目的:分析子痫前期(PE)孕妇外周血辅助性T细胞和自然杀伤(NK)细胞表达水平。方法:选择近期住院确诊的PE妇女42例,入选对象均接受辅助性T细胞和自然杀伤(NK)细胞(CD3^+、CD4^+、CD8^+、CD4^+/CD8^+、CD56^+)检测,并与同... 目的:分析子痫前期(PE)孕妇外周血辅助性T细胞和自然杀伤(NK)细胞表达水平。方法:选择近期住院确诊的PE妇女42例,入选对象均接受辅助性T细胞和自然杀伤(NK)细胞(CD3^+、CD4^+、CD8^+、CD4^+/CD8^+、CD56^+)检测,并与同期住院正常怀孕产妇(对照组,20例)的检测结果比较。结果:PE组血清CD4^+、CD4^+/CD8^+、CD56^+表达水平明显高于对照组,而CD8^+表达水平则显著低于后者(P均〈0.05-0.01)。结论:子痫前期孕妇外周血辅助性T细胞和自然杀伤(NK)细胞的水平异常表达。 展开更多

目的:探讨α-硫辛酸(α-lipoic acid,ALA)对溃疡性结肠炎小鼠Th17相关炎症因子表达的影响。方法:将雄性Balb/c小鼠(22-25g)随机分为对照组(Control)、ALA对照组(ALA)、溃疡性结肠炎组(UC)、ALA治疗组(UC+ALA)四组,每组10... 目的:探讨α-硫辛酸(α-lipoic acid,ALA)对溃疡性结肠炎小鼠Th17相关炎症因子表达的影响。方法:将雄性Balb/c小鼠(22-25g)随机分为对照组(Control)、ALA对照组(ALA)、溃疡性结肠炎组(UC)、ALA治疗组(UC+ALA)四组,每组10只,观察各组小鼠疾病活动指数(DAI)评分,比较结肠质量指数及结直肠组织病理变化;酶联免疫吸附(ELISA)检测小鼠血清中IL-17、IL-21分泌情况,RT-PCR检测结肠组织TNF-α、TGF-βmRNA表达情况。结果:结肠质量指数、DAI及组织病理学评分在对照组及ALA组中无明显差异,但在UC组中显著升高,在UC+ALA组中明显降低(P〈0.05);IL-17、IL-21在对照组及ALA组小鼠血清中的水平无明显差异,但在UC组中显著升高,在UC+ALA组中明显降低(P〈0.05);TNF-α、TGF-βmRNA在对照组及ALA组小鼠结肠组织中的表达水平无明显差异,但在UC组中显著升高,在UC+ALA组中明显降低(P〈0.05)。结论:α-硫辛酸能够降低TNBS引起的溃疡性结肠炎小鼠IL-17、IL-21、TNF-α、TGF-β等Th17相关炎症因子的表达水平,改善炎症反应及病理损伤。 展开更多

Function of Helper T Cells in the Memory CTL-mediated Anti-tumor Immunity

To investigate the role of CD4 + helper T (Th) cells in the memory CTL-mediated anti-tumor immunity, the RAG-1 gene knock out mice were adoptively transferred with OT-1 cells to generate the memory CTL, the C57BL/6 mice immunized with the epitope peptide of OVA specific Th cells and with different adjuvants were adoptively transferred with these memory-CTLs, and then the animals were challenged with tumor cells EG7. It was found that although the simple immunization of mice with the epitope peptide of the OVA specific Th cells could generate more effect CTL, but this effect was not so strong enough to resist completely the challenges with tumor cells. Nevertheless, the memory CTL-mediated anti-tumor immune effect required the helps of Th1 and Th2 cells. The cross-regulation between Th1 and Th2 cells seemed to be beneficial for the host to generate more effector CTL for mounting an efficient anti-tumor response. It concluded that the interaction between Th1 and Th2 cells might be more important than the single subset of Th cells in the memory CTL-mediated anti-tumor immune response. More attention should be paid in this regard for the future studies. 展开更多

Immune recovery after fluid resuscitation in rats with severe hemorrhagic shock 被引量:3

Objective: To investigate the effects of resuscitation with normal saline(NS), hypertonic saline(HTS), and hydroxyethyl starch(HES) on regulatory T cells(Tregs), helper T 1(Th1)/Th2 and cytotoxic T 1(Tc1)... Objective: To investigate the effects of resuscitation with normal saline(NS), hypertonic saline(HTS), and hydroxyethyl starch(HES) on regulatory T cells(Tregs), helper T 1(Th1)/Th2 and cytotoxic T 1(Tc1)/Tc2 profiles in the treatment of hemorrhagic shock. Methods: Rats subjected to severe hemorrhagic shock were resuscitated for 30 min with NS(n=8), HTS(n=8), or HES(n=8); sham(n=8) and naive control(n=8) groups were used for comparison. Following fluid resuscitation, the whole shed blood was reinfused for 30 min, and the rats were observed with continuous hemodynamic monitoring for 120 min. CD4^+CD25^+Foxp3^+ Treg proportions, Th1/Th2 and Tc1/Tc2 profiles in spleen were analyzed by three-color flow cytometry. Results: The proportion of CD4^+CD25^+Foxp3^+ Tregs and ratios of Th1/Th2 and Tc1/Tc2 did not differ among control, sham, and HTS groups, but were significantly lower in NS and HES groups(both P0.05 vs. sham); NS and HES levels were similar. The level of Tc1 was significantly increased in HTS(P0.05 vs. sham), and levels of Tc2 were increased in NS, HES, and HTS groups compared to sham(all P0.05), but did not differ from each other. Conclusions: HTS resuscitation has a greater impact on immune system recovery than NS or HES by preserving the proportion of Tregs and maintaining the balance between Th1/Th2 and Tc1/Tc2 cells in the spleen. Thus, HTS resuscitation provides potential immunomodulatory activity in the early stage after hemorrhagic shock. 展开更多
