Phaeocystis globosa is an important unicellular eukaryotic alga that can also form colonies.P.globosa can cause massive harmful algal blooms and plays an important role in the global carbon or sulfur cycling.Thus far,...Phaeocystis globosa is an important unicellular eukaryotic alga that can also form colonies.P.globosa can cause massive harmful algal blooms and plays an important role in the global carbon or sulfur cycling.Thus far,the ecophysiology of P.globosa has been investigated by numerous studies.However,the proteomic response of P.globosa to nitrogen depletion remains largely unknown.We compared four protein preparation methods of P.globosa for two-dimensional electrophoresis(2-DE)(Urea/Triton X-100 with trichloroacetic acid(TCA)/acetone precipitation;TCA/acetone precipitation;Radio Immuno Precipitation Assay(RIPA)with TCA/acetone precipitation;and Tris buffer).Results show that the combination of RIPA with TCA/acetone precipitation had a clear gel background and showed the best protein spot separation effect,based on which the proteomic response to nitrogen depletion was studied using 2-DE.In addition,we identified six differentially expressed proteins whose relative abundance increased or decreased more than 1.5-fold(P<0.05).Most proteins could not be identified,which might be attributed to the lack of genomic sequences of P.globosa.Under nitrogen limitation,replication protein-like,RNA ligase,and sn-glycerol-3-phosphate dehydrogenase were reduced,which may decrease the DNA replication level and ATP production in P.globosa cells.The increase of endonucleaseⅢand transcriptional regulator enzyme may affect the metabolic and antioxidant function of P.globosa cells and induce cell apoptosis.These findings provide a basis for further proteomic study of P.globosa and the optimization of protein preparation methods of marine microalgae.展开更多
The current design philosophy for submarine hulls,in the preliminary design stage,generally considers as governing limit states material yielding along with various buckling modes.It is common belief that,beyond the d...The current design philosophy for submarine hulls,in the preliminary design stage,generally considers as governing limit states material yielding along with various buckling modes.It is common belief that,beyond the design pressure,material yielding of the shell plating should occur first,eventually followed by local buckling,while global buckling currently retains the highest safety factor.On the other hand,in the aeronautical field,in some cases structural components are designed in such a way that local instability may occur within the design loads,being the phenomena inside the material elastic range and not leading to a significant drop in term of stiffness.This paper is aimed at investigating the structural response beyond a set of selected limit states,using nonlinear FE method adopting different initial imperfection models,to provide the designers with new information useful for calibrating safety factors.It was found that both local and global buckling can be considered as ultimate limit states,with a significant sensitivity towards initial imperfection,while material yielding and tripping buckling of frames show a residual structural capacity.In conclusion,it was found that the occurrence of local buckling leads to similar sudden catastrophic consequences as global buckling,with the ultimate strength capacity highly affected by the initial imperfection shape and amplitude.展开更多

The main discussion is on how to predict employees’ job satisfaction through the implementation of interpersonal emotion management strategies and the main source of research is review of existing literature. This review addresses the gap in the literature by reviewing the role of interpersonal emotional management on followers’ job performance. This review finds the relationship between interpersonal emotional management which consists of four dimensions: circumstance adjustment,attentional placement,cognitive change,modulation of emotional response and job satisfaction. Overall,this literature review contributes to the interpersonal emotion management and job satisfaction literatures by providing meaningful management implications to the organizations.展开更多

Especially under certain conditions, it shows that existence and uniqueness of non-small amplitude stable limit cycle is equivalent to the local un-stability of positive equilibrium and the local stability of positive equilibrium implies its global stability.
