文摘目的比较蛇黄散与其改良剂型蛇黄凝胶的体外释放度与透皮特性。方法分别采用微孔滤膜与离体小鼠鼠皮及改进的Franz扩散池进行体外释放与透皮试验,以HPLC法定量测定蛇黄凝胶与蛇黄散中君药—蛇鳞草中triphyllin A与大黄中大黄酸的量为指标,测定2种制剂的体外释放量(Qs)、释放率(Q)、透皮量(Ls)与透皮率(L),并采用3种方程对蛇黄凝胶的体外释放与透皮结果进行模拟拟合。结果蛇黄凝胶中triphyllin A 24 h的Qs=72.1885μg·cm^(-2),Q=30.4583%,Ls=52.0867μg·cm^(-2),L=21.9768%,大黄酸24 h的Qs=15.2200μg·cm^(-2),Q=9.4290%,Ls=13.5456μg·cm^(-2),L=8.3917%,蛇黄散中triphyllin A 24 h的Qs=45.2395μg·cm^(-2),Q=17.1249%,Ls=27.0434μg·cm^(-2),L=10.2370%,大黄酸24 h的Qs=6.3763μg·cm^(-2),Q=1.9921%,Ls=5.8751μg·cm^(-2),L=1.8355%。结论改良后的蛇黄凝胶体外Qs、Q、Ls与L均优于蛇黄散,释放与透皮特征明显优于传统剂型蛇黄散,且蛇黄凝胶不易过敏,贴敷方便,便于临床使用;蛇黄凝胶释放与透皮模型均符合Higuchi方程,说明蛇黄凝胶体外释放和透皮均属于骨架型缓释制剂。
