In order to determine the best loading density of tobacco leaf with cage clamp in bulk-curing barns, a curing test was conducted on middle-part tobacco leaves. The results showed that for good appearance quality of cured tobacco, the loading density at 59.52 kg/m2 (4 000 kg/barn) was the best, fol owed by 66.96 kg/m2 (4 500 kg/barn) and 74.40 kg/m2 (5 000 kg/barn). As leaf loading density increased, the contents of leaf nicotine and total nitrogen increased, sugar content had smal changes, the ratio of sugar to nicotine decreased and tended to be balanced and the content of neutral aroma substances was decreasing, but the difference among treatments was not significant. With the increase of loading density, the sensory quality of cured leaf was better. When leaf loading density was 66.96 kg/m2 (4 500 kg/barn), apperance quality of cured leaf improved considerably, with prominent aro-ma. Therefore, the best loading density of tobacco leaf in cage clamp of bulk-curing barn would be 66.96 kg/m2 (4 500 kg/barn) . 展开更多

The appearance quality, physical properties, chemical components and sensory quality of 69 flue-cured tobacco leaf samples from 6 counties of Fuzhou City in 2013 were analyzed and evaluated, and were compared with those in southeast area of China. The results showed that the flue-cured tobacco leaves in Fuzhou in 2013 had better hygroscopicity, toughness and filling ability, as well as suitable dragging force. However, some of the tobacco leaves were slightly thin, and had relatively high stem ratio. The tobacco leaves in Fuzhou had higher potassium content, lower starch content, and higher potassium to chlorine ratio. The middle tobacco leaves had better coordination of chemical components and general sensory quality(medium to medium-super level). Most upper and middle flue-cured tobacco leaf samples had medium-super aroma quality, relatively sufficient aroma quantity, and relatively concentrated smoke; while the lower flue-cured tobacco leaves had little aroma quantity and insufficient smoke. The appearance quality score of lower tobacco leaf samples in Fuzhou in 2013 was slightly lower than that in southeastand whole China. The scores of maturity and leaf structure of middle tobacco leaves in Fuzhou were equivalent to those in southeast China and the national averages. The upper leaves in Fuzhou obtained an equivalent score in oil content and slightly lower scores in other indexes as compared with those in southeast China. The middle-lower tobacco leaves in Fuzhou had better hygroscopicity and toughness compared with those in southeastand whole China. The total alkaloid content per plant andreducing sugar content and potassium content in the middle and lower parts were higher than those in southeast and whole China. However, the scores of coordination of chemical components of lower and upper tobacco leaves in Fuzhou were slightly lower than those in southeast and whole China. The sensory quality of the 3 parts of tobacco leaves in Fuzhou were similar to that in southeast and whole China, but the score of consistency of upper tobacco leaves in Fuzhou was slightly higher. 展开更多

[Objective] This study was conducted to compare the aroma quality of up-per leaves of K326 flue-cured in different barns, and provide theoretical references for the development and optimization of bulk curing barn and the supporting tech-niques. [Method] The upper leaves of K326 were flue-cured in a commonness barn, a four-shelf bulk curing barn and a three-shelf bulk curing barn separately, and then the contents of aroma components and taste quality indices of the finished leaves were measured and compared. [Result] The contents of total aroma components, chromoplast pigment degradation products, phenylalanine aroma components and neophytadiene showed no significant differences in the leaves flue-cured in four-shelf bulk curing barn and commonness barn, but were significantly or very significantly higher than those in the leaves flue-cured in three-shelf bulk curing barn. The con-tents of carotenoid degradation products, Mail ard reaction products, cembranes aro-ma components and total aroma components (except neophytadiene) showed no significant difference among the three treatments. The results of sensory evaluation revealed that the sensory taste of the leaves flue-cured in four-shelf bulk curing barn was the best, as the finished leaves had rich, delicate aroma, comfortable and clean aftertaste. The leaves flue-cured in commonness barn was better than those flue-cured in three-shelf bulk curing barn in aroma quality, amount and aftertaste. [Conclusion] The four-shelf bulk curing barn had a large volume for tobacco leaf processing, but a similar baking environment to the commonness barn, which is conducive to the formation and accumulation of aroma components, so the finished leaves had similar appearance and quality with those flue-cured in commonness barn. The upper flue-cured leaves in three-shelf bulk curing bran had poorer quality than those flue-cured in the other two barns. 展开更多

In order to provide a more suitable environment for tobacco leaf growth, this study collected meteorological information and tobacco leaf quality information from the main tobacco-producing counties in the province and conducted relevant analysis to find out the main meteorological factors affecting the growth, development, yield, and quality of upper tobacco leaves. Clarify the suitable climatic conditions for the growth and development of the upper leaves, and clarify the influence of various meteorological factors on the quality of the upper leaves. The results showed that the nicotine and total nitrogen content of the upper leaves were significantly positively correlated with the average daily temperature during the maturity period, and negatively correlated with the rainfall during the maturity period;the rainfall in July was significantly positively correlated with the upper leaf clutter score, which was sufficient in the mature period. Moisture conditions are conducive to alleviating the upper leaf clutter;the number of sunshine hours in August is the main factor affecting the content of aroma substances in the upper leaves, and it is also significantly positively correlated with the content of aroma substances. 展开更多

Nitrogen(N)supply is the most important factor affecting yield and quality of flue-cured tobacco(FCT).A field experiment and an in situ incubation method were used to study the effects of soil N mineralization in the later stages of growth on yield and nicotine content of FCT in Fenggang and Jinsha,Guizhou Province. The yield and market value of FCT at Fenggang were much lower than those at Jinsha. However, the nicotine content of middle and upper leaves was much higher at Fenggang than at Jinsha when the same rate of fertilizer N was applied, which might be due to a higher N supply capacity at the Fenggang site.At later stages of growth(7-16 weeks after transplanting),the soil net N mineralization at Fenggang(56 kg N ha^(-1))was almost double that at Jinsha(30 kg N ha^(-1)).While soil NH_4-N and NO_3-N were almost exhausted by the plants or leached 5 weeks after transplanting, the N taken up at the later growth stages at Fenggang were mainly derived from soil N mineralization, which contributed to a high nicotine content in the upper leaves. The order of soil N contribution to N buildup in different leaves was:upper leaves>middle leaves>lower leaves. Thus, soil N mineralization at late growth stages was an important factor affecting N accumulation and therefore the nicotine content in the upper leaves. 展开更多

Quinolinate phosphoribosyltransferase(QPRTase),a key enzyme in ensuring nicotinic acid is available for the synthesis of defensive pyridine alkaloids in Nicotiana species, also plays an important role in nicotinamide adenine dinucleotide(NAD)biosynthesis. In this study, the morphological traits, the quality characteristics and photosynthetic parameters in QPT-overexpressing/interfering tobacco plants were investigated, respectively. Results showed that the interference of QPT gene not only reduced significantly the morphological traits including plant height, stem girth, leaf number and leaf length, etc.at 20 days after transplanting(DAT),but the flowering period was delayed 10-15 d in interfered tobaccos compared with the overexpressed, control and wild-type counterparts. However,at 40 DAT and 60 DAT,only three indexes(plant height, stem girth and leaf number)in QPT-interfering plants appeared significant difference in comparison with other three types of tobacco lines.Meanwhile, the determination results from nicotine, sugar,K+and Cl-content showed the nicotine content in interfered plants was always significantly lower than that in overexpressed plants, control and the wild-type ones respectively whatever toppling or not.At the same time, the toppling treatment also caused the increasement of K+content among the four different tobacco lines, but the maximum increase amplitude of K+content was found in QPT-overexpressing tobaccos while the minimum appeared in QPT-interfering plants.Finally,QPT-interference in transgenic tobaccos likewise affected the photosynthesis by reducing net photosynthetic rate(Pn),stomata conductance(Gs)and transpiration rate(Tr),while there was no significant difference between QPT-overexpressing plants and the controls and the wild-types. 展开更多
