Gaseous detonation propagating in a toroidal chamber was numerically studied for hydrogen/oxygen/nitrogen mixtures. The numerical method used is based on the three-dimensional Euler equations with detailed finiterate chemistry. The results show that the calculated streak picture is in qualitative agreement with the picture recorded by a high speed streak camera from published literature. The three-dimensional flow field induced by a continuously rotating detonation was visualized and distinctive features of the rotating detonations were clearly depicted. Owing to the unconfined character of detonation wavelet, a deficit of detonation parameters was observed. Due to the effects of wall geometries, the strength of the outside detonation front is stronger than that of the inside portion. The detonation thus propagates with a constant circular velocity. Numerical simulation also shows three-dimensional rotating detonation structures, which display specific feature of the detonation- shock combined wave. Discrete burning gas pockets are formed due to instability of the discontinuity. It is believed that the present study could give an insight into the interest- ing properties of the continuously rotating detonation, and is thus beneficial to the design of continuous detonation propulsion systems. 展开更多

Compared with those of the relative theoretical formula, the results are satisfactory. The model mentioned above has been applied to the water area of the Lianzhou Bay, Guangxi Province. On the basis of the analysis and comparison with the field data, it shows clearly that the model calculation results are reasonable. 展开更多

目的:探讨微重力环境对肝干细胞增殖及分化的影响。方法:从人肝组织中分离、培养肝干细胞,进行肝干细胞鉴定后,分别进行平面培养、微载体培养、模拟微重力下三维培养。利用倒置相差显微镜、扫描电镜观察不同培养条件下细胞形态学变化;... 目的:探讨微重力环境对肝干细胞增殖及分化的影响。方法:从人肝组织中分离、培养肝干细胞,进行肝干细胞鉴定后,分别进行平面培养、微载体培养、模拟微重力下三维培养。利用倒置相差显微镜、扫描电镜观察不同培养条件下细胞形态学变化;检测上清液中葡萄糖消耗量和乳酸脱氢酶含量以比较细胞增殖和活性情况;荧光定量PCR法检测细胞ALB、EpCAM、CK19、AFP、HNF6、CYP3A4、CYP3A7 mRNA表达变化。结果:模拟微重力环境下,肝干细胞以微载体球面为基底呈三维立体结构生长,形成大小不等细胞团;扫描电镜下见细胞团表面有丰富的细胞外基质及微绒毛;从9 d开始葡萄糖消耗量明显增加,至17 d达高峰,而LDH含量始终保持低水平;培养至21 d细胞仍高表达EpCAM、ALB、CK19、HNF6、CYP3A7,不表达AFP、CYP3A4,符合肝干细胞表型特征,与微载体培养结果相比有统计学差异(P<0.05)。结论:模拟微重力环境能促进肝干细胞呈三维立体结构生长,有利于细胞快速增殖并维持细胞活性和表型。 展开更多

目的:体外研究琼脂糖用于肺腺癌细胞株A549三维培养,并观察细胞在三维环境中的行为改变。方法:用琼脂糖作为三维培养基质,观察细胞在三维环境中的生长现象;黏附实验用以检测细胞对胞外基质的黏附率;MTT比色法检测细胞对DDP的药物敏感... 目的:体外研究琼脂糖用于肺腺癌细胞株A549三维培养,并观察细胞在三维环境中的行为改变。方法:用琼脂糖作为三维培养基质,观察细胞在三维环境中的生长现象;黏附实验用以检测细胞对胞外基质的黏附率;MTT比色法检测细胞对DDP的药物敏感性和细胞增殖抑制率;流式细胞术检测细胞自然凋亡情况。同时比较二维及三维培养细胞的差异。结果:在琼脂糖中,A549细胞生长呈现球形的团块状生长,细胞团块大小不一,生长状态良好。随着细胞的增长,3D培养的A549细胞对DDP的抗药性增加,药敏性降低;二维和三维培养下的A549细胞黏附率存在显著差异;3D培养下的细胞自然凋亡明显减少。结论:A549细胞在琼脂糖三维培养中,其细胞行为与二维培养时发生显著改变,二者具有显著性的差异。因此,琼脂糖可用于构建A549三维培养模型,此模型可为进一步研究提供研究基础。 展开更多

Exact solutions of three-dimensional(3D)crack problems are much less in number than those of two-dimensional ones,especially for multi-field coupling media exhibiting a certain kind of material anisotropy.An exact3Dthermoelastic solution has been reported for a uniformly heated penny-shaped crack in an infinite magnetoelectric space,with impermeable electromagnetic conditions assumed on the crack faces.Exact 3Dsolutions for the penny-shaped crack subjected to uniform or point temperature load are further presented here when the crack faces are electrically and magnetically permeable.The solutions,obtained by the potential theory method,are exact in the sense that all field variables are explicitly derived and expressed in terms of elementary functions.Along with the previously reported solution,the limits or bounds of the stress intensity factor at the crack-tip for a practical crack can be identified. 展开更多

Accuracy of Low-level Surface in Hierarchical Construction of Potential Energy Surface

The effects of low-level PES on the overall accuracy of the final surface constructed by using hierarchical construction were investigated with the constructions of a number of global surfaces for the H3 system at UHF (UMP2, DFT-B3LYP, UCCSD(T))/vtz, and UCCSD(T)/avqz levels of theory. The total reaction probabilities for the H+H2 reaction calculated on these surfaces revealed that the accuracy of UCCSD(T)/avqz surface is very close to the well-known BKMP2 surface, while the UCCSD(T)/vtz PES has a slightly higher barrier. In contrast, the low-level theories (UHF, UMP2, DFT-B3LYP) with vtz basis set can only provide a qualitative description of this simplest reaction despite the fact that they are widely used to study reactions in complex systems. On the other hand, although these theories are not accurate on describing the reaction, they can be used to provide the low- level PESs for hierarchical construction of the UCCSD(T)/avqz PES with the number of UCCSD(T)/avqz energies substantially reduced. 展开更多

目的观察三维培养条件下不同转移潜能人肝癌细胞株MHCC97-H/L形成血管生成拟态的差异,并探讨其血管生成相关机制。方法建立MHCC97.H、MHCC97-L三维培养体系,倒置显微镜下观察24h血管样结构形成差异。应用血管生成基因芯片筛选三维培... 目的观察三维培养条件下不同转移潜能人肝癌细胞株MHCC97-H/L形成血管生成拟态的差异,并探讨其血管生成相关机制。方法建立MHCC97.H、MHCC97-L三维培养体系,倒置显微镜下观察24h血管样结构形成差异。应用血管生成基因芯片筛选三维培养24h后两种细胞差异表达的基因,并用逆转录.聚合酶链反应(RT—PCR)和蛋白质印迹(Western blot)方法验证部分差异基因的表达。结果三维培养24h,MHCC97-H和MHCC97-形成的血管样结构长度分别为(474.00±16.37)、(319.67±40.50)inm/cm2,MHCC97-H形成的血管样结构长度明显较MHCC97-L长(P〈0.01)。在113个血管生成相关基因中,MHCC97-H表达较MHCC97-L上调2倍以上的有4个,包括Angiopoietin-like 4、Jagged1、Endoglin和VE—cadherin。RT-PCR和Westernblot方法验证结果与基因芯片结果一致。结论高转移潜能人肝癌细胞株MHCC97-H体外形成血管生成拟态能力明显较低转移潜能人肝癌细胞株MHCC97-L强,其原因可能与MHCC97-H高表达某些血管生成基因有关。 展开更多

目的探讨脉动式张应力环境下,在生物反应器内进行组织工程血管(TEBV)培养的可行性。方法从人脐动脉获取血管平滑肌细胞(VSMC),经体外培养扩增后接种到聚乙醇酸(PGA)支架并置于生物反应器内进行三维培养。实验组利用气动式左心... 目的探讨脉动式张应力环境下,在生物反应器内进行组织工程血管(TEBV)培养的可行性。方法从人脐动脉获取血管平滑肌细胞(VSMC),经体外培养扩增后接种到聚乙醇酸(PGA)支架并置于生物反应器内进行三维培养。实验组利用气动式左心循环辅助泵对VSMC施加模拟人体内血管环境(压力120mmHg,1mmHg=0.133kPa,频率60次/rain)的脉动式张应力作用。对照组无张力环境下进行三维培养。2周后收获TEBV分别行扫描电镜(SEM)检测、组织切片HE染色和Masson染色。结果实验组TEBV外观色泽鲜亮,具有一定厚度和弹性,能自我维持管腔形状。SEM显示实验组TEBV管壁表面光滑平整,横切面有丰富的细胞外基质(ECM),ECM分布均匀,把PGA完全包裹。HE染色管壁均匀致密,ECM及细胞的排列具有一定方向性,其中含有未降解的PGA碎片。Masson染色管壁胶原纤维丰富。对照组TEBV外观色泽暗淡,管壁薄,弹性差,从支架取下后管腔塌陷。SEM显示管壁表面欠光滑,横切面上ECM含量少,大部分PGA没有被包裹。组织切片染色管壁结构疏松,ECM和胶原纤维含量较少,层次感差。结论在生物反应器内的脉动式张应力环境下,可培养出具有良好形态结构的组织工程血管样组织。 展开更多

Application of spiral CT image 3D reconstruction in severe talar neck fracture 被引量:15

Objective: To explore the application of the spiral computerized tomography (CT) image three-dimensional ( 3D ) reconstruction technique associated with the conventional radiography in the diagnosis and treatment of severe talar neck fracture. Methods: Using the multi-slice spiral CT image 3D reconstruction technique, we analysed Ⅱ cases of talar neck fracture. The fractures were reduced and fixed through a minimal incision and internal fixation with titanium cannulated lag screws. Results. In the Ⅱ cases, the results of CT image 3D reconstruction were in concordance with plain radiograph in 6 case of Hawkins type Ⅱ. And the remaining 5 cases of Hawkins types Ⅲ and Ⅳ could not be classified exactly only by radiographs, one of whom was misdiagnosed. After using the CT image 3D reconstruction, the 5 cases were classified exactly before osteosynthesis. The classifications of these Ⅱ cases were confirmed finally by surgical findings. The duration of operation were 45-Ⅰ40 min, averaging 8Ⅰ min (including the duration of C-arm fluoroscopy). X-ray exposure time was 6-58 seconds, averaging 22 seconds. The blood loss was less than Ⅰ00 ml. The fracture union was achieved in 3 months. No nonunion, talus avascular necrosis or joint surface collapse occurred. Postoperative follow-up was from Ⅰ to 25 months. According to Hawkins score, excellent result was found in 6 type Ⅱ cases and Ⅰ type Ⅲ case; good result in I type Ⅲ case with both medial and lateral malleolar fracture, Ⅰ type Ⅲ with medial malleolus fractures and Ⅰ open type Ⅲ; fair result in Ⅰ open type Ⅳ with lateral malleolus fracture. Conclusions : By using the multl-slice spiral CT image 3D reconstruction associated with radiography to diagnose and treat severe talar neck fractures, the accuracy of diagnosis can be improved obviously. Based on this technique, more consummate operational plan can be designed and performed so as to achieve a better therapeutic effect. 展开更多
