Total phenolic, flavonoid content, and antioxidant activity of flour, noodles, and steamed bread made from different colored wheat grains by three milling methods 被引量:9

The objective of this study was to evaluate the effects of wheat variety, food processing, and milling method on antioxidant properties. Black wheat variety Heibaoshi 1 had the highest total phenolic content(659.8 μg gallic acid equivalents g-1), total flavonoid content(319.3 μg rutin equivalents g-1), and antioxidant activity, whereas light purple wheat variety Shandongzimai 1 had the lowest total flavonoid content(236.2 μg rutin equivalents g-1) and antioxidant activity. Whole wheat flour and partially debranned grain flour had significantly higher total phenolic contents, total flavonoid contents, and antioxidant activity than refined flour(P < 0.05). Compared with flour, total phenolic contents, total flavonoid contents and antioxidant activity decreased in noodles and steamed bread, whereas noodles had slightly higher total phenolic and flavonoid content than steamed bread. Antioxidant activities(by ferric reducing ability of plasma assay) of steamed bread made from whole wheat flour, partially debranned grain flour, and refined flour were 23.5%, 21.1%, and 31.6% lower, respectively, than the corresponding values of flour. These results suggested that black whole wheat flour and partially debranned grain flour are beneficial to human health. 展开更多

Objective:To prepare various crude extracts using different polarities of solvent and to quantitatively evaluate their total phenol,flavonoids contents and phytochemical screening of Thymus vulgaris collected from Al Jabal Al Akhdar,Nizwa.Sultanate of Oman.Methods:The leave sample was extracted with methanol and evaporated.Then it was defatted with water and extracted with different polarities organic solvents with increasing polarities.The prepare hexane,chloroform,ethyl acetate,hutanol and methanol crude extracts were used for their evaluation of total phenol,flavonoids contents and phytochemical screening study.The established conventional metliods were used for quantitative determination of total phenol,flavonoids contents and phytochemical screening.Results:Phytochemical screening for various crude extracts were tested and shown positive result for flavonoids,saponins and steroids compounds.The result for total phenol content was the highest in hutanol and the lowest in methanol crude extract whereas the total flavonoids contents was the highest in methanol and the lowest hexane crude extract.Conclusions:The crude extracts from locally grown Thymus vulgaris showed high concentration of flavonoids and it could be used as antibiotics for different curable and uncurable diseases. 展开更多

Objective:To perform phytochemical screening,estimate total phenolics,flavonoids and to evaluate antioxidant potential of Moringa peregrina(M.peregrina) leaves.Methods:The dried powdered leaves of M.peregrina(150 g) were extracted exhaustively by Soxhlet with ethanol and then fractionated into hexane,chloroform,ethy alacetate and methanol.All the prepared extracts were also analyzed by gas chromatography-mass spectrometry to identify and characterize the chemical compounds present in the crude extracts.Folin- Ciocalteu reagent and aluminium chloride colorimetric methods were used to estimate total phenolic and flavonoid content of extracts.Hydrogen peroxide and 1,1 diphenyl-2-picrylhydrazyl were used to determine in vitro antioxidant activity.Results:Phytochemical analysis of ethanol extract showed presence of major classes of phytochemicals.Gas chromatography-mass spectrometry results revealed presence of 19 phytoconstituents in hexane extract,6 in ethyl acetate and 7 compounds in methanolic extract.Methanol extract was found to contain the highest phenolic content and flavonoids.In vitro antioxidant activities of all crude extracts were significant and comparable with the standard ascorbic acid.Conclusions:Results of this study show that the leaves of M.peregrina are the rich source of phenolic compounds that can play an important role in preventing the progression of many diseases. 展开更多

[Objectives] To optimize the extraction process of total flavonoids from Sanguisorbae Radix and carbonized Sanguisorba root,compare quality of different batches of Sanguisorbae Radix,study the effects of processing on the content of flavonoids,and provide scientific basis for reasonable utilization of Sanguisorbae Radix. [Methods] Test samples were prepared by heating,refluxing,and extraction,the extraction process was optimized by orthogonal experiment design,color was developed by NaNO_2-Al( NO_3)3-NaOH,and total flavonoids were measured by UV method at the wavelength of 510 nm. [Results] The linear relationship of rutin was excellent in the concentration range of 0. 1248 mg/mL-0. 5712 mg/mL,R^2= 0. 9997; the average recovery was 99. 67% and the RSD was 0. 70%. The optimum extraction conditions were as follows: the volume fraction of ethanol was 50%,the extraction temperature was 90℃,the extraction time was 90 min,and the solid-to-liquid ratio was 1∶ 20( g/mL). [Conclusions] After optimization of the extraction process,the extraction rate of total flavonoids in samples of Sanguisorbae Radix was significantly increased; there was certain difference in the content of total flavonoids between different batches of Sanguisorbae Radix and processed products; the total flavonoids significantly declines in carbonized sanguisorba root,and the influence of processing on its curative effect was to be further studied. 展开更多

[Methods]The ultraviolet-visible spectrophotometry was used to determine the content of total flavonoids in Pueraria lobata( Willd.) Ohwi. With the puerarin as index,the reflux extraction and single factor test were employed to investigate the effects of temperature,time,ethanol concentration and solid-liquid ratio on the content of total flavonoids in Pueraria lobata( Willd.) Ohwi,respectively. Under the optimal extraction technology,the content of total flavonoids in Pueraria lobata( Willd.) Ohwi at different altitudes was determined.[Results] The optimum extraction process was as follows: 70%ethanol; solid-liquid ratio of 1∶ 30; 1 h reflux extraction. Under these conditions,the extraction rate of flavonoids in Pueraria lobata( Willd.) Ohwi was 11. 48%,the total flavonoids content of different kudzu parts was in the order of roots > stems > leaves,and the total flavonoids content of the sample at about an altitude of 1000 m was significantly higher than at the altitudes of 1400 m and 1700 m.[Conclusions]It was suggested that the Pueraria lobata( Willd.) Ohwi should not be cultivated as medicinal plant in too high mountains,and the stems and leaves of Pueraria lobata( Willd.) Ohwi could be used as raw materials for extracting total flavonoids. 展开更多

[Results] A method for determining the content of total flavonoids was established,the content of total flavonoids was 11.35-21.35 mg·g^(-1),and the average content was 17.64 mg·g^(-1). In this study, in vitro antioxidant activity, total phenolic content and concentration of flavonoids of four different extracts of Adina cordifolia (Roxb.) were determined using spectrophotometric methods. Antioxidant activity of extracts was expressed as percentage of DPPH radicals’ inhibition and IC50 values (μg/ml). IC50 values ranged from 20.39 to 38.96 μg/ml. The total phenolic content ranged from 17.48 to 20.83 mg/g of dry weight of extract, expressed as gallic acid equivalents. The total flavonoid concentrations varied from 17.49 to 22.48 mg/g, expressed as quercetin equivalents. The significant linear correlation was confirmed between the values for the total phenolic content and antioxidant activity of plant extracts. Adina cordifolia (Roxb.) can be regarded as promising candidates for natural plant sources of antioxidants with high value. 展开更多

[目的]探究大黄不同极性部位有效成分含量与体外抗氧化和抑菌活性的相关性。[方法]采用90%乙醇回流提取大黄,采用氯仿、乙酸乙酯和正丁醇依次分级萃取,得到不同极性部位;以高效液相色谱(high performance liquid chromatography,HPLC)法测定其蒽醌类物质,以紫外分光度法测定总酚和总黄酮含量;以1,1-二苯基-2-三硝基苯肼(1,1-diphenyl-2-picrylhydrazyl,DPPH)法、2,2-联氮-二(3-乙基-苯并噻唑-6-磺酸)二铵盐[2,2'-azinobis-(3-ethylbenzthiazoline-6-sulphonate),ABTS]法和总还原力法测定抗氧化活性,采用抑菌圈直径和生长曲线测定法探究大黄不同极性部位对大肠杆菌、金黄色葡萄球菌和枯草芽孢杆菌的影响。[结果]正丁醇部位芦荟大黄素-8-O-葡萄糖苷和大黄酸-8-O-葡萄糖苷含量最多,峰面积分别为(2.72±0.07)mAU*min和(2.59±0.06)mAU*min,总酚和总黄酮含量最高,分别为(29.95±4.95)mg·g^(-1)和(35.71±3.26)mg·g^(-1);正丁醇部位的抗氧化活性和抑菌活性最强;芦荟大黄素-8-O-葡萄糖苷、大黄酸-8-O-葡萄糖苷、总酚和总黄酮含量与抗氧化活性正相关(P<0.05)。[结论]大黄总提物正丁醇部位的抗氧化和抑菌活性最强,可能与芦荟大黄素-8-O-葡萄糖苷、大黄酸-8-O-葡萄糖苷等成分有关,该结果对后续的深入研究具有指导意义。 展开更多

目的:比较仿野生种植三叶青不同部位中总黄酮含量、抗氧化活性以及抗炎能力差异。方法:采用醇提工艺对仿野生种植三叶青茎叶、块根、根须进行提取,比较不同部位间总黄酮含量差异;通过DPPH自由基、ABTS^(+)自由基、羟自由基清除实验以及... 目的:比较仿野生种植三叶青不同部位中总黄酮含量、抗氧化活性以及抗炎能力差异。方法:采用醇提工艺对仿野生种植三叶青茎叶、块根、根须进行提取,比较不同部位间总黄酮含量差异;通过DPPH自由基、ABTS^(+)自由基、羟自由基清除实验以及铁离子还原力测定,比较仿野生种植三叶青不同部位间抗氧化能力的强弱;采用脂多糖(LPS)诱导RAW264.7细胞作为炎症模型,通过细胞NO的释放量比较仿野生种植三叶青不同部位间的抗炎能力差异。结果:在最佳提取工艺条件下,测得仿野生种植三叶青茎叶、块根、根须总黄酮含量分别为11.86±0.23、8.48±0.10、7.52±0.02 mg·g^(-1);建立三叶青HPLC指纹图谱,共标定了10个共有峰,通过对照品指认出6号峰为芦丁、8号峰为槲皮素、9号峰为山奈酚,并对其含量进行检测,结果显示块根中三种指标成分含量均最高,其次根须,茎叶最低。检测仿野生种植三叶青不同部位对DPPH自由基、ABTS^(+)自由基以及羟自由基的IC_(50)值,茎叶分别为0.2107、0.2315、0.7625 mg/mL;块根分别为0.3134、0.3758、0.8967 mg/mL;根须分别为0.2058、0.2587、0.7856 mg/mL。三叶青茎叶、块根、根须对铁离子还原能力的吸光度值分别为0.172、0.153、0.184。当仿野生种植三叶青三部位的质量浓度在25~200μg/mL之间时,它们不会对RAW264.7细胞造成毒性,能够有效地抑制LPS诱导的NO释放,从而达到良好的抗炎效果。结论:本研究结果为仿野生种植三叶青不同部位的质量评价提供参考,并对仿野生种植三叶青非药用部位废物再利用及后续全资源开发提供理论依据。 展开更多

低温等离子体预处理对超声辅助提取姜粉总黄酮的影响及机制初探

目的:探究低温等离子体预处理对超声辅助提取姜粉总黄酮的提取效果,并对其可能机制进行初步探讨,为姜粉总黄酮的提取提供一种新方法。方法:以姜粉中总黄酮得率为考察指标,以低温等离子体放电电源功率、处理时间、进气量为考察因素,进行... 目的:探究低温等离子体预处理对超声辅助提取姜粉总黄酮的提取效果,并对其可能机制进行初步探讨,为姜粉总黄酮的提取提供一种新方法。方法:以姜粉中总黄酮得率为考察指标,以低温等离子体放电电源功率、处理时间、进气量为考察因素,进行单因素实验和三因素三水平响应面优化试验。并应用高效液相色谱法测定低温等离子体处理前后姜粉中21种多酚类化合物的含量,通过扫描电镜(scan electronic microscope,SEM)和傅里叶变换红外光谱(Fourier transform infrared spectroscopy,FTIR)对低温等离子体处理前后姜粉的结构进行分析。结果:姜粉中总黄酮的最佳提取条件为:真空度小于100 Pa,放电电源功率295 W、处理时间60 s,进气量150 cm^(3)/min,总黄酮得率为26.9 mg/g。色谱结果表明21种多酚类化合物得到较好的分离,且精密度高(relative standard deviation,RSD≤3.75%)、稳定性好(RSD≤4.25%)、重复性好(RSD≤4.75%)、加标回收结果准确可靠(平均回收率84.11%~100.86%)。低温等离子体处理后姜粉总黄酮的含量增加,酚酸的含量下降;低温等离子体处理增加了姜粉中淀粉颗粒以及细胞壁碎片的表面粗糙度,并且导致木质素的分解,加大了提取溶剂与姜粉的接触面积,因此有利于溶剂的渗透。结论:超声法辅助低温等离子体处理是提取姜粉总黄酮的一种有效方法。 展开更多

目的:建立蒙药复方沙棘颗粒剂中甘草酸单铵盐、总黄酮的含量测定方法。方法:采用高效液相色谱法,C_(18)(250 mm×4.6 mm,5μm)为色谱柱,以乙腈-0.05%磷酸溶液(38∶62)为流动相,体积流量为1.0 mL/min,进样量为10μL,在252 nm波长下... 目的:建立蒙药复方沙棘颗粒剂中甘草酸单铵盐、总黄酮的含量测定方法。方法:采用高效液相色谱法,C_(18)(250 mm×4.6 mm,5μm)为色谱柱,以乙腈-0.05%磷酸溶液(38∶62)为流动相,体积流量为1.0 mL/min,进样量为10μL,在252 nm波长下测定甘草酸单铵盐含量;利用紫外可见分光光度法,以芦丁为对照品,在510 nm波长下检测蒙药复方沙棘颗粒剂总黄酮含量。结果:甘草酸单铵盐对照品在0.053~0.530μg范围内与峰面积呈良好的线性关系,Y=11.003X+0.0031,r=1,平均回收率为105.9%,RSD=1.22%(n=6);芦丁对照品在2.0~80.0μg/mL范围内与吸光度值呈良好的线性关系,线性回归方程为Y=11.537X+0.0014,r=0.9999,平均回收率为99.20%,RSD值为0.54%(n=6)。结论:该方法操作简便,准确,具有良好的精密度、重复性和稳定性,适用于蒙药复方沙棘颗粒剂的质量控制。 展开更多

广西产火炭母总黄酮提取工艺优化及含量测定

采用紫外可见分光光度法测定火炭母(Polygonum chinense L.)中总黄酮的含量,以总黄酮含量为考察指标,以提取方式、提取溶剂、料液比、提取时间、提取次数为考察因素,在单因素试验的基础上,采用L9(33)正交设计优化火炭母中总黄酮的提取... 采用紫外可见分光光度法测定火炭母(Polygonum chinense L.)中总黄酮的含量,以总黄酮含量为考察指标,以提取方式、提取溶剂、料液比、提取时间、提取次数为考察因素,在单因素试验的基础上,采用L9(33)正交设计优化火炭母中总黄酮的提取工艺。最优提取工艺:70%乙醇为提取溶剂,料液比1∶70(m/V),超声波提取45 min,提取2次。以芦丁为对照品,在500 nm波长处测定其吸光度,平均加样回收率为98.76%,相对标准偏差为1.72%。结果表明,13个产地火炭母药材的总黄酮含量为22.205 4~52.173 7 mg/g,相对标准偏差(RSD)为1.07%~2.81%。13个产地总黄酮含量由高到低依次为桂林市、南宁市、玉林市、百色市、贺州市、河池市、贵港市、防城港市、钦州市、梧州市、来宾市、崇左市、柳州市。该方法精密度、稳定性、重复性良好,可作为广西产火炭母药材质量评价的依据。 展开更多

目的牡丹(Paeonia suffruticosa Andr.)是毛茛科芍药属的多年生落叶灌木,其根皮被称为“牡丹皮”,药用历史悠久,其传统非药用部位“叶”在民族地区应用广泛,研究牡丹叶的药用价值是提高中药资源综合利用的有益尝试。牡丹叶含有的多酚、... 目的牡丹(Paeonia suffruticosa Andr.)是毛茛科芍药属的多年生落叶灌木,其根皮被称为“牡丹皮”,药用历史悠久,其传统非药用部位“叶”在民族地区应用广泛,研究牡丹叶的药用价值是提高中药资源综合利用的有益尝试。牡丹叶含有的多酚、黄酮类活性成分,在抑菌、抗氧化和抑制酪氨酸酶等方面作用显著。方法采用甲醇提取法获取牡丹叶提取物,建立同时测定牡丹叶提取物中5种活性成分(没食子酸、芍药苷、山奈素、槲皮素和异鼠李素)含量的高效液相色谱(HPLC)法。同时,采用Folin-Ciocalteu法和硝酸铝-亚硝酸钠比色法测定了牡丹叶提取物中总黄酮和总多酚的含量。立足羧甲基壳聚糖(CMCS)抗菌活性和良好的载药特性,研究其与牡丹叶提取叶的协同机制。结果含量测定结果显示,牡丹叶中没食子酸、芍药苷、山奈素、槲皮素和异鼠李素的含量分别为17.780%、0.470%、1.150%、1.920%和3.160%,其线性关系、稳定性、重复性和精密度等均良好。体外抗菌活性结果显示,牡丹叶提取物(PLE)与CMCS联用的FICI系数均小于0.500,其对金黄色葡萄球菌和大肠埃希菌的抑菌效果均表现为协同作用。结论牡丹叶提取物中含有较高的多酚类和黄酮类成分,并且与CMCS具有较强的协同作用。为进一步开发牡丹叶与羧甲基壳聚糖相关抗菌制剂的研制提供联用数据支撑,为牡丹药用资源的综合利用奠定基础。 展开更多

The current interest in the health beneifts of whole wheat grain has prompted breeders to further increase the concentration of antioxidants in wheat. The objective of this study was to investigate the variation in antioxidant content among Chinese wheat grains and the relationship between antioxidants and grain color and morphological characteristics. A wide variation was observed in the total phenolic, carotenoid and lfavonoid contents, as well as the antioxidant activity (AOA), of Chinese wheat varieties. Black wheat had the highest mean total phenolic, carotenoid and lfavonoid contents and the highest AOA, followed by red and white wheats. The grain color parameters were signiifcantly negatively correlated with total phenolic, carotenoid and lfavonoid contents and AOA among all of the wheat varieties examined, and grain weight was also signiifcantly negatively correlated with these traits. The same correlation between grain weight and antioxidant traits was also observed within individual groups of wheat, which indicates that grain weight may be used as an index for selecting wheat varieties with high AOA. Landraces had signiifcantly higher lfavonoid content than commercial wheat varieties. The results of this study may be useful for breeding nutrient-rich wheat varieties. 展开更多

葛花菊花水煎液中总黄酮含量的测定方法研究

以葛花和菊花为原料,以水煎法作为提取方法,采用可见分光光度法和紫外分光光度法对水煎液中总黄酮含量进行测定,从而探索合适的测量葛花菊花水煎液中总黄酮浓度的方法。可见分光光度法以芦丁对照品,采用NaNO_(2)-Al(NO_(3))_(3)-NaOH显... 以葛花和菊花为原料,以水煎法作为提取方法,采用可见分光光度法和紫外分光光度法对水煎液中总黄酮含量进行测定,从而探索合适的测量葛花菊花水煎液中总黄酮浓度的方法。可见分光光度法以芦丁对照品,采用NaNO_(2)-Al(NO_(3))_(3)-NaOH显色法,紫外分光光度法分别以芦丁和鸢尾苷为对照品,分别进行可见光区和紫外的光谱扫描。结果表明,芦丁对照品溶液显色后在506 nm处有吸收峰位,而样品溶液同条件显色后在可见光范围内无吸收峰位。在紫外分光光度法中,芦丁对照品溶液在258 nm处有最大峰值,而样品溶液在264 nm处有最大吸收峰值,两者的吸收峰位相差6 nm。而鸢尾苷对照品溶液和样品溶液在波长264 nm均有吸收峰,两者吸收峰位一致。以NaNO_(2)-Al(NO_(3))_(3)-NaOH的显色法不适用于测定样品溶液中总黄酮的含量,而以鸢尾苷对照品的紫外分光光度法最适用于葛花菊花水煎液总黄酮含量的测定。通过方法学考察表明该法是一种稳定、精密性和重复性俱佳的方法。该研究为以葛花和菊花为主要原料的解酒药方的开发与应用提供了基础。 展开更多

目的 建立一种适用于相思子叶黄酮部位中5个黄酮成分含量测定的一测多评法(QAMS)。方法 采用高效液相色谱法,以滨蓟黄苷为内标,通过外标法(ESM)建立其与柚葡糖苷、(2R)-柚皮素6-C-β-D-吡喃葡萄糖苷、异柚葡糖苷和高车前素-4′-O-β-D... 目的 建立一种适用于相思子叶黄酮部位中5个黄酮成分含量测定的一测多评法(QAMS)。方法 采用高效液相色谱法,以滨蓟黄苷为内标,通过外标法(ESM)建立其与柚葡糖苷、(2R)-柚皮素6-C-β-D-吡喃葡萄糖苷、异柚葡糖苷和高车前素-4′-O-β-D葡糖苷的相对校正因子(RCF),以该RCF计算对应4种成分的含量(计算值),分析计算值与实测值之间的差异,实现一测多评。结果 柚葡糖苷、(2R)-柚皮素6-C-β-D-吡喃葡萄糖苷、异柚葡糖苷和高车前素-4′-O-β-D葡糖苷与滨蓟黄苷的RCF分别为0.76、0.77、1.38和0.33,且该RCF具有良好稳定性和重现性。12批相思子叶总黄酮样品经QAMS与ESM测定比较,结果无显著性差异,计算值与实测值之间的RSD均小于4%。结论 所建立的QAMS具有良好的准确性和可靠性,可用于相思子叶总黄酮部位的品质分析和评价。 展开更多
