●AIM:To identify the differential methylation sites(DMS)and their according genes associated with diabetic retinopathy(DR)development in type 1 diabetes(T1DM)children.●METHODS:This study consists of two surveys.A to...●AIM:To identify the differential methylation sites(DMS)and their according genes associated with diabetic retinopathy(DR)development in type 1 diabetes(T1DM)children.●METHODS:This study consists of two surveys.A total of 40 T1DM children was included in the first survey.Because no participant has DR,retina thinning was used as a surrogate indicator for DR.The lowest 25%participants with the thinnest macular retinal thickness were included into the case group,and the others were controls.The DNA methylation status was assessed by the Illumina methylation 850K array BeadChip assay,and compared between the case and control groups.Four DMS with a potential role in diabetes were identified.The second survey included 27 T1DM children,among which four had DR.The methylation patterns of the four DMS identified by 850K were compared between participants with and without DR by pyrosequencing.●RESULTS:In the first survey,the 850K array revealed 751 sites significantly and differentially methylated in the case group comparing with the controls(|Δβ|>0.1 and Adj.P<0.05),and 328 of these were identified with a significance of Adj.P<0.01.Among these,319 CpG sites were hypermethylated and 432 were hypomethylated in the case group relative to the controls.Pyrosequencing revealed that the transcription elongation regulator 1 like(TCERG1L,cg07684215)gene was hypermethylated in the four T1DM children with DR(P=0.018),which was consistent with the result from the first survey.The methylation status of the other three DMS(cg26389052,cg25192647,and cg05413694)showed no difference(all P>0.05)between participants with and without DR.●CONCLUSION:The hypermethylation of the TCERG1L gene is a risk factor for DR development in Chinese children with T1DM.展开更多

In recent years,karst construction projects in the built-up area of Wuhan(capital of Hubei Province,China)are increasing,and the karst geological disasters have aroused social concerns.The actual engineering projects ...In recent years,karst construction projects in the built-up area of Wuhan(capital of Hubei Province,China)are increasing,and the karst geological disasters have aroused social concerns.The actual engineering projects usually use shallow geophysical exploration methods to explore karst.This paper uses Spatial Auto-Correlation Method(SPAC)and electromagnetic Computerized Tomography(CT)to detect karst in urban built-up areas.Depending on the different physical properties of rock and soil,the SPAC method can better reveal the interface between soil and rock strata and the interface between soil layers.The electromagnetic CT method can identify strata according to the apparent absorption coefficient,which can better reveal the interface between soil and rock,the interface between the more intact and weathered rock.The SPAC method is mainly qualitative to measure the low-speed area,namely,the wrong geological body i.e.,karst cave,but also can detect the fracture zone or filling mode of karst cave,and at the same time,cannot use exploration holes or logging observation.The electromagnetic CT method can accurately detect the location and scale of the karst caves and has a higher accuracy detecting karst bands.In addition,exploration holes or well logging observations are also expected to be conducted,and their detection effect is greatly affected by lithology.展开更多

Understanding of the molecular characteristics of the different histologic types of thym...Background The prognostic relevance of World Health Organization (WHO) subtypes within type B thymomas is still controversial. Understanding of the molecular characteristics of the different histologic types of thymomas will provide meaningful information for diagnosis and therapeutic management in type B thymoma. Methods Proteins extracted from twelve type B thymoma tissue specimens (six type B1 and six type B2) were analyzed by two-dimensional electrophoresis (2-DE) coupled with MALDI-TOF-MS. Differentially expressed proteins were then assayed in sixty-nine type B thymoma tissues (including B1, B2 and B3) by tissue array analysis with immunohistochemistry staining. The relationship of their expression with clinicopathological parameters, such as tumor stage or WHO classification, was estimated by Spearman's Rank Correlation Test. Results Sixteen differentially expressed proteins between type B1 and B2 thymoma tissues were identified. The differential levels of ezrin and glutathione S-transferase pi (GSTP1) were validated using immunohistochemistry staining. A statistically significant difference was observed in the positive rate of ezrin expression between type B1 thymoma and type B3 thymoma (Z= -2.963, P 〈0.01). Ezrin showed a tendency to be expressed in higher classification tumors from type B1 to B3. A statistical analysis demonstrated that type B2 and B3 tumors had significantly higher positive expression of GSTP1 than the B1 group (type B2 vs. BI: Z= -2.582, P 〈0.01; type B3 vs. BI: Z= -4.012, P 〈0.001). The results also showed a strong correlation between GSTP1 and WHO type staging of B1 to B3 tumors (Spearman's correlation coefficient: 0.633, P 〈0.001). Statistical analysis showed that there was close correlation between GSTP1 and ezrin expression with the clinical stage (Spearman's correlation coefficients, ezrin: 0.481, P 〈0.05; GSTPI: 0.484, P 〈0.01). Conclusions Differentially expressed proteins between type B1 and B2 thymoma tissues were analyzed by comparative proteomic analysis. The techniques of proteomic analysis and tissue array provide a potential tool for screening of key molecules in type B thymoma histological sub-classifications. Like many of the tropical islands, the population of Andaman and Nicobar Islands, though not directly, relies predominantly upon rain water harvesting to quench their need and also depends on the groundwater sources. ...Like many of the tropical islands, the population of Andaman and Nicobar Islands, though not directly, relies predominantly upon rain water harvesting to quench their need and also depends on the groundwater sources. In the background of climate change, severity of hydrological cycle is much anticipated which may cause more extreme and unusual precipitation. It is quite essential to have other alternatives. Accordingly, groundwater could be exploited as a potential alternative. The present study intends to find out the potential groundwater source and estimate aquifer parameters in Kodiyaghat (KD) and Burmanallah (BN). As these areas are composed of very hard rock, Wenner-Schlumberger array has been used to carry out a 2D Electrical Resistivity Tomography survey to find out the fracture zone as well as to delineate the aquifer. KD and BN show maximum resistivity of 25,416 Ωm and 5985 Ωm indicate very hard rock terrain. Similarly, the minimum values of resistivity (21.6 Ωm and 30.4 Ωm) were observed at KD and BN define the presence of freshwater aquifers respectively. The aquifer identified was found to be at a depth of 5 m to 19.9 m at KD and 2.5 m to 20 m at BN. The calculated Hydraulic conductivity (14.85 m/day and 30.14 m/day), transmissivity (86.25 m2/day and 271.27 m2/day) and porosity (28.7% and 31.24%) values at KD and BN confirmed that, the located aquifer was of fresh ground water quality and can be utilized for drinking and house hold purposes. According to the results, almost 70% of the study area is hard rock terrain and 30% comes under potential aquifer zone. The results also show that, both the areas were characterized by Horst and Graben topography and suggest possible groundwater sources for future exploration.展开更多
Purpose-The purpose of this paper is to propose radiating system by avoiding electromagnetic interference in unwanted directions and to radiate the energy in the required direction with an optimization technique.Desig...Purpose-The purpose of this paper is to propose radiating system by avoiding electromagnetic interference in unwanted directions and to radiate the energy in the required direction with an optimization technique.Design/methodology/approach-Practically,multiple,incompatible variables require concurrent boost on a synthesis of systematic antenna assemblage.The authors have worked out the main statistic penalty function to ensure all the restrictions.Here,MBPSO(Modified Binary Particle Swarm Optimization)is developed and introduced thin planar synthesis restriction.The sigmoid function is used to update the particle position.Different analytical demonstrations have been carried out,and the exhibited methods are predominant than the algorithms.Findings-A 20310 planar antenna array is synthesized using modified BPSO.The authors have suppressed the PSLL in two principal planes and as well as in the entire f plane.Numerical results state that MBPSO outperforms the other binary BPSO,BCSO,ACO,RGA,GAoptimization techniques.MBPSO achieved a51.84 dB PSLL level,whereas BPSO achieved48.57 dB with the same 50%thinning.Originality/value-Planar array antenna formation is one of the most complex syntheses because the array gets filled with more antenna elements.The machine-like complication and implementation of such an antenna arrangement with a broad opening would be expensive.It is not easy to control the required radiation patterns shape by using a uniform feeding network.To get better flexibility for sustaining the sidelobe levelheaded along with consistent amplitude distribution.So as far as prominence has been given to the evolutionary algorithm,find an ideal solution for objective array combinational problems.展开更多
OBJECTIVE: To investigate the clinical differentiation of spleen-deficiency pattern(SDP), a group of symptoms and signs defined in terms of Traditional Chinese Medicine for its clinical practice.METHODS: Peripheral ve...OBJECTIVE: To investigate the clinical differentiation of spleen-deficiency pattern(SDP), a group of symptoms and signs defined in terms of Traditional Chinese Medicine for its clinical practice.METHODS: Peripheral venous blood(> 3 m L) was collected from each of six type 2 diabetes mellitus(T2DM)-SDP patients and six healthy volunteers. After the isolation of peripheral white blood cells(PWBCs), total RNA was extracted, and quality control was performed on all RNA samples. Microarray experiments were conducted using the Agilent human whole genome gene chip, and genes demonstrating differential expression were screened. Bioinformatics analysis was conducted on these genes using several online databases.RESULTS: We screened a total of 175 differentially expressed genes(DEGs), of which 111(63%) were down-regulated and 64(37%) were up-regulated in T2DM-SDP patients compared with healthy controls. Among the 175 genes, 158 had biological function annotations: 46(29%) were directly related to an individual's immune regulation or response, 25(16%) were associated with substance and energy metabolism of PWBCs which could also indirectly influence immunity, and the remaining87(55%) were involved in a variety of PWBC biological processes that might eventually influence the immune function. Kyoto Encyclopedia of Genes and Genomes pathway enrichment analysis revealed that the DEGs were predominantly enriched in seven immune-related pathways. Hierarchical cluster analysis identified gene expression patterns that were distinguishable between the two study groups.CONCLUSION: Our results suggest that T2DM-SDP patients experience significant hypoimmunity and/or immune dysfunctions, and possess a specific gene expression profile. These findings offer new insights into SDP and the clinical pattern differentiation of T2DM-SDP.展开更多

The auditory brainstem implant(ABI)was originally developed to provide rehabilitation of retrocochlear deafness caused by neurofibromatosis type 2(NF2).Recent studies of the ABI have investigated outcomes in non-NF2 c...The auditory brainstem implant(ABI)was originally developed to provide rehabilitation of retrocochlear deafness caused by neurofibromatosis type 2(NF2).Recent studies of the ABI have investigated outcomes in non-NF2 cohorts,such as patients with cochlear nerve aplasia or cochlear ossification and more recently,intractable tinnitus.New technologies that improve the ABI-neural tissue interface are being explored as means to improve performance and decrease side effects.Innovative discoveries in optogenetics and bioengineering present opportunities to continually evolve this technology into the future,enhancing spatial selectivity of neuronal activation in the cochlear nucleus and preventing side effects through reduction in activation of non-target neuronal circuitry.These advances will improve surgical planning and ultimately improve patients1 audiological capabilities.ABI research has rapidly increased in the 21st century and applications of this technology are likely to continually evolve.Herein,we aim to characterize ongoing clinical,basic science,and bioengineering advances in ABIs and discuss future directions of this technology.展开更多

After the isolation of peripheral white blood cells(PWBCs), total RNA was extracted, and quality control was performed on all RNA samples. Microarray experiments were conducted using the Agilent human whole genome gene chip, and genes demonstrating differential expression were screened. Bioinformatics analysis was conducted on these genes using several online databases.RESULTS: We screened a total of 175 differentially expressed genes(DEGs), of which 111(63%) were down-regulated and 64(37%) were up-regulated in T2DM-SDP patients compared with healthy controls. Among the 175 genes, 158 had biological function annotations: 46(29%) were directly related to an individual's immune regulation or response, 25(16%) were associated with substance and energy metabolism of PWBCs which could also indirectly influence immunity, and the remaining87(55%) were involved in a variety of PWBC biological processes that might eventually influence the immune function. Kyoto Encyclopedia of Genes and Genomes pathway enrichment analysis revealed that the DEGs were predominantly enriched in seven immune-related pathways. Hierarchical cluster analysis identified gene expression patterns that were distinguishable between the two study groups.CONCLUSION: Our results suggest that T2DM-SDP patients experience significant hypoimmunity and/or immune dysfunctions, and possess a specific gene expression profile. These findings offer new insights into SDP and the clinical pattern differentiation of T2DM-SDP.

文摘The auditory brainstem implant(ABI)was originally developed to provide rehabilitation of retrocochlear deafness caused by neurofibromatosis type 2(NF2).Recent studies of the ABI have investigated outcomes in non-NF2 cohorts,such as patients with cochlear nerve aplasia or cochlear ossification and more recently,intractable tinnitus.New technologies that improve the ABI-neural tissue interface are being explored as means to improve performance and decrease side effects.Innovative discoveries in optogenetics and bioengineering present opportunities to continually evolve this technology into the future,enhancing spatial selectivity of neuronal activation in the cochlear nucleus and preventing side effects through reduction in activation of non-target neuronal circuitry.These advances will improve surgical planning and ultimately improve patients1 audiological capabilities.ABI research has rapidly increased in the 21st century and applications of this technology are likely to continually evolve.Herein,we aim to characterize ongoing clinical,basic science,and bioengineering advances in ABIs and discuss future directions of this technology.
