Traditional Chinese medicine (TCM) prescriptions containing Chrysanthemi Flos are commonly used for brightening eyes and have shown good clinical value for treating retinal diseases. This study aimed to screen a novel TCM prescription against retinal diseases based on patents containing Chrysanthemi Flos and explores its medication rule and mechanism using data mining, network pharmacology, molecular docking and molecular dynamics simulation. Methods: Data mining approach was used to screen a novel TCM prescription against retinal diseases and explore its medication rule. The mechanism of the novel TCM prescription against retinal diseases was revealed by constructing the network of TCMs-core active ingredients-core targets-core pathways using network pharmacology. Molecular docking and molecular dynamics simulation were performed to confirm the binding ability between core active ingredients and core targets. Results: TCM prescriptions with clearing heat and supplementing activities were applied most to treat three types of retinal diseases including glaucoma, diabetic retinopathy and age-related macular degeneration. These TCM prescriptions mainly possessed bitter and cold properties and were associated with the liver and lung meridians. A novel TCM prescription consisting of Chrysanthemi Flos, Cassiae Semen, Lycii Fructus, Angelicae Sinensis Radix and Salviae Miltiorrhizae Radix et Rhizoma was screened for the first time. Eighteen compounds were found to be the core active ingredients, modulating sixteen core targets including STAT3, MAPK3, AKT1, PIK3CA, EGFR and TP53, which may involve fifteen core pathways such as the AGE-RAGE signaling pathway in diabetic complications, MAPK signaling pathway, PI3K-Akt signaling pathway, HIF-1 signaling pathway and EGFR tyrosine kinase inhibitor resistance. Molecular docking and molecular dynamics simulation showed a strong binding affinity between almost all of the core active ingredients and targets. Conclusion: The novel TCM prescription has the potential to treat retinal diseases by inhibiting the inflammatory response, oxidative stress, retinal neovascularization, and apoptosis of retinal ganglion cells and retinal pigment epithelium cells as well as ameliorating retinal structural impairment through core targets and signaling pathways. This is the first study to screen a novel TCM prescription against retinal diseases, and provides reliable evidence and promising directions for new drug development.展开更多

METHODS: The State Intellectual Property Office Patents Database, China Patent Information Center Patents Database, Soopat Patent Search Platform, and CNIPR Search Platform of Chinese and Foreign Patent Database were searched to identify Traditional Chinese Medicine compound prescription patents for mammary gland hyperplasia that had been maintained for at least 5 years. Logistic regression analysis was used to detect the factors influencing the maintenance of valid patents. RESULTS: From 1 January 1993 to 31 December 2019, a total of 1228, Traditional Chinese Medicine compound prescription patents for mammary gland hyperplasia were authorized. Of these applications, 381 patents were granted and only 176 patents were maintained for more than 5 years. The five independent variables assessed in this study all had significant explanatory functions. CONCLUSIONS: The maintenance of valid patents for Traditional Chinese Medicine compound prescriptions for treating mammary gland hyperplasia is of great importance. The maintenance of such patents depends on many factors, such as the concentration of effective herbs, number of dependent claims, provision of new dosage forms and detailed experimental data, and transfer of patent rights.展开更多

This is the first study to screen a novel TCM prescription against retinal diseases, and provides reliable evidence and promising directions for new drug development.

文摘Objective There are many clinical reports on traditional Chinese medicine(TCM)combined with nucleoside(acid)analogues(NAs)for the treatment of chronic hepatitis B(CHB),but its efficacy and safety are not completely clear. This meta-analysis aims to evaluate the clinical efficacy and safety thus providing evidence for clinical applications. Methods We searched Chinese databases the China National Knowledge Infrastructure(CNKI),Wanfang Data, and China Science and Technology Journal Database(VIP),as well as English databases Pub Med and Cochrane Library, from time of establishment to April 14,2021.Literature quality was evaluated according to the bias risk assessment criteria of Cochrane Collaboration network.Rev Man 5.3 and Stata 12.0 software were used to perform this research. Results A total of 23 articles,3282 patients, and 25 TCM prescriptions were included in this study.NAs plus TCM remarkably improved the clinical total effective rate[Odds ratio(OR)=3.92,P<0.00001],TCM syndrome score(Mean difference=-3.73,P<0.00001),hepatitis B virus(HBV)DNA negative conversion rate(OR=1.49,P=0.0001),hepatitis Be antigen(HBe Ag)negative conversion rate(OR=2.03,P<0.00001),alanine aminotransferase levels[Std mean difference(SMD)=-0.95,P<0.00001],and aspartate aminotransferase levels(SMD=-0.70,P=0.0004).Adverse reaction rates did not increase in the combined treatment group(OR=0.97,P=0.84).A comprehensive analysis of the 25 TCM prescriptions suggested that the combination of spleen-strengthening prescriptions with NAs showed better effects than other prescriptions. Conclusion TCM in combination with NAs,demonstrated better clinical efficacy against CHB than NAs alone. In addition, the combination of spleen-strengthening prescriptions and NAs was identified as the best therapeutic strategy. However, more randomized controlled trials of high quality are needed to provide more reliable clinical basis for the application of TCM.

文摘OBJECTIVE: To explore the value and factors influencing the maintenance of valid Traditional Chinese Medicine compound prescription patents for the management of mammary gland hyperplasia. The maintenance of such patents depends on many factors, such as the concentration of effective herbs, number of dependent claims, provision of new dosage forms and detailed experimental data, and transfer of patent rights.
