The Yellow Sea Cold Water Mass(YSCWM)is a distinct hydrographic phenomenon of the Yellow Sea,and the distribution pattern of meio-and macrobenthos diff ers inside and outside of the YSCWM.However,such a pattern has never been observed in the microbenthic ciliate communities.Therefore,we hypothesized that benthic ciliates followed a similar distribution pattern as meio-and macrobenthos,but this pattern has not been uncovered by morphological methods.We evaluated the diversity and distribution of benthic ciliates at fi ve stations along hydrographic gradients across the YSCWM and adjacent shallow water by using morphology and DNA and complementary DNA(cDNA)high-throughput sequencing of the V4 region of 18S rRNA gene.Results showed that the diversity of benthic ciliates detected by DNA(303 OTUs),and the cDNA(611 OTUs)sequencing was much higher than that detected by the morphological method(79 species).Morphological method detected roughly diff erent ciliate communities inside and outside of the YSCWM,but without statistical signifi cance.No clear pattern was obtained by DNA sequencing.In contrast,cDNA sequencing revealed a distinct distribution pattern of benthic ciliate communities like meioand macrobenthos,which coincided well with the results of the environmental parameter analysis.More than half of the total sequences detected by DNA sequencing belonged to planktonic ciliates,most(if not all)of which were recovered from historic DNA originating through the sedimentation of pelagic forms because none of them were observed morphologically.The irrelevant historic DNA greatly infl uenced the recovery of rare species and thus limited the understanding of the benthic ciliate diversity and distribution.Our research indicates that the methods used have signifi cant eff ects on the investigation of benthic ciliate communities and highlights that cDNA sequencing has great advantages in estimating the diversity and distribution of benthic ciliates,as well as the potential for benthic environmental assessments. 展开更多

By thecoefficient of difference test, 3 and 4 characters are found to be beyond the subspecies levelbetween Chinese Ayu and nominate subspecies (Plecoglossus altivelis altivelis) and between ChineseAyu (Plecoglossus altivelis chinensis) and Ryukyus subspecies (Plecoglossus altivelis ryukyuensis),respectively, which shows that they belong to different subspecies. We described a new species, Microhyla beilunensis sp. nov., from Zhejiang Province of China. Phylogenetic analyses based on the mitochondrial 12 S, 16 S and CO1 gene sequences suggested that the new taxon was distinctly separated from its congeners and closed to M. mixtura and M. okinavensis. Morphologically, the new species could be identified from its congeners except M. mixtura by several characters:(1) rudimentary webs on toe base;(2) absence of disks and dorsal median longitudinal grooves on finger tips;(3) presence of disks and dorsal median longitudinal grooves on toe tips. As well, the new species could be identified from topotype M. mixtura by the combination of characters:(1) apart from the stripes, bar-shaped and oval-shaped patterns, the rounded spots present on the dorsum of body and legs;(2) the outer metacarpal tubercles prominently larger than the inner one;(3) of males, the ratios of HW, IND, UEW and LAW to SVL of the new species were significantly larger than those of M. mixtura(P 〈 0.01), and the ratios of SL, IOD, LAHL, HLL, TL, TFL and FL to SVL of the new species were significantly less than those of M. mixtura(P 〈 0.05). 展开更多

As a fundamental category of river system, comparisons in diverse regions and types with quantitative parameters on meander morphology will help to distinguish different characteristics and approach formations and evolutions. It also can provide scientific basis of geomorphology for their protections and exploitation of the meander resources. Based on the images of Google Earth, meanders’ parame- ters are proposed and then measured and calculated. The result suggests that incised meanders in the Jialing River are among the most complex, irregular and sinuous channels. Special river gradient, geological structures, strata and lithology, backwater effect of landform are key factors to shape me- anders of the Jialing River. The evolution of meander in the Jialing River obeys the principle of mini- mum-maximum energy dissipation. Easy acquisitions of remote sensing data make regional and global comparisons possible, and then the differences of formations can be explored. The morphology can probably provide an evidence of antecedent river and demonstrate the channel incisions and crustal uplifts. Meander core in Chinese is named after the isolated hills in the Jialing River. The incised me- ander is a kind of resource and needs to be protected. 展开更多

In this paper, we tried morphological and molecular analyses to separate these two species. C. aerea and C. linum can be distinguished from morphological characteritics, such as frond dimension, cells size and shape, their mean length/width ratios(LWR), and cell walls constriction. Thalli of C. aerea attenuate basipetally, with diameter 270–500 μm at upper portion, 160–360 μm at middle portion, 100–160 μm at basal portion. For the upper part, the length of cells is less than their diameter. Cell walls usually constrict at the dissepiments, which are pellucid or colorless and give the filament beaded appearance. In contrast, thalli of C. linum often have a constant diameter of 90–300 μm within the same individual, cell walls usually do not constrict and cells are cylindrical or barrel shaped. The LWR is larger than that of C. aerea. Results show that the pairwise distance between two species is 3.6%–3.7% for 18 S r RNA gene and 53.5%–54.3% for ITS region. In phylogeny, they distribute at distant clades, which confirms a genetic divergence at molecular level. In addition, morphological data indicates that filament diameter of C. linum samples is highly variable, ranging from 90 μm to 300 μm. 本研究通过构建半夏品系的杂交体系并研究杂交种苗的扩繁周期,以解决半夏自然杂交成功率低、杂交种苗繁育效率低的问题。对消毒过的半夏花粉和雌蕊以TTC法测定其活力,并通过对比实验确定雌蕊的最佳培养基,对3种新的半夏品系持续培养出F_... 本研究通过构建半夏品系的杂交体系并研究杂交种苗的扩繁周期,以解决半夏自然杂交成功率低、杂交种苗繁育效率低的问题。对消毒过的半夏花粉和雌蕊以TTC法测定其活力,并通过对比实验确定雌蕊的最佳培养基,对3种新的半夏品系持续培养出F_(1)代杂交苗,通过杂交苗继代探究转接周期和成苗周期。结果表明,半夏雌蕊和花粉的最佳消毒时间为6 min;雌蕊的最佳培养基配方是半糖无琼脂MS培养基20.00 g/L+蔗糖15.00 g/L+琼脂5.00 g/L+6-BA 1.00 mg/L+NAA 0.01 mg/L,pH=5.8。以此为基础,培育出F_(1)代苗;4种半夏品系的F_(1)代杂交苗与亲本组培苗叶型有明显区别,经过扩繁发现了两个杂交种苗的扩繁周期。本研究建立了基于植物组织培养技术的半夏杂交体系并获得杂交苗,并探明杂交种苗的繁育周期。 展开更多

干旱胁迫对不同木薯苗期生理特性及气孔形态的影响

【目的】为探究干旱胁迫下木薯苗期光合特性及生理响应,比较木薯品种间耐旱性。【方法】选择5个木薯品种为试验材料,采用盆栽试验,设置正常供水(CK)和重度干旱胁迫(D)处理,研究干旱胁迫及复水后木薯苗期光合、渗透调节物质、膜脂过氧化... 【目的】为探究干旱胁迫下木薯苗期光合特性及生理响应,比较木薯品种间耐旱性。【方法】选择5个木薯品种为试验材料,采用盆栽试验,设置正常供水(CK)和重度干旱胁迫(D)处理,研究干旱胁迫及复水后木薯苗期光合、渗透调节物质、膜脂过氧化物含量、抗氧化酶活性等生理指标以及气孔形态的变化。【结果】①干旱胁迫后各品种木薯的净光合速率、气孔导度、胞间CO_(2)浓度和蒸腾速率与对照相比均降低;②干旱胁迫后,木薯叶片中的可溶性糖和脯氨酸积累,同时超氧化物歧化酶和过氧化氢酶活性显著升高,丙二醛含量均有不同程度的升高,复水后上述指标都有所降低或恢复到对照水平;③相较于其他品种,干旱胁迫后1-3-4的气孔密度增大,开度明显减小。【结论】干旱胁迫下木薯通过改变自身的渗透调节能力、酶促抗氧化活力以及叶片保水能力来抵抗干旱,其中木薯品种1-3-4的光合性能、生理特性和抗旱能力优于其他品种,并且在恢复水分供应后能迅速恢复植物生长势,从而表现出较强抗旱能力。 展开更多

Comparison of an automated digital cell morphology analysis system with manual counting

Background:Bionovation's CSFA800 is a new automated digital cell imaging analyzer.We evaluated the performance of the CSFA800 by comparing it with artificial peripheral blood white blood cell counting.Methods:According to inclusion and exclusion criteria,131 randomly selected samples(77 abnormal samples and 54 normal samples)were compared.Correlations between automated and manual counting results were analyzed.Manual counting was carried out according to the guidelines of the Association of Clinical and Laboratory Standards.Results:Counts of neutrophils,lymphocytes,monocytes,eosinophils,baso-phils,and immature granulocytes obtained from CSFA800 and artificial methods were linearly and positively correlated,with R values of 0.73,0.65,0.24,0.2,0.4,and 0.63,respectively,all p<0.05.Therefore,correlations be-tween CSFA800 and manual counting are acceptable.Compared with the DI‐60 Automated Digital Cell Morphology System(DI‐60;Sysmex),CSFA800 is more efficient and can analyze 20,000 cells in 1 min.However,the overall accuracy of CSFA800 is not as good as DI‐60,although its counting perfor-mance is better for basophils.Conclusions:The performance of CSFA800 for WBC counts is acceptable,and it displayed good performance for neutrophils,lymphocytes,and imma-ture granulocytes.Compared to DI‐60,CSFA800 is more efficient but has slightly lower overall accuracy.To some extent,CSFA800 is helpful to opti-mize the clinical laboratory workflow and improve the working efficiency of inspectors. 展开更多

以北京、河南和陕西不同产地的栓皮栎、麻栎、锐齿栎、房山栎、辽东栎、蒙古栎、夏栎、沼生栎等8种落叶栎类种子为研究对象,测定每粒种子的重量、种长、直径、包被长、碗疤直径和体积等形态指标,分析了不同种类、不同产地种子形态差异... 以北京、河南和陕西不同产地的栓皮栎、麻栎、锐齿栎、房山栎、辽东栎、蒙古栎、夏栎、沼生栎等8种落叶栎类种子为研究对象,测定每粒种子的重量、种长、直径、包被长、碗疤直径和体积等形态指标,分析了不同种类、不同产地种子形态差异以及种内各因子之间的相互关系。结果表明:8种落叶栎类种子百粒重、长度、直径、体积、包被长和碗疤直径平均值分别为(266.18±80.23)g、(20.70±1.51)mm、(14.65±2.07)mm、(2.82±0.91)cm3、(8.90±2.27)mm和(9.93±2.02)mm,不同种类、不同产地、不同个体栎类种子形态指标差异明显,指标间相关性较强;因子分析提出了落叶栎类种子3个公因子分别为:与直径相关的指标、与包被长相关的指标以及与种长相关的指标。为植物数量分类及相关研究提供参考。 展开更多

对5种禾本科植物种子中的淀粉粒(包括粟、黍、青狗尾草、高粱及薏苡等)进行了形态学观察和描述,结合颗粒长度的测量和统计学分析,建立了区分这些作物淀粉粒的一般方法。结果显示,粟和黍的淀粉粒以多边形居多,比例分别为75%和96%;青狗尾... 对5种禾本科植物种子中的淀粉粒(包括粟、黍、青狗尾草、高粱及薏苡等)进行了形态学观察和描述,结合颗粒长度的测量和统计学分析,建立了区分这些作物淀粉粒的一般方法。结果显示,粟和黍的淀粉粒以多边形居多,比例分别为75%和96%;青狗尾草中的淀粉粒以圆形为主;高粱和薏苡的淀粉粒形态具有较高的多样性。表现在消光臂上,高粱淀粉粒的消光臂呈现弯曲特征,而其他4种作物淀粉粒的消光臂则较为平直。在颗粒的长度分布上,粟淀粉粒为2.77~18.40μm,黍淀粉粒为3.93~12.85μm,青狗尾草淀粉粒为2.19~11.90μm,高粱淀粉粒为4.11~30.30μm,薏苡淀粉粒为5.48~25.44μm。研究表明,粟、黍、青狗尾草、高粱及薏苡等5种禾本科植物的淀粉粒形态特征存在相似性,但是也有差异。综合运用二维形状、脐点位置、表面特征、消光臂特征及长度等多种指标,可以对这几种作物的淀粉粒进行一定程度的区分。这一研究结果可用于辅助对古代淀粉粒的科属鉴定。 展开更多
