With the expansion of herbal medicine (HM) market, the issue on how to apply up-to- date analytical tools on qualitative analysis of HMs to assure their quality, safety and efficacy has been arousing great attention. Due to its inherent characteristics of accurate mass measurements and multiple stages analysis, the integrated strategy of liquid chromatography (LC) coupled with time-of-flight mass spectrometry (TOF-MS) and ion trap mass spectrometry (IT-MS) is well-suited to be performed as qualitative analysis tool in this field. The purpose of this review is to provide an overview on the potential of this integrated strategy, including the review of general features of LC-IT-MS and LC-TOF-MS, the advantages of their combination, the common procedures for structure elucidation, the potential of LC-hybrid-IT-TOF/MS and also the summary and discussion of the applications of the integrated strategy for HM qualitative analysis (2006-2011). The advantages and future developments of LC coupled with IT and TOF-MS are highlighted. 展开更多

建立了超高效液相色谱-四极杆飞行时间质谱法(UPLC-Q-TOF-MS)对小鼠大脑中的多巴胺(DA)、5-羟色氨(5-HT)、二羟基苯乙酸(DOPAC)和高香草酸(HVA)4种单胺类神经递质的含量进行同时测定。样品以30%乙醇溶液(甲酸调至p H 5.0)... 建立了超高效液相色谱-四极杆飞行时间质谱法(UPLC-Q-TOF-MS)对小鼠大脑中的多巴胺(DA)、5-羟色氨(5-HT)、二羟基苯乙酸(DOPAC)和高香草酸(HVA)4种单胺类神经递质的含量进行同时测定。样品以30%乙醇溶液(甲酸调至p H 5.0)匀浆处理后,冷冻离心,过滤。在优化色谱-质谱条件下,采用ESI和APCI复合源,正/负离子扫描方式进行数据采集。4种单胺类神经递质在0.5-1 000μg/L范围内呈良好线性关系,相关系数(r)大于0.999 3,方法的检出限为0.5-5.0μg/L,平均回收率为93.3%-101.8%,相对标准偏差为1.1%-3.3%。该方法具有操作简便、高效快速、回收率良好、灵敏度高、重现性好和准确度高等优点,可用于小鼠脑组织内4种单胺类神经递质的含量测定。 展开更多

To increase spatial resolution and signal-to-noise ratio in PET imaging,we present in this paper the design and performance evaluation of a PET detector module combining both depth-of-interaction(DOI) and time-offlight(TOF) capabilities.The detector module consists of a staggered dual-layer LYSO block with2 mm × 2 mm × 7 mm crystals.MR-compatible SiPM sensors(MicroFJ-30035-TSV,SensL) are assembled into an 8× 8 array.SiPM signals from both fast and slow outputs are read out by a 128-channel ASIC chip.To test its performance,a flood histogram is acquired with a ^(22)Na point source on top of the detector,and the energy resolution and the coincidence resolving time(CRT) value for each individual crystal are measured.The flood histogram shows excellent crystal separation in both layers.The average energy resolution at 511 keV is 14.0 and 12.7%at the bottom and top layers,respectively.The average CRT of a single crystal is 635 and 565 ps at the bottom and top layers,respectively.In conclusion,the compact DOI-TOF PET detector module is of excellent crystal identification capability,good energy resolution and reasonable time resolution and has promising application prospective in clinical TOF PET,PET/MRI,and brain PET systems. 展开更多

Detecting feature points on the human body in video frames is a key step for tracking human movements. There have been methods developed that leverage models of human pose and classification of pixels of the body image. Yet, occlusion and robustness are still open challenges. In this paper, we present an automatic, model-free feature point detection and action tracking method using a time-of-flight camera. Our method automatically detects feature points for movement abstraction. To overcome errors caused by miss-detection and occlusion, a refinement method is devised that uses the trajectory of the feature points to correct the erroneous detections. Experiments were conducted using videos acquired with a Microsoft Kinect camera and a publicly available video set and comparisons were conducted with the state-of-the-art methods. The results demonstrated that our proposed method delivered improved and reliable performance with an average accuracy in the range of 90 %.The trajectorybased refinement also demonstrated satisfactory effectiveness that recovers the detection with a success rate of 93.7 %. Two variants of application of a transaxial mirror with stigmatic spatial time-of-flight focusing in the time-of-flight mass spectrometer have been considered. In the first variant, the transaxial mirror is used as an ion reflector in the ordinary scheme of the time-of-flight mass reflectron. In the second variant, the transaxial mirror simultaneously fulfills the function of the ion reflector and corrector of aberrations caused by the energy spread of ions in the package, formed by the ion source of the mass reflectron. The expressions defining the conditions of stigmatic spatial time-of-flight focusing in the transaxial mirror and the combined system consisting of an ion source and a mirror have been derived. The relationships between geometrical and electrical parameters of three- and four-electrode transaxial mirrors realizing these conditions have been obtained by numerical calculations. 展开更多

建立了一种气相色谱(GC)与单光子电离-飞行时间质谱(SPI-TOF MS)联用(GC/SPI-TOF MS)的分析方法。首先,设计了一种双层套管的传输管用于连接GC与SPI-TOF MS,实现了GC与单光子电离离子源的无缝连接。在此基础上,以n-十五烷标准品和苯/甲... 建立了一种气相色谱(GC)与单光子电离-飞行时间质谱(SPI-TOF MS)联用(GC/SPI-TOF MS)的分析方法。首先,设计了一种双层套管的传输管用于连接GC与SPI-TOF MS,实现了GC与单光子电离离子源的无缝连接。在此基础上,以n-十五烷标准品和苯/甲苯/二甲苯的标准气为对象,对电离源的重要电压参数进行了优化,得到了纯净的分子离子峰,实现了对各类有机物的快速和准确定性。最后,将该方法用于分析柴油中的挥发性与半挥发性有机物,获得了柴油组分的二维GC×SPI-TOF MS谱图。不需要复杂的谱图解析和数据处理,根据谱图中离子的质荷比(m/z)归纳了柴油的主要成分,包括脂肪烃、芳香烃和含量很低的苯并吡咯等含氮化合物;根据色谱的保留时间将柴油中的同分异构体区分开来。结果表明GC/SPI-TOF MS法是一种简单、有效的分析方法,非常适于柴油及复杂环境样品等的分析表征。 展开更多

乳腺癌组织N-连接糖链的分析

目的: 解析乳腺癌组织的特异性糖链结构,为乳腺癌的早期诊断提供可能标志物。方法:收集2014年5月至2014年9月在陕西省肿瘤医院乳腺科手术切除的乳腺癌患者组织标本15例及距癌组织边缘3-5 cm的癌旁组织标本5例,另选取乳腺小叶增生组... 目的: 解析乳腺癌组织的特异性糖链结构,为乳腺癌的早期诊断提供可能标志物。方法:收集2014年5月至2014年9月在陕西省肿瘤医院乳腺科手术切除的乳腺癌患者组织标本15例及距癌组织边缘3-5 cm的癌旁组织标本5例,另选取乳腺小叶增生组织标本4例,提取组织蛋白质,利用滤膜辅助的糖蛋白糖链释放方法获得N-连接糖链,应用基质辅助激光解析电离飞行时间质谱(MALDITOF/TOFMS)技术分析乳腺癌组织、癌旁组织和正常乳腺组织中糖蛋白N连接糖链类型分布和含量变化情况。结果:乳腺癌组织、癌旁组织和正常乳腺组织中分别鉴定到38、34和22个糖链。与癌旁组织及正常乳腺组织相比,癌症组织中复杂型、高甘露糖型、平分型、岩藻糖化和二天线型糖链的含量除高甘露糖型明显升高外,其他均显著降低(P〈0.05或P〈0.01)。乳腺癌组织中相对分子质量分别为1 580.455和1 742.490的2种高甘露糖型N糖链的含量显著高于正常乳腺组织(P〈0.05或P〈0.01)、相对分子质量分别为1 647.473和1 850.583的2种复杂型N-糖链的含量则显著低于正常乳腺组织(P〈0.05或P〈0.01)。这4种糖链经ROC曲线分析显示具有较高检测准确性(P〈0.05或P〈0.01)。结论:发现的4种乳腺癌组织特异性的N糖链有成为早期诊断和靶向治疗标志物的潜力。 展开更多

An ionic liquid(IL)electrospray thruster was developed for application in micro-nano satellites or gravitational wave detectors.The thruster employed a porous ceramic emitter with seven emitter strips located on its emission surface.Without any liquid-supply device,IL was delivered through porous media to emitter strips via capillary effect.Multiple emission sites then formed at the tip of each strip.A charged beam of up to 350μA(with a current density of 540μA cm^(-2))was stably produced in the negative mode.However,in the positive mode,a corona was observed which could prevent the thruster from emitting larger current.A time-of-flight mass spectrometer with significantly improved signal-to-noise ratio was built,which was used to obtain the mass distribution of the beam of the thruster.A retarding potential analysis was also performed.The test results showed that the thruster worked in the pure-ion regime,and delivered a maximum thrust of 67.1μN with specific impulses of 3952 s and 3117 s in the positive and negative modes,respectively. 展开更多

AIM: To investigate the relationship between urinary peptide changes and Helicobacter pylori (H. pylorl) infection using urinary peptidome profiling. METHODS: The study was performed in volunteers (n = 137) who gave informed consent. Urinary peptides were enriched by magnetic beads based weak cation exchange chromatography'and spectrums acquired by matrix-assisted laser desorption/ionization time-of-flight (MALDI-TOF) mass spectrometry (MS). ClinProTools bioinformatics software was used for statistical analysis and the recognition of peptide patterns. The marker peptides were identified by LTQ Obitrap XL tandem MS. RESULTS: Approximately 50 proteins or peptides which loaded onto the magnetic beads were detected by MAL-DI-TOF MS. By optimizing the parameters of the model, the Genetic Algorithm model had good recognition capability (97%) and positive predictive value (94%). Based on the model, 2 markers with molecular masses of 6788 and 1912 Da were found that differentiated between H. pylori positive and negative volunteers. The m/z 1912 sequence was parsed as SKQFTSSTSYN- RGDSTF. The peptide was identified as isoform 1 of the fibrinogen a chain precursor, whose concentration in urine was markedly higher in H. pylori infected volunteers than in H. pylori non-infected ones. CONCLUSION: The appearance of urinary fibrinogen degradation products is caused by an active H. pyloriinduced process. 展开更多

Genetic algorithm and kernel partial least square (GA-KPLS) and Levenberg- Marquardt artificial neural network (L-M ANN) techniques were used to investigate the correlation between retention time (tR) and descriptors for 150 organic contaminants in natural water and wastewater, which are obtained by gas chromatography coupled to high-resolution time-of-flight mass spectrometry (GC-TOF MS). The L-M ANN model gave a significantly better performance than the GA-KPLS model. This indicates that L-M ANN can be used as an alternative modeling toot for quantitative structure-retention relationship (QSRR) studies. 展开更多

In this paper, the theory of CFD method was analyzed in reality condition. The design, simulation and printed-circuit-board (PCB) performance of CFD circuit were shown. Finally, an over amplified method was introduced, by which the influence of direct-current (DC) bias error could be reduced. The experimental results showed that timing discriminator could set the timing point to a certain point on echo pulse, which did not depend on the amplitude of echo pulse. 展开更多

In this review we introduce three main models for tran- sient imaging with a time-of-flight (ToF) camera: correlation model, frequency-domain model, and compressive sensing model. Transient imaging applications usually involve resolving the problem of light transport and separating the light rays arriving along different paths. We discuss two of the applications: imaging objects inside scattering media and recovering both the shape and texture of an object around a corner. 展开更多

The double-threshold method has been widely used in ultrasonic flow measurement to determine time-of-flight(TOF)due to its low cost and ease of implementation.Performance of this method is negatively affected by the cycle-skip phenomenon which occurs frequently under inconstant working conditions,especially varied fluid temperature.This paper proposes a method to suppress the phenomenon to facilitate reliable determination of TOF in ultrasonic flow measurement.First,the double-threshold method is used to generate a feature point to segment the signal.Second,based on the correlation coefficient and signal power,judgement factors of individual signal periods are calculated to determine signal onset.Finally,a valid zero crossing which has a constant lag from the onset is selected to determine the TOF.Thus,the cycle-skip phenomenon is suppressed.Two additional modifications are proposed to eliminate the influence of varied signal frequency and low sampling rate.The proposed method was validated by an experiment based on an ultrasonic water flow sensor.Results showed that the frequently appearing cycle-skip phenomenon can be successfully suppressed by the proposed method. 展开更多

The photoneutron source (PNS, phase 1), an electron linear accelerator (linac)-based pulsed neutron facility that uses the time-of-flight (TOF) technique, was constructed for the acquisition of nuclear data from the Thorium Molten Salt Reactor (TMSR) at the Shanghai Institute of Applied Physics (SINAP). The neutron detector signal used for TOF calculation, with information on the pulse arrival time, pulse shape, and pulse height, was recorded by using a waveform digitizer (WFD). By using the pulse height and pulse-shape discrimination (PSD) analysis to identify neutrons and "y-rays, the neutron TOF spectrum was obtained by employing a simple electronic design, and a new WFD-based DAQ system was developed and tested in this commissioning experiment. The DAQ system developed is characterized by a very high efficiency with respect to millisecond neutron TOF spectroscopy. 展开更多

目的探索电针对胃黏膜损伤大鼠胃黏膜细胞损伤修复的作用机制。方法将大鼠随机分为正常组、模型组、针刺治疗组,每组10只,应用乙醇灌胃法造成胃黏膜损伤大鼠模型,用电针对胃黏膜损伤大鼠治疗10天,采用链霉蛋白酶消化法分离胃黏膜细胞,... 目的探索电针对胃黏膜损伤大鼠胃黏膜细胞损伤修复的作用机制。方法将大鼠随机分为正常组、模型组、针刺治疗组,每组10只,应用乙醇灌胃法造成胃黏膜损伤大鼠模型,用电针对胃黏膜损伤大鼠治疗10天,采用链霉蛋白酶消化法分离胃黏膜细胞,用表面加强激光解析电离飞行时间质谱技术(SELDI-TOF-MS)及WCX2蛋白芯片获得各组大鼠胃黏膜细胞的蛋白质指纹图谱,对比分析各组的差异蛋白质质/荷比峰。结果模型组大鼠胃黏膜细胞蛋白质SELDI-TOF-MS蛋白指纹图谱与正常组比较有4个蛋白质质/荷比峰差异有统计学意义(P<0.05或P<0.01),其中两个蛋白质质/荷比峰明显升高(P<0.05或P<0.01),两个蛋白质质/荷比峰明显降低(P<0.05);针刺治疗组与正常组比较共有4个蛋白质质/荷比峰差异有统计学意义(P<0.05或P<0.01),其中1个蛋白质质/荷比峰明显升高(P<0.05),3个蛋白质质/荷比峰明显降低(P<0.05或P<0.01);针刺治疗组与正常组比较亦有4个蛋白质质/荷比峰差异有统计学意义(P<0.05或P<0.01),且4个蛋白质质/荷比均为降低(P<0.05或P<0.01)。结论电针对胃黏膜损伤的修复可能不是单纯针地对某一个或两个蛋白的调节发挥治疗作用的,而可能是多个蛋白质参与的复杂的链环反应。 展开更多
