Lack of Tocopherol Inhibits Rice Growth by Triggering an Ectopic Stress Response and the Accumulation of DELLA Protein

Although tocopherols are essential for rice development,the molecular details by which their absence affects development remain to be determined.To study how tocopherols function during rice development,we performed a transcriptome deep sequencing(RNA-seq)analysis of the rice cultivar Nipponbare(Nip)and the tocopherol-deficient mutant small grain and dwarf 1-2(sgd1-2).We identified 563 differentially expressed genes that were enriched in Gene Ontology categories associated with metabolism,stress,cellular responses,and transcriptional regulation.We determined that the total fatty acid composition of Nip and sgd1-2 was comparable,although cell membrane penetrability in sgd1-2 was significantly higher than in Nip under optimal growth conditions,indicating that tocopherol deficiency induces cell membrane damage.The expression levels of dehydration-responsive element binding 1(DREB1)genes and free proline content in sgd1-2 were also higher than those in Nip.We also showed that the DELLA protein SLENDER RICE1(SLR1)accumulated in sgd1-2,resulting in significant changes in the global transcriptome.Our study confirms that the lack of tocopherol accumulation in rice induced ectopic stress responses and limited growth by enhancing SLR1 abundance through increasing SLR1 transcript levels.These results provide new insights into tocopherol during rice development. 展开更多

美国红仁核桃及其油脂营养品质评价

为了系统评价美国红仁核桃及其油脂的营养品质及特性,以传统栽培品种“西洛3号”作为对照,分别测定两种核桃的主要营养成分、氨基酸和矿质元素的组成和含量,分析核桃油的脂肪酸含量和组成,比较脂质伴随物VE、植物甾醇和多酚含量,测定油... 为了系统评价美国红仁核桃及其油脂的营养品质及特性,以传统栽培品种“西洛3号”作为对照,分别测定两种核桃的主要营养成分、氨基酸和矿质元素的组成和含量,分析核桃油的脂肪酸含量和组成,比较脂质伴随物VE、植物甾醇和多酚含量,测定油脂质量指标。结果表明:美国红仁核桃的粗脂肪质量分数为(50.40±0.72)%和粗蛋白质量分数(19.60±0.56)%,均显著高于西洛核桃(P<0.05),前者谷氨酸、精氨酸和天冬氨酸含量也显著高于后者,分别为(166.24±0.26)、(94.10±2.91)、(86.26±1.94)mg/g,2种核桃矿质元素含量相近;美国红仁核桃内种皮总酚[(727.08±0.24)mg/g]、总黄酮[(57.67±0.02)mg/g]和花色苷[(1.21±0.55)mg/g]含量均显著高于西洛核桃种皮(P<0.05);美国红仁核桃油中亚油酸含量[(67.78±0.01)%]显著高于西洛核桃油,前者多不饱和脂肪酸质量分数[(76.74±0.20)%]也显著高于后者(P<0.05),但前者的总VE质量分数[(169.88±0.12)mg/kg]、总植物甾醇质量分数[(2095.06±2.26)mg/kg]和多酚质量分数[(30.93±0.01)mg/g]都略低于西洛核桃油。红仁核桃油皂化值(171.78 mg KOH/g)、过氧化值(3.94 mmol/kg)、酸价(0.91mg/g)、茴香胺值(1.95)和羰基价(0.83 mmol/kg)均比西洛核桃油的低。美国红仁核桃富含脂肪和蛋白质,种皮颜色鲜红,富含多酚、黄酮和花色苷类物质,其亚油酸含量高于其他常见核桃品种,ω-6和ω-3系脂肪酸的比值更为合理,为核桃品种改良提供了良好的种质资源。 展开更多

复配抗氧化剂在牡丹籽油乳液中的抗氧化作用研究

滩羊肉TEAC含量的高光谱快速检测技术

生育酚当量抗氧化能力(TEAC)是评估肌肉内源抗氧化程度的指标之一,可用于评估亲水化合物的抗氧化活性及清除自由基的能力。为探究快速检测滩羊肉中TEAC的可行性,采用可见近红外(Vis/NIR)高光谱成像技术,建立基于光谱信息融合图像纹理特... 生育酚当量抗氧化能力(TEAC)是评估肌肉内源抗氧化程度的指标之一,可用于评估亲水化合物的抗氧化活性及清除自由基的能力。为探究快速检测滩羊肉中TEAC的可行性,采用可见近红外(Vis/NIR)高光谱成像技术,建立基于光谱信息融合图像纹理特征(TFS)的TEAC定量预测模型。将不同部位样本集根据3∶1的比例随机划分成校正集和预测集,在400~1000 nm范围内采集反射光谱图像,提取每个样本图像的感兴趣区域(ROI)以获取原始光谱数据;采用中值滤波(MF)、基线校准(Baseline)、卷积平滑(S-G)和多元散射校正(MSC)四种算法对原始光谱中散射及干扰信息进行校正,并建立偏最小二乘回归(PLSR)模型,将光谱数据与TEAC值进行关联。采用间隔随机蛙跳(IRF)、变量组合集群分析(VCPA)、竞争性自适应加权抽样(CARS)和迭代变量子集优化(IVSO)算法提取TEAC浓度的特征波长。采用灰度共生矩阵(GLCM)算法对肉样的主要TFS依次进行提取。基于特征光谱及图谱融合信息建立反向传播人工神经网络(BP-ANN)和最小二乘支持向量机(LSSVM)模型,对滩羊肉中TEAC含量预测并进行对比分析。结果表明,(1)最优预处理为Baseline建立的PLSR模型,其Rc为0.9121,RMSEC为0.9635,Rp为0.8683,RMSEP为1.2770;(2)采用IRF、VCPA、CARS和IVSO分别提取出了71、9、22和39个特征波长,占全光谱的56.8%、7.2%、17.6%和31.2%;(3)基于多元特征提取算法建立的BP-ANN和LSSVM模型,对TEAC含量进行预测时Baseline-IVSO-LSSVM(Rc=0.9132,RMSEC=0.9620,Rp=0.8646,RMSEP=1.2883)具有最优预测性能;(4)相比于单一的光谱信息模型,TEAC相关的TFS图谱融合模型IVSO-TF1-BP-ANN显示出更好的效果,其Rp为0.8916,较特征波长数据建模提高了0.0286。 展开更多

维生素E对半滑舌鳎垂体α-生育酚转移蛋白基因表达的影响

生育酚转移蛋白(α-tocopherol transfer protein,α-TTP)是一种可以结合维生素E的蛋白,在协助机体转运和调控维生素E含量等方面起到重要作用。维生素E能够影响鱼类垂体中生长和繁殖相关激素的分泌,这是否与α-TTP有关,值得进一步探讨... 生育酚转移蛋白(α-tocopherol transfer protein,α-TTP)是一种可以结合维生素E的蛋白,在协助机体转运和调控维生素E含量等方面起到重要作用。维生素E能够影响鱼类垂体中生长和繁殖相关激素的分泌,这是否与α-TTP有关,值得进一步探讨。本实验旨在探究维生素E对半滑舌鳎(Cynoglossus semilaevis)垂体α-TTP基因表达的影响。在基础饲料中分别添加维生素E(DL-α-生育酚乙酸酯)0、200、400、800和1600 mg/kg,对半滑舌鳎成鱼(464.0±2.6)g进行持续60 d的养殖实验;另外,在L-15培养基中添加0、18和54μmol/L的维生素E,对半滑舌鳎垂体细胞进行为期3 d的体外原代培养实验。分别取垂体组织和垂体原代细胞,克隆α-TTP基因,并通过实时荧光定量PCR分析α-TTP基因相对表达量。结果显示,α-TTP基因全长3964 bp,编码293个氨基酸;α-TTP基因在半滑舌鳎各组织均有表达,其中,在脾脏中表达最高,其次是肾脏,在胃中表达量最低;α-TTP基因表达量随着饲料中维生素E含量的增加而呈先升高后下降的变化趋势,400 mg/kg组显著高于其他各组(P<0.05);随着细胞培养液中维生素E浓度的升高,α-TTP基因相对表达量显著增加。综上所述,本研究首次报道维生素E能够影响垂体中α-TTP基因的表达,饲料中添加400 mg/kg的维生素E能够显著促进半滑舌鳎垂体α-TTP基因的表达。本研究为探讨α-TTP在介导维生素E影响鱼类生长与繁殖方面的潜在机制提供新思路。 展开更多

目的探索以维生素E聚乙二醇琥珀酸酯(TPGS)做乳化剂,制备α-生育酚琥珀酸酯纳米乳(T-NE),对纳米乳进行处方优化、制备方法以及制剂学性质考察,并且评价其体外抗肿瘤细胞活性。方法自2022年1―4月,先后通过水滴定法绘制伪三元相图,根据... 目的探索以维生素E聚乙二醇琥珀酸酯(TPGS)做乳化剂,制备α-生育酚琥珀酸酯纳米乳(T-NE),对纳米乳进行处方优化、制备方法以及制剂学性质考察,并且评价其体外抗肿瘤细胞活性。方法自2022年1―4月,先后通过水滴定法绘制伪三元相图,根据纳米乳区域确定最优处方,再采用乳化蒸发法制备,并对其进行了理化性质评价,包括类型确定、形态学考察、粒径大小和分布、Zeta电位、载药量和稳定性,并通过细胞活力实验(MTT)进行体外抗肿瘤细胞活性评价。结果T-NE的最优处方为α-生育酚琥珀酸酯(α-TOS)、维生素E、TPGS、聚乙二醇400质量比1∶1∶2∶1;所制得T-NE为O/W型,外观呈类球形且无粘连;载药量为(20.15±1.91)g/L,粒径为(258.8±3.48)nm,多分散指数(PDI)为0.136±0.03,Zeta电位值为(-27.0±0.46)mV;4℃和25℃条件下放置稳定性良好,且适宜室温长期贮存;与游离的α-TOS药物溶液相比,T-NE对人乳腺癌细胞系(MCF-7)及人卵巢癌细胞系(A2780)细胞具有更强的抑制作用,IC50值分别为16.68μmol/L和7.50μmol/L,且空白纳米乳基本无毒性作用。结论该实验制备的T-NE外观均一呈类球形,粒径较小且分布均匀,载药量高稳定性好,同时表现出比游离α-TOS更为优异的抗肿瘤细胞活性作用,具有良好的开发前景。 展开更多

两种生育酚酯衍生物对D-半乳糖致衰老小鼠的抗衰老作用

为探究两种生育酚酯衍生物(D-α-生育酚醋酸酯和DL-α-生育酚醋酸酯)的抗衰老作用,本实验测定其体外抗氧化能力,并采用D-半乳糖构建衰老小鼠模型,同时用两种生育酚酯衍生物干预42 d,分析衰老相关指标变化规律。结果表明:在0.1~0.3 mg/m... 为探究两种生育酚酯衍生物(D-α-生育酚醋酸酯和DL-α-生育酚醋酸酯)的抗衰老作用,本实验测定其体外抗氧化能力,并采用D-半乳糖构建衰老小鼠模型,同时用两种生育酚酯衍生物干预42 d,分析衰老相关指标变化规律。结果表明:在0.1~0.3 mg/mL质量浓度范围内,两种生育酚酯衍生物的羟自由基清除能力、2,2’-联氮双(3-乙基苯并噻唑啉-6-磺酸)阳离子自由基清除能力、Fe2+螯合能力、总还原能力均优于VC。与衰老模型组相比,经过生育酚酯衍生物干预后,小鼠血清中丙二醛含量显著下降(P<0.05,P<0.01,P<0.001,P<0.0001),谷胱甘肽过氧化物酶和总抗氧化能力显著升高(P<0.05,P<0.01,P<0.001,P<0.0001);血清中炎症因子白细胞介素-6、白细胞介素-1β、肿瘤坏死因子-α和肝功能指标天冬氨酸氨基转移酶、丙氨酸氨基转移酶水平显著降低(P<0.05,P<0.01,P<0.001,P<0.0001);小鼠肝脏中核因子-E2-相关因子(nuclear factor E2 related factor 2,Nrf2)、醌氧化还原酶1(quinone oxidoreductase,NQO1)、血红素氧合酶1(heme oxygenase-1,HO-1)的mRNA及蛋白相对表达量提升,其中DL-α-生育酚醋酸酯的Nrf2、NQO1、HO-1 mRNA相对表达量分别增加了514.08%、461.78%、515.32%,蛋白相对表达量分别增加了620.00%、988.89%、1200.00%(P<0.0001)。综上所述,两种生育酚酯衍生物对D-半乳糖致衰老小鼠具有抗衰老作用,且DL-α-生育酚醋酸酯效果更好,二者抗衰老作用可能与调控Nrf2信号通路相关。 展开更多

Background Heat stress has severe negative consequences on performance and health of pigs,leading to significant economic losses.The objective of this study was to investigate the effects of supplemental vitamin E and a botanical extract in feed or drinking water on growth performance,intestinal health,and oxidative and immune status in grow-ing pigs housed under heat stress conditions.Methods Duplicate experiments were conducted,each using 64 crossbred pigs with an initial body weight of 50.7±3.8 and 43.9±3.6 kg and age of 13-week and 12-week,respectively.Pigs(n=128)were housed individually and assigned within weight blocks and sex to a 2×4 factorial arrangement consisting of 2 environments(thermo-neutral(21.2℃)or heat-stressed(30.9℃))and 4 supplementation treatments(control diet;control+100 IU/L of D-α-tocopherol in water;control+200 IU/kg of DL-α-tocopheryl-acetate in feed;or control+400 mg/kg of a botanical extract in feed).Results Heat stress for 28 d reduced(P≤0.001)final body weight,average daily gain,and average daily feed intake(-7.4 kg,-26.7%,and-25.4%,respectively)but no effects of supplementation were detected(P>0.05).Serum vitamin E increased(P<0.001)with vitamin E supplementation in water and in feed(1.64 vs.3.59 and 1.64 vs.3.24),but not for the botanical extract(1.64 vs.1.67 mg/kg)and was greater when supplemented in water vs.feed(P=0.002).Liver vitamin E increased(P<0.001)with vitamin E supplementations in water(3.9 vs.31.8)and feed(3.9 vs.18.0),but not with the botanical extract(3.9 vs.4.9 mg/kg).Serum malondialdehyde was reduced with heat stress on d 2,but increased on d 28(interaction,P<0.001),and was greater(P<0.05)for antioxidant supplementation compared to control.Cellular proliferation was reduced(P=0.037)in the jejunum under heat stress,but increased in the ileum when vitamin E was supplemented in feed and water under heat stress(interaction,P=0.04).Tumor necrosis factor-αin jejunum and ileum mucosa decreased by heat stress(P<0.05)and was reduced by vitamin E sup-plementations under heat stress(interaction,P<0.001).Conclusions The addition of the antioxidants in feed or in drinking water did not alleviate the negative impact of heat stress on feed intake and growth rate of growing pigs. 展开更多

旨在为大豆油的精准适度加工以保留微量营养成分提供参考,对巴西和美国大豆原油、中和油、脱色油、脱臭油、脱臭馏出物的生育酚含量及其异构体占比进行了分析,探讨和研究生育酚在精炼各工段的变化及生育酚异构体的占比规律。结果表明:... 旨在为大豆油的精准适度加工以保留微量营养成分提供参考,对巴西和美国大豆原油、中和油、脱色油、脱臭油、脱臭馏出物的生育酚含量及其异构体占比进行了分析,探讨和研究生育酚在精炼各工段的变化及生育酚异构体的占比规律。结果表明:大豆油中生育酚在精炼各工段均有损失,脱臭工段损失最多,生育酚和α-、γ-、δ-生育酚异构体损失率分别为11.68%~19.37%、6.10%~17.64%、11.56%~19.68%、17.29%~25.51%,在脱臭和整个精炼过程中3种生育酚异构体的损失率均表现为δ-生育酚>γ-生育酚>α-生育酚;与原油相比较,精炼后大豆油中α-、γ-生育酚占比分别增加了0.4~2.1百分点和0.1~1.2百分点,δ-生育酚占比减少了1.3~2.7百分点,说明大豆油中的δ-生育酚更容易被高温分解或挥发而被汽提蒸汽夹带进脱臭馏出物中;美国大豆原油酸值(KOH)在1.5 mg/g以下,其脱臭馏出物中生育酚含量在97419~109123 mg/kg,为原油的80~89倍,巴西大豆原油酸值(KOH)在1.5~3.3 mg/g,其脱臭馏出物中生育酚含量在34678~83803 mg/kg,为原油的59~84倍,美国和巴西的大豆油脱臭馏出物中α-、γ-、δ-生育酚占比分别为6.5%~9.3%、61.0%~62.4%、26.7%~31.4%和8.4%~9.9%,58.0~61.8%,28.0%~30.4%,与对应脱臭油相比较,美国和巴西的大豆油脱臭馏出物中α-、γ-生育酚占比分别减少了0.1~2.5百分点、4.5~6.7百分点和0.5~2.9百分点、2.9~6.9百分点,δ-生育酚占比分别增加了5.5~8.2百分点和5.6~7.8百分点,说明脱臭工段生育酚因挥发而被汽提蒸汽夹带的速率大小为δ-生育酚>α-生育酚>γ-生育酚。因此,应该优化脱臭工段的工艺条件,以最大程度避免生育酚的过度损失。 展开更多

The variation among Chinese genotypes of Brassica napus L. for seed tocopherols content and their analysis using gas chromatography has not been comprehensively reported till to date. In the present study, the tocopherol contents of four Chinese genotypes of Brassica napus L., namely, Gaoyou 605, Zhejiang 619, Zheshuang 758, and Zheshuang 72, were evaluated using three modified sample preparation protocols (P1, P2, and P3) for tocopherol extraction. These methods were distinguished as follows. Protocol one (P1) included the evaporation of solvent after extraction without silylation. Protocol two (P2) followed the direct supernatant collection after overnight extraction without drying and silylation. Protocol three (P3) included trimethylsilylation with N,O-bis(trimethylsilyl) trifluoroacetamide. Genotypic comparison of tocopherol and its isoforms revealed that Gaoyou 605 was dominant over the other genotypes with (140.5+ 10.5), (316.2+ 9.2), and (559.1+ 24.3) ~tg g-~ of seed meal ct-, 7-, and total (T-) tocopherol, respectively, and a 0.44+0.04 ^- to 7-tocopherol ratio. The comparison of the sample preparation protocols, on the other hand, suggests that P3 is the most suitable method for the tocopherol extraction from Brassica oilseeds and for the analysis of tocopherols using gas chromatography flame ionization detector (GC-FID). Trimethylsilylation is the key step differentiating P3 from P1 and P2. Variations detected in tocopherol contents among the Chinese rapeseed (B. napus) genotypes signify the need to quantify a wide range of rapeseed germplasm for seed tocopherol dynamics in short and crop improvement in long. 展开更多

In the present study, the impact of chromium in presence and absence of α-tocopherol was studied on plasma membrane of liver and kidney in male Wistar rats (80—100 g body weight). Significant increase in membrane cholesterol level as well as significant decrease in membrane phospholipid level in chromium exposed ( 0.8 mg /100 g body weight/d, i.p., for 4 weeks) animals suggest structural alteration of both liver and kidney plasma memebrane. The alkaline phosphatase, total ATPase and Na +-K +-ATPase activities of plasma membrane were significantly decreased in both liver and kidney after chromium treatment. However, α-tocopherol (30 mg/100 g diet) supplementation can restrict the changes in these membrane-bound enzyme activities. Thus, the usefulness of dietary supplementation of α-tocopherol to restrain the chromium-induced membrane damage is suggested. 展开更多

The objective of this study was to determine whether hydroponicallygrown lettuce contains as much ascorbic acid and tocopherol as soil-grown lettuce. We analyzed four varieties of lettuce, including: Waldmann’s Dark Green, Red Lollo Antago, Red Romaine Annapolis, and Butterleaf. The tocopherol content of hydroponically-grown Waldmann’s Dark Green, Red Lollo Antago, and Red Romaine Annapolis was 229%, 497% and 492% higher, respectively, compared to their soil-grown counterpart. The ascorbic acid content of hydroponically-grown Waldmann’s Dark Green, Red Lollo Antago, and Red Romaine Annapolis was 93%, 171% and 216% higher, respectively, compared to their soil-grown counterparts. Hydroponically grown lettuce varieties are significantly higher in both ascorbic acid and tocopherol content than their soil-grown counterparts, and hydroponic gardening is a viable option for producing nutritious fruits and vegetables. 展开更多

Walnut biological and nutritional value is also enhanced by its valuable protein and rich in nutrient composition such as vitamins and minerals. The most important characteristic of walnut oil is the abundance of polyunsaturated fatty acids, which makes it a unique food because of high amount of linoleic acid. Due to having valuable protein, vitamins and minerals it enhances biological and nourishment value, also. Recent epidemiological studies showed that consumption of walnut reduce cardiovascular diseases due to the rich in antioxidant properties, valuable fatty acids and tochopherols contents. In Turkey, walnut production and consumption increases year by year. The kernel of walnut genotypes shows variability in terms of their fat, fatty acid and tocopherol profiles. In this paper, it was aimed to characterize 10 walnut (Juglans regia L.) cultivars (Bilecik, Chandler, Hartley, Howard, Maras 12, Maras 18, Midland, Pedro, Sen and Serr) based on their fatty acid profiles using GC (Gas Chromatography), tocopherol and its isomers by HPLC (High Performance Liquid Chromatography) and total phenol content with spectrometric methods. Among the walnut cultivars “Hartley” was the highest linoleic acid (64.56%) and “Howard” was the α-linolenic acid 13.26 (%). The highest values of α (38.76 μg/g), β + γ (312.19 μg/g) and δ-tocopherol (40.77 μg/g) and total phenol (349 mg GAE/100 g ext) content were detected in “Sen” cultivar. Obtained results might be significant for further breeding programme to improve rich in especially γ-tocopherol linoleic acids and total phenolic compounds. 展开更多

Non-a-tocopherols are hydroxymethylated and hydrogenated to produce a-tocopherol in one pot process by simultaneously reacting with paraformaldehyde and hydrogen in the presence of catalysts of benzenesulfonic acid and 5% Pd/C in an autoclave. Effects of various operation conditions have been studied. The preferable reaction conditions are: temperature 180℃ to 200℃, pressure 5.0 MPa, acid concentration 0.5 g/100 ml ethanol, mass ratio of Pd/C to tocopherols 7.1g/100g, and reaction time 5.0 h. A product with a-tocopherol content of 80% was obtained by using a raw material with a total tocopherols content of 80.54%. The conversion of non-a-tocopherols is almost 100%, and the mole yield of a-tocopherol is more than 90%. 展开更多
