Objective:To use high-frequency ultrasound to evaluate the efficacy ofα-lipoic acid combined with traditional Chinese medicine in the treatment of diabetic peripheral neuropathy(DPN),so as to provide a basis for clin...Objective:To use high-frequency ultrasound to evaluate the efficacy ofα-lipoic acid combined with traditional Chinese medicine in the treatment of diabetic peripheral neuropathy(DPN),so as to provide a basis for clinical medication and evaluation of therapeutic effect.Methods:From December 2018 to December 2019,110 patients with DPN who met the inclusion and exclusion criteria were divided into observation group and control group according to different treatment plans.The control group was treated withα-lipoic acid,and the observation group was treated withα-lipoic acid combined with traditional Chinese medicine encapsulation treatment,and used high-frequency ultrasound,nerve conduction velocity and serological examination to comprehensively evaluate the effectiveness of the two treatment methods.Results:The mean amplitude of glycemic excursions(MAGE)value of the observation group and the control group after treatment was significantly lower than before treatment(P<0.05);After treatment,the SCV of the ulnar nerve,median nerve,and common peroneal nerve in the two groups was significantly faster than before treatment(P<0.05).Similar to SCV,the MCV of the three nerves measured after treatment in the two groups was significantly faster than before treatment(P<0.05).Compared between the groups after treatment,the three kinds of nerve SCV and MCV in the observation group were significantly faster than those in the control group(P<0.05).The cross-sectional area(CSA)value of the ulnar nerve,median nerve and common peroneal nerve in the observation group after treatment was significantly lower than before treatment(P<0.05).Compared with the control group after treatment,the three nerves CSA in the observation group was significantly lower than that in the control group(P<0.05).Abnormal ultrasound performance:The proportion of abnormal ultrasound performances of the ulnar nerve,median nerve,and common peroneal nerve in the observation group after treatment was significantly lower than before treatment(P<0.05).The composition ratio of internal echo reduction and ambiguity in the observation group after three nerve treatments was significantly lower than that in the control group after treatment(P<0.05).In the correlation analysis,the three kinds of nerve CSA before and after treatment were negatively correlated with SCV and amplitude(P<0.05),and positively correlated with latency(P<0.05).Conclusion:The combination ofα-lipoic acid and Chinese medicine encapsulation technology has a good effect on the treatment and repair of DPN nerve damage,and can be routinely applied in clinical treatment.High-frequency ultrasound can intuitively observe changes in peripheral nerves and can be used to evaluate the prognosis of DPN.展开更多
Disorder of energy metabolism is a major pathological change in the progression of heart failure.This process leads to insufficient myocardial energy and further aggravates cardiac dysfunction.Disorders of metabolic s...Disorder of energy metabolism is a major pathological change in the progression of heart failure.This process leads to insufficient myocardial energy and further aggravates cardiac dysfunction.Disorders of metabolic substrate utilization,mainly glucose and fatty acids,play an important role in this process.Research over the years has shown that some traditional Chinese medicines or compound prescriptions whose main role is to replenish qi and warm yang have good effects in regulating energy metabolism disorders.It has been found that some active ingredients in traditional Chinese medicine can regulate the uptake and utilization of myocardial cell metabolic substrate,so that the metabolism of myocardial cells can be adjusted in a direction that is beneficial to the body under hypoxic conditions,increasing the overall energy supply of the myocardium and improving heart function.This article reviewed the research of traditional Chinese medicine intervention on glucose and lipid metabolism of heart failure myocardial cells,and preliminarily summarizes the law and mechanism of traditional Chinese medicine intervention in heart failure myocardial glucose and fatty acid metabolism,hoping to provide clues for energy metabolism therapy research from the perspective of traditional Chinese medicine.展开更多

These included 0.180 g/mL of Herba Epimedi (Epimedium), Semen Cuscutae (Dodder Seed), or Herba Cistanches (Desertliving Cistanche), 0.04 mg/mL monoamine oxidase-B inhibitor selegiline, or distil ed water for 14 consecutive days to prepare drug-containing serum or blank serum. MES23.5 cells in the logarithmic phase were cultured in media supplemented with 15%drug-containing serum for 24 hours, fol owed by incubation in culture solution containing 100μmol/L H2O2 for 3 hours. 3-(4,5-Dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) and flow tometry results showed that al drug-containing serums improved the survival rate of H 2 O 2-injured MES23.5 cells, inhibited pro-apoptotic FasL and caspase-3 expression, promoted anti-apoptotic Bcl-2 expression. However, drug-containing serums had little influence on Fas expression in H 2 O 2-injured MES23.5 cells. Enzyme-linked immunosorbent assay results showed that serum containing Herba Cistanches or Herba Epimedi increased the expression of nerve growth factor, brain-derived neurotrophic factor, and glial cellline-derived neurotrophic factor in injured MES23.5 cells;serum containing Semen Cuscutae only increased brain-derived neurotrophic factor expres-sion; while expression of the above neurotrophic factors remained the same in cells treated with serum containing selegiline. Objective Our objective was to screen drugs with good protective effects on gefitinib-induced hepatotoxicity.Methods Fifty-four specific pathogen-free-grade male mice of the Institute of Cancer Research were randomly ...Objective Our objective was to screen drugs with good protective effects on gefitinib-induced hepatotoxicity.Methods Fifty-four specific pathogen-free-grade male mice of the Institute of Cancer Research were randomly divided into a normal group,gefitinib group,glutathione group,ligustrazine group,silymarin group,glycyrrhizic acid group,baicalin group,paeoniflorin group,and matrine group,with six mice in each group.Except for the normal group,the remaining groups of mice were intragastrically administered 400 mg·kg^(-1)of gefitinib for 16 days to induce liver injury.Mice in each treatment group were intragastrically administered 100 mg·kg^(-1)of the corresponding drug 30 minutes after gefitinib administration each day.The normal group and model group mice were intragastrically administered with an equal volume of 0.5%carboxy-methylcellulose sodium(CMC-Na),and the administration volume was 10 mg·kg^(-1).Then,30 minutes after the last administration,blood was collected from the retro-orbital venous plexus,and the mice were killed by cervical dislocation to obtain liver weight and calculate the liver index.Liver pathological changes were observed by hematoxylin–eosin(HE)staining;the levels of alanine aminotransferase(ALT)and aspartate aminotransferase(AST)in serum were detected using biochemical kits.AML12 cells were cultured in a medium containing drugs for 30 minutes.Except for the normal group,the remaining groups were induced cell damage with 20μmoL·L1 gefitinib for 24 hours.Cell viability was detected using a cell counting kit-8,and the levels of ALT,AST,and lactate dehydrogenase(LDH)in the cell culture supernatant were measured using biochemical kits.Results Animal experiments showed that compared with the gefitinib group,glycyr-rhizic acid and baicalin significantly increased the body weight of mice(p<0.01)and decreased the liver index and levels of ALT and AST(p<0.05);ligustrazine and silymarin significantly increased the body weight of mice(p<0.01)and decreased the level of AST(p<0.05);paeoniflorin and matrine significantly decreased the levels of ALT and AST(p<0.01).HE staining showed that the liver tissue of mice in the gefitinib group presented a large number of inflammatory cell infiltrations and disordered arrange-ment of hepatic cords;glutathione,glycyrrhizic acid,baicalin,paeoniflorin,and matrine significantly alleviated pathological damage to mouse liver tissue.Cell experi-ments showed that all drugs could alleviate gefitinib-induced damage to AML12 cells to varying degrees,among which glycyrrhizic acid,baicalin,and paeoniflorin had better protective effects,with cell survival rates increased to 96.4,81.1,and 78.2%,respectively.Compared with the gefitinib group,glycyrrhizic acid significantly reduced the levels of ALT,AST,and LDH(p<0.05);silymarin,baicalin,and paeoniflorin significantly reduced the levels of ALT and LDH(p<0.05).Conclusion Glycyrrhizic acid,baicalin,and paeoniflorin all have good protective effects against gefitinib-induced hepatotoxicity,among which glycyrrhizic acid has the best effect.展开更多
