Objective:To evaluate the alteration of chemical behavior of L-ascorbic acid(vitamin C) with metal ion(nickel) at different pH solutions in vitro.Methods:Spectra of pure aqueous solution of L-ascorbic acid(E mark) com...Objective:To evaluate the alteration of chemical behavior of L-ascorbic acid(vitamin C) with metal ion(nickel) at different pH solutions in vitro.Methods:Spectra of pure aqueous solution of L-ascorbic acid(E mark) compound and NiSO_4(H_2O)(sigma USA) were evaluated by UV visible spectrophotometer.Spectral analysis of L-ascorbic acid and nickel at various pH(2.0, 7.0,7.4 and 8.6) at room temperature of 29℃ was recorded.In this special analysis,combined solution of L-ascorbic acid and nickel sulfate at different pH was also recorded.Results:The result revealed that λ_(max)(peak wavelength of spectra) of L-ascorbic acid at pH 2.0 was 289.0 run whereas at neutral pH 7.0,λ_(max) was 29S.4 run.In alkaline pH 8.6,λ_(max) was 295.4 nm and at pH 7.4 the λ_(max) of L-ascorbic acid remained the same as 295.4 nm.Nickel solution at acidic pH 2.0 was 394.5 nm,whereas at neutral pH 7.0 and pH 7.4 were the same as 394.5 nm.But at alkaline pH 8.6,λ_(max) value of nickel sulfate became 392.0 nm.The combined solution of L-ascorbic acid and nickel sulfate(6 mg/mL each) at pH 2.0 showed 292.5 nm and 392.5 nm,respectively whereas at pH 7.0,L-ascorbic acid showed 296.5 nm and nickel sulfate showed 391.5 nm.At pH 7.4,L-ascorbic acid showed 297.0 nm and nickel sulfate showed 394.0 nm in the combined solution whereas at pH 8.6(alkaline) L-ascorbic acid and nickel sulfate were showing 297.0 and 393.5 nm,respectively. Conclusions:Results clearly indicate an altered chemical behavior of L-ascorbic acid either alone or in combination with nickel sulfate in vitro at different pH.Perhaps oxidation of L-ascorbic acid to L-dehydro ascorbic acid via the free radical(HSc*) generation from the reaction of H,ASc + Ni(Ⅱ) is the cause of such alteration of λ_(max),value of L-ascorbic acid in the presence of metal nickel.展开更多
Western diet(rich in highly refined sugar and fat)can induce a range of metabolic dysfunctions in animals and humans,including neuroinflammation and cognitive function decline.Neuroinflammation and cognitive impairmen...Western diet(rich in highly refined sugar and fat)can induce a range of metabolic dysfunctions in animals and humans,including neuroinflammation and cognitive function decline.Neuroinflammation and cognitive impairment,two critical pathological characteristics of Alzheimer’s disease,have been closely associated with microbial alteration via the gut-brain axis.Thus,the present study aimed to investigate the influence of 2-O-β-D-glucopyranosyl-L-ascorbic acid(AA-2βG)isolated from the fruits of Lycium barbarum on preventing the high-fructose diet(HFrD)induced neuroinflammation in mice.It was found that AA-2βG prevented HFr D-induced cognitive deficits.AA-2βG also predominantly enhanced the gut barrier integrity,decreased lipopolysaccharide entry into the circulation,which subsequently countered the activation of glial cells and neuroinflammatory response.These beneficial effects were transmissible by horizontal fecal microbiome transplantation,transferring from AA-2βG fed mice to HFr D fed mice.Additionally,AA-2βG exerted neuroprotective effects involving the enrichment of Lactobacillus and Akkermansia,potentially beneficial intestinal bacteria.The present study provided the evidence that AA-2βG could improve indices of cognition and neuroinflammmation via modulating gut dybiosis and preventing leaky gut.As a potential functional food ingredient,AA-2βG may be applied to attenuate neuroinflammation associated with Western-style diets.展开更多

Ascorbi...The aim of this study was to investigate the effect of storage conditions on the sensory quality, colour and texture of fresh-cut cabbage during the addition of ascorbic acid, citric acid and calcium chloride. Ascorbic acid maintained the overall quality for 14 days at 0℃ and 7 days at 5℃;no difference, however, was observed regarding browning of cut surface compared to the control sample at both storage temperatures. Calcium chloride maintained the overall quality and cut surface browning for 14 days at both storage temperatures. It was also found that citric acid 1% can be used for minimally processed cabbage. Soaking with citric acid helped retain the color and increased the overall acceptance and organoleptic quality of fresh cut cabbage;it reduced browning of the cut surface and protected against formation of black specks. Citric acid treatment combined with low temperature storage (0℃) prolonged the shelf life of minimally processed cabbage for 22 days, time sufficient for acceptable marketing of the product. The lightness of minimally processed cabbage decreased linearly from 70.94 ± 6 to 63.8 ± 8.5 - 61.3 ± 8 units for the chemical treatments during 22 days of storage at 0℃. Hue angle values during storage time were also significantly influenced by chemical treatments mainly at 0℃.展开更多

The conversion efficiency of the cell, fill factor and the cell performance were observed 1.0257%, 0.2598% and 170.0 minutes in dark respectively. These data suggested that Asc2P played an important role in the regulation of porcine albumin mRNA level. b). The enhancing effect of Asc2P on ammonium metabolic activity was also observed in porcine, but not rat, mature hepatocytes. The porcine ornithine transcarbamylase (OTC) and arginase mRNAs were augmented by Asc2P, indicating that Asc2P had a direct effect on the urea cycle. c). The porcine collagen type I and type III mRNA, but not type XII mRNA, were detected as well, sugessting that Asc2P did not have the effect on the non-parenchymal hepatocytes to induce collagen type I and III mRNA expression. d). Our RT-PCR analysis demonstrated that the porcine hepatocytes expressed the sodium-ascorbate co-transporters SVCT1 and SVCT2, however, the intensities of porcine sodium-ascorbate co-transporters SVCT1 and SVCT2 bands were not changed markedly. These findings indicated that the Asc2P had no effect on SVCT1 and SVCT2 mRNA expression. e). The enhancing effect of Asc2P on porcine albumin mRNA was inhibited by staurosporine, a portein kinase inhibitor. We conclude that the enhanced albumin mRNA by Asc2P might be due to activation of tyrosine protein kinase and/or PKC and the Asc2P enhanced porcine albumin mRNA mainly at the transcriptional step.展开更多

Results from a lab study on the effect of sucrose (5 g per cup, 150 mL) and/or ascorbic acid (2 mL per cup, 150 mL) on dissolved aluminum compounds during the infusion of two commercial types of dry tea leaves (black, green) with boiling water (5, 15 min infusion time) are presented. Factors influencing the presence of dissolved aluminum in the infusions of both tea leaves were infusion time and sugar contents, as well as the interaction between ascorbic acid and sucrose (p < 0.05). Aluminum contents found after 15 min of infusion were 0.7 mg L–1 for black tea infusions added with sugar, and 0.69 mg L–1 for green tea added with both sugar and ascorbic acid. Both concentrations are higher than the level accepted in Mexico for drinking water (there is no act concerning tea infusions), that is 0.2 mg L–1.展开更多

Topical AA was applied to both patients at least one month before and after with a frequency of four times per day. After the surgery, both eyes improved best-corrected visual acuity(BCVA) and there was limited human corneal endothelial cell loss without signs of corneal endothelial decompensation, such as deteriorated BCVA or persistent corneal edema during the follow-up of at least two years.CONCLUSION Perioperative administration of topical AA may be an alternative therapy to the triple procedure in patients expecting to undergo cataract surgery.展开更多

Using this technique, the catalytic ability of cobalt hexacyanoferrate(CoHCF) microcrystalline toward AA oxidation was also studied. It was found that the dispersed CoHCF powder in the PEG paste can generate well shaped thin layer cyclic voltammetric waves with the peak height proportional to the scan rate, corresponding to the Fe centered redox reactions. This oxidation step catalyzed the AA oxidation. Also, this pasted CoHCF powder generated well resolved in situ MFTIRs spectra, by which a chemical interaction between CC bond of AA ring and CoHCF lattice was revealed. A corresponding surface docking mechanism for the catalytic reaction has been proposed.展开更多

The fabrication process involved a previous electropolymerization of aspartic acid and followed by the eletrodeposition of gold nano-particles on the glassy carbon electrode. The resulting poly (aspartic acid)-nanogold modified electrode (PAA- nano-Au/GCE) was characterized by scanning electron microscopy (SEM) and electrochemical impedance spectros-copy (EIS). A higher catalytic activity was obtained to electrocatalytic oxidation of dopamine (DA), ascorbic acid (AA) and uric acid (UA) due to the enhanced peak current and well-defined peak separations compared with three, bare GCE, PAA/GCE and nano-Au/GCE. Simultaneous determination of DA, AA, and UA were studied by voltammetry. The linear range of 5.0 × 10-7 - 1.0 × 10-4 M for DA, 5.0 × 10-6 - 2.0 × 10-3 M for AA and 5.0 × 10-6 - 1.0 × 10-3 M for UA was obtained. The detection limit was calculated for DA, AA and UA as being 6.5 × 10-8 M, 5.6 × 10-7 M and 3.0 × 10-7 M, respectively (S/N = 3). The practical application of the present modified electrode was demonstrated by the determination of DA, AA and UA in calf serum and fetal calf serum samples.展开更多

CO2 release through reaction of sodium carbonate and oxalic acid, created deoxygenated atmosphere for reduction of dye which greatly boosted the reaction rate. Initially slow reaction in presence of atmospheric oxygen proceeded rapidly when sodium carbonate was added. The reaction followed fractional order kinetics with AA and zero order kinetics with MG. The rate of reaction shows no linear dependence on [H+] concentration as an acidic medium. The rate of reaction is directly related with the elevated concentration of salt, which suggests that the two same charged species are involved in the rate determining step. Secondary reactions at elevated temperature showed complex kinetics.展开更多

Methods: The testicular △~5, 3β-hydroxysteroid dehydrogenase (HSD), 17β-HSD, peroxidase and catalase activities along with the levels of malondialdehyde (MDA) and conjugated dienes in testicular tissue were measured for the evaluation of testicular oxidative stress. The plasma testosterone (T) level was measured by immunoassay. Various germ cells at stage Ⅶ of spermatogenic cycle were quantified from testicular stained sections. Results: Cyclophosphamide treatment results in a significant inhibition in the testicular △~5, 3β-HSD and 17β-HSD activities, a decrease in plasma T level and a diminution in the counts of various germ cells. Moreover, this treatment was also associated with a significant inhibition of the peroxidase and catalase activities along with high levels of MDA and conjugated dienes in the testis. All these changes were reversed by ascorbic acid co-administration. Conclusion: Cyclophosphamide treatment at the dosage used caused testicular gametogenic and androgenic disorders as well as induced testicular oxidative stress that can be reversed by ascorbic acid co-administration.展开更多
Background: Laying hens over 75 weeks of age commonly show great declines in immunity and production performance.It is unclear whether these declines can be relieved by supplementing with ascorbic acid(AA) in feed.Two...Background: Laying hens over 75 weeks of age commonly show great declines in immunity and production performance.It is unclear whether these declines can be relieved by supplementing with ascorbic acid(AA) in feed.Two trials were conducted to investigate the synthesis and metabolism of AA in layers of different ages and the effects of dietary supplemental AA on the performance and the immune and antioxidant statuses of 78 weeks old hens.Methods: In Exp.1,equal numbers(24 hens) of 35 weeks old(Young) and 75 weeks old(Old) layers were fed the same diet without AA supplementation for 4 weeks.In Exp.2,360 healthy 78 weeks old laying hens were randomly assigned to 4 treatments(basal diet supplemented with 0,0.25,0.5,or 1 g AA/kg diet) in an 8-week feeding trial.Results: The old hens tended to have decreased L-gulonolactone oxidase(GLO) synthase activity in the kidney and liver than that of the young hens(P = 0.07 and P = 0.05,respectively).Compared with the young hens,the old hens had lower hepatic antioxidant capacity allowing for the lower thioredoxin(TXN),thioredoxin reductase(TXNR) and cytochrome b5 reductase(CYB5 R) gene expression(P < 0.05),whereas increased sodium-dependent vitamin C transporter(SVCT) 1 expression levels in the ileum and kidney and enhanced splenic and hepatic AA concentrations(P < 0.05).Dietary supplementation with AA significantly decreased GLO enzyme activity but increased splenic AA concentration and anti-bovine serum albumin IgG levels(P < 0.05) and tended to increase CD4+T lymphocyte numbers(P = 0.06) in serum.Supplementation of 0.25 g AA/kg diet significantly increased hepatic total antioxidant capacity(T-AOC,P < 0.05) relative to the control group.Conclusions: Laying hens could synthesize AA in both the kidney and the liver,though the GLO enzyme activities were 100 times greater in kidneys than in livers.The old laying hens had greater absorption and reabsorption capacity and higher AA retention in some tissues that did the young hens.Dietary supplementation of AA can improve the health of old layers by enhancing immunity and antioxidant capacity.展开更多
Actinidia eriantha is one of the species of kiwifruit with a particularly high ascorbic acid (AsA) content.However, the molecular mechanism driving AsA richness in fruit remains unclear.In order to reveal the molecula...Actinidia eriantha is one of the species of kiwifruit with a particularly high ascorbic acid (AsA) content.However, the molecular mechanism driving AsA richness in fruit remains unclear.In order to reveal the molecular mechanism of AsA richness in A. eriantha, this study constructed a regulatory network related to AsA metabolism by combining genomics,metabolomics and transcriptomics.We assembled a high-quality genome of A. eriantha ‘Ganlv 1’ with only five remaining gaps.The assembly is comprised of 29 pseudochromosomes with a total size of 615.95 Mb, and contig N50 of 20.35 Mb.Among them,24 of the pseudochromosomes were obtained directly from telomere-to-telomere.The LTR assembly index score and consensus quality value were 21.34 and 39.90%, respectively.Subsequently, 61 metabolites and 2 092genes were found to be differentially accumulated/expressed during fruit development by metabolome and transcriptome assays, respectively.AsA metabolism and the cyclic regeneration pathway were found to have high expression levels throughout fruit growth and development,suggesting its crucial role in the regulation of AsA.Furthermore,the AsA contents are highly associated with ascorbate peroxidase genes.The genome obtained in this study provides genomic resources for the genetic and breeding research ofA. eriantha, and the constructed regulatory network can provide a public data platform for future research on kiwifruit.展开更多
To counteract the effects of drought stress,scientists have adopted several approaches including the use of different chemicals both inorganic and organic,which is contemplated as a highly efficient and cost-effective...To counteract the effects of drought stress,scientists have adopted several approaches including the use of different chemicals both inorganic and organic,which is contemplated as a highly efficient and cost-effective shot-gun approach.Ascorbic acid(AsA)is a potential organic substance,which widely occurs in plants,and is considered to be an effective antioxidant to counteract reactive oxygen species(ROS).Thus,a pot experiment was performed to assess the relative mitigating impacts of synthetic AsA and naturally occurring AsA in the form of lemon juice(LJ)and orange juice(OJ)on two cultivars of okra(Abelmoschus esculentus L.)namely Sabz Pari and Bhindi Sanwali under varying water deficit conditions.After 30 days of seed germination,okra seedlings were subjected to different irrigation regimes,i.e.,water deficit stress[(65%and 50%F.C.)and control conditions(100%F.C.)].Different levels of AsA[control(no spray),14 mg L^(−1)LJ,24 mg L^(−1)OJ and 150 mg L^(−1)AsA]obtained from different sources were applied as a foliar spray to control and water-stressed plants.Drought stress prominently reduced plant growth and yield attributes of the okra cultivars.Water-deficit conditions(65%and 50%F.C.)substantially decreased the fruit chlorophyll(a,b)pigments and the activity of superoxide dismutase(SOD)enzyme,while an increase was observed in the contents of fruit’s hydrogen peroxide(H_(2)O_(2)),malondialdehyde(MDA),total phenolics,total soluble sugars,AsA,and total soluble proteins.Drought stress also increased the activities of antioxidant enzymes like peroxidase(POD)and catalase(CAT).However,plant growth and yield attributes,fruit chlorophyll pigments,total phenolics,total soluble sugars,total free amino acids,total soluble proteins,AsA,GB,H_(2)O_(2),and the activities of antioxidant enzymes(POD and CAT)were increased by the AsA exogenous treatment in both okra cultivars under water deficit and control conditions.Overall,LJ and OJ were more effective than the synthetic AsA in upregulating the physiological and metabolic processes of okra plants.So,cost-effective as well as multi-nutrient natural sources of AsA could be suggested for alleviating the harmful effects of water deficit stress on plants.展开更多
BACKGROUND A low-volume polyethylene glycol(PEG) solution that combines ascorbic acid with PEG-based electrolyte solution(PEG-ASC) is gaining mainstream acceptance for bowel preparation due to reduced volume and impro...BACKGROUND A low-volume polyethylene glycol(PEG) solution that combines ascorbic acid with PEG-based electrolyte solution(PEG-ASC) is gaining mainstream acceptance for bowel preparation due to reduced volume and improved taste.Although several reports showed that bowel preparation with PEG-ASC volume lower than 2.0 L with laxative agents could be an alternative to traditional preparation regimen, the cleansing protocols have not been fully investigated.AIM To evaluate the cleansing efficacy of 1.2 L PEG-ASC solution comparing with 2.0 L PEG electrolyte(PEG-ELS) for bowel preparations.METHODS A randomized, single-blinded, open-label, single-center, non-inferiority study was conducted. In total, 312 Japanese adult patients(aged > 18 years) who underwent colonoscopy were enrolled. Patients were randomly allocated to bowel lavage with either 1.2 L of PEG-ASC solution with at least 0.6 L of an additional clear fluid(1.2 L PEG-ASC group) or 2.0 L of PEG-ELS(PEG-ELS group). Then, 48 mg of sennoside was administered at bedtime on the day before colonoscopy, and the designated drug solution was administered at the hospital on the day of colonoscopy. Bowel cleansing was evaluated using the Boston Bowel Preparation Scale(BBPS). The volume of fluid intake and required time for bowel preparation were evaluated. Furthermore, compliance, patient tolerance,and overall acceptability were evaluated using a patient questionnaire, which was assessed using a visual analog scale.RESULTS In total, 291 patients(1.2 L PEG-ASC group, 148; PEG-ELS group, 143) completed the study. There was no significant difference in successful cleansing, defined as a BBPS score ≥ 2 in each segment, between the two groups(1.2 L PEG-ASC group, 91.9%; PEG-ELS group, 90.2%; 95%CI:-0.03-0.09). The required time for bowel preparation was significantly shorter(164.95 min ± 68.95 min vs 202.16 min± 68.69 min, P < 0.001) and the total fluid intake volume was significantly lower(2.23 L ± 0.55 L vs 2.47 L ± 0.56 L, P < 0.001) in the 1.2 L PEG-ASC group than in the PEG-ELS group. Palatability, acceptability of the volume of solution, and overall acceptability evaluated using a patient questionnaire, which was assessed by the visual analog scale, were significantly better in the 1.2 L PEG-ASC group than in the PEG-ELS group(7.70 cm ± 2.57 cm vs 5.80 cm ± 3.24 cm, P < 0.001). No severe adverse event was observed in each group.CONCLUSION The 1.2 L PEG-ASC solution was non-inferior to the 2.0 L PEG-ELS solution in terms of cleansing efficacy and had better acceptability among Japanese patients.展开更多

The characteristics of microcapsules such as the content of core material, the yield, the water proof degree, the microencapsulation efficiency and the mean diameter were estimated. The yield, the microencapsulation efficiency and the water proof degree gradually increased with the feed ratio, the coating time and the concentration of pulverizing solvent. The microcapsules showed the thermal responsibility and induced polymerization of methyl methacrylate monomer.展开更多
Wallerian degeneration occurs after peripheral nerve injury and provides a beneficial microenvironment for nerve regeneration.Our previous study demonstrated that ascorbic acid promotes peripheral nerve regeneration,p...Wallerian degeneration occurs after peripheral nerve injury and provides a beneficial microenvironment for nerve regeneration.Our previous study demonstrated that ascorbic acid promotes peripheral nerve regeneration,possibly through promoting Schwann cell proliferation and phagocytosis and enhancing macrophage proliferation,migration,and phagocytosis.Because Schwann cells and macrophages are the main cells involved in Wallerian degeneration,we speculated that ascorbic acid may accelerate this degenerative process.To test this hypothesis,400 mg/kg ascorbic acid was administered intragastrically immediately after sciatic nerve transection,and 200 mg/kg ascorbic acid was then administered intragastrically every day.In addition,rat sciatic nerve explants were treated with 200μM ascorbic acid.Ascorbic acid significantly accelerated the degradation of myelin basic protein-positive myelin and neurofilament 200-positive axons in both the transected nerves and nerve explants.Furthermore,ascorbic acid inhibited myelin-associated glycoprotein expression,increased c-Jun expression in Schwann cells,and increased both the number of macrophages and the amount of myelin fragments in the macrophages.These findings suggest that ascorbic acid accelerates Wallerian degeneration by accelerating the degeneration of axons and myelin in the injured nerve,promoting the dedifferentiation of Schwann cells,and enhancing macrophage recruitment and phagocytosis.The study was approved by the Southern Medical University Animal Care and Use Committee(approval No.SMU-L2015081)on October 15,2015.展开更多
The kiwifruit trees(Actinidia deliciosa cv.Bruno)were sprayed with 5 mmol L-1 oxalic acid(OA)or water(as control)at 130,137 or 144 d after full-blossom,and then the fruit were harvested at commercial maturity and stor...The kiwifruit trees(Actinidia deliciosa cv.Bruno)were sprayed with 5 mmol L-1 oxalic acid(OA)or water(as control)at 130,137 or 144 d after full-blossom,and then the fruit were harvested at commercial maturity and stored at room temperature(20±1)℃ for 13 d.The effect of pre-harvest spraying of OA on postharvest quality of kiwifruit was evaluated during storage.The OA spraying slowed the increase in soluble solids content(SSC)and decrease in titratable acid(TA),as well as increased contents of ascorbic acid(AsA)and total-AsA accompanied with higher AsA/DHA ratio in kiwifruit during storage.Moreover,the OA spraying significantly reduced the contents of acetaldehyde and ethanol in kiwifruit,along with significant decrease in activities of enzymes involved in ethanol fermentation metabolism during the later period of storage,which was beneficial to control off-flavor associated with over accumulation of ethanol during postharvest.It was suggested that pre-harvest spraying of OA might maintain the postharvest quality of kiwifruit in relation to delay in fruit ripening,AsA maintenance and regulation of ethanol fermentation.展开更多

文摘Objective:To evaluate the alteration of chemical behavior of L-ascorbic acid(vitamin C) with metal ion(nickel) at different pH solutions in vitro.Methods:Spectra of pure aqueous solution of L-ascorbic acid(E mark) compound and NiSO_4(H_2O)(sigma USA) were evaluated by UV visible spectrophotometer.Spectral analysis of L-ascorbic acid and nickel at various pH(2.0, 7.0,7.4 and 8.6) at room temperature of 29℃ was recorded.In this special analysis,combined solution of L-ascorbic acid and nickel sulfate at different pH was also recorded.Results:The result revealed that λ_(max)(peak wavelength of spectra) of L-ascorbic acid at pH 2.0 was 289.0 run whereas at neutral pH 7.0,λ_(max) was 29S.4 run.In alkaline pH 8.6,λ_(max) was 295.4 nm and at pH 7.4 the λ_(max) of L-ascorbic acid remained the same as 295.4 nm.Nickel solution at acidic pH 2.0 was 394.5 nm,whereas at neutral pH 7.0 and pH 7.4 were the same as 394.5 nm.But at alkaline pH 8.6,λ_(max) value of nickel sulfate became 392.0 nm.The combined solution of L-ascorbic acid and nickel sulfate(6 mg/mL each) at pH 2.0 showed 292.5 nm and 392.5 nm,respectively whereas at pH 7.0,L-ascorbic acid showed 296.5 nm and nickel sulfate showed 391.5 nm.At pH 7.4,L-ascorbic acid showed 297.0 nm and nickel sulfate showed 394.0 nm in the combined solution whereas at pH 8.6(alkaline) L-ascorbic acid and nickel sulfate were showing 297.0 and 393.5 nm,respectively. Conclusions:Results clearly indicate an altered chemical behavior of L-ascorbic acid either alone or in combination with nickel sulfate in vitro at different pH.Perhaps oxidation of L-ascorbic acid to L-dehydro ascorbic acid via the free radical(HSc*) generation from the reaction of H,ASc + Ni(Ⅱ) is the cause of such alteration of λ_(max),value of L-ascorbic acid in the presence of metal nickel.

文摘Western diet(rich in highly refined sugar and fat)can induce a range of metabolic dysfunctions in animals and humans,including neuroinflammation and cognitive function decline.Neuroinflammation and cognitive impairment,two critical pathological characteristics of Alzheimer’s disease,have been closely associated with microbial alteration via the gut-brain axis.Thus,the present study aimed to investigate the influence of 2-O-β-D-glucopyranosyl-L-ascorbic acid(AA-2βG)isolated from the fruits of Lycium barbarum on preventing the high-fructose diet(HFrD)induced neuroinflammation in mice.It was found that AA-2βG prevented HFr D-induced cognitive deficits.AA-2βG also predominantly enhanced the gut barrier integrity,decreased lipopolysaccharide entry into the circulation,which subsequently countered the activation of glial cells and neuroinflammatory response.These beneficial effects were transmissible by horizontal fecal microbiome transplantation,transferring from AA-2βG fed mice to HFr D fed mice.Additionally,AA-2βG exerted neuroprotective effects involving the enrichment of Lactobacillus and Akkermansia,potentially beneficial intestinal bacteria.The present study provided the evidence that AA-2βG could improve indices of cognition and neuroinflammmation via modulating gut dybiosis and preventing leaky gut.As a potential functional food ingredient,AA-2βG may be applied to attenuate neuroinflammation associated with Western-style diets.
文摘The aim of this study was to investigate the effect of storage conditions on the sensory quality, colour and texture of fresh-cut cabbage during the addition of ascorbic acid, citric acid and calcium chloride. Ascorbic acid maintained the overall quality for 14 days at 0℃ and 7 days at 5℃;no difference, however, was observed regarding browning of cut surface compared to the control sample at both storage temperatures. Calcium chloride maintained the overall quality and cut surface browning for 14 days at both storage temperatures. It was also found that citric acid 1% can be used for minimally processed cabbage. Soaking with citric acid helped retain the color and increased the overall acceptance and organoleptic quality of fresh cut cabbage;it reduced browning of the cut surface and protected against formation of black specks. Citric acid treatment combined with low temperature storage (0℃) prolonged the shelf life of minimally processed cabbage for 22 days, time sufficient for acceptable marketing of the product. The lightness of minimally processed cabbage decreased linearly from 70.94 ± 6 to 63.8 ± 8.5 - 61.3 ± 8 units for the chemical treatments during 22 days of storage at 0℃. Hue angle values during storage time were also significantly influenced by chemical treatments mainly at 0℃.
文摘Photogalvanic cells are photoelectrochemical cells chargeable in light for solar energy conversion and storage. They may be energy source for the future, if their electrical performance is increased. In this study, a photosensitizer Lissamine green B, a reductant Ascorbic acid and a surfactant NaLS have been used in the photogalvanic cell. The generated photopotential and photocurrent are 850.0 mV and 375.0 μA respectively. The conversion efficiency of the cell, fill factor and the cell performance were observed 1.0257%, 0.2598% and 170.0 minutes in dark respectively. The effects of different parameters on the electrical output of the photogalvanic cell were observed. A mechanism was proposed for the photogeneration of electrical energy.
文摘In this study, the effect of ascorbic acid 2-phosphate (Asc2P) was tested on porcine and rat mature hepatocytes in vitro. a). Asc2P increased the porcine, but not rat, albumin secretion and mRNA expression. The enhancing effect of Asc2P on porcine C/EBP alpha mRNA was observed in porcine mature hepatocytes. These data suggested that Asc2P played an important role in the regulation of porcine albumin mRNA level. b). The enhancing effect of Asc2P on ammonium metabolic activity was also observed in porcine, but not rat, mature hepatocytes. The porcine ornithine transcarbamylase (OTC) and arginase mRNAs were augmented by Asc2P, indicating that Asc2P had a direct effect on the urea cycle. c). The porcine collagen type I and type III mRNA, but not type XII mRNA, were detected as well, sugessting that Asc2P did not have the effect on the non-parenchymal hepatocytes to induce collagen type I and III mRNA expression. d). Our RT-PCR analysis demonstrated that the porcine hepatocytes expressed the sodium-ascorbate co-transporters SVCT1 and SVCT2, however, the intensities of porcine sodium-ascorbate co-transporters SVCT1 and SVCT2 bands were not changed markedly. These findings indicated that the Asc2P had no effect on SVCT1 and SVCT2 mRNA expression. e). The enhancing effect of Asc2P on porcine albumin mRNA was inhibited by staurosporine, a portein kinase inhibitor. We conclude that the enhanced albumin mRNA by Asc2P might be due to activation of tyrosine protein kinase and/or PKC and the Asc2P enhanced porcine albumin mRNA mainly at the transcriptional step.
文摘Sepsis and septic shock remain a major cause of morbidity and mortality among patients admitted in the intensive care unit.Diabetes is a major risk factor for the development of sepsis.The global mortality of sepsis remains high,despite significant interventions and guidelines.It has been known for decades that patients with sepsis have reduced levels of antioxidants,most notably vitamin C.Furthermore,experimental data has demonstrated multiple beneficial effects of vitamin C in sepsis.In addition,corticosteroids and thiamine may have synergistic biological effects together with vitamin C.Preliminary data suggests that therapy with hydrocortisone,ascorbic acid and thiamine improves the outcome of patients with sepsis with the potential to save millions of lives.However,this intervention has met with much resistance and has not been widely adopted.Ultimately,we await the final jury verdict on this simple,safe and cheap intervention.
文摘Tea consumption has increased due to its beneficial effects. Results from a lab study on the effect of sucrose (5 g per cup, 150 mL) and/or ascorbic acid (2 mL per cup, 150 mL) on dissolved aluminum compounds during the infusion of two commercial types of dry tea leaves (black, green) with boiling water (5, 15 min infusion time) are presented. Factors influencing the presence of dissolved aluminum in the infusions of both tea leaves were infusion time and sugar contents, as well as the interaction between ascorbic acid and sucrose (p < 0.05). Aluminum contents found after 15 min of infusion were 0.7 mg L–1 for black tea infusions added with sugar, and 0.69 mg L–1 for green tea added with both sugar and ascorbic acid. Both concentrations are higher than the level accepted in Mexico for drinking water (there is no act concerning tea infusions), that is 0.2 mg L–1.

文摘BACKGROUND The current case report describes successful phacoemulsification with the aid of perioperative topical ascorbic acid(AA) in two patients with corneal endothelial disorders to prevent postoperative corneal endothelial decompensation.CASE SUMMARY Two eyes of two patients underwent phacoemulsification with pre-existing corneal endothelial disorders including Fuchs corneal endothelial dystrophy(Patient 1) and endotheliitis(Patient 2). Topical AA was applied to both patients at least one month before and after with a frequency of four times per day. After the surgery, both eyes improved best-corrected visual acuity(BCVA) and there was limited human corneal endothelial cell loss without signs of corneal endothelial decompensation, such as deteriorated BCVA or persistent corneal edema during the follow-up of at least two years.CONCLUSION Perioperative administration of topical AA may be an alternative therapy to the triple procedure in patients expecting to undergo cataract surgery.
文摘In-situ microscopic FTIR spectroelectrochemical technique(MFTIRs) was applied to studying the electrochemical oxidation of ascorbic acid(AA) in poly(ethylene glycol)(PEG) paste at a 100 μm diameter Pt disk electrode. Using this technique, the catalytic ability of cobalt hexacyanoferrate(CoHCF) microcrystalline toward AA oxidation was also studied. It was found that the dispersed CoHCF powder in the PEG paste can generate well shaped thin layer cyclic voltammetric waves with the peak height proportional to the scan rate, corresponding to the Fe centered redox reactions. This oxidation step catalyzed the AA oxidation. Also, this pasted CoHCF powder generated well resolved in situ MFTIRs spectra, by which a chemical interaction between CC bond of AA ring and CoHCF lattice was revealed. A corresponding surface docking mechanism for the catalytic reaction has been proposed.
文摘A nanostructured polymer film incorporated gold nanoparticles modified electrode was fabricated. The fabrication process involved a previous electropolymerization of aspartic acid and followed by the eletrodeposition of gold nano-particles on the glassy carbon electrode. The resulting poly (aspartic acid)-nanogold modified electrode (PAA- nano-Au/GCE) was characterized by scanning electron microscopy (SEM) and electrochemical impedance spectros-copy (EIS). A higher catalytic activity was obtained to electrocatalytic oxidation of dopamine (DA), ascorbic acid (AA) and uric acid (UA) due to the enhanced peak current and well-defined peak separations compared with three, bare GCE, PAA/GCE and nano-Au/GCE. Simultaneous determination of DA, AA, and UA were studied by voltammetry. The linear range of 5.0 × 10-7 - 1.0 × 10-4 M for DA, 5.0 × 10-6 - 2.0 × 10-3 M for AA and 5.0 × 10-6 - 1.0 × 10-3 M for UA was obtained. The detection limit was calculated for DA, AA and UA as being 6.5 × 10-8 M, 5.6 × 10-7 M and 3.0 × 10-7 M, respectively (S/N = 3). The practical application of the present modified electrode was demonstrated by the determination of DA, AA and UA in calf serum and fetal calf serum samples.
文摘Reduction kinetics of the methylene green (MG) with ascorbic acid (AA) in acidic medium at λmax 660 nm was monitored through visible spectrophotomtry in absence and presence of sodium carbonate. CO2 release through reaction of sodium carbonate and oxalic acid, created deoxygenated atmosphere for reduction of dye which greatly boosted the reaction rate. Initially slow reaction in presence of atmospheric oxygen proceeded rapidly when sodium carbonate was added. The reaction followed fractional order kinetics with AA and zero order kinetics with MG. The rate of reaction shows no linear dependence on [H+] concentration as an acidic medium. The rate of reaction is directly related with the elevated concentration of salt, which suggests that the two same charged species are involved in the rate determining step. Secondary reactions at elevated temperature showed complex kinetics.
文摘Aim: To study the detrimental effects of cyclophosphamide on the testicular androgenic and gametogenic activities through endocrine inhibition and/or induction of oxidative stress in male albino rats and to evaluate the protective effect of ascorbic acid. Methods: The testicular △~5, 3β-hydroxysteroid dehydrogenase (HSD), 17β-HSD, peroxidase and catalase activities along with the levels of malondialdehyde (MDA) and conjugated dienes in testicular tissue were measured for the evaluation of testicular oxidative stress. The plasma testosterone (T) level was measured by immunoassay. Various germ cells at stage Ⅶ of spermatogenic cycle were quantified from testicular stained sections. Results: Cyclophosphamide treatment results in a significant inhibition in the testicular △~5, 3β-HSD and 17β-HSD activities, a decrease in plasma T level and a diminution in the counts of various germ cells. Moreover, this treatment was also associated with a significant inhibition of the peroxidase and catalase activities along with high levels of MDA and conjugated dienes in the testis. All these changes were reversed by ascorbic acid co-administration. Conclusion: Cyclophosphamide treatment at the dosage used caused testicular gametogenic and androgenic disorders as well as induced testicular oxidative stress that can be reversed by ascorbic acid co-administration.

文摘Background: Laying hens over 75 weeks of age commonly show great declines in immunity and production performance.It is unclear whether these declines can be relieved by supplementing with ascorbic acid(AA) in feed.Two trials were conducted to investigate the synthesis and metabolism of AA in layers of different ages and the effects of dietary supplemental AA on the performance and the immune and antioxidant statuses of 78 weeks old hens.Methods: In Exp.1,equal numbers(24 hens) of 35 weeks old(Young) and 75 weeks old(Old) layers were fed the same diet without AA supplementation for 4 weeks.In Exp.2,360 healthy 78 weeks old laying hens were randomly assigned to 4 treatments(basal diet supplemented with 0,0.25,0.5,or 1 g AA/kg diet) in an 8-week feeding trial.Results: The old hens tended to have decreased L-gulonolactone oxidase(GLO) synthase activity in the kidney and liver than that of the young hens(P = 0.07 and P = 0.05,respectively).Compared with the young hens,the old hens had lower hepatic antioxidant capacity allowing for the lower thioredoxin(TXN),thioredoxin reductase(TXNR) and cytochrome b5 reductase(CYB5 R) gene expression(P < 0.05),whereas increased sodium-dependent vitamin C transporter(SVCT) 1 expression levels in the ileum and kidney and enhanced splenic and hepatic AA concentrations(P < 0.05).Dietary supplementation with AA significantly decreased GLO enzyme activity but increased splenic AA concentration and anti-bovine serum albumin IgG levels(P < 0.05) and tended to increase CD4+T lymphocyte numbers(P = 0.06) in serum.Supplementation of 0.25 g AA/kg diet significantly increased hepatic total antioxidant capacity(T-AOC,P < 0.05) relative to the control group.Conclusions: Laying hens could synthesize AA in both the kidney and the liver,though the GLO enzyme activities were 100 times greater in kidneys than in livers.The old laying hens had greater absorption and reabsorption capacity and higher AA retention in some tissues that did the young hens.Dietary supplementation of AA can improve the health of old layers by enhancing immunity and antioxidant capacity.

文摘Actinidia eriantha is one of the species of kiwifruit with a particularly high ascorbic acid (AsA) content.However, the molecular mechanism driving AsA richness in fruit remains unclear.In order to reveal the molecular mechanism of AsA richness in A. eriantha, this study constructed a regulatory network related to AsA metabolism by combining genomics,metabolomics and transcriptomics.We assembled a high-quality genome of A. eriantha ‘Ganlv 1’ with only five remaining gaps.The assembly is comprised of 29 pseudochromosomes with a total size of 615.95 Mb, and contig N50 of 20.35 Mb.Among them,24 of the pseudochromosomes were obtained directly from telomere-to-telomere.The LTR assembly index score and consensus quality value were 21.34 and 39.90%, respectively.Subsequently, 61 metabolites and 2 092genes were found to be differentially accumulated/expressed during fruit development by metabolome and transcriptome assays, respectively.AsA metabolism and the cyclic regeneration pathway were found to have high expression levels throughout fruit growth and development,suggesting its crucial role in the regulation of AsA.Furthermore,the AsA contents are highly associated with ascorbate peroxidase genes.The genome obtained in this study provides genomic resources for the genetic and breeding research ofA. eriantha, and the constructed regulatory network can provide a public data platform for future research on kiwifruit.
文摘To counteract the effects of drought stress,scientists have adopted several approaches including the use of different chemicals both inorganic and organic,which is contemplated as a highly efficient and cost-effective shot-gun approach.Ascorbic acid(AsA)is a potential organic substance,which widely occurs in plants,and is considered to be an effective antioxidant to counteract reactive oxygen species(ROS).Thus,a pot experiment was performed to assess the relative mitigating impacts of synthetic AsA and naturally occurring AsA in the form of lemon juice(LJ)and orange juice(OJ)on two cultivars of okra(Abelmoschus esculentus L.)namely Sabz Pari and Bhindi Sanwali under varying water deficit conditions.After 30 days of seed germination,okra seedlings were subjected to different irrigation regimes,i.e.,water deficit stress[(65%and 50%F.C.)and control conditions(100%F.C.)].Different levels of AsA[control(no spray),14 mg L^(−1)LJ,24 mg L^(−1)OJ and 150 mg L^(−1)AsA]obtained from different sources were applied as a foliar spray to control and water-stressed plants.Drought stress prominently reduced plant growth and yield attributes of the okra cultivars.Water-deficit conditions(65%and 50%F.C.)substantially decreased the fruit chlorophyll(a,b)pigments and the activity of superoxide dismutase(SOD)enzyme,while an increase was observed in the contents of fruit’s hydrogen peroxide(H_(2)O_(2)),malondialdehyde(MDA),total phenolics,total soluble sugars,AsA,and total soluble proteins.Drought stress also increased the activities of antioxidant enzymes like peroxidase(POD)and catalase(CAT).However,plant growth and yield attributes,fruit chlorophyll pigments,total phenolics,total soluble sugars,total free amino acids,total soluble proteins,AsA,GB,H_(2)O_(2),and the activities of antioxidant enzymes(POD and CAT)were increased by the AsA exogenous treatment in both okra cultivars under water deficit and control conditions.Overall,LJ and OJ were more effective than the synthetic AsA in upregulating the physiological and metabolic processes of okra plants.So,cost-effective as well as multi-nutrient natural sources of AsA could be suggested for alleviating the harmful effects of water deficit stress on plants.
文摘BACKGROUND A low-volume polyethylene glycol(PEG) solution that combines ascorbic acid with PEG-based electrolyte solution(PEG-ASC) is gaining mainstream acceptance for bowel preparation due to reduced volume and improved taste.Although several reports showed that bowel preparation with PEG-ASC volume lower than 2.0 L with laxative agents could be an alternative to traditional preparation regimen, the cleansing protocols have not been fully investigated.AIM To evaluate the cleansing efficacy of 1.2 L PEG-ASC solution comparing with 2.0 L PEG electrolyte(PEG-ELS) for bowel preparations.METHODS A randomized, single-blinded, open-label, single-center, non-inferiority study was conducted. In total, 312 Japanese adult patients(aged > 18 years) who underwent colonoscopy were enrolled. Patients were randomly allocated to bowel lavage with either 1.2 L of PEG-ASC solution with at least 0.6 L of an additional clear fluid(1.2 L PEG-ASC group) or 2.0 L of PEG-ELS(PEG-ELS group). Then, 48 mg of sennoside was administered at bedtime on the day before colonoscopy, and the designated drug solution was administered at the hospital on the day of colonoscopy. Bowel cleansing was evaluated using the Boston Bowel Preparation Scale(BBPS). The volume of fluid intake and required time for bowel preparation were evaluated. Furthermore, compliance, patient tolerance,and overall acceptability were evaluated using a patient questionnaire, which was assessed using a visual analog scale.RESULTS In total, 291 patients(1.2 L PEG-ASC group, 148; PEG-ELS group, 143) completed the study. There was no significant difference in successful cleansing, defined as a BBPS score ≥ 2 in each segment, between the two groups(1.2 L PEG-ASC group, 91.9%; PEG-ELS group, 90.2%; 95%CI:-0.03-0.09). The required time for bowel preparation was significantly shorter(164.95 min ± 68.95 min vs 202.16 min± 68.69 min, P < 0.001) and the total fluid intake volume was significantly lower(2.23 L ± 0.55 L vs 2.47 L ± 0.56 L, P < 0.001) in the 1.2 L PEG-ASC group than in the PEG-ELS group. Palatability, acceptability of the volume of solution, and overall acceptability evaluated using a patient questionnaire, which was assessed by the visual analog scale, were significantly better in the 1.2 L PEG-ASC group than in the PEG-ELS group(7.70 cm ± 2.57 cm vs 5.80 cm ± 3.24 cm, P < 0.001). No severe adverse event was observed in each group.CONCLUSION The 1.2 L PEG-ASC solution was non-inferior to the 2.0 L PEG-ELS solution in terms of cleansing efficacy and had better acceptability among Japanese patients.

In the experiment, the feed ratio of the amount of tripalmitin to that of ascorbic acid, the coating time and the concentration of ethyl alcohol of pulverizing solvent were changed stepwise. The characteristics of microcapsules such as the content of core material, the yield, the water proof degree, the microencapsulation efficiency and the mean diameter were estimated. The yield, the microencapsulation efficiency and the water proof degree gradually increased with the feed ratio, the coating time and the concentration of pulverizing solvent. The microcapsules showed the thermal responsibility and induced polymerization of methyl methacrylate monomer.

文摘Wallerian degeneration occurs after peripheral nerve injury and provides a beneficial microenvironment for nerve regeneration.Our previous study demonstrated that ascorbic acid promotes peripheral nerve regeneration,possibly through promoting Schwann cell proliferation and phagocytosis and enhancing macrophage proliferation,migration,and phagocytosis.Because Schwann cells and macrophages are the main cells involved in Wallerian degeneration,we speculated that ascorbic acid may accelerate this degenerative process.To test this hypothesis,400 mg/kg ascorbic acid was administered intragastrically immediately after sciatic nerve transection,and 200 mg/kg ascorbic acid was then administered intragastrically every day.In addition,rat sciatic nerve explants were treated with 200μM ascorbic acid.Ascorbic acid significantly accelerated the degradation of myelin basic protein-positive myelin and neurofilament 200-positive axons in both the transected nerves and nerve explants.Furthermore,ascorbic acid inhibited myelin-associated glycoprotein expression,increased c-Jun expression in Schwann cells,and increased both the number of macrophages and the amount of myelin fragments in the macrophages.These findings suggest that ascorbic acid accelerates Wallerian degeneration by accelerating the degeneration of axons and myelin in the injured nerve,promoting the dedifferentiation of Schwann cells,and enhancing macrophage recruitment and phagocytosis.The study was approved by the Southern Medical University Animal Care and Use Committee(approval No.SMU-L2015081)on October 15,2015.

文摘The kiwifruit trees(Actinidia deliciosa cv.Bruno)were sprayed with 5 mmol L-1 oxalic acid(OA)or water(as control)at 130,137 or 144 d after full-blossom,and then the fruit were harvested at commercial maturity and stored at room temperature(20±1)℃ for 13 d.The effect of pre-harvest spraying of OA on postharvest quality of kiwifruit was evaluated during storage.The OA spraying slowed the increase in soluble solids content(SSC)and decrease in titratable acid(TA),as well as increased contents of ascorbic acid(AsA)and total-AsA accompanied with higher AsA/DHA ratio in kiwifruit during storage.Moreover,the OA spraying significantly reduced the contents of acetaldehyde and ethanol in kiwifruit,along with significant decrease in activities of enzymes involved in ethanol fermentation metabolism during the later period of storage,which was beneficial to control off-flavor associated with over accumulation of ethanol during postharvest.It was suggested that pre-harvest spraying of OA might maintain the postharvest quality of kiwifruit in relation to delay in fruit ripening,AsA maintenance and regulation of ethanol fermentation.
