Common bean (Phaseolus vulgaris L.) is an economic important crop and one of the major grain legumes for human consumption in Latin America, Africa and Asia. A morphological study of shoot induced from embryonic axes development in four Costa Rican bean varieties (Brunca, Huetar, Guaymi and Bribri) cultivated on MS media with or without 5 mg·L–1 de N6-benzylaminopurine (BAP) by scanning electron microscopy (SEM) was developed in the present work. Micrographs showed similarities and differences in the ultrastructure of the apical dome, epidermal surface, stomata and different types of trichomes in the varieties cultivated on organogenesis media. Genotypes with advantageous morphological characteristics for genetic transformation, in particular an exposed apical dome, were identified. This work will contribute to the optimization of the in vitro regeneration of four common bean varieties. 展开更多

Electrocoagulation is progressively becoming an ecologically friendly water treatment method owing to its lack of secondary pollution,high active ingredient concentration,high treatment effectiveness,simple equipment,and simplicity of automated control implementation.Herein,electrocoagulation is offered as a method for treating wastewater containing azo dyes using a revolutionary flexible electronic fabric that can be mass-producible at a reasonable price.A computer-controlled machine embroiders 316L stainless steel fiber(316L SSF)onto an insulating fabric to manufacture a flexible electronic device of cathode and anode with a monopolar arrangement on the fabric surface.Using methyl orange(MO)solution to simulate azo dye wastewater,the decolorization rate of 500 ml MO reached 99.25% under the conditions of 50 mg·L^(-1)initial mass concentration,120 min electrolysis time,15 mA·g^(-1)current density,1 cm electrode spacing,0.1 mol·L^(-1)NaCl,pH 7.6,200 r·min^(-1)rotational speed of the stirrer,and 22-25℃ room temperature.In addition,it is feasible to embroider flexible electronic fabrics with varied sizes and numbers of electrodes based on the amount of treated sewage to increase the degradation rate,which has significant practical application value. 展开更多

The influence of the local environment on the Co magnetic moments is discussed by comparing the magnetic and electronic properties of ThCo4B to its parent ThCo5 compound. The total magnetic moment in these two compounds is dominated by the Co moment. The Spin orbit interaction affects the electronic structure and spin-density maps of the p-state of Th. 展开更多

Third-order nonlinear optical (NLO) materials have broad application prospects in high-density data storage, optical computer, modern laser technology, and other high-tech industries. The structures and frequencies of Dinaphtho[2,3-b:2’,3’-d]thiophene-5,7,12,13-tetraone (DNTTRA) and its 36 derivatives containing azobenzene were calculated by using density functional theory B3LYP and M06-2X methods at 6-311++g(d, p) level, respectively. Besides, the atomic charges of natural bond orbitals (NBO) were analyzed. The frontier orbitals and electron absorption spectra of A-G5 molecule were calculated by TD-DFT (TD-B3LYP/6-311++g(d, p) and TD-M06-2X/6-311++g(d, p)). The NLO properties were calculated by effective finite field FF method and self-compiled program. The results show that 36 molecules of these six series are D-π-A-π-D structures. The third-order NLO coefficients γ (second-order hyperpolarizability) of the D series molecules are the largest among the six series, reaching 10<sup>7</sup> atomic units (10<sup><span style="color:#4F4F4F;font-family:-apple-system, " font-size:14px;white-space:normal;background-color:#ffffff;"="">-</span>33</sup> esu) of order of magnitude, showing good third-order NLO properties. Last, the third-order NLO properties of the azobenzene ring can be improved by introducing strong electron donor groups (e.g. -N(CH<sub>3</sub>)<sub>2</sub> or -NHCH<sub>3</sub>) in the azobenzene ring, so that the third-order NLO materials with good performance can be obtained. 展开更多

Many instruments have helped traders to effectively communicate,establish contracts,manage risk and protect their benefits while collaborating with people who have diverse cultural backgrounds.The B/L(bill of lading)has been one of the most significant documents in the shipping industry since it was invented in the 13th century in Italy.It is an evidence of existing contracts between the shipper and the carrier,in which both of them agreed to deliver the goods safely from the port of loading to the port of discharge.However,many problems are triggered by requiring the presentation of the original paper B/L at the discharging port so that the buyer gets the cargo in time.As of today,many attempts were carried out to develop an electronic equivalent to the paper B/L.Additionally,many practices were formed in order for these electronic equivalents to be legally reinforced and enhanced.This article presents the main transitional stages of these efforts and the status of this progress.It analyses the legal background which has been established to validate these ventures and the option of using an electronic B/L in order for the ships to be able to deliver the goods without the presentation of the original document.In the meantime,this article highlights potential areas for further investigation. 展开更多

The mechanism of the proton_transfer_coupled electron transfer (PT_ET) reactions between the menaquinone Q A (MQ 1) and ubiquinone Q B (UQ 1) in the bacterial photosynthetic reaction center of Rhodopseudomona viridis was studied by using the B3LYP/6_31G(d) method. The changes of standard Gibbs free energy ΔG 0 of all possible reactions followed the ET reaction (1) were calculated. The results indicated that: (1) according to the ΔG 0 values of corresponding reactions, UQ 1 could not accept two electrons from MQ - 1 continually without the coupled proton transfer reactions. Because of ΔG 0 2b 0, ΔG 0 3b 0 and ΔG 0 4b 0, the corresponding PT_ET reactions could take place along with reactions (2b), (3b) and (4b) sequentially; (2) on the gaseous condition, the first and second transferred protons (H +(1) and H +(2)) from the surrounding amino acid residues or water molecules will combine with the oxygen No.7 and oxygen No.8 of UQ 1, respectively. On the condition of protein surroundings (by SCRF model, ε =4.0), the results are converse but the energy difference between the combination of H +(1) and H +(2) with UQ - 1 is quite small. 【目的】非洲猪瘟病毒(African swine fever virus,ASFV)是对生猪养殖业危害极大的一种病原体,B602L蛋白作为其重要的非结构蛋白,是衣壳蛋白p72折叠为正确的三级结构的伴侣分子之一,在病毒组装成二十面体结构的过程中发挥着关键作用。... 【目的】非洲猪瘟病毒(African swine fever virus,ASFV)是对生猪养殖业危害极大的一种病原体,B602L蛋白作为其重要的非结构蛋白,是衣壳蛋白p72折叠为正确的三级结构的伴侣分子之一,在病毒组装成二十面体结构的过程中发挥着关键作用。特异性抗体对于抗非洲猪瘟免疫应答机制及血清学检测方法研究具有重要意义,本研究旨在获取ASFV B602L蛋白,并制备抗ASFV B602L蛋白的多克隆抗体,为ASFV B602L蛋白功能研究及相关诊断试剂的研发提供材料。【方法】以ASFV China/2018/AnhuiXCGQ株的B 602 L基因为研究对象,应用生物信息学软件对B602L蛋白的理化性质、信号肽、跨膜结构、抗原性和蛋白结构进行分析后,利用同源重组的方法将其克隆至原核表达载体pET-28a(+)中,转化大肠杆菌BL21(DE3)感受态细胞后经诱导表达获得B602L重组蛋白,利用镍柱亲和层析进行蛋白纯化,通过SDS-PAGE和Western blotting对B602L蛋白纯化效果进行验证。将纯化后的B602L蛋白免疫BALB/c小鼠制备多克隆抗体,通过间接ELISA方法测定多克隆抗体效价,通过Western blotting和Dot blotting检测多克隆抗体特异性。【结果】生物信息学分析表明,B602L蛋白属于稳定性亲水蛋白,无跨膜区和信号肽。二级结构主要含有α-螺旋(54.91%)、延伸链(9.81%)和无规则卷曲(32.08%)。成功构建了ASFV B602L重组质粒,SDS-PAGE和Western blotting结果显示,重组菌pET-28a-B602L-BL21在16℃、0.5 mmol/L IPTG诱导下高效表达,获得的重组蛋白主要表达在BL21菌株上清中,大小约为70 ku,且能与ASFV阳性血清特异性结合。间接ELISA检测结果显示,制备的抗B602L鼠源多克隆抗体效价达1∶409600,Western blotting和Dot blotting试验结果表明,抗B602L鼠源多克隆抗体能够特异性识别B602L蛋白。【结论】原核表达的B602L重组蛋白具有良好的免疫原性,能够与ASFV阳性血清特异性结合,制备的鼠抗ASFV B602L蛋白多克隆抗体具备较好的生物学活性。本试验结果可为进一步研究ASFV B602L蛋白的生物学功能和非洲猪瘟诊断方法的建立提供生物材料和理论支撑。 展开更多

An E//B neutral particle analyzer(NPA)has been designed and is under development at Sichuan University and Southwestern Institute of Physics.The main purpose of the E//B NPA is to measure the distribution function of fast ions in the HL-2A/3 tokamak.The E//B NPA contains three main units,i.e.the stripping unit,the analyzing unit and the detection unit.A gas stripping chamber was adopted as the stripping unit.The results of the simulations and beam tests for the stripping chamber are presented.Parallel electric and magnetic fields provided by a NdFeB permanent magnet and two parallel electric plates were designed and constructed for the analyzing unit.The calibration of the magnetic and electric fields was performed using a 50 kV electron cyclotron resonance ion source(ECRIS)platform.The detection unit consists of 32lutetium-yttrium oxyorthosilicate(LYSO)detector modules arranged in two rows.The response functions ofα,hydrogen ions(H^(+),H_(2)^(+)and H_(3)^(+))andγfor a detector module were measured with^(241)Am,^(137)Cs and^(152)Eu sources together with the 50 kV ECRIS platform.The overall results indicate that the designed E//B NPA device is capable of measuring the intensity of neutral hydrogen and deuteron atoms with energy higher than 20 keV. 展开更多

目的探究MyD88L265P及CD79B基因突变对原发性中枢神经系统淋巴瘤(PCNSL)患者预后的影响。方法收集2018年8月-2020年11月于兰州大学第二医院经开颅手术或立体定位活检术确诊的18例免疫功能正常(无HIV感染及免疫抑制药服用病史)的PCNSL患... 目的探究MyD88L265P及CD79B基因突变对原发性中枢神经系统淋巴瘤(PCNSL)患者预后的影响。方法收集2018年8月-2020年11月于兰州大学第二医院经开颅手术或立体定位活检术确诊的18例免疫功能正常(无HIV感染及免疫抑制药服用病史)的PCNSL患者临床资料进行回顾性分析。分别采用实时荧光定量PCR和一代基因测序技术检测18例PCNSL患者肿瘤组织中MyD88L265P和CD79B基因突变情况。针对可能与PCNSL首次无疾病进展生存期(PFS)和总生存期相关的指标行单因素分析,并采用Cox回归进行多因素分析。结果18例PCNSL肿瘤组织MyD88L265P突变率为38.9%,CD79B突变率为33.3%,MyD88L265P/CD79B共突变率为27.8%。单因素分析结果显示,病灶多发、病灶深部受累、肿瘤组织CD79B突变患者的PFS时间更短(P<0.05);多因素分析结果显示,病灶深部受累(HR=0.135,95%CI 0.023~0.799,P<0.05)、肿瘤组织CD79B突变(HR=0.149,95%CI 0.028~0.800,P<0.05)是PCNSL患者预后不良的独立危险因素。结论本组PCNSL患者肿瘤组织MyD88L265P、CD79B突变频率较高,这两种基因突变尤其是CD79B突变可能与PCNSL不良预后相关。 展开更多

Objective:To determine the major changes in the microstructure of Candida albicans(C. albicans) after treatment with Euphorbia hirta(E.hirta) L.leaf extract.Methods:Transmission electron microscopy was used to study the ultrastructural changes caused by E.hirta extract on C. albicans cells al various exposure time.Results:It was found that the main abnormalities were the alterations in morphology,lysis and complete collapse of the yeast cells after 36 h of exposure to the extract.Whereas the control cultures showed a typical morphology of Candida with a uniform central density,typically structured nucleus,and a cytoplasm with several elements of endomembrane system and enveloped by a regular,intact cell wall.Conclusions:The significant antifungal activity shown by this methanol extract of E.hirta L.suggests its potential against infections caused by C.albicans.The extract may be developed as an anticandidal agent. 展开更多

High energy particles are the main target of satellite space exploration; particle storm events are closely related to solar activity,cosmic ray distribution, and magnetic storms. The commonly seen energetic particle(electron) precipitation anomalies include mainly the inner and outer Van Allen radiation belts, the South Atlantic Anomaly, and the anomalous stripes excited by artificial electromagnetic waves. The China Seismo-Electromagnetic Satellite(CSES), launched in February of 2018, provides a platform for studying ionospheric particle disturbances. This paper reports the first studies of electron precipitation phenomenon based on high energy particle data from the CSES. We find that the global distribution of electron fluxes in the low energy band(0.1–3 MeV) can relatively well reflect the anomalous precipitation belt, which is consistent with results based on the DEMETER satellite, indicating that the quality of the lowenergy band payload of the CSES is good. In addition, this paper makes an in-depth study of the electron precipitation belt excited by the NWC artificial VLF electromagnetic transmitter located in Australia, which appears as a typical wisp structure on the energy spectrum. The magnetic shell parameter L corresponding to the precipitation belt ranges from 1.44 to 1.74, which is close to the L value(~1.45) of the NWC transmitter; the energy of the precipitation electrons is between 100 keV and 361.57 keV, among which the precipitation of 213.73 keV electrons is most conspicuous. 展开更多

In this study, we evaluated the oligosaccharide content, resistant starch and the g... “Pinhão”, the seed of Araucaria angustifolia, is an important food, being part of the eating habits of Indigenous communities. In this study, we evaluated the oligosaccharide content, resistant starch and the growth of probiotic bacteria. GF4 (1-fructofuranosylnystose) was the main fructo-oligosaccharides found, in higher contents compared to other food sources. Maltooligosaccharides (MOS) represented the main part of the oligosaccharides profile of Brazilian pine seeds. In descending order of importance was maltoheptaose (G7), maltohexose (G6) and maltotriose (G3). The starches from the variety Sanct josephi presented the highest amount of resistant starch that could stimulate probiotic strains, mainly B. breve and L. plantarum, and may have a prebiotic effect, potentially promoting health benefits. This study advances the understanding of the chemical composition of the main portion of the “pinhão” enhancing awareness of its potential as a healthy food source, contributing to different uses and indirectly with the species preservation. 展开更多
