目的:观察虎杖醇提物、水提物对阿尔茨海默病小鼠模型学习记忆能力的影响,并初步探讨其作用机制。方法:采用侧脑室注射β-淀粉样蛋白片段1-42(Aβ1-42)建立AD小鼠模型,通过Morris水迷宫实验检测小鼠学习记忆能力,使用试剂盒检测小... 目的:观察虎杖醇提物、水提物对阿尔茨海默病小鼠模型学习记忆能力的影响,并初步探讨其作用机制。方法:采用侧脑室注射β-淀粉样蛋白片段1-42(Aβ1-42)建立AD小鼠模型,通过Morris水迷宫实验检测小鼠学习记忆能力,使用试剂盒检测小鼠脑组织总抗氧化能力、乙酰胆碱酯酶活性、丙二醛和一氧化氮含量。结果:虎杖醇提物、水提物均不同程度地改善模型小鼠的学习记忆能力,提高总抗氧化能力,以醇提物效果更好,醇提物还能抑制乙酰胆碱酯酶活性,降低丙二醛和一氧化氮含量。结论:虎杖能改善Aβ致AD小鼠模型的学习记忆能力,其作用机制可能与抗氧化、减少炎症介质的产生和调节胆碱能系统等多方面综合作用有关。 展开更多

目的:观察我院自制复方虎杖敛疮液治疗慢性下肢溃疡对其表面细菌、真菌所致感染的疗效。方法:慢性下肢溃疡合并表面细菌、真菌感染者分别以数字随机法纳入治疗组72例,对照组40例。治疗组采用复方虎杖敛疮液加温擦洗患处后溻渍治疗,并给... 目的:观察我院自制复方虎杖敛疮液治疗慢性下肢溃疡对其表面细菌、真菌所致感染的疗效。方法:慢性下肢溃疡合并表面细菌、真菌感染者分别以数字随机法纳入治疗组72例,对照组40例。治疗组采用复方虎杖敛疮液加温擦洗患处后溻渍治疗,并给予必要的病因和清创治疗。对照采用生理盐水清洁创面,再以l%乳酸依沙吖啶纱布外敷包扎固定,亦给予必要的病因和清创治疗。观察期间两组均不使用抗生素、手术及介入疗法,观察溃疡表面感染情况。结果:治疗组溃疡表面感染治疗前后积分差10.0±1.8,对照组积分差6.7±1.3。结论:复方虎杖敛疮液具有清热解毒、利湿化浊、活血化瘀、敛疮生肌的功效,对慢性下肢溃疡表面细菌、真菌抗菌效果明显,对其所致的感染疗效显著。 展开更多

Aim To establish reliable methods for evaluating the quality of rhizoma of Polygonum cuspidatum( Huzhang in Chinese). Methods TLC and HPLC were employed for the chemical identification and content determination,respectively. Results A qualitative TLC method and a quantitative HPLC method with piceid as the reference substance were established, respectively. With piceid as the reference substance and ethyl acetate-methanol-formic acid-water ( 19:3:0.5:1) as the mobile phase, a TLC method for the identification of Huzhang from the commonly used crude drugs of the same family was also set up. Conclusion The established TLC method can reasonably appraise the quality of the drug and easily distinguish Huzhang from the other commonly used crude drugs of the same family. The HPLC method for determining piceid is simple, reproducible, accurate, and feasible. 展开更多

Optimization of extraction and determination of emodin from Polygonum cuspidatum Sieb. et Zucc. products by HPLC-DAD 被引量:1

A uniform experimental design procedure was used to investigate the effects of some operating parameters on the extraction of emodin from Polygonum cuspidatum Sieb. et Zucc. products. Variables tested were volume ratio of material to solvent, size of material, extraction time and temperature and composition of extraction solvent (mixtures of acetone-water). Each variable was tested at seven levels; 7 experiments were performed in random order. Analyses of the extracts were performed by high-performance liquid chromatography with diode array detection(HPLC-DAD). Analytical responses were processed by using a forward regression analysis, in order to find polynomial function describing the relationship between variables and responses. For all the analytes the experimental conditions for providing the highest extraction yield inside the experimental domain considered were found. Finally, a simple, rapid and accurate analytical method was developed for the determination of emodin by high performance liquid chromatography. The separation is achieved within 25 rain on an ODS column using methanol and water as gradient mobiles. Emodin can be quantified by using external standard method detecting at 436 nm. Good linearity is obtained with correlation coefficient exceeding 0.9986 and the detection limit and the quantification limit are 1.53 and 3.23 mg/L respectively. This method shows good reproducibility for the quantification of the emodin with intra-day and inter-day relative standard deviation less than 2.3% and 5.6% respectively. Under optimized extraction conditions, the recovery of the standard is 96.5%. The validated method is successfully applied to quantify the emodin in seven Polygonum cuspidatum sieb. Et zucc. products, which provided an idea for the pre-treatment of determination of active compounds in traditional Chinese medicines. 展开更多

However, the genetic basis for their activity is virtually unknown. In this study, 25600002 short reads (2.3 Gb) of P. cuspidatum root transcriptome were obtained via lllumina HiSeq 2000 sequencing. A total of 86418 urtigenes were assembled de novo and annotated. Twelve, 18, 60 and 54 unigenes were respectively mapped to the mevalonic acid (MVA), methyl-D-erythritol 4-phosphate (MEP), shikimate and resveratrol biosynthesis pathways, suggesting that they are involved in the biosynthesis of pharmaceutically important anthra- quinone and resveratrol. Eighteen potential UDP-glycosyltransferase unigenes were identified as the candidates most likely to be involved in the biosynthesis of glycosides of secondary metabolites. Identification of relevant genes could be important in eventually increasing the yields of the medicinally useful constituents of the P. cuspidatum root. From the previously published transcriptome data of 19 non-model plant taxa, 1127 shared orthologs were identified and characterized. This information will be very useful for future functional, phylogenetic and evolutionary studies of these plants. 展开更多

Methods: Thirty-two healthy male Wistar rats were employed in this study. A total of 8 rats received normal saline intravenously (control grou p),8 rats received 10 mg/kg of endotoxin (endotoxic shock group),8 rats re ceived 1 mg/kg of polydatin after endotoxin injection (polydatin treatment g roup),and 8 rats received 1 mg/kg of polydatin (polydatin prevention group) 30 minutes before endotoxin injection. Mean arterial pressure was measured once hal f an hour. Lung tissues were collected 6 hours later. Phospholipase A2 activit y was measured with acid titration. The gene expression of secretory phospholipa se A2 type IIA was detected with reverse transcription polymerase chain reacti on. Meanwhile,the histological changes of the lungs among four groups were comp ared through microscopic examination.Results: Phospholipase A2 activity and the gene expression of secretory phospholipase A2 type IIA increased after endotoxin injection,but polydatin could inhibit these effects of endotoxin. Obvious morphological eviden ce could be found in the lung pathological sections and the protective effect of polydatin was most significant in the polydatin prevention group.Conclusions: Polydatin has prophylactic and therapeutic effects (the former is more distinct than the latter) on acutely injured lungs in rats with endotoxic shock and which suggests that polydatin may be a phospholipase A 2 inhibitor. 展开更多

Poaceae species such as silver grass or reed are commonly used in traditional mulch farming in Japan,where the Nishi-Awa area is a Globally Important Agricultural Heritage Systems(GIAHS)site.Farmers here have traditionally used silver grass for mulch farming;furthermore,local farmers have learned from long-standing experience that Japanese knotweed(Fallopia japonica,Polygonaceae)is better for cultivation of solanaceous crops in this area.However,it is unclear why Japanese knotweed mulching is beneficial for cultivation of solanaceous crops.Thus,in this study,we hypothesized that Japanese knotweed mulching may be effective in avoiding hazards associated with continuous potato cropping,as native potato used to be cultivated twice a year in the past.Therefore,we investigated the effects of Japanese knotweed mulching on continuous potato cropping and after tomato cropping,which is another solanaceous crop species.Field experiments were conducted in 2018.First,we compared Japanese knotweed mulching,silver grass mulching and no grass mulching(control)in a soil under continuous potato cultivation and in an uncultivated soil.As a result,the extent of the potato yield decrease was reduced by Japanese knotweed mulching.Secondly,we compared Japanese knotweed-mulching and no grass mulching in a soil after a tomato crop and in an uncultivated soil.The extent of decrease in potato growth and yield was also reduced by Japanese knotweed mulching.These findings indicate that mulching with Japanese knotweed helped to avoid the risks associated with continuous potato cropping. 展开更多

Poaceae plant species,such as silver grass,are commonly used in mulching activities Japan.In contrast,local farmers have traditionally used Japanese knotweed(Fallopia japonica)mulch in the cultivation of solanaceous crops in the Nishi-Awa area of Japan,which is a Globally Important Agricultural Heritage Systems site.We have previously evaluated the positive effects of Japanese knotweed mulching on solanaceous crops,such as eggplants,tomato,and potato.In the present study,we observed that the naturally occurring diseases in the solanaceous crops tended to decrease when the knotweed mulching system was adopted,in comparison to when Poaceae mulch was adopted.In eggplants,leaf mold and powdery mildew decreased under Japanese knotweed mulching.We further evaluated the effects of Japanese knotweed mulching by inoculating test plants with Pseudomonas cichorii.We observed suppression of bacterial disease and tomato mosaic virus under Japanese knotweed mulching and following spraying with Japanese knotweed extracts.In addition,disease-resistance genes were expressed at high levels in Arabidopsis thaliana,a model plant,following treatment with Japanese knotweed extracts.The results suggest that Japanese knotweed has potential applications in future sustainable agriculture activities. 展开更多
