AIM: To identify the disease-associated mutations in a Chinese Stargardt disease(STGD) family, extend the existing spectrum of disease-causing mutations and further define the genotype-phenotype correlations.METHODS: ...AIM: To identify the disease-associated mutations in a Chinese Stargardt disease(STGD) family, extend the existing spectrum of disease-causing mutations and further define the genotype-phenotype correlations.METHODS: A Chinese STGD family and 200 normal controls were collected. Whole exome sequencing(WES) and bioinformatics analysis were performed to find the pathogenic gene mutation. Physico-chemical parameters of mutant and wildtype proteins were computed by Prot Param tool. Domains analysis was performed by SMART online software. HOPE online software was used to analyze the structural effects of mutation. Immunofluorescence, quantitative real-time polymerase chain reaction and Western blotting were used for expression analysis.RESULTS: Using WES, a novel homozygous mutation(NM_000350: c.G3190 C, p.G1064 R) in ABCA4 gene was identified. This mutation showed co-segregation with phenotype in this family. It was not found in the 200 unrelated health controls and absent from any databases. It was considered "Deleterious" as predicted by five function prediction softwares, and was highly conserved during evolution. ABCA4 was expressed highly in the human eye and mouse retina. The p.G1064 R was located in AAA domain, may force the local backbone into an incorrect conformation, disturb the local structure, and reduce the activity of ATPase resulting in the disease pathology. CONCLUSION: We define a novel pathogenic mutation(c.G3190 C of ABCA4) of STGD. This extends the existing spectrum of disease-causing mutations and further defines the genotype-phenotype correlations.展开更多

The state of charge (SOC) and state of health (SOH) are two of the most important parameters of Li-ion batteries in industrial production and in practical applications. The real-time estimation for these two param...The state of charge (SOC) and state of health (SOH) are two of the most important parameters of Li-ion batteries in industrial production and in practical applications. The real-time estimation for these two parameters is crucial to realize a safe and reliable battery application. However, this is a great problem for LiFePO4 batteries due to the large constant potential plateau in the charge/discharge process. Here we propose a combined SOC and SOH co-estimation method based on the experimental test under the simulating electric vehicle working condition. A first-order resistance-capacitance equivalent circuit is used to model the battery cell, and three parameter values, ohmic resistance (Rs), parallel resistance (Rp) and parallel capacity (Cp), are identified from a real-time experimental test. Finally we find that Rp and Cp could be utilized to make a judgement on the SOIl. More importantly, the linear relationship between Cp and the SOC is established to make the estimation of the SOC for the first time.展开更多

Type IV secretion system (T4SS) is one of the secretion systems and it usually consists of 12 genes: VirB1, VirB2 ...VirB11, and VirD4[2]. The structure and molecular mechanisms of these genes have been well analyzed in Gram-negative strains[3] and Gram-positive strains were once believed to be lack of T4SS. However, some recent studies revealed that one or more virB/D genes also exist in some kinds of Gram-positive bacteria and play similar role, and form a T4SS-like system[3]. The VirBl-like, VirB4, VirB6, and VirD4 genes were identified in the chromosome of Gram-positive bacterium Streptococcus suis in our previous studies and their role as important mobile elements for horizontal transfer to recipients in an 89 K pathogenicity island (PAl) was demonstrated[45]. Barley Fusarium head blight(FHB),caused by species of the Fusarium fungus,is a devastating disease that is reemerging worldwide in recent years.In this study,a novel gene,HvORG4,was cloned from barley by using cDNA li...Barley Fusarium head blight(FHB),caused by species of the Fusarium fungus,is a devastating disease that is reemerging worldwide in recent years.In this study,a novel gene,HvORG4,was cloned from barley by using cDNA library and suppression subtractive hybridization(SSH) library strategies.The SSH library and cDNA library were constructed from the Chinese barley cultivar Jing02-461(resistance to FHB) infected by Fusarium graminearum isolate Huanggang-1.For the SSH analysis,more than 120 differentially expressed cDNAs were identified and sequenced.One of them showed high homology to the AtORG4 gene and was used as a probe to screen the cDNA library of Jing02-461.Six positive clones were identified and one of them contained a full-length cDNA,which was named HvORG4.Sequence analysis showed that HvORG4 encoded a deduced basic protein of 197 amino acids.Northern blotting analysis showed that HvORG4 was constitutively expressed in root and stalk,not in leaf or spike,and strongly induced in barley spikelets in response to infection with F.graminearum isolate Huanggang-1.Its homology and expression profile suggest that the HvORG4 might function as a transcription factor,playing an important role in signal transduction pathway for defense against FHB in barley.展开更多

LiNi_(0.5)Mn_(1.5)O_(4) and LiMn_(2)O_(4) with novel spinel morphology were synthesized by a hydrothermal and postcalcination process.The synthesized LiMn_(2)O_(4) particles(5–10 lm)are uniform hexahedron,while the L...LiNi_(0.5)Mn_(1.5)O_(4) and LiMn_(2)O_(4) with novel spinel morphology were synthesized by a hydrothermal and postcalcination process.The synthesized LiMn_(2)O_(4) particles(5–10 lm)are uniform hexahedron,while the LiNi_(0.5)Mn_(1.5)O_(4) has spindle-like morphology with the long axis 10–15 lm,short axis 5–8 lm.Both LiMn_(2)O_(4) and LiNi_(0.5)Mn_(1.5)O_(4) show high capacity when used as cathode materials for Li-ion batteries.In the voltage range of 2.5–5.5 V at room temperature,the LiNi_(0.5)Mn_(1.5)O_(4) has a high discharge capacity of 135.04 mA·h·g^(-1) at 20 mAg^(-1),which is close to 147 mA·h·g^(-1)(theoretical capacity of LiNi_(0.5)Mn_(1.5)O_(4)).The discharge capacity of LiMn_(2)O_(4) is 131.08 mA·h·g^(-1) at 20 mAg^(-1).Moreover,the LiNi_(0.5)Mn_(1.5)O_(4) shows a higher capacity retention(76%)compared to that of LiMn_(2)O_(4)(61%)after 50 cycles.The morphology and structure of LiMn_(2)O_(4) and LiNi_(0.5)Mn_(1.5)O_(4) are well kept even after cycling as demonstrated by SEM and XRD on cycled LiMn_(2)O_(4) and LiNi_(0.5)Mn_(1.5)O_(4) electrodes.展开更多

The novel concept of the unique airway diseases its only supported by the similarities within pathogenetic process. Antagonists of H1 and H2 receptors are quite effective in allergic rhinitis, but not effective enough in asthma. In an era of corticosteroids, leucotriene antagonists and Anti-IgE treatment, there is still a challenge to search for more effective, more acurate and more safe treatment option. Antagonists (inversive agonists) of histamine receptors H4 seems to be one of the promising targets in the allergic rhinitis and asthma treatment. The first H4 antagonist entered to clinics and the results from a proof-of-concept Phase II clinical study is expected to be disclosed soon. This review article summarizes current knowledge on H4R that have been collected in various studies sharing evidences about efficacy of H4R as a reasonable target for diseases with histamine involved pathogenetic pathways.展开更多
