Methods: Glioma C6 cells were divided into four groups: A group, treated without IGF-1; B group, treated with 0.1 ng/mL dose of IGF-1; C group, treated with 1 ng/mL dose of IGF-1; D group, treated with 10 ng/mL dose of IGF-1. PTTG mRNA was detected by reverse transcription polymerase chain reaction (RT-PCR), western blotting was used to detect the expression of PTTG protein. Results: The expressions of PTTG mRNA were 1.370 ± 0.212, 2.198 ± 0.354, 3.452 ± 0.332, and 4.576 ± 0.387 respectively in the four groups, and there was a significantly difference between any two groups (P < 0.01). The protein expressions of PTTG in the four groups were 1.407 ± 0.334, 1.813 ± 0.465, 2.412 ± 0.576, and 3.128 ± 0.665 respectively, and there was a significantly difference between any two groups (P < 0.01). Conclusion: IGF-1 can up-regulate the expression of PTTG significantly in dosage-dependent manner.展开更多

Methods: The protein expressions of PTTG and PCNA were detected by immunostaining assay using streptavidin-peroxidase (SP) method in 80 cases of glioma. Results: The positive rates of PTTG in grades Ⅰ-Ⅳ gliomas were 56.3%, 68.2%, 80.8%, and 100.0% respectively, and the protein expression of PTTG increased with the increasing of the pathological grade (X^2= 9.602, P 〈 0.05); The positive rates of PCNA protein were 37.5%, 54.5%, 69.2%, and 93.8% respectively, and the protein expression of PCNA increased with the increasing of the pathological grade (X2 = 12.147, P 〈 0.01). The expression of PTTG had positive correlation with the expression of PCNA protein (~s = 0.557, P 〈 0.01). Conclusion: The expressions of PTTG and PCNA proteins were related to malignant degree of glioma, and may cooperate with each other in the tumorigenesis and progression and can be considered as the indicators of the biological behaviors in glioma.展开更多
Objective To investigate the dynamic changes observed in serum levels of interleukins (ILs), tumor necrosis factor-α (TNF-α) and transforming growth factor-β1 (TGF-β1 ) in severe acute respiratory syndrome (SARS) ...Objective To investigate the dynamic changes observed in serum levels of interleukins (ILs), tumor necrosis factor-α (TNF-α) and transforming growth factor-β1 (TGF-β1 ) in severe acute respiratory syndrome (SARS) patients.Methods Sixty-one cases of SARS with positive antibodies to SARS coronavirus (SARS-CoV) were classified into the following categories: initial stage (3-7 days), peak stage (8-14 days), and remission and recovery stage (15 -27 days). Forty-four healthy individuals were used as controls. Serum levels of ILs, TNF-a and TGF-p, were measured in all subjects. Serum antibodies to SARS-CoV were detected only in SARS cases.Results The mean concentration of serum IL - 6 in SARS patients did not differ from that in the control group in initial and peak stages, but became significantly higher in remission and recovery stage compared with the control group, initial and peak stages ( P<0. 01). The mean concentration of serum IL-8 in SARS patients did not differ from that of the control group in initial stage, but was significantly higher than control group in peak stage and remission and recovery stage ( P < 0. 05). And it was more significantly higher in remission and recovery stage than in peak stage ( P<0. 01). The mean concentrations of IL-16 and TNF-αin SARS patients were higher than those of the control group for every length of the clinical courses investigated, and were especially high in remission and recovery stage (P<0. 01). The purpose of this study was to investigate the expression of pituitary tumor transforming gene(PTTG)and basicfibroblast growth factor(bFGF)in oral squamous cell carcinoma(OSCC)and tongue cancer cell line Tca8113,as w...The purpose of this study was to investigate the expression of pituitary tumor transforming gene(PTTG)and basicfibroblast growth factor(bFGF)in oral squamous cell carcinoma(OSCC)and tongue cancer cell line Tca8113,as well as their effects on each other.We detected PTTG protein and bFGF in OSCC tissues from 56 cases using the streptavidin-biotin peroxidase(S-P)method;additionally,after being treated with different concentrations of anti bFGF or PTTG antibody,PTTG or bFGF expression in Tca8113 was examined by immuno-cytochemistry.The results were as follows:(1)Positive rates of PTTG protein and bFGF were 78.2%and 67.3%in OSCC,respectively,which were significantly higher than those in normal mucosal tissues(P<0.05).PTTG protein was significantly up-regulated in poorly and moderately differentiated tumors compared to well differentiated tumors(P<0.05),and there was also a significant difference between tumors with lymph node metastasis and tumors without lymph node metastasis(P<0.05).PTTG protein expression was positively correlated with bFGF(r=0.382,P<0.05);(2)PTTG protein emitted strongfluorescence in Tca8113,and it decreased after being treated with anti-bFGF antibody.Anti-PTTG anti-body also had an inhibitive effect on bFGF expression.In summary,the overexpression of PTTG protein is closely related with OSCC differentiation and lymph node metastasis.PTTG protein expression conforms to bFGF in OSCC tissues and Tca8113 cells.Detection of both PTTG and bFGF may help to judge the degree of malignancy and prognosis of patients with OSCC.展开更多
