Bone marrow niche-mediated survival of leukemia stem cells in acute myeloid leukemia: Yin and Yang

Acute myeloid leukemia （AML） is characterized by the accumulation of circulating immature blasts that exhibit uncontrolled growth, lack the ability to undergo normal differentiation, and have decreased sensitivity to apoptosis. Accumulating evidence shows the bone marrow （BM） niche is critical to the maintenance and retention of hematopoietic stem cells （HSC）, including leukemia stem cells （LSC）, and an increasing number of studies have demonstrated that crosstalk between LSC and the stromal cells associated with this niche greatly influences leukemia initiation, progression, and response to therapy. Undeniably, stromal cells in the BM niche provide a sanctuary in which LSC can acquire a drug-resistant phenotype and thereby evade chemotherapy- induced death. Yin and Yang, the ancient Chinese philosophical concept, vividly portrays the intricate and dynamic interactions between LSC and the BM niche. In fact, LSC-induced microenvironmental reprogramming contributes significantly to leukemogenesis. Thus, identifying the critical signaling pathways involved in these interactions will contribute to target optimization and combinatorial drug treatment strategies to overcome acquired drug resistance and prevent relapse following therapy. In this review, we describe some of the critical signaling pathways mediating BM niche-LSC interaction, including SDFI/CXCL12, Wnt/β-catenin, VCAM/VLA-4/NF-κB, CD44, and hypoxia as a newly-recognized physical determinant of resistance, and outline therapeutic strategies for overcoming these resistance factors.

Gamma-aminobutyric acid(GABA)and glutamate are two important amino acid neurotransmitters widely present in the nervous systems of mammals,insects,round worm,and platyhelminths,while their receptors are quite diversified across different animal phyla.However,the evolutionary mechanisms between the two conserved neurotransmitters and their diversified receptors remain elusive,and antagonistic interactions between GABA and glutamate signal transduction systems,in particular,have begun to attract significant attention.In this review,we summarize the extant results on the origin and evolution of GABA and glutamate,as well as their receptors,and analyze possible evolutionary processes and phylogenetic relationships of various GABAs and glutamate receptors.We further discuss the evolutionary history of Excitatory/Neutral Amino Acid Transporter(EAAT),a transport protein,which plays an important role in the GABA-glutamate“yin and yang”balanced regulation.Finally,based on current advances,we propose several potential directions of future research.

Influence of Moxibustion of Complementary Acupoints on Analgesic Effect and Mast Cell Function in CIA Rats

[Objectives]To explore the moxibustion effect and mechanism of"Complementary Acupoints"on rheumatoid arthritis(RA).[Methods]A total of 32 Wistar rats were randomly divided into 4 group:normal control(K),model group(M group),"the Complementary Acupoints"Yanglingquan and Yinlingquan group(AB group),Shenshu and Zusanli group(CD group),with 8 rats in each group.Moxibustion was performed using refined moxa floss(2 mg/pc)on acupoints in both sides every other day,5 pieces for each acupoint,and implemented for 10 times.Before and after the modeling and after the intervention,the left foot sole thermal pain threshold of each group was measured.After the intervention,the rats'left paws were examined by X-ray to detect the number of mast cells(MCs)and degranulation in the affected area of the foot.[Results]The thermal pain threshold of the model group was lower than that of the K group(P<0.05).The number of MC in the affected area and the degranulation were increased.After the moxibustion intervention,the thermal pain threshold of the AB and CD groups was higher than that of the M group(P<0.05);the number of MCs and degranulation were decreased compared with M group,and the degree of reduction was better than that of CD group.The data of foot and claw showed that the foot and paw soft tissue swelling,bone and joint deformity and bone destruction were better in AB and CD groups were more relieved than that in M group.[Conclusions]The moxibustion of Complementary Acupoints can effectively intervene with RA of rats,and its mechanism may be closely related to the MC degranulation.

Clinical application of Bushen Jianpi Gutai Decoction in threatened abortion

Objective:To discuss the clinical application of the"seven,five and three"odd-numbered law theory and analyze the effect on serum sex hormone level.Methods:A total of 100 patients with early threatened abortion admitted from January 2021 to October 2021 were randomly divided into study group and control group,with 50 patients in each group.Patients in the control group were treated with dydrogesterone tablets,while patients in the study group were treated with Bushen JianpiGutai Decoction combined with dydrogesterone tablets.Clinical efficacy and progesterone level before and after treatment were observed and compared between the two groups,as well as clinical symptom relief time of the two groups,including hemostasis time,abdominal pain relief time and lumbar acid relief time,and sex hormone level of the two groups were observed and compared.Results:The clinical efficacy was significantly better than control patients(P<0.05),no significant difference in progesterone level comparison before treatment(P>0.05),significantly better than control patients(P<0.05),hemostasis,abdominal pain remission and waist acid remission were significantly shorter than control patients(P<0.05),and both significantly better sex hormone levels after treatment(P<0.05).Conclusion:Traditional Chinese medicine has certain advantages in the treatment of threatened abortion,odd-numbered law theoryguidesthe regulation of Yin and Yang in the female menstrual cycle and reproductive function is mainly the regular change of Yin and Yang.Under the guidance of"seven,five,three"odd-numbered law theoryof Traditional Chinese medicine master Xia Guicheng,master the key date and the best time of its transformation,regulate Yin and Yang in the female body,to prevent the occurrence and development of menstrual diseases and adverse childbirth.Bushen Jianpi Gutai decoction can effectively treat threatened abortion,significantly improve the clinical symptoms,improve the level of serum sex hormone,worthy of clinical promotion.

Nature has endowed gaseous molecules such as O_(2),CO_(2),CO,NO,H2 S,and N2 with critical and diverse roles in sustaining life,from supplying energy needed to power life and building blocks for life ’s physical structure to mediating and coordinating cellular functions.In this article,we give a brief introduction of the complex functions of the various gaseous molecules in life and then focus on carbon monoxide as a specific example of an endogenously produced signaling molecule to highlight the importance of this class of molecules.The past twenty years have seen much progress in understanding CO’s mechanism(s) of action and pharmacological effects as well as in developing delivery methods for easy administration.One remarkable trait of CO is its pleiotropic effects that have few parallels,except perhaps its sister gaseous signaling molecules such as nitric oxide and hydrogen sulfide.This review will delve into the sophistication of CO-mediated signaling as well as its validated pharmacological functions and possible therapeutic applications.

Detwmination of Blood Flow in the Lower Leg of Patients withDiabetes Mellitus and the Effects of Treatment with thePrinciple of Vitalizing Blood and Solubilizing Thrombus

Blood flow in the lower leg was determined in 50 cases of non-insulin dependent diabetesmellitus using an XLJ-2 bipolar rheoencephalometry impedance rheogram. In patients with leg pain (85legsi and diabetic foat (15 legs3, the blood flow of the lower leg was decreased as compared withhealthy legs (P<0. 01￣0. 001) . The group with diabetic foot showed a bigger decrease compared withthe leg pain group (PMO. 051 . Atter treatment with the principle of vitalizing blood and solubilizing throm-bus, 22 cases with leg pain (32 legs) showed a significant increase of blood flow in the lower leg (P<0. 001). The group with diabetic foot failed to show any significant increase, indicating the importance ofearly treatment. This method of determining blood flow in the lower leg will help early discovery of abnor-mal changes of blood supply in the lower leg. Treatment with the principle of vitalizing blood and solubiliz-ing thrombus can help improve the conditions of the patients.

Discussion about the Treatment of Alzheimer's Disease with Yisui Fuyong Decoction Based on Synaptic Plasticity
