Antibiotic-Resistant Bacterial Group in Field Soil Evaluated by a Newly Developed Method Based on Restriction Fragment Length Polymorphism Analysis 被引量:1

Spreading of antibiotic resistant bacteria into environment is becoming a major public health problem, implicating affair of the indirect transmission of antibiotic resistant bacteria to human through drinking water, or vegetables, or daily products. Until now, the risk of nosocomial infection of antibiotic resistant bacteria has mainly been evaluated using clinical isolates by phenotypic method. To evaluate a risk of community-acquired infection of antibiotic resistant bacteria, a new method has been developed based on PCR-RFLP without isolation. By comparing restriction fragment lengths of the 16S rDNA gene from bacterial mixture grown under antibiotic treatment to those simulated from the DNA sequence, bacterial taxonomies were elucidated using the method of Okuda and Watanabe [1] [2]. In this study, taxonomies of polymyxin B resistant bacteria group in field soils, paddy field with organic manure and upland field without organic manure were estimated without isolation. In the both field soils, the major bacteria grown under the antibiotic were B. cereus group, which had natural resistance to this antibiotic. In field applied with organic manure, Prevotella spp., and the other Cytophagales, which were suggested to be of feces origin and to acquire resistance to the antibiotic, were detected. When numbers of each bacterial group were roughly estimated by the most probable number method, B. cereus group was enumerated to be 3.30 × 106 MPN/g dry soil in paddy field soil and 1.32 × 106 MPN/g dry soil in upland filed. Prevotella spp. and the other Cytophagales in paddy field were enumerated to be 1.31 × 106 MPN, and 1.07 × 106 MPN·g-1 dry soil. 展开更多

Although the Japanese Standards for manure recommended that it took at least 6 months to complete the maturing phase, there was no reliable ground. In order to find out shortening method of the maturing phase, the microorganisms concerned with a progress of the maturing was determined by using the most probable number method (MPN) and PCR-RFLP of the 16S rDNA, which was found effective to provide numbers and taxonomy of polymyxin B resistant bacterial groups in the former paper [1]. Compared to the numbers after thermophilic phase, those of Actinobacteria, δ-proteobacteria, and the other gram negative bacteria increased to 50 times, 20 times, and 105 times, respectively, after maturing phase, while those of Bacillus spp., and α and β-proteobacteria decreased to 1/10, and 1/105 after maturing phase. Numbers of the other Fumicutes, and γ-proteobacteria remained in the same revel. Actinobacteria, δ-proteobacteria, and the other gram negative bacteria might be concerned with a progress of the maturing phase, because these bacterial groups were detected and enumerated due to their proliferation ability. Although number of Acitinobacteria might be underestimated because of a PCR bias, the method was found effective for the purpose to monitor bacteria actively proliferated in culture medium. 展开更多

Evaluation of the Method Based on Restriction Fragment Length Polymorphism Analysis as Simple Analysis Method of Lactic Acid Bacteria in Foods

Lactic acid bacteria have not only been used to produce various kinds of fermented food, but also used as probiotic products. As lactic acid bacterial group was consisted from diverse genera, a simple inspection method by which numbers and contained microorganisms could be automatically analyzed without any preliminary information was required to use them more effectively. In this manuscript, lactic acid bacterial groups in commercial products of kimuchi, komekouji-miso, and yoghurt were identified and enumerated by our newly developed method [1]-[3], to evaluate whether the method could be used as an inspection method of various food samples. In kimuchi, numerically dominant bacteria were Lactobacillus sakei, and L. casei (1.4 × 104 MPN g<sup>-1</sup>) and Leuconostoc spp. (l.4 × 104 MPN). In kouji-miso, numerically dominant bacteria was Bacillus spp. (3 × 103 MPN), which mainly included B. subtilis group and B. cereus group. Lactic acid bacteria such as Lactobacillus spp., or Lactococcus spp., included in the komekouji-miso, could be enumerated after 3 days incubation (1.24 × 104 MPN), but not detected after 7 days incubation. In yoghurt A and C, Lactococcus lactis was detected as numerically dominant lactic acid bacteria (3.0 × 105 MPN). In yoghurt B, Lactobacillus spp., or Lactococcus spp., was detected not only by a culturebased method but also by an unculture-based method, although there was a difference between the both estimated numbers. The present results suggested that the method might become useful as a simple inspection method of food microorganisms, because time and labor of the analysis could be reduced by using an unculture-based method and MCE-202 MultiNA. In this study, Bifidobacteriium spp. was not detected in B and C yoghurt, in spite of indicating their existence, and numbers of lactic acid bacteria were lower than the level of the daily product regulation, because 16S rDNA of Bifidobacteriium spp. might not be amplified by the used PCR condition. The PCR condition must be changed so as to amplify Bifidobacterium spp., before the method will be used as an inspection method for lactic acid bacteria. 展开更多

有限回采空间内贫矿资源无底柱分段崩落法低贫损开采方案研究

做好无底柱分段崩落法的贫损控制对于矿山经济效益及环境保护具有重要意义,但当矿床开采条件复杂、矿石品位较低时,矿石贫损控制难度将会大幅增加,在此条件下如何做好矿石贫损控制一直是崩落法开采方向的重要关注点。金川龙首矿西二采... 做好无底柱分段崩落法的贫损控制对于矿山经济效益及环境保护具有重要意义,但当矿床开采条件复杂、矿石品位较低时,矿石贫损控制难度将会大幅增加,在此条件下如何做好矿石贫损控制一直是崩落法开采方向的重要关注点。金川龙首矿西二采区无底柱分段崩落法采场仅可布置4个分段,回采空间十分有限,不利于矿石的贫损控制,加之西二采区矿石品位较低但经济价值较高,因此在开采过程中需要严格控制矿石贫损。针对西二采区的复杂开采条件及贫损控制需求,提出了一种“首采分段总量控制出矿、中间分段低贫化出矿、最末分段低贫化出矿+辅助进路回收残留矿石”的组合式低贫损回采方案。以西二采区为工程背景,设计并制作了一种高仿真度的物理放矿模型并对该方案展开了试验研究。结果表明:即便是在暂时不考虑回收最末分段残留矿石的情况下,该方案可使西二采区崩落法采场的矿石回收率达到75.6%,而废石混入率仅为6.0%,在确保矿石回收率的基础上显著降低了矿石贫化。该方案在现场应用后,实际的矿石回收指标与试验结果基本接近,表明该方案在西二采区有限回采空间内实现了贫矿资源低成本、高质量回收的总体目标。 展开更多

路径规划是煤矿机器人在煤矿井下狭小巷道空间中应用亟待解决的关键技术之一。针对传统人工势场(APF)算法在狭小巷道环境中规划出的路径可能离巷道边界过近,以及在障碍物附近易出现目标不可达和路径振荡等问题,提出了一种基于改进APF算... 路径规划是煤矿机器人在煤矿井下狭小巷道空间中应用亟待解决的关键技术之一。针对传统人工势场(APF)算法在狭小巷道环境中规划出的路径可能离巷道边界过近,以及在障碍物附近易出现目标不可达和路径振荡等问题,提出了一种基于改进APF算法的煤矿机器人路径规划方法。参考《煤矿安全规程》有关规定建立了巷道两帮边界势场,将机器人行驶路径尽量规划在巷道中间,以提高机器人行驶安全性;在障碍物斥力势场中引入调节因子,以解决目标不可达问题;引入转角限制系数以平滑规划出的路径,减少振荡,提高规划效率,保证规划路径的安全性。仿真结果表明:当目标点离障碍物很近时,改进APF算法可成功规划出能够抵达目标点的路径;改进APF算法规划周期数较传统算法平均减少了14.48%,转向角度变化累计值平均减少了87.41%,曲率绝对值之和平均减少了78.09%,表明改进APF算法规划的路径更加平滑,路径长度更短,规划效率和安全性更高。 展开更多

基于PRECEDE模式的居民减盐行为影响因素的混合方法研究

背景中国居民高血压患病率高而控制率低,大量流行病学研究表明减少膳食盐的摄入有助于降低血压水平,因此探索居民减盐行为的影响因素对于居民自身的健康管理及减轻社会负担意义重大。目的基于PRECEDE模式,探索山东居民减盐行为的倾向因... 背景中国居民高血压患病率高而控制率低,大量流行病学研究表明减少膳食盐的摄入有助于降低血压水平,因此探索居民减盐行为的影响因素对于居民自身的健康管理及减轻社会负担意义重大。目的基于PRECEDE模式,探索山东居民减盐行为的倾向因素、强化因素和促成因素,为后续减盐工作提供科学建议。方法运用混合研究方法,采用目的抽样法,先后于2016年和2018年采访居民共计114名,访谈提纲主要包含对控盐的态度、在控盐过程中遇到的困难以及获得的帮助等。定量数据来源于2016年山东省减盐防控高血压项目终末调查鲁东、鲁中南地区的问卷数据,从数据库中选取与高血压相关调查内容,共纳入研究样本6697份。结果多分类Logistic回归结果显示,在控制其他变量影响下,性别、职业、文化程度对减盐行为依从性有影响(P<0.05)。在控制个人一般情况的影响下,个人口味、减盐知识及对低盐饮食的态度对减盐行为依从性有影响(P<0.05)。医生的干预能强化居民减盐行为。通过医务人员的健康教育,不仅能增强高血压患者的减盐行为与意识,还能加强家人的督促,提升家人的减盐行为与意识,进而达到促进居民减盐的目的。家人在日常生活中的督促能强化居民控盐行为。山东省减盐行动制定了项目推进政策、餐饮业政策、食品业政策以及减盐健康教育政策,打造了浓厚的减盐大环境。低盐产品中对居民影响较大的是限盐勺,低盐食品及低钠盐的可及性相对较低影响了其控盐作用。大众媒体宣传是居民获得减盐信息的主要途径,目前作用较为明显的大众传播方式包括电视和在公共场所的宣传。结论对低盐饮食的态度及知识水平因素是影响居民减盐行为的倾向因素;医生的劝诫和家人的提醒能帮助居民开始减盐,也能强化居民减盐行为,属于强化因素;减盐相关政策、大众传播为居民减盐创造了社会氛围,减盐相关产品则提供了现实条件,属于促成因素。 展开更多

长期利尿剂与限钠法在治疗慢性肾病高血压患者中的应用

目的 探讨长期利尿剂与限钠法在治疗慢性肾病(CKD)高血压患者的应用效果。方法 选取2019年9月至2021年3月我院肾内科收治的26例CKD高血压患者,所有患者均接受阿米洛利/氢氯噻嗪(每天5 mg/50 mg)和膳食钠(60 mmol/d)限制治疗。分析两种... 目的 探讨长期利尿剂与限钠法在治疗慢性肾病(CKD)高血压患者的应用效果。方法 选取2019年9月至2021年3月我院肾内科收治的26例CKD高血压患者,所有患者均接受阿米洛利/氢氯噻嗪(每天5 mg/50 mg)和膳食钠(60 mmol/d)限制治疗。分析两种干预措施对血压、肾功能和体液平衡的影响,将其与利尿剂的肾脏清除率相关联。结果 饮食钠限制将钠排泄从每天160 mmol/L减少到64 mmol/L;与钠限制(SBP从134 mmHg到129 mmHg)相比,利尿剂使24 h平均收缩压(SBP从138到124 mmHg)产生更大的降低作用,并且对细胞外水、肾小球滤过率(eGFR)、血浆肾素和醛固酮有显著影响;两种干预措施都导致体质量和氨基末端脑钠肽前体下降,没有显著降低白蛋白尿,而利尿剂却显著降低了尿血管紧张素原和β2-微球蛋白的排泄;尽管较低的eGFR和较高的血浆硫酸吲哚酚与较低的利尿剂清除率相关,但在较低的eGFR下仍维持了对体质量和血压的利尿作用。在利尿剂治疗期间,较高的前列腺素E2排泄与较低的游离水清除率相关,4例患者出现轻度低钠血症。结论 长期利尿剂在降低CKD的血压和细胞外容量方面不劣于饮食钠盐限制。尽管利尿剂清除率较低,但CKD患者仍能维持对利尿剂的敏感性。 展开更多

This paper proposes a new power grid investment prediction model based on the deep restricted Boltzmann machine(DRBM)optimized by the Lion algorithm(LA).Firstly,two factors including transmission and distribution price reform(TDPR)and 5G station construction were comprehensively incorporated into the consideration of influencing factors,and the fuzzy threshold method was used to screen out critical influencing factors.Then,the LA was used to optimize the parameters of the DRBM model to improve the model’s prediction accuracy,and the model was trained with the selected influencing factors and investment.Finally,the LA-DRBM model was used to predict the investment of a power grid enterprise,and the final prediction result was obtained by modifying the initial result with the modifying factors.The LA-DRBMmodel compensates for the deficiency of the singlemodel,and greatly improves the investment prediction accuracy of the power grid.In this study,a power grid enterprise was taken as an example to carry out an empirical analysis to prove the validity of the model,and a comparison with the RBM,support vector machine(SVM),back propagation neural network(BPNN),and regression model was conducted to verify the superiority of the model.The conclusion indicates that the proposed model has a strong generalization ability and good robustness,is able to abstract the combination of low-level features into high-level features,and can improve the efficiency of the model’s calculations for investment prediction of power grid enterprises. 展开更多

The separated penalty parameters method and the optimal hybrid element model with penalty balance were also presented. The penalty balance method can effectively refrain the parasitical stress on the premise of no additional degrees of freedom. The numeric experiment shows that the presented element not only is effective in improving greatly the numeric calculation precision of distorted grids but also has the universality. 展开更多

The advantage of solar sails in deep space exploration is that no fuel consumption is required. The heliocentric distance is one factor influencing the solar radiation pressure force exerted on solar sails. In addition, the solar radiation pressure force is also related to the solar sail orientation with respect to the sunlight direction. For an ideal flat solar sail, the cone angle between the sail normal and the sunlight direction determines the magnitude and direction of solar radiation pressure force. In general, the cone angle can change from 0° to 90°. However, in practical applications, a large cone angle may reduce the efficiency of solar radiation pressure force and there is a strict requirement on the attitude control. Usually, the cone angle range is restricted less more than an acute angle (for example, not more than 40°) in engineering practice. In this paper, the time-optimal transfer trajectory is designed over a restricted range of the cone angle, and an indirect method is used to solve the two point boundary value problem associated to the optimal control problem. Relevant numerical examples are provided to compare with the case of an unrestricted case, and the effects of different maximum restricted cone angles are discussed. The results indicate that (1) for the condition of a restricted cone-angle range the transfer time is longer than that for the unrestricted case and (2) the optimal transfer time increases as the maximum restricted cone angle decreases. 展开更多

The present manuscript examines the circular restricted gravitational three-body problem (CRGTBP) by the introduction of a new approach through the power series method. In addition, certain computational algorithms with the aid of Mathematica software are specifically designed for the problem. The algorithms or rather mathematical modules are established to determine the velocity and position of the third body’s motion. In fact, the modules led to accurate results and thus proved the new approach to be efficient. 展开更多

In addition to the SITE roads METEOROLOGICAL conditions are an important part of AUTOMOTIVE ROAD testing and there are also clear requirements in the GB/T12534-1990 General Rules for AUTOMOTIVE ROAD Test methods. For example the test should be conducted in rain and fog free weather with a relative humidity of less than 95% a temperature of 0-40 and a wind SPEED of no more than 3m/s. At the same time METEOROLOGICAL conditions also serve as an important basis for ROAD control in the test site. Real time information such as wind SPEED rainfall and visibility provides necessary reference for SITE managers to issue notices on SPEED limits SITE closures and traffic restrictions directly affecting the timeliness of ROAD testing safety control. Therefore this article conducts research on the application of METEOROLOGICAL observation SITES in AUTOMOTIVE testing SITES from the construction of METEOROLOGICAL observation SITES METEOROLOGICAL services and abnormal weather ROAD control.

This paper deals with generation of halo orbits in the three-dimensional photogravitational restricted three-body problem, where the more massive primary is considered as the source of radiation and the smaller primary is an oblate spheroid with its equatorial plane coincident with the plane of motion. Both the terms due to oblateness of the smaller primary are considered. Numerical as well as analytical solutions are obtained around the Lagrangian point L1, which lies between the primaries, of the Sun-Earth system. A comparison with the real time flight data of SOHO mission is made. Inclusion of oblateness of the smaller primary can improve the accuracy. Due to the effect of radiation pressure and oblateness, the size and the orbital period of the halo orbit around L1 are found to increase. 展开更多

Identification and Enumeration Method of Both Eukaryotic and Prokaryotic Microorganisms in Food Sample

The method to analyze both eukaryotic and prokaryotic microorganisms without preliminary microbial information of sample seemed to be useful not only for research and investigation of microorganisms but also for industry using microorganisms. In the present manuscript, preparation of a new DNA primers, new reference database for 18S rDNA for our newly developed method [1]- [3], and analyses of eukaryotic and prokaryotic microorganisms in fermentation products were presented. In komekouji, Aspergillus spp., was enumerated to be 46.5 × 106 MPN g<sup>-1</sup>, and Penicillium spp., was enumerated to be 1.5 × 106 MPN g<sup>-1</sup>. In dry yeast, Saccharomyces group, were enumerated to be 8600 × 106 MPN g<sup>-1</sup>. In komekouji-miso, no eukaryotic microorganism was detected, while the other Bacillus spp., was numerically dominant (21.5 × 106 MPN g<sup>-1</sup>) as prokaryotic microorganisms, followed by B. subtilis group (4.65 × 106 MPN g<sup>-1</sup>), and the other Firmicutes (3.7 × 106 MPN g<sup>-1</sup>). The komekouji-miso included lower number of Actinobacteria (0.15 × 106 MPN g<sup>-1</sup>), Burkhokderia sp. (1.5 × 106 MPN g<sup>-1</sup>), and the other α,β,γ-proteobacteria (0.12 × 106 MPN g<sup>-1</sup>). In sake-kasu, both prokaryote and eukaryote were not detected by the method. Present results indicated that using both universal primers for eukaryotic and prokaryotic microorganisms, each groups of prokaryotic and eukaryotic microorganisms were enumerated without any preliminary information nor setting up standard curve, which were required for real time PCR. 展开更多
