Extraction and Refining Process of Polysaccharide from Tricholoma matsutake and Verification of Its Whitening Effect

[Objectives]The paper was to determine the extraction and refining process of polysaccharide from Tricholoma matsutake,and to verify its whitening effect.[Methods]With T.matsutake as the research object,the effects of eluent concentration,loading amount and diameter-to-height ratio on the refining process of T.matsutake polysaccharide were explored by orthogonal test,to optimize the dynamic elution conditions of T.matsutake polysaccharide.The survival rate of mouse melanoma cells(B16),inhibition of melanin synthesis and inhibition of tyrosinase activity of crude extract A1 and refined extracts A2-A4 were tested to verify the whitening effect of T.matsutake.[Results]The best extraction and refining process for T.matsutake polysaccharide was:eluent 40%ethanol(volume fraction),loading amount 30 mg/g(polysaccharide-resin),diameter-to-height ratio 1∶8.Under this condition,the extraction rate of T.matsutake can reach up to 33%(A3).The whitening efficacy test showed that crude extract A1 and refined extracts A2-A4 had good whitening effects,and the whitening effects were A3≥A4>A1>A2.[Conclusions]The study provides a reference for the application of T.matsutake polysaccharide in cosmetics. 展开更多

Two undescribed Tricholoma triterpenoids,namely tricholopardins C(1)and D(2),were isolated from the wild mushroom Tricholoma pardinum.Their structures with absolute configurations were elucidated by spectroscopic methods,as well as the single crystal X-ray diffraction.Compounds 1 and 2 were further obtained by chemical conversions from the known analogues.Compound 1 showed significant cytotoxicity to MCF-7 and Hela cell lines with IC_(50)values of 4.7μM and 9.7μM,respectively.Its mechanism of inducing MCF-7 cell apoptosis was studied briefly. 展开更多

The results showed that the genetic characteristics of Tricholoma matsutake str... A total of 10 Tricholoma matsutake strains isolated from Yajiang County, Sichuan Province of China were analyzed by using AFLP technique. The results showed that the genetic characteristics of Tricholoma matsutake strains can be properly revealed by AFLP, and among the 10 Tricholoma matsutake strains, the genetic aspects were very similar. At the level of 90% similarity, all the strains collected together, and at the similarity of 92 % , three AFLP groups formed. Simultaneously, the effects of quantity of the primers on the second AFLP PCR were determined. The optimal quantity of Eco +1/Mse+1, Eco + 2/Mse + 2 and Eco + 3/ Mse+3 primer pairs used in the AFLP was 1 pmol/5 pmol, 0. 6 pmol/10 pmol and 0.1 pmol/8 pmol, respectively. 展开更多

Two membranes of different molecular weight cut-off, namely, 30 kD and 10 kD were chosen with ultrafiltration. The antioxidant activity of the purification polysaccharides was evaluated by various antioxidant assay, including DPPH (l,l-diphenyl-2-picrylhydrazyl) radical scavenging, superoxide anion radical scavenging and hydrogen peroxide scavenging. The scavenging effect on hydroxyl radical and DPPH radical, which may be comparable to Vitamin C, while the scavenging effect on superoxide anion radicals was not obvious. 展开更多

In order to achieve the transformation from the conventional soil-covering cultivation of Tricholoma giganteum with bags to the soilless cultivation with bottles and understand the mechanism of primordium formation of Tricholoma giganteum,with Tricholoma giganteum mycelia as experimental materials,this paper studied the effect of different fruiting treatments on the activity of three enzymes in different time.The results showed that from the mycelial recovery to primardial formation and budding under three treatment groups which could form primordium,the tyrosinase activity was relatively stable,and under two treatment groups which could not form primordium,the tyrosinase activity dropped after the rise and reached a maximum on the 9 th day,significantly higher than under the other three groups,indicating that too high tyrosinase inhibited primardial formation; the prolease and amylase activity was effectively activated before primardial formation,and the enzymic activity was significantly higher than under the two treatment groups which could not form primordium. 展开更多

Tricholoma giganteum also being called Tricholoma lobayense, belongs to Basidiomycotina, Hymenomycetes, Agaricales, Tricholomataceae and Tricholoma. It has high economic and medicinal values and is a kind of rare edible mushroom. Modern pharmacological studies have shown that T. giganteum have antitumor, antioxidant, antimicrobial and free radicals scavenging activity, and it can inhibit high blood pressure and HIV-1 reverse transcriptase. In this study, the technologies of isolation and purification of polysaccharides of T. giganteum were systematically studied. T. giganteum polysaccharides were obtained by hot-water extraction method. The infection of the extraction temperature, the extraction time, the solvent-solid ratio and the volume of solvent used during precipitation on the polysaccharides extraction ratio were studied. The extraction process was optimized by the uniform design (UD) method on the basis of the results of the single-factor experiments. The optimum extraction conditions of the mushroom polysaccharides (MP) were as follows: Extraction temperature was 100℃;extraction time was 3 hours;solvent-solid ratio was 20:1 and 5 times volume of 95% ethanol. The yield of polysaccharides of T. giganteum was 12.92%. 展开更多

Evaluation for human body whitening effcacy of cosmetics with extract from Tricholoma matsutake Sing as main effective constituent was conducted. Thirty female volunteers with age between 20 and 45 years old were kindly invited to use the cosmetic product on the inner side of their crank arm for a testing period of 8 weeks continuously. Values of MI, L* and ITA° of the skin were measured and the results display signifcant difference ( P〈0. 05) before and after the 8 weeks testing period. Duncan multiple comparisons showed that the whitening effcacy of cosmetic product formulated with the extract from Tricholoma matsutake Sing is signifcantly greater than that of the formulation matrix and equivalent with that of ascorbyl ethyl ether. Via the data collection and statistical analysis, this method can directly refect the whitening effect of active additives in cosmetics, and it is suitable for the evaluation of the whitening effcacy of cosmetics with additives. 展开更多

松茸和姬松茸含有多种活性成分,不但具有较高的营养价值,还具有良好的保健功效.为构建松茸和姬松茸挥发性成分保留时间神经网络的定量结构—保留关系模型,根据松茸和姬松茸挥发性化合物中原子的特性和连接状态,提出了一种新的分子结构参... 松茸和姬松茸含有多种活性成分,不但具有较高的营养价值,还具有良好的保健功效.为构建松茸和姬松茸挥发性成分保留时间神经网络的定量结构—保留关系模型,根据松茸和姬松茸挥发性化合物中原子的特性和连接状态,提出了一种新的分子结构参数(原子指数A),并计算了松茸和姬松茸挥发性成分的电性拓扑状态指数(Em)和电性距离矢量(Mm).选取电性拓扑状态指数的E_(2)和电性距离矢量的M_(15)和M_(21)等3种指数,结合原子指数A共4种结构参数,作为BP神经网络法的输入层变量,松茸挥发性成分的色谱保留时间作为输出层变量,采用4-8-1的网络结构,构建的模型总相关系数r为0.9919,根据该模型计算得到的保留时间预测值与文献实验值的平均误差为0.57;同样采用这4种参数,与姬松茸挥发性成分的保留时间进行研究,采用4-7-1的网络结构,所得模型总相关系数r为0.9929,保留时间预测值与实验值的平均误差为0.58.结果表明:—CH_(3),—CH_(2)—和氧氮杂原子等基团片段及连接方式是影响松茸和姬松茸挥发性成分色谱保留时间的主要因素. 展开更多

野生松茸的发生与其土壤的生态环境特点密切相关。试验以国家地理标志产品“南华松茸”为对象,比较菌塘土壤与非菌塘土壤的理化性质及可培养微生物数量,并基于高通量测序技术分析土壤微生物多样性及菌群结构差异,以明确菌塘的微生态区... 野生松茸的发生与其土壤的生态环境特点密切相关。试验以国家地理标志产品“南华松茸”为对象,比较菌塘土壤与非菌塘土壤的理化性质及可培养微生物数量,并基于高通量测序技术分析土壤微生物多样性及菌群结构差异,以明确菌塘的微生态区系特征。结果表明,菌塘土壤各理化参数高于非菌塘土,其中速效钾(含量增加151.22%)、锰(增加156.38%)、铜(增加105.88%)含量差异极显著,磷、锌、镁含量差异显著。菌塘土壤中主要三大微生物类群的数量与非菌塘相比差异极显著;高通量测序结果表明菌塘土壤微生物群落结构差异显著,优势细菌类群为卡氏伯克霍尔德菌属(Burkholderia-Caballeronia-Paraburkholderia)、分支杆菌属(Mycobacterium.)、酸杆菌属(Acidipila)、热酸菌属(Acidothermus);优势真菌类群为暗梗单孢霉属(Chloridium)、Sagenomella、Glarea、锁瑚菌科未分类的属unclassified-Clavulinaceae、口蘑属(Tricholoma)。 展开更多
