In vitro recovery studies are essential to calibrating the microdialysis system for in vivo studies. The effect of a combination of antibiotics on recovery into microdialysate requires investigation. In vitro microdialysis recovery studies were conducted on a combination of vancomycin and tobramycin, in a simulated in vivo model. Comparison was made between recoveries for three different concentrations and three different perfusate flow rates. The overall relative recovery for vancomycin was lower than that of tobramycin. For tobramycin, a concentration of 20μg/mL and flow rate of 1.0μL/min had the best recovery. A concentration of 5.0μg/mL and flow rate of 1.0μL/min yielded maximal recovery for vancomycin. Large molecular size and higher protein binding resulted in lower relative recoveries for vancomycin. Perfusate flow rates and drug concentrations affected the relative recovery when a combination of vancomycin and tobramycin was tested. Low perfusate flow rates were associated with higher recovery rates. For combination antibiotic measurement which includes agents that are highly protein bound, in vitro studies performed prior to in vivo studies may ensure the reliable measurement of unbound concentrations.展开更多

The aim of this study was to investigate the effect of exercise test on corrected QT dispersion (QTcd) and QTcd recovery in PEX. Patients and Methods: A total of 37 patients diagnosed as PEX were included in the study. The control group consisted of 38 patients. The Electrocardiographies (ECG) were taken. The patients were subjected to an exercise test. Results: In the study group, baseline, peak and third minute recovery QTcd values were 41.35 ± 7.27 ms, 65.19 ± 7.83 ms, and 53.61 ± 6.45 ms, respectively. In the control group, baseline, peak and third minute recovery QTcd values were 29.92 ± 4.23 ms, 38.31 ± 4.22 ms and 28.76 ± 3.09 ms, respectively. There was statistically significant?difference between the baseline and peak QTcd values of the study group and?the control group (t = 8.34224/p 0.00001, t = 18.55079/p 0.00001, respectively). In addition, while there was a statistically significant?difference between the baseline QTcd and the third minute recovery QTcd in the study group, there was no statistically significant difference between the baseline QTcd value and the third minute recovery QTcd value in the control group as heart rate recovery (t = 8.388558/p 0.00001, t = 1.967315/p = 0.056668, respectively).?Conclusion: All of them were found to have a higher risk in terms of malignant ventricular arrhythmia in the study group, both at rest and with effort, in a statistically significant way compared to the control group.展开更多

Des...The development of pollution-free dyeing technology, including anhydrous dyeing and non-aqueous dyeing technologies, has always been an important way and research hot in energy conservation and emission reduction. Designing new structural dye molecules is the key to water-saving dyeing processes.Herein, three reactive dyes were designed and synthesized, which contained large planar multiconjugated systems and multi-reactive groups. The designed reactive dyes are expected to have high affinity and high fixations in non-aqueous or small bath dyeing processes. The reactive dyes were applied in the decamethylcyclopentasiloxane(DMCS) reverse micelle dyeing for cotton fabric. High exhaustion rate of 99.35%, 98.10% and 98.80%, and fixation rate of 95.15%, 96.34% and 94.40% for three dyes, R1,R2 and R3, could be respectively obtained. The dyes can be fully utilized and had excellent dyeing performance, fastness and levelling properties under the revere micelle dyeing. The cotton fabric is like an oil-water separator in the dyeing process, where the dye micelles rapidly absorb and permeate into the cotton fibers. DMCS circulates around the fabric to transfer mass and energy. After dyeing, the solvent can be separated quickly and reused. The new reactive dyes containing large planar and multi-conjugated systems have potential application in green and sustainable dyeing technology with less wastewater and higher utilization.展开更多

We studied the CatWalk XT■gait analysis for evaluating hindlimb functional recovery in a widely used and clinically relevant thoracic contusion/compression spinal cord injury model in rats. Rats were randomly assigned to either a T9 spinal cord injury or sham laminectomy. Locomotion recovery was assessed using the Basso, Beattie, and Bresnahan open field rating scale and the CatWalk XT■gait analysis. To determine the potential bias from weight changes, corrected hindlimb(H) values(divided by the unaffected forelimb(F) values) were calculated. Six weeks after injury, cyst formation, astrogliosis, and the deposition of chondroitin sulfate glycosaminoglycans were assessed by immunohistochemistry staining. Compared with the baseline, a significant spontaneous recovery could be observed in the CatWalk XT■parameters max intensity, mean intensity, max intensity at%, and max contact mean intensity from 4 weeks after injury onwards. Of note, corrected values(H/F) of CatWalk XT■parameters showed a significantly less vulnerability to the weight changes than absolute values, specifically in static parameters. The corrected CatWalk XT■parameters were positively correlated with the Basso, Beattie, and Bresnahan rating scale scores, cyst formation, the immunointensity of astrogliosis and chondroitin sulfate glycosaminoglycan deposition. The CatWalk XT■gait analysis and especially its static parameters, therefore, seem to be highly useful in assessing spontaneous recovery of hindlimb function after severe thoracic spinal cord injury. Because many CatWalk XT■parameters of the hindlimbs seem to be affected by body weight changes, using their corrected values might be a valuable option to improve this dependency.展开更多

Methods: We enrolled 102 RA patients treated with Tofacitinib in combination with MTX from a multicenter, exploratory, short-term, prospective and observational ultrasound cohort study of patients who received biologic or targeted synthetic disease-modifying antirheumatic drug (DMARD) therapy. We evaluated the patients’ clinical disease activity and musculoskeletal ultrasound (MSUS) scores. The serum concentrations of two venous blood inflammatory indicators were evaluated (c-reactive protein [CRP], erythrocyte sedimentation rate [ESR]) by multiplex bead assays at baseline, 3, and 6?months: the change over 6?months was defined as the P value. Before MSUS score treatment, an associate chief physician tested the wrist joints of each patient at the first treatment, and the attending physician with 3 years of MSUS experience and 7 years of work experience also performed semi-quantitative scores on the same RA patients and tested the consistency of the results. Results: Tofacitinib in combination with MTX significantly improved the clinical disease activity and MSUS score over 6?months. Serum ESR and CRP were significantly elevated at 6?months after the Tofacitinib in combination with MTX introduction (P P P Conclusions: MSUS scores may be useful for predicting RA patients’ therapeutic responses to abatacept.展开更多

At the breakdown voltage of 450 kV, with the vacuum diode voltage of about 200 kV, and a current of 30 kA, recovery characteristics of H2, N2, SF6 were studied. The recovery percentages of the gas breakdown voltage and vacuum diode voltage were determined. The results show that hydrogen has the best recovery characteristics. At a pulse interval of 8.8 ms, the recovery percentages of both the gas breakdown voltage and vacuum diode voltage for hydrogen exceed 95%. For SF6 and N2 with an interval of 25 ms and 50 ms respectively, a 90% voltage recovery was obtained. The experiments also proved that the repetitive rate of the high power accelerator with a pulse forming line is mainly restricted by the gas switch repetitive rate; the recovery percentages of the vacuum diode voltage are limited by the recovery percentages of the gas switch breakdown voltage. The hydrogen switch can be employed for a high repetitive rate-high power accelerator with a pulse forming line.展开更多

The web- server is called Pred-PFR (Predicting Protein Folding Rate). Pred-PFR is featured by fusing multiple individual predictors, each of which is established based on one special feature derived from the protein sequence. The ensemble pre-dictor thus formed is superior to the individual ones, as demonstrated by achieving higher correlation coefficient and lower root mean square deviation between the predicted and observed results when examined by the jack-knife cross-validation on a benchmark dataset constructed recently. This multistudy report examined changes and the doseàresponse relationships between recoveryàstress psychological states, training load(TL), heart rate(HR), heart rate recovery(HRR), and heart rate variability(HRV) while providing evidence for the factorial validity of a short French version of the RecoveryàStress Questionnaire for Athletes(RESTQ-36-R-Sport).Methods: Four hundred and seventy-three university athletes(Study 1), 72 full expert swimmers(Study 2), and 11 national to international swimmers(Study 3) participated in the study. Data were analyzed through confirmatory factor analyses(Study 1), repeated ANOVAs and correlational analyses(Study 2), t tests and correlational analyses(Study 3).Results: Multiple-group confirmatory factor analyses showed that the RESTQ-36-R-Sport scores were partially invariant across gender, type of sport, and practice level(Study 1). A doseàresponse relationship was performed between TL and RESTQ-36-R-Sport scores during an ecological training program(Study 2). Finally, relationships were found between physiological(HRR) and psychological(RESTQ-36-R-Sport) states during an ecological tapering period leading to a national championship(Study 3).Conclusion: As a whole, these findings provided evidence for the usefulness of the short version of the RESTQ-36-R-Sport for regular monitoring to prevent potential maladaptation due to intensive competitive sport practice.展开更多

Generally, they are either elevated or depressed at the same time. This study was aimed at evaluating the clinical ability of these markers in the early diagnosis of pulmonary tuberculosis. Materials and Methods: A total of 50 (male 39, female 21) patients who tested positive to Acid Alcohol Fast Bacilli (AAFB) were studied. ESR and CRP values were estimated using standard procedures. Results: The mean ESR value among the AAFB positive patients was 53.16 ± 4.92 mm/hr while the CRP value was 0.273 ± 0.035 mg/L. Gender and age were not found to have any influence on the ESR and CRP values. No relationship was found to exist between ESR and CRP (r = 0.17;p = 0.235) and age (r = 0.125;p = 0.388). Conclusion: A moderately elevated ESR and low CRP values exist at the early diagnosis of tuberculosis. There is no correlation between ESR and CRP at the onset of tuberculosis;hence CRP cannot be used as a screening tool for early diagnosis of tuberculosis.展开更多

Protein content increased with N application rate and with N proportion applied at late stage. β-glucan content also responded significantly to N application rate and timing, but with different pattern with protein content. Of three N rate treatments, the medium N rate (135 kg ha-1) had the highest β-glucan content, being significantly higher than low N rate (90 kg ha-1) and no difference with high N rate (180 kg ha-1). A-mong three N timing treatments, two times of N top-dressing at both tillering and booting stage had significantly higher β-glucan content than once N top-dressing at tillering or booting stage. Sowing date has the dramatic effect on both β-glucan and protein content. Protein content decreased with the delayed sowing, and kernel weight showed opposite tendency. Either earlier or later sowing caused increased β-glucan content relative to sowing in early November, which is the normal date for barley sowing locally. Seeding rate had no significant influence on both β-glucan and protein content.展开更多

It is the key point that how much the count of bacteria and how long it can preserve. Freeze-drying skill is one of the best preservation methods, but it's defect that damage to biological systems can be attributed to changes in the physical state of membrane lipids or/and changes in the structure of sensitive proteins and the decreased survival rate. This work used pretreatment methods-cold shock and hot shock to reduce the damage to bacteria during freeze-drying. Using unpretreated samples as control, aspects of cold shock or heat shock. The response of Enterococcous faecalis A31 was investigated during lag phase, the middle of exponential growth phase and the terminal of exponential growth phase. The results suggested that when cold shock, the shock protein concentration produced by treated sample within 2 h during the middle of exponential growth phase was higher than 4, 8, 24 h, The concentration of shock protein produced by treated sample at 10 ℃ was higher than 20℃, The concentration of sample protein at the middle of exponential growth phase was higher than that at the terminal of exponential growth phase. After one-month storage, the survival rate at 10℃ was better than 20 ℃ and 4 ℃comparatively. The survival rate at (6 h) 4 ℃/24 h (lag period) was higher than (10 h) 4 ℃/24 h (the middle of exponential growth phase), and the effect of 10℃/8 h shock treatments was best during the middle of exponential growth phase, when hot shock, The concentration of heat shock proteins produced was not obvious, and survival rate was higher at 45 ℃, 30 min than at 45 ℃, 60 min.展开更多

文摘Ischemic stroke can cause blood-brain barrier(BBB)injury, which worsens brain damage induced by stroke.Abnormal expression of tight junction proteins in endothelial cells(ECs)can increase intracellular space and BBB leakage.Selective inhibition of mitogen-activated protein kinase, the negative regulatory substrate of mitogen-activated protein kinase phosphatase(MKP)-1,improves tight junction protein function in ECs,and genetic deletion of MKP-1 aggravates ischemic brain injury. However, whether the latter affects BBB integrity, and the cell type-specific mechanism underlying this process, remain unclear. In this study,we established an adult male mouse model of ischemic stroke by occluding the middle cerebral artery for 60 minutes and overexpressed MKP-1 in ECs on the injured side via lentiviral transfection before stroke. We found that overexpression of MKP-1 in ECs reduced infarct volume, reduced the level of inflammatory factors interleukin-1β,interleukin-6,and chemokine C-C motif ligand-2,inhibited vascular injury, and promoted the recovery of sensorimotor and memory/cognitive function.Overexpression of MKP-1 in ECs also inhibited the activation of cerebral ischemia-induced extracellular signal-regulated kinase(ERK)1/2 and the downregulation of occludin expression. Finally,to investigate the mechanism by which MKP-1 exerted these functions in ECs,we established an ischemic stroke model in vitro by depriving the primary endothelial cell of oxygen and glucose, and pharmacologically inhibited the activity of MKP-1 and ERK1/2.Our findings suggest that MKP-1 inhibition aggravates oxygen and glucose deprivation-induced cell death, cell monolayer leakage, and downregulation of occludin expression, and that inhibiting ERK1/2 can reverse these effects. In addition,co-inhibition of MKP-1 and ERK1/2 exhibited similar effects to inhibition of ERK1/2.These findings suggest that overexpression of MKP-1 in ECs can prevent ischemia-induced occludin downregulation and cell death via deactivating ERK1/2,thereby protecting the integrity of BBB,alleviating brain injury, and improving post-stroke prognosis.

文摘Microdialysis is a technique used to measure the unbound antibiotic concentration in the interstitial spaces, the target site of action. For combination antibiotic measurement which includes agents that are highly protein bound, in vitro studies performed prior to in vivo studies may ensure the reliable measurement of unbound concentrations.
文摘Background: In pseudoexfoliative syndrome (PEX), extraocular accumulation is associated with cardiovascular morbidity and mortality. In addition, while there was a statistically significant?difference between the baseline QTcd and the third minute recovery QTcd in the study group, there was no statistically significant difference between the baseline QTcd value and the third minute recovery QTcd value in the control group as heart rate recovery (t = 8.388558/p 0.00001, t = 1.967315/p = 0.056668, respectively).?Conclusion: All of them were found to have a higher risk in terms of malignant ventricular arrhythmia in the study group, both at rest and with effort, in a statistically significant way compared to the control group.

文摘Conformance control and water plugging are a widely used EOR method in mature oilfields. However, majority of conformance control and water plugging agents are unavoidable dehydrated situation in high-temperature and high-salinity low permeability reservoirs.Consequently,a novel conformance control system HPF-Co gel, based on high-temperature stabilizer(CoCl_(2)·H_(2)O,CCH)is developed. The HPF-Co bulk gel has better performances with high temperature(120℃)and high salinity(1×10^(5)mg/L).According to Sydansk coding system, the gel strength of HPF-Co with CCH is increased to code G.The dehydration rate of HPF-Co gel is 32.0%after aging for 150 d at 120℃,showing excellent thermal stability. The rheological properties of HPF gel and HPF-Co gel are also studied. The results show that the storage modulus(G′)of HPF-Co gel is always greater than that of HPF gel.The effect of CCH on the microstructure of the gel is studied. The results show that the HPF-Co gel with CCH has a denser gel network, and the diameter of the three-dimensional network skeleton is 1.5-3.5μm.After 90 d of aging,HPF-Co gel still has a good three-dimensional structure.Infrared spectroscopy results show that CCH forms coordination bonds with N and O atoms in the gel amide group, which can suppress the vibration of cross-linked sites and improve the stability at high temperature.Fractured core plugging test determines the optimized polymer gel injection strategy and injection velocity with HPF-Co bulk gel system, plugging rate exceeding 98%.Moreover,the results of subsequent waterflooding recovery can be improved by 17%.

文摘The development of pollution-free dyeing technology, including anhydrous dyeing and non-aqueous dyeing technologies, has always been an important way and research hot in energy conservation and emission reduction. The new reactive dyes containing large planar and multi-conjugated systems have potential application in green and sustainable dyeing technology with less wastewater and higher utilization.
文摘Measuring the tanning skin can provide a scientific method to evaluate the efficacy of cosmetics exfoliator.Dihydroxyacetone(DHA,5.0%(w/w))was used to stain the inner forearms cuticle of subjects. The skin surface L*and b*were determined, then ITA and its recovery rate were calculated to evaluate the exfoliating efficacy of cosmetics. The results showed that no significant(P≥0.05)differences were observed for L*,b*and ITA on different skin surface of unstained subjects. After staining with DHA,no adverse reaction was found in the stained areas, and the changes of L*,b*and ITA on different skin surface were consistent(P≥0.05).Two different cosmetics were used continuously to exfoliate the skin in different areas. After treatment for 7 days, the corresponding areas of skin L*,b*and ITA had significant(P<0.05)improvement in comparison with negative control area. But there were differences in the evaluation results of the exfoliation efficacy using these three parameters. The recovery rate of ITA of two samples improved significantly(P<0.05)than the negative control area. The work indicated that using 5.0%DHA for skin browning and measuring the skin surface color change by ITA recovery rate can accurately and sensitively assess the skin cuticle renewal rate difference. This method can be used as one of the effective way to evaluate the efficacy of cosmetics exfoliation.
文摘Assessment of locomotion recovery in preclinical studies of experimental spinal cord injury remains challenging. Because many CatWalk XT■parameters of the hindlimbs seem to be affected by body weight changes, using their corrected values might be a valuable option to improve this dependency.
文摘Background: To evaluate the effect of treatment on musculoskeletal ultrasound (MSUS) and explore whether MSUS are associated with therapeutic response in rheumatoid arthritis (RA) patients treated with Tofacitinib in combination with methotrexate (MTX). Serum ESR and CRP were significantly elevated at 6?months after the Tofacitinib in combination with MTX introduction (P P P Conclusions: MSUS scores may be useful for predicting RA patients’ therapeutic responses to abatacept.

文摘A two-pulse method is used to determine the insulation recovery time of the gas spark gap switch with different types of gas applied in a high power accelerator with a water dielectric pulse forming line. The hydrogen switch can be employed for a high repetitive rate-high power accelerator with a pulse forming line.

文摘Acupuncture has been shown to lessen the inflammatory reaction after acute spinal cord injury and reduce secondary injury. However, the mechanism of action remains unclear. In this study,a rat model of spinal cord injury was established by compressing the T8-9 segments using a modified Nystrom method.Twenty-four hours after injury,Zusanli(ST36),Xuanzhong(GB39),Futu(ST32)and Sanyinjiao(SP6)were stimulated with electroacupuncture.Rats with spinal cord injury alone were used as controls. At 2,4 and 6 weeks after injury, acetylcholinesterase(ACh E)activity at the site of injury, the number of medium and large neurons in the spinal cord anterior horn, glial cell line-derived neurotrophic factor(GDNF)m RNA expression, and Basso,Beattie and Bresnahan locomotor rating scale scores were greater in the electroacupuncture group compared with the control group. These results demonstrate that electroacupuncture increases ACh E activity,up-regulates GDNF m RNA expression, and promotes the recovery of motor neuron function in the anterior horn after spinal cord injury.
文摘We have developed a web-server for predicting the folding rate of a protein based on its amino acid sequence information alone. As a user-friendly web- server, Pred-PFR is freely accessible to the public at www.csbio.sjtu.edu.cn/bioinf/Folding Rate/.

文摘Background: Psychological stress and recovery monitoring is a key issue for increasing athletes' health, well-being, and performance. Finally, relationships were found between physiological(HRR) and psychological(RESTQ-36-R-Sport) states during an ecological tapering period leading to a national championship(Study 3).Conclusion: As a whole, these findings provided evidence for the usefulness of the short version of the RESTQ-36-R-Sport for regular monitoring to prevent potential maladaptation due to intensive competitive sport practice.
文摘Background: ESR and CRP measurements reflect different aspects of systemic inflammation. There is no correlation between ESR and CRP at the onset of tuberculosis;hence CRP cannot be used as a screening tool for early diagnosis of tuberculosis.

文摘The influences of N application rate, timing, sowing date and seeding rate on β-glucan and protein content in barley grains were studied through the field experiments in Hangzhou, China during 1997 -2001. Seeding rate had no significant influence on both β-glucan and protein content.

文摘Probiotics which can provide potential health benefits for consumers and prevent disease, is the most important research field for functional food in the future. The survival rate at (6 h) 4 ℃/24 h (lag period) was higher than (10 h) 4 ℃/24 h (the middle of exponential growth phase), and the effect of 10℃/8 h shock treatments was best during the middle of exponential growth phase, when hot shock, The concentration of heat shock proteins produced was not obvious, and survival rate was higher at 45 ℃, 30 min than at 45 ℃, 60 min.
