The fatstoring cells were confirmed by the presentation of the lipid drop in the cytoplasm and the visualization of dismin with anti-dismin antibody by using indirect immunofluorescence method. By the videotape recorder (VIR) and the imagine analysis system, we observed the wandering immigration, the abrupt contraction of fat-storing cells with spike, then becoming a ball-like shape in its division phase. After that the cell began to extend and the contraction of these cells can be induced by the presence of 10-2 mmol/L endothelin-1 , 1 mmol/L of substance P and 2×10-5 mmol/L noradrenalin. After removal of these agents the contracted cells would become extended. All these findings indicate that the fat-storing cells have ability of contraction and movement.展开更多

The color of the bacterial colony on 2216E medium was from coccineus to rose bengal. Optimum culture conditions were sodium chloride concentration(g/dm3),10~30;pH,3~8;temperature,25~28℃;tryptone and yeast extract as nitrogen sources and glucose as carbon source. Under optimum conditions, pigment accumulation started after 12 h,reaching a maximum rate of synthesis at 36 h.展开更多

The axillary buds of its dormant branches were used as explants. A fourfactor and four-level orthogonal test was designed in order to choose the best differentiation medium for providing the technical support of Lonicera edulis micropropagation. The results showed that the culture medium and concentration of 6-BA were the main factors, and the best differentiation condition was MS culture medium containing 2.0 mg · L-1 6-BA, 0.3 mg · L-1 IBA and 1.5 mg · L-1 GA3.展开更多

Assurance of genetic stability over long term maintenance of hPSCs is pivotal in this endeavor, but hPSCs can adapt to life in culture by acquiring non-random genetic changes that render them more robust and easier to grow. In separate studies between 12.5% and 34% of hPSC lines were found to acquire chromosome abnormalities over time, with the incidence increasing with passage number. The predominant genetic changes found in hPSC lines involve changes in chromosome number and structure (particularly of chromosomes 1, 12, 17 and 20), remi- niscent of the changes observed in cancer cells. In this review, we summarize current knowledge on the causes and consequences of aneuploidy in hPSCs and highlight the potential links with genetic changes observed in human cancers and early embryos. We point to the need for comprehensive characterization of mechanisms underpinning both the acquisition of chromosomal abnormalities and selection pressures, which allow mutations to persist in hPSC cultures. Elucidation of these mechanisms will help to design culture conditions that minimize the appearance of aneuploid hPSCs. Moreover, aneuploidy in hPSCs may provide a unique platform to analyse the driving for- ces behind the genome evolution that may eventually lead to cancerous transformation.展开更多
