A total of 16 Friesian cows were used in the study to determine the peripheral concentration of hydroxyproline(HYPRO) and 3 methyl histidine(3 MEHIS).The objectives of the experiment were to find out the relationship between collagen and myosin degradation and uterine involution in the postpartum cow.The results showed tht the mean concentrations of HYPRO and 3 MEHIS were 13.47±2.38 and 18.35±2.77 n mole/mL,respectively,for the cows during the days 60~90 postpartum.And for the cows immediately after calving the concentrations of HYPRO and 3 HEHIS reduced from 34.29 and 28.06 nmole/mL on the day one after calving to the normal lever of 13.23 and 17.97 n mole on the day 50 postpartum,respectively.There was a good correlation between the peripheral concentrations of both HYPRO and 3 MEHIS and the diameters of the involuting uterus(r=0.8128). 展开更多

The objectives of the experiment were to find out thhe relationship between collagen and myosin ddegradation and uterine involution in the postpartum cow. The results in the experiment showed that the mean concentrations of HYPRO and 3-MEHIS were 138.32±22.99 and 37.09 ±3.90 nmol·mL-1,respectively,for the cows during the days between 60-90 postpartum,and for the cows immediately after calving the concentrations of HYPRO and 3 -MEHIS reduced from 284.30 and 65.48 nmol ·mL-1 on the day one after calving to the normal level of 109.18 and 33.51 nmol·mL^-1 on the day 50 post- partum,respectively. There was a good correlation between the urinary concentrations of both HYPRO and 3 -MEHIS and the diameters of the involuting uterus (r = 0. 79). 展开更多

CHANGES OF URINARY HYDROXYPROLINE IN FRACTURE PATIENTS

Fifteen cases of fresh fractures have been studied, including 14 cases of tubu-lar bone fractures with age from 28 to 60 years and one case of vertebral fracture ofdorsolumbar region, 16 years of age. There were 13 male and 2 female patients. Theyhad no endocrine disturbances or other bone diseases. The ratio ofhydroxyproline/creatinine (HP/Cr) in these cases increased gradually within 20-30 daysafter fracture and then reached the peak value which was about 3-4 times of normal, thepeak value maintained for a long time. If the trauma was mild or the patient was young,the peak began to decline on the 60th day after fracture. While in those severe cases withprolonged confinement to bed, the peak only began to come down about 110th dayafter fracture. The ratio of HP/Cr decreased rapidly from the peak at first, then pursueda slow course. Generally, the ratio of HP/Cr returned to normal on the 240th to 310thday. Bone union began at a time when peak value started to decline, and with completionof the remodelling process, the HP/Cr value returned to normal. 展开更多

Vitamin C is a water soluble vitamin found in many natural sources, citric fruits, vegetables, particularly as antioxidants and as a factor to catabolism and metabolism. The aim of this article is to correlate the level of vitamin C and the hydroxyproline level in urine samples among young palestinian children from age 5 - 14 years. Materials and Methods: Urine samples of 34 individuals of both sexes, their ages ranged from 5 - 14 years were collected and analyzed for determination of Urinary Hydroxyproline by using Modified Neuman and Logan Method. The result of this study shows a significant correlation between Vitamin C and Hydroxyproline at level 0.01. The conclusion of this study was that low vitamin C intake was found among young palestinian children from age 5 - 14 years, Vitamin C should be supplemented in the drinks taken by young palestinian children from age 5 - 14 years as well as further research and investigation with large samples required to include all children from middle and southern Palestine. 展开更多

BACKGROUND: Hepatic fibrosis is the common patholo gical change in various chronic liver diseases,and its major cause is the imbalance between the production and degra dation of the extracellular matrix, which is mainly com posed of collagens. Dan-Shao-Hua-Xian ( DSHX) cap sule, a traditional Chinese herbal compound, has shown marked preventive effects on hepatic fibrosis in rats in our previous studies. The present study was designed to further investigate its therapeutic actions on hepatic fibrosis in rats and its possible mechanisms. METHODS: Eighty male Wistar rats were randomly divid- ed into normal control group, hepatic fibrosis group, non DSHX-treated group, low-dose-treated group, and high dose-treated group. The rat models of hepatic fibrosis were established by subcutaneous injecton of CC14 , drinking al- cohol , giving diet of hyperliprosis and hypoprotein for 8 weeks. The two DSHX-treated groups were treated respec- tively with low dose (0.5 g/kg) and high dose (1.0 g/kg) of DSHX capsule p. o. everyday for 8 weeks. At the end of the experiment, liver indexes were calculated in each group in addition to the levels of the serum hyaluronic acid and alanine aminotransferase. Their degree of hepatic fibrosis and urinary excretion of hydroxyproline and expression of collagen , were detected. RESULTS: Comparison of the indexes of the hepatic fibro- sis group and non-DSHX-treated group revealed that the liver indexes, levels of serum hyaluronic acid and alanine aminotransferase, and stage of hepatic fibrosis were all sig- nificantly reduced in the two DSHX treated groups. The urinary excretion of hydroxyproline was increased and the expression of collagen I and HI in liver tissue was lessened. These alterations were more obviously observed in the high-dose-treated group. CONCLUSION: DSHX capsule has certain therapeutic effect on hepatic fibrosis in rats. 展开更多

Objective:To investigate the antifibrotic effects of Chrysanthemum indicum ethanol extract(CIEE)against activated hepatic stellate cells(HSC)and thioacetamide(TAA)-induced hepatofibrosis in rats.Methods:Cell viability and proliferation of HSC-T6 cells were measured using MTT assay.Primary HSCs were used to study morphology.TAA(200 mg/kg)was used to induced hepatic fibrosis in rats.CIEE(100 and 500 mg/kg)and silymarin(50 mg/kg)were administered orally.Liver functions including alanine transaminase,aspartate transaminase,glutathione,and hydroxyproline levels were measured using commercial kits.Liver sections and fibrotic biomarker expression were measured using hematoxylin and eosin staining and real-time polymerase chain reaction.Results:In vitro study revealed that CIEE(0.1,0.25,and 0.5 mg/mL)inhibited the proliferation of activated HSCs exposed to transforming growth factor(TGF)-β and restored the activated primary HSC morphology.In in vivo studies,TAA-induced increase in liver/body weight ratio(5.46±0.26)was significantly reduced(4.13±0.22)by CIEE(P<0.05 at 500 mg/kg).CIEE(100 and 500 mg/kg)improved the liver functions by significantly attenuating changes in alanine transaminase,aspartate transaminase,glutathione,and hydroxyproline levels(P<0.05).Further,CIEE(100 and 500 mg/kg)ameliorated the histological changes in liver tissue and TGF-β expression significantly(P<0.05)in TAA-induced rats.Conclusions:CIEE significantly protects against TAA-induced liver damage in rats and can be used in the treatment of liver fibrosis. 展开更多

Delonix elata L.is a Ceasalpinaceae species and is traditionally used in India for treatment of skin diseases,liver diseases and rheumatic problems.However,systematic evaluation of its wound healing activity is lacking.Thus,in the present study,we aimed to assess the wound healing activity of D.elata stem bark extract(DSE) and its isolated constituent quercetin-3-rhamnopyranosyl-(1-6) glucopyranoside(QRPG) in rats.The formulations effects on wound healing were assessed by the wound contraction rate,epithelialization period,tensile strength,content of the hydroxyproline,hexosamine and uronic acid in granulation tissue,histopathological studies and Col 1 α(I) expression level in wound tissue by reverse transcription polymerase chain reaction(RT-PCR) study.The topical application of DSE ointment caused faster epithelialization,significant wound contraction(100%),and better tensile strength(710.5 ± 10.5 g/cm^2),while QRPG showed wound epithelialization with 98.2%contraction,better than that of the control group(78.18%).The biochemical analysis of granulation tissue revealed that DSE and QRPG significantly increased hydroxyproline,hexosamine and uronic acid content.A significant increase in the expression of Col 1 α(I) was observed in the wound tissue of DSE and QRPG treated rats.DSE and QRPG were shown to enhance wound healing by increasing collagen synthesis through upregulation of Col 1 α(I),thus validating ethnomedicinal uses. 展开更多

Objective:To screen the wound healing activity of aqueous and ethanolic extract of roots of Ficus benghalensis.Methods:Both the extracts were evaluated for wound healing by three models ie. incision,excision and dead space wound.In incision and dead space the extracts were applied ilaily topically till the 10th post wounding days while in excision model it was till the complete epithelialization process.Standard group were administered Povidone iodine ointment topically daily.The breaking strength,percentage of wound contraction,period of epithelialization,dry granulation weight and hydroxyproline content were observed.Results:The result of the present study showed that both extracts were able to increase the breaking strength(incision model), decrease period of epithelialization.increase percentage wound contraction(excision model), increase hydroxyproline content(Dead space wound model) significantly compared with control group(P【0.05).Conclusions:Based on result we conclude that aqueous extract is more effective than ethanolic extract.However,it is needed more research to be carried out especially on toxicity studies of ethanolic extract. 展开更多

Objective:To evaluate the in vivo wound healing activity of Acanthus leucostachyus leaf extracts using an excision wound model in mice.Methods:Mice were divided into two groups of six animals in each group:the control group and the Acanthus leucostachyus extract-treated group.Healing potential was evaluated by determination of physical parameters(contraction rate,epithelialization period,and tensile strength),biochemical parameters(protein,DNA,and hydroxyproline content),the expression of growth factor and pro-inflammatory cytokines,as well as histological and ultrastructural observations.Results:Treatment with Acanthus leucostachyus leaf extracts markedly increased the rate of wound contraction,tensile strength,the concentrations of protein,DNA,and hydroxyproline,and the expression of growth factor,as well as promoted epithelialization,compared to the control.In addition,Acanthus leucostachyus leaf extracts significantly reduced the expression of pro-inflammatory cytokines.Histological and ultrastructural studies revealed the presence of thicker epithelial layer and smoother surface topography in the extract-treated group compared to the control.Conclusions:Acanthus leucostachyus leaf extracts show potent wound-healing activity and can be used as a wound healing agent. 展开更多

We examined whether the powerful sodium ferulate(SF) could improve myocardial ischemia fibrosis degree and gain the information of myocardial energy metabolism via experimental model of myocardial ischemia fibrosis. The model of myocardial ischemia fibrosis was made for the wistar rats induced with 15 mg/kg isoproterenol(Iso) subcutaneous injection. In experiment l, Iso effective model drug of myocardial ischemia fibrosis at present was administrated to the rats with myocardial ischemia fibrosis and 2, 4, 6, 12, 24, 48 and 72 h, 7 and 21 d later, the changes of adenosine content in rat myocardial tissue and hydroxyproline in blood serum were compared. In experiment 2, SF was administrated to the rats with myocardial ischemia fibrosis and 21 d later, the effect of SF on benazepril as reference drug curing myocardial ischemia fibrosis was evaluated by measuring the changes of adenosine in rat myocardial tissue and hydroxyproline content in blood serum. In the model group, there was a remarkable increase in hydroxyproline content and a decrease in adenosine content of myocardial tissue in experiment 1; there was a remarkable decrease in hydroxyproline in blood serum and a increase in adenosine content in myocardial tissue, which were recovered gradually to control in experiment 2(P〈0.05). The results of experiment 1 show that with the increasing of pathological change degree, the rat adenosine content reduced gradually and arrived at the lowest value three weeks later(P〈0.05); the hydroxyproline content was obviously higher compared with that of control group(P〈0.05). The results of experiment 2 show that rat adenosine content in myocardial tissue and hydroxyproline content in blood serum were recovered gradually to normal level after injection SF. The effect of SF against myocardial ischemia fibrosis in preventive group is better than that of improving the myocardial ischemia fibrosis drug. Meanwhile, hinting HPLC is a good method to measure the adenosine content. 展开更多

Background:Green coffee beans contain biochemical compounds including caffeine,chlorogenic acid,trigonelline,and diterpenoid alcohols.In traditional Iranian medicine,coffee bean powder is used for the treatment of wounds.Previous studies have shown that green coffee bean extract has a number of health benefits,including induction of weight loss,reduced blood pressure,and hepatoprotective and anti-Parkinson effects.The aim of this study was to investigate the wound-healing activity of green coffee bean extract on a full-thickness wound model.Methods:Full-thickness wounds of 20×20 mm were created on the back of New Zealand white rabbits.The animals were divided into 6 groups.Three concentrations of green coffee bean extract(5%,10%,and 15%w/w)in a eucerin base were applied over the wounds in 3 test groups.One percent phenytoin cream was used in one group as a positive control.The rabbits of negative control and vehicle groups received no treatment and eucerin,respectively.For the evaluation of green coffee bean extract’s wound-healing effects,measures included:(1)wound-closure rate,by daily measuring of the wound surface area and calculating the reduction in area;(2)period of epithelialization,the number of days until the scab dropped from the wound;(3)hydroxyproline content,measured by a hydroxyproline assay kit;(4)lipid peroxidation,measured by a lipid peroxidation assay kit;and(5)histopathological state(hematoxylin-eosin stain)of wound-tissue samples on days 7 and 14 post-wounding.Results:The results of this study showed significantly enhanced wound-closure rate,shorter period of epithelialization(P<0.01),increased hydroxyproline content(P<0.001)and suppressed lipid peroxidation(P<0.001 on day 14)of wounds on animals treated with the 10%green coffee bean extract compared to the negative control and eucerin-treated groups.Moreover,the therapeutic effects of 10%green coffee bean were significantly superior than those of phenytoin on enhancing wound-closure rate,decreasing period of epithelialization(P<0.05),increasing hydroxyproline content(P<0.001 on day 7 and P<0.0001 on day 14)and suppressing lipid peroxidation(P<0.0001 on day 14).In addition,histopathological study supported the wound healing activity of green coffee bean extract.Conclusion:Our results showed that green coffee bean extract has a potential for promoting wound healing,thus supporting its traditional use for this purpose. 展开更多

ResuIts 1. The content of hydroxyproline in the affected skin was decreased. 2. The thickness of skin was decreased and the water content was reduced. 3. The microcirculation of local tissues was enhanced. 4. The activity of the macrophages was increased. Purpose The purpose of this study is to review the urine products of bone breakdown as markers of bone resorption and usefulness of urinary hydroxyproline.Data Related researches published in 1985 -2000 were systematically reviewed.Results Bone markers could be used for early diagnosis of bone metabolic diseases. Biochemical markers of bone resorption that reflect osteoclast activity and/or collagen degradation provide a new and potentially important clinical tool for the assessment and monitoring of bone metabolism. Assessment of bone resorption can be achieved with measurement of urinary hydroxylysine glycosides, urinary excretion of the collagen pyridinium cross-links, urinary excretion of type I collagen telopeptide breakdown products (cross-linked telopeptides) and urinary hydroxyproline.Conclusion Urinary hydroxyproline has been in use as a marker of bone resorption, but it lacks sensitivity and specificity. It is a modified aminoacid that is a metabolic product of collagen breakdown. Hydroxyproline may be released either free or with fragments of the collagen molecule attached during bone resorption, and it is also liberated by the breakdown of complement and nonskeletal collagen. 展开更多

Oxidative stress has been confirmed in relation to intestinal mucosa damage and multiple bowel diseases.Hydroxyproline (Hyp) is an imino acid abundant in sow's milk. Compelling evidence has beengathered showing the potential antioxidative properties of Hyp. However, the role and mechanism ofHyp in porcine intestinal epithelial cells in response to oxidative stress remains unknown. In this study,small intestinal epithelial cell lines of piglets (IPEC-1) were used to evaluate the protective effects of Hypon 4-hydroxy-2-nonenal (4-HNE)-induced oxidative DNA damage and apoptosis. IPEC-1 pretreated with0.5 to 5 mmol/L Hyp were exposed to 4-HNE (40 mmol/L) in the presence or absence of Hyp. Thereafter,the cells were subjected to apoptosis detection by Hoechst staining, flow cytometry, and Western blot orDNA damage analysis by comet assay, immunofluorescence, and reverse-transcription quantitative PCR(RT-qPCR). Cell apoptosis and the upregulation of cleaved-caspase-3 induced by 4-HNE (40 mmol/L) wereinhibited by 5 mmol/L of Hyp. In addition, 5 mmol/L Hyp attenuated 4-HNE-induced reactive oxygenspecies (ROS) accumulation, glutathione (GSH) deprivation and DNA damage. The elevation in transcriptionof GADD45a (growth arrest and DNA-damage-inducible protein 45 alpha) and GADD45b(growth arrest and DNA-damage-inducible protein 45 beta), as well as the phosphorylation of H2AX(H2A histone family, member X), p38 MAPK (mitogen-activated protein kinase), and JNK (c-Jun N-terminalkinase) in cells treated with 4-HNE were alleviated by 5 mmol/L Hyp. Furthermore, Hyp supplementationincreased the protein abundance of Krüppel like factor 4 (KLF4) in cells exposed to 4-HNE.Suppression of KLF4 expression by kenpaulone impeded the resistance of Hyp-treated cells to DNAdamage and apoptosis induced by 4-HNE. Collectively, our results indicated that Hyp serves to protectagainst 4-HNE-induced apoptosis and DNA damage in IPEC-1 cells, which is partially pertinent with theenhanced expression of KLF4. Our data provides an updated explanation for the nutritional values ofHyp-containing animal products. 展开更多

Objective: To observe the metabolic, regulatory and anti-oxidative effects of modified Bushen Huoxue Decoction (BSHXD, 补肾活血方), a Chinese herbal medicine for Kidney (Shen)-reinforcement and bloodactivation, on an osteoarthritis (OA) rabbit model. Methods: A rabbit model for knee joint OA was established by the classic Huith's method. The OA model rabbits were randomized into 5 groups: the model control group, the positive control group treated with glucosamine sulfate, and the three BSHXD treated groups treated respectively with low, moderate, and high doses of BSHXD. In addition, a normal control group and a sham-operated group were set up. Experimental animals were sacrificed after a 7-week treatment, and pathological changes in cartilaginous tissue were estimated using the Mankin criteria. Hydroxyproline (Hyp) and malonaldehyde (MDA) contents in blood serum and urine, as well as superoxide dismutase (SOD) activity and nitric oxide (NO) content in blood serum and knee joint synovial homogenates were detected. Results: Mankin scoring showed insignificant statistical differences between the various treatment groups (P〉0.05), but all were better than the model control group (P〈0.05). Serum and udnary contents of Hyp and MDA as well as serum and synovial levels of NO were significantly lower, but the SOD activity in blood serum and synovial tissue was higher in the BSHXD treated groups than in the model group (P〈0.01); the effect of BSHXD was dose-dependent to some extent. Coaclasloa: The modified BSHXD shows an effect of improving cartilage metabolism in experimental rabbits with OA, and possesses osteo-chondric protective effects in antagonizing peroxidation injury. 展开更多

BACKGROUND: The purposes of this study were to confirm the changes in myocardial collagen level after left ventricular assist device (LVAD) support in dilated cardiomyopathy (DCM), find the relation between these changes and prognosis, and test a practical method to assess the level of myocardial collagen. METHODS: Left ventricular samples were collected from DCM patients with different prognosis (transplanted group n = 8, weaning group n = 10) at the time when the LVADs were implanted and again during cardiac transplantation (n = 8). The level of neutral salt soluble collagen (NSC) and acid soluble collagen (ASC) was measured by Sircol collagen assay, and that of total collagen and insoluble collagen (ISC) by quantification of hydroxyproline (Hyp). Serum samples were collected from a portion of these patients (transplanted group, n = 6; weaning group n = 7) at the time the LVADs were implanted, 1 month after implantation and on explantation. Circulating concentration of carboxy-terminal propeptide of type I procollagen (P I CP), amino-terminal propeptide of type I procollagen (P I NP), amino-terminal propeptide of type III procollagen (P III NP) and type I collagen telopeptide (I CTP) were measured by the equilibrium type radioimmunoassay. Background Gastrojejunostomy is one of the most frequently used procedures for general surgeons. The creation of anastomosis between various parts of the gastrointestinal tract is a basic technical component and major task in the daily practice of almost all gastrointestinal procedures. This research evaluated a new procedure of making gastrointestinal anastomosis with stent.Methods Twenty experimental mini-pigs were randomized into two groups. In stent anastomosis group (SA), the anastomoses were constructed with a poly-levolactic acid stent. In hand-sewn group (HA), the anastomoses were performed with a single-layer continuous suture. Abdominal X-ray with intraluminal contrast was performed on the 10th postoperative day. Five pigs of each group were sacrificed on the postoperative days 3 and 14 to determine anastomotic bursting pressure in situ, hydroxyproline concentration, and histopathological evaluation of the anastomotic sites.Results There was no intraoperative morbidity or mortality. The median time needed for the sutured anastomosis was (21.7±2.3) minutes and for the stent anastomosis was (11.9±1.9) minutes (P 〈0.001). Abdominal X-ray with intraluminal contrast demonstrated normal gas distribution and showed no evidence of leakage or obstruction. Macroscopic appearance at the longitudinal opening of anastomosis was always good in both groups. The median anastomotic bursting pressure was (18.2±1.6) kPa in SA group on postoperative day 3, compared with (11.7±3.2) kPa in HA group (P=0.003). The anastomotic bursting pressure on day 14 was not significantly different between SA group ((27.1±2.6)kPa) and HA group ((28.3±1.7) kPa) (P=0.388). The hydroxyproline concentrations were not significantly different.Conclusions The stent anastomosis was not considered to be more difficult than a sutured anastomosis. This method is proved to be safe and feasible compared with the traditional hand-sewn method in the porcine model. The method increases early anastomotic strength in this study. 展开更多
