[ Method] Effects of various factors on mycelial growth and spore germination were stud- ied, and the lethal temperature of spores and indoor toxicity of fungicides were also determined. [Result] The fungal mycelium had a better growth rate on water- melon juice medium than the others. The optimum temperature for mycelial growth and spore germination were 28 ℃ and 30 ℃, and the optimum pH values were 7 - 8 and 7 - 9, respectively. Light-dark alternation was conducive for mycelial growth. The lethal temperature of spore was 60 ℃ for 5 min. The fungal mycelium grew better while taking fructose and glucose as carbon sources and yeast extract and peptone as nitrogen sources. Toxicity determination showed that 50% pro- chloraz-manganese chloride complex WP had the greatest inhibition effect on mycelial growth, with the ECso value of 0.730 9 μg/mL; followed by 32.5% benzoic azoxystrobin SC, 30% benzoylate· propieanazol EC, 10% difenoconazole WG, 25% bromothalonil · carbendazim WP and 25% bromothalonil WP, with the ECso values from 1.884 7μg/mL to 8. 161 0μg/mL. Arx, and accomplish the preliminary screening the test of fungicides. [Method]The pathogen was isolated from the infected leaves of watermelon collected from greenhouse in Hainan Province. The biological characteristics of C. orbiculare and indoor toxicity of fungicides were studied. [Results] The mycelium grew fastest on the medium with leaf extract of watermelon. The optimum temperature for mycelial growth and spore germination was 28 and 30 ℃, and the optimum pH was 7-9 and 6-8, respectively. The continuous darkness condition was most suitable for mycelial growth. The lethal temperature of spore was 55 ℃ water bath for 10 min in water bath. The suitable carbon sources were maltose, L-arabinose and glucose, and the suitable nitrogen sources were yeast extract, beef extract and peptone. In the indoor toxicity determination, 32.5% Benzoic azoxystrobin SC, 50% Dichlorotetrakis WP and 10% Difenoconazole WG showed better inhibition effects against the mycelial growth of C. orbiculare, with the EC50 values of 0.018 0, 0.060 2 and 0.471 5 μg/ml, respectively. Water toxicity determination with electrochemically active bacteria(EAB)shows promise for providing early warnings for heavy metal pollution in water.However,thus far,only idealized tests with a few types of heavy met...Water toxicity determination with electrochemically active bacteria(EAB)shows promise for providing early warnings for heavy metal pollution in water.However,thus far,only idealized tests with a few types of heavy metals have been conducted.In this study,an automatic water-toxicitydetermination system with high technical maturity was established,and the toxicological properties of common heavy metals were systematically assessed.The results demonstrated that the common heavy metals linearly inhibited EAB currents in the range of 0.1 mg/L to 0.5 mg/L.The toxicity ranking of the tested heavy metals was Pb^(2+)>Tl^(3+)>Cu^(2+)>Cd^(2+)>Zn^(2+)>Ni^(2+)>Hg^(2+)>As^(3+).The toxicity interaction mainly exhibited an antagonistic effect in binary heavy metal mixtures.The system can accurately determine surface water toxicity and rapidly monitor heavy metal pollution,with good repeatability and a long lifetime.Overall,this study demonstrates that EAB are capable of long-term(>60 d)surface water quality monitoring and on-site early warning of heavy metal pollution.展开更多
[Objective] This study was conducted to isolate and screen new Bacillus thuringiensis (Bt) strains against Lepidoptera insecticides. [Method] Bt strains were isolated from soil of Changbai Mountain area by temperatu...[Objective] This study was conducted to isolate and screen new Bacillus thuringiensis (Bt) strains against Lepidoptera insecticides. [Method] Bt strains were isolated from soil of Changbai Mountain area by temperature screening method, and highly-toxic Bt stains were then selected by biological determination and toxicity de- termination. [Result] From 150 soil samples, 18 Bt isolates were isolated, with an average isolation rate of 12.0%. Specifically, the isolation rate from mountain field was 8.5 %, and the isolation rate from farmland was 16.2%. The results of activity determination showed that there were17, 5 and 4 strains showing lethality rate over 90% against Plutel/a xylosrel/a, Spodoptera litura and Spodptera exigua, respectively, and among them, strain YNI-1 exhibited high activity against all the 3 kinds of in- sects. The results of toxicity determination showed that strain YNl-lhad the best fast-acting property against S.litura and S. exigua; strains with high toxicity against S. exigua were YN6-2〉YN1-1〉YN4-2〉YN2-6 sequentially; and toxicity of strains against S. /itura was in order of YN4-4〉YN1-1 〉YN6-1 〉YN4-1 〉YN2-1. The toxicities of sever-al germicides to Col etotrichum fragariae were determined with indoor mycelial growth inhibition assay. Field control test was preformed to further verify the control efficacy of germicides. [Result] The isolated strawberry anthracnose pathogenic strains were identified as C. gloeosporioide from genus Col etotrichum of subphylum Deuteromycotina. Prochloraz, pyrazole Kresoxim and hexaconazole al exhibited good indoor control effects; 25% pyrazole Kresoxim EC2000-fold solution, 24% azoxys-trobin·hexaconazole (azoxystrobin∶hexaconazole=1∶2) SC1500-fold solution and 25%prochloraz EC1000-fold solution al exhibited significant field control efficacy. The results of indoor toxicity test showed that LC^o of reagent 1, reagent 2 and reagent 3 reached 10 270, 12 810 and 13 040 mg/L, respectively. The results of field efficacy test showed that control efficacy of reagent 1, reagent 2 and reagent 3 reached 60.52%, 34.60% and 46.99%, respectively. Overall, reagent 1 (91% ultrafine powder of tobacco leaves, 3% silicone oil, 2% sasangua cake, 2% emulsifier, 2% ascorbic acid) exhibited the highest control efficiency against pomegranate aphids, which can be further popularized and utilized.展开更多
