The polymorphisms of adiponectin promoter from 290 pigs, including 5 breeds of Lantang pig, Large spotted pig, Large white pig, Landrace and Duroc, were analyzed with PCR-RFLP. [ Result] At SNP site of adiponectin 5'-flanking region -1 010 bp (G/A), GG genotype frequency in Chinese indigenous pigs was significantly higher than that in exotic pigs. At SNP site of adiponectin 5'-flanking region -394 bp (T/C), the genotype distribution of Chi- nese indigenous pigs was abundant, while no CC genotype was detected in exotic pigs, and T allele frequency was higher in exotic pigs. [ Conclusion] SNP site mutation of - 1 010 bp (G/A) may lead to changes of the gene transcription level, while SNP site of -394 bp (T/C) properly has no relationship with gene transcription level and fat deposition.展开更多

[Result] The constructed library has a total capacity of 6.5×10^6 recombinant clones, and a low proportion of clones without inserted frag- ments; the size of inserted fragments ranges between 0.3-5.0 kb, with strict classifi- cation and ideal consistency. Furthermore, the proportion of clones harboring long in- serted fragments (1.0-5.0 kb) is as high as 30%, achieving the standard for high- quality full-length cDNA library. [Conclusion] The full-length cDNA library of germinat- ing seeds of P. heterocycla was successfully constructed, which laid important foun- dation for the functional genomics research of bamboo plants.展开更多

A genomic library contained 400-800 bp inserts was constructed and screened for the presence of (GA/CT) n and (CA/GT) n repeats. The proportion of positive clones yielded estimated of 3.72×10 4 such dinucleotide repeats per genome, roughly comparable to the abundance reported in other eukaryotic genomes. Twenty_six positive clones were sequenced. In contrast to mammalian genomes, the (GA/CT) n motif was much more abundant than the (CA/GT) n motif in these clones. The (GA/CT) n repeats also showed longer average repeat length (mean n =10.4 versus 6.5), suggesting that they are better candidates for yielding polymorphic genetic markers in the snap bean genome.展开更多

Eight acid mine water samples were collected from five areas in Tongling. Phylogenetic analyses revealed that bacteria mainly fell into ten divisions, which were Betaproteobacteria, Gammaproteobacteria, Alphaproteobacteria, Deinococcus-Thermus, Nitrospira, Firmicutes, Actinobacteria, Deltaproteobacteria, Bacteroidetes, Chloroflexi. Archaea fell into three phylogenetic divisions, Thermoplasma, Ferroplasma and Thermogymnomonas. The unweighted pair group method with arithmetic mean(UPGMA) cluster analysis based on the microbial communities’ compositions revealed that five samples shared similarity with the dominance of Meiothermus and Thermomonas. Two samples had the preponderant existence of Acidithiobacillus and Leptospirillum. The remaining sample owned higher microbial communities’ diversity with the Shannon-Weaver H up to 2.91. Canonical correlation analysis(CCA) suggested that microbial community structures had great association with p H and the concentration of Hg2+, Pb2+, Fe3+, Cl-, SO2- 4in water.展开更多

Vertebrate GATAs can be divided into two subgroups, the GATAI/2/3 and the GATA4/5/6 ...GATA factors are evolutionarily conserved and play crucial roles during embryonic development in both vertebrates and invertebrates. Vertebrate GATAs can be divided into two subgroups, the GATAI/2/3 and the GATA4/5/6 classes. Through genomic analysis, we have identified three GATA factors, representing the GATA1/2/3 and GATA4/5/6 subfamilies respectively, and one GATA like protein in the genome of the basal chordate amphioxus (Branchiostoma floridae, cephalochordata). Partial sequence of GATA123 in the amphioxus Branchiostoma belcheri (BbGATA123) was cloned and its expression pattern during early embryonic development was studied. Expression of BbGATA123 is first detected in the mesendoderm during gastrulation. Interestingly, in the late neurula and early larva stages, it is expressed strongly in the cerebral vesicle and the mid gut region. Its expression is compared to Otx, a gene known crucial for the development of anterior structures. Our observations suggest that GATA123, together with Otx, might play an important role in the development of amphioxus cerebral vesicle, the counterpart of the vertebrate brain.展开更多

I ...According to Kripke, traditional approaches to presupposition and anaphora do not work in some compound sentences because traditionally, they have been thought to assign presuppositions to each clause in isolation. I agree with this criticism, but also think that context is required in order to determine the presupposition included in a complex sentence. To show the context role in fixing the utterance truth-conditions in those cases, this paper introduces the Kripkean anaphoric account on presuppositions (section 2), and then criticizes this approach because it is very restrictive (section 3). The paper concludes that to solve those difficulties, one should appeal to a two-dimensional framework, including a new parameter that focuses on the sentence according to the concrete features of the context: the speaker's point of view (section 4).展开更多

Amid the recent global upsurge of studies on the philosophy of emotion, a reinterpretation of traditional Confucian philosophy, especially Mencius's ideas on the siduan (Four Original Sources) of morality, may shed...Amid the recent global upsurge of studies on the philosophy of emotion, a reinterpretation of traditional Confucian philosophy, especially Mencius's ideas on the siduan (Four Original Sources) of morality, may shed new light on the subject in comparative philosophy. This paper probes the structure of moral feeling and reason described by Mencius's from five perspectives: (1) In view of the relationship between feeling and reason, is it better to use the expression "siduan" or the expression "sixin (Four Heart-minds)"? (2) In view of dispositional feeling, what are the four original sources? (3) In view of moral feeling, what are the structural order of the four sources and the corresponding procedure of reasoning of four heart-minds of Humanity? (4) In view of positive feeling, how does moral feeling grow out of the goodness of human nature? (5) What is the global value of Mencius's ideas on human moral feeling? The author concludes that Mencius's thought on moral feeing has a global value and cross-cultural significance, and that Chinese wisdom is more than regional but universally applicable. The structure of moral feeling and reason that Mencius identified is in accordance with the principles ofzhiliang (grasping the two poles of the beginning and the end) and yongzhong (emloying the middle). The principles ofzhiliang and yongzhong are true universal wisdoms of Confucianism, which should be rekindled today. From a practical point of view, "the Way begins from moral feelings," and Confucius and Yah Hui's seeking the simple, virtuous life is an ideal model of emotional well-being.展开更多
Acrochaete leptochaete (Huber) Nielsen (Chaetophoraceae, Chlorophyta) was isolated from the macroalgae Chaetomorpha collected from intertidal pools in Rongcheng, Shandong, China. 18S rDNA combined with ITS regions...Acrochaete leptochaete (Huber) Nielsen (Chaetophoraceae, Chlorophyta) was isolated from the macroalgae Chaetomorpha collected from intertidal pools in Rongcheng, Shandong, China. 18S rDNA combined with ITS regions were used to ascertain the morphological identification of the isolated material. Based on the unialgal culture, asexual reproduction and growth characteristics of A. leptochaete were investigated over wide ranges of temperature and irradiance. Results revealed that asexual reproduction of A. leptochaete could be realized by biflagellate zoospores. The zoospores germinated directly to give self- replicating generations. Zoospore germination was bipolar. A temperature range from 13-21 ℃ and a lower irradiance of 36 μmol/(m2.s) were most favorable for the growth ofA. leptochaete. Thallus organization, an important taxonomic criterion for the genus Acrochaete, was affected markedly by temperature and irradiance. Our results extend the knowledge about the species' general biology and its morphological plasticity. For classification and identification of a simple microphytic algae like A. leptochaete, which are traditionally placed in the class Chaetophoraceae, we propose that molecular tools associated with culture observations are applied.展开更多

Blooming flowers and dead trees, burning urns, and cloudy landscapes also accompany this gloomy imagery, which alludes to a warning about moral behavior. The first part of this essay deals with a brief history about the symbolism of the skull in vanitas imagery (1590-1630). And the second part of the essay focuses on two prints by the Dutch artist and printmaker from Haarlem, Hendrick Goltzius (1558-1617), who honored this proverb Homo bulla est in Allegory of Transience as a remembrance of the brevity of life.展开更多

Electron microscopic and molecular phylogenetic ...During a survey for marine microalgal resources, we isolated a rare marine euglenoid from the coastal waters of Qingdao, China in 2009, and established a pure culture. Electron microscopic and molecular phylogenetic (18S rDNA and 16S rDNA sequences) analyses revealed a close affinity with Eutreptiella gymnastica, a bloom-forming species. Different culture conditions were monitored to understand optimal E. gymnastica growth characteristics. The optimal growth conditions in a batch culture of this isolate were 20C, 160 gmol photons/(mE.s) of white light, and a salinity of 10-31. Nutrient experiments demonstrated that growth increased dramatically with a phosphorus concentration greater than 72 gmol/L. Understanding the effect of culture conditions on E. gymnastica may help understanding the blooming mechanism of this alga in its natural environment.展开更多

First, which projec...This paper furthers the understanding of capital budgeting (CB) processes. It emphasizes the assumed disposition of the forecast cash benefits from projects. The two reasons for CB are discussed. First, which projects are and are not acceptable from a financial prospective? Second, of financially acceptable projects, CB methods can rank or order projects relative to desirability. Under conditions of certainty, the paper describes and contrasts three primary methods: pay back (PB), net present value (NPV), and internal rate of return (IRR) and three secondary methods: profitability index (PI), terminal value (TV), and modified internal rate of return (MIRR). The presentation lists the strengths and weaknesses of each method in order to direct attention to some of the many aspects of CB decisions that firm managers should consider. Next, approaches for taking risk into consideration are discussed. The discussion raises the important issues of what to do with the net cash benefits from various projects when and if they are received. Finally, the paper provides flexible tools and example problems for teaching CB in a variety of contexts, such as professional corporate training or academic coursework.展开更多

A large number of microsatellite markers are required for the whole genome survey in the fields of molecular ecology,quantitative genetics and genomics. Therefore,it is extremely necessary to select several versatile,low-cost,efficient and time-and labor-saving methods to develop a large panel of microsatellite markers. In this study,we used Zhikong scallop(Chlamys farreri) as the target species to compare the efficiency of the five methods derived from three strategies for microsatellite marker development. The results showed that the strategy of constructing small insert genomic DNA library resulted in poor efficiency,while the microsatellite-enriched strategy highly improved the isolation efficiency. Although the mining public database strategy is time-and cost-saving,it is difficult to obtain a large number of microsatellite markers,mainly due to the limited sequence data of non-model species deposited in public databases. Based on the results in this study,we recommend two methods,microsatellite-enriched library construction method and FIASCO-colony hybridization method,for large-scale microsatellite marker development. Both methods were derived from the microsatellite-enriched strategy. The experimental results obtained from Zhikong scallop also provide the reference for microsatellite marker development in other species with large genomes.展开更多
To determine whether reforestation efforts in the denuded hills have significant impacts on hydrology in the Xinjiang River watershed, the authors examined eight land-cover scenarios to compare hydrologic responses an...To determine whether reforestation efforts in the denuded hills have significant impacts on hydrology in the Xinjiang River watershed, the authors examined eight land-cover scenarios to compare hydrologic responses and to provide a conceptual basis for restoration practices. The authors analyzed a 17-year time period using remote sensing to develop land-cover classification for the watershed. Climate, soil and terrain data for the watershed were used as input in the SWAT (soil and water analysis tool) to quantify and compare the impacts on hydrologic processes. The model was calibrated to a two-year record of stream discharge measurements. The results show significant increase in forest-cover on hills (13%). However, the hydrological response is not very significant considering the changes in forest-cover, the surface runoff and percolation ratios only changed by 2% and 1% over time. Installment of earthen irrigation ponds in the outlets of sub-basin with maximum runoff had provided the most significant hydrologic improvements and could provide irrigation water to increase crop yield on remaining cropland. The study will provide information to the local government to aid decision-making in sustainable reforestation programs resulting in better hydrologic functioning for sustainable water resource management.展开更多

Through prediction calculation of eight groups of naphtha samples and eight groups of gasoline samples, it is verified that the normal distribution hypothesis is more applicable than gamma distribution hypothesis for the prediction model. According to the prediction results of the samples, the restrain range of normal distribution parameters during model computing process is summarized. With the bulk properties of naphtha samples and the value range of distribution parameters as input conditions, this study utilizes the improved novel molecular matrix to predict the composition of naphtha samples. As the results show, the novel molecular matrix can predict more detailed composition information of naphtha and improve prediction accuracy with less unknown parameters.展开更多

文摘In Dutch vanitas imagery of Homo bulla est there is a sad child (homo) holding a scalloped shell filled with soap and water. Although the child is amused and playing with the formation of beautiful transparent water circles, his expression is mischievous and melancholic. Blooming flowers and dead trees, burning urns, and cloudy landscapes also accompany this gloomy imagery, which alludes to a warning about moral behavior. The first part of this essay deals with a brief history about the symbolism of the skull in vanitas imagery (1590-1630). And the second part of the essay focuses on two prints by the Dutch artist and printmaker from Haarlem, Hendrick Goltzius (1558-1617), who honored this proverb Homo bulla est in Allegory of Transience as a remembrance of the brevity of life.

文摘During a survey for marine microalgal resources, we isolated a rare marine euglenoid from the coastal waters of Qingdao, China in 2009, and established a pure culture. Electron microscopic and molecular phylogenetic (18S rDNA and 16S rDNA sequences) analyses revealed a close affinity with Eutreptiella gymnastica, a bloom-forming species. Different culture conditions were monitored to understand optimal E. gymnastica growth characteristics. The optimal growth conditions in a batch culture of this isolate were 20C, 160 gmol photons/(mE.s) of white light, and a salinity of 10-31. Nutrient experiments demonstrated that growth increased dramatically with a phosphorus concentration greater than 72 gmol/L. Understanding the effect of culture conditions on E. gymnastica may help understanding the blooming mechanism of this alga in its natural environment.
文摘This paper furthers the understanding of capital budgeting (CB) processes. It emphasizes the assumed disposition of the forecast cash benefits from projects. The two reasons for CB are discussed. First, which projects are and are not acceptable from a financial prospective? Second, of financially acceptable projects, CB methods can rank or order projects relative to desirability. Under conditions of certainty, the paper describes and contrasts three primary methods: pay back (PB), net present value (NPV), and internal rate of return (IRR) and three secondary methods: profitability index (PI), terminal value (TV), and modified internal rate of return (MIRR). The presentation lists the strengths and weaknesses of each method in order to direct attention to some of the many aspects of CB decisions that firm managers should consider. Next, approaches for taking risk into consideration are discussed. The discussion raises the important issues of what to do with the net cash benefits from various projects when and if they are received. Finally, the paper provides flexible tools and example problems for teaching CB in a variety of contexts, such as professional corporate training or academic coursework.

文摘Microsatellite markers have become one kind of the most important molecular tools used in various researches. A large number of microsatellite markers are required for the whole genome survey in the fields of molecular ecology,quantitative genetics and genomics. Therefore,it is extremely necessary to select several versatile,low-cost,efficient and time-and labor-saving methods to develop a large panel of microsatellite markers. In this study,we used Zhikong scallop(Chlamys farreri) as the target species to compare the efficiency of the five methods derived from three strategies for microsatellite marker development. The results showed that the strategy of constructing small insert genomic DNA library resulted in poor efficiency,while the microsatellite-enriched strategy highly improved the isolation efficiency. Although the mining public database strategy is time-and cost-saving,it is difficult to obtain a large number of microsatellite markers,mainly due to the limited sequence data of non-model species deposited in public databases. Based on the results in this study,we recommend two methods,microsatellite-enriched library construction method and FIASCO-colony hybridization method,for large-scale microsatellite marker development. Both methods were derived from the microsatellite-enriched strategy. The experimental results obtained from Zhikong scallop also provide the reference for microsatellite marker development in other species with large genomes.
文摘To determine whether reforestation efforts in the denuded hills have significant impacts on hydrology in the Xinjiang River watershed, the authors examined eight land-cover scenarios to compare hydrologic responses and to provide a conceptual basis for restoration practices. The authors analyzed a 17-year time period using remote sensing to develop land-cover classification for the watershed. Climate, soil and terrain data for the watershed were used as input in the SWAT (soil and water analysis tool) to quantify and compare the impacts on hydrologic processes. The model was calibrated to a two-year record of stream discharge measurements. The results show significant increase in forest-cover on hills (13%). However, the hydrological response is not very significant considering the changes in forest-cover, the surface runoff and percolation ratios only changed by 2% and 1% over time. Installment of earthen irrigation ponds in the outlets of sub-basin with maximum runoff had provided the most significant hydrologic improvements and could provide irrigation water to increase crop yield on remaining cropland. The study will provide information to the local government to aid decision-making in sustainable reforestation programs resulting in better hydrologic functioning for sustainable water resource management.

文摘To improve the naphtha composition prediction model based on molecular type homologous series matrix (MTHS), this paper puts forward a novel molecular matrix to characterize the naphtha composition and the norreal distribution hypothesis to better describe the molecular composition distribution within each homologous series of the molecular matrix. Through prediction calculation of eight groups of naphtha samples and eight groups of gasoline samples, it is verified that the normal distribution hypothesis is more applicable than gamma distribution hypothesis for the prediction model. According to the prediction results of the samples, the restrain range of normal distribution parameters during model computing process is summarized. With the bulk properties of naphtha samples and the value range of distribution parameters as input conditions, this study utilizes the improved novel molecular matrix to predict the composition of naphtha samples. As the results show, the novel molecular matrix can predict more detailed composition information of naphtha and improve prediction accuracy with less unknown parameters.
