritance,the Patriarch’s Hall Anthology is dedicated to constructing an exclusive lineage of the Dharma transmission and establishing Shitou Sect as the direct line of transmission of the Sixth Patriarch Huineng.In terms of the image of the founder,the Patriarch’s Hall Anthology makes use of the literary methods of fictionalization and dichotomy to portray Shitou Xiqian(石头希迁)as the legitimate successor to the“treasury of the true Dharma-eye(正法眼藏).”In terms of Chan ideology,the Patriarch’s Hall Anthology,through criticizing Mazu’s Chan teaching that“the mind is precisely the Buddha(即心即佛),”established Shitou Sect as the true inheritor of the spirit of the Tathāgatagarbha(Rulaizang如来藏).It is through these three aspects that the Patriarch’s Hall Anthology consciously and systematically highlights the orthodox position of Shitou Sect in the Southern School of Chan.展开更多
