We generated OsAAP6 and OsAAP10 knockout mutants in three high-yielding japonica varieties and one japonica line using the CRISPR/Cas9 system. Mutation efficiency varied with genetic background in the T_0 generation, and GPC in the T_1 generation decreased significantly,owing mainly to a reduction in glutelin content. Amylose content was down-regulated significantly in some Osaap6 and all Osaap10 mutants. The increased taste value of these mutants was supported by Rapid Visco Analysis(RVA) profiles, which showed higher peak viscosity and breakdown viscosity and lower setback viscosity than the wild type. There were no significant deficiencies in agronomic traits of the mutants. Targeted mutagenesis of OsAAP6 and OsAAP10, especially OsAAP10, using the CRISPR/Cas9 system can rapidly reduce GPC and improve ECQ of rice, providing a new strategy for the breeding cultivars with desired ECQ.展开更多

Methods: Using cell ...Objective: To determine the effect of cis-9, trans-1 1-conjugated linoleic acid on the cell cycle of mammary cancer cells (MCF-7) and the possible mechanism of the inhibitory effect of c9,t11-CLA. Methods: Using cell culture and immunocytochemical techniques, we examined the cell growth, DNA synthesis, expression of PCNA, cyclin A, B1, D1, p16ink4a and p21cip/waf1 of MCF-7 cells at various c9,t11-CLA concentrations (25μM, 50μM, 100μM and 200μM), at 24h and 48h. 96% ethand was used as negative control. Results: The cell growth and DNA synthesis of MCF-7 cells were inhibited by c9,t11-CLA. After treatment with various doses of c9,t11-CLA mentioned above for 8 days, the inhibition frequency was 27.18%, 35.43%, 91.05%, and 92.86%, respectively. Inhibitory effect of c9,t11-CLA on DNA synthesis (except for 25μM, 24h) was demonstrated by significantly less incorporation of 3H-TdR than the negative control (P<0.05 and P<0.01). To further investigate the influence of the cell cycle progression, we found that c9,t11-CLA may arrest the cell cycle of MCF-7 cells. Immunocytochemical staining demonstrated that incubation with different concentration of c9,t11-CLA at various times significantly decreased the expression of PCNA, Cyclin A, B1, D1 in MCF-7 cells compared to the negative control (P<0.01), whereas the expression of p16ink4a and p21cip/waf1, cyclin-dependent kinases inhibitors (CDKI), were increased. Conclusions: The cell growth and proliferation of MCF-7 cells is inhibited by c9,t11-CLA via blocking cell cycle, accompanying reduced expression of cyclin A, B1, D1 and enhanced expression of CDKI (p16ink4a and p21cip/waf1).展开更多
The effect of ethanedioic acid(Ed A) functionalization on Al2O3 supported Ni catalyst was studied on the hydrodeoxygenation(HDO), isomerization, kinetics and Arrhenius parameters of octadec-9-enoic acid(OA) into...The effect of ethanedioic acid(Ed A) functionalization on Al2O3 supported Ni catalyst was studied on the hydrodeoxygenation(HDO), isomerization, kinetics and Arrhenius parameters of octadec-9-enoic acid(OA) into biofuel in this report. This was achieved via synthesis of two catalysts; the first, nickel alumina catalyst(Ni/Al2O3) was via the incorporation of inorganic Ni precursor into Al2O3; the second was via the incorporation nickel oxalate(Ni Ox) prepared by functionalization of Ni with Ed A into Al2O3 to obtain organometallic Ni Ox/Al2O3 catalyst. Their characterization results showed that Ni species present in Ni/Al2O3 and Ni Ox/Al2O3 were 8.2% and 9.3%, respectively according to the energy dispersive X-ray result. Ni Ox/Al2O3 has comparably higher Ni content due to the Ed A functionalization which also increases its acidity and guarantees high Ni dispersion with weaker metal-support-interaction leading to highly reducible Ni as seen in the X-ray diffraction, X-ray photoelectron spectroscopy, TPR and Raman spectroscopy results. Their activities tested on the HDO of OA showed that Ni Ox/Al2O3 did not only display the best catalytic and reusability abilities, but it also possesses isomerization ability due to its increased acidity. The Ni Ox/Al2O3 also has the highest rate constants evaluated using pseudo-first-order kinetics,but the least activation energy of 176 k J/mol in the biofuel formation step compared to 244 k J/mol evaluated when using Ni/Al2O3. The result is promising for future feasibility studies toward commercialization of catalytic HDO of OA into useful biofuel using organometallic catalysts.展开更多

This study aims at assessing the impact of H9N2 viral pass...Low pathogenic Avian Influenza (AI) virus has the ability to evolve to high pathogenic viruses resulting in significant economic losses in the poultry sector. This study aims at assessing the impact of H9N2 viral passaging in broilers and its relatedness to pathogenicity and amino acid (a.a) sequences of the hemagglutinin (HA) cleavage site and neuraminidase (NA) stalk. The original H9N2 AI virus (P0) was used to challenge ten-21 days old broilers. Individual recovery of H9N2 virus from homogenates of trachea, lungs and airsacs was attempted in 9 days old chicken embryos, as a conclusion of the first passage (P1). Tracheal isolates of H9N2 were passaged for a second (P2) and a third (P3) time in broilers, followed by a similar embryonic recovery procedure. The a.a. sequence of a part of HA1 cleavage site and Neuraminidase stalk were compared among the differently passaged viruses;an assessement of the relatedness of the determined a.a. sequences to the pathogenicity in broilers, based on frequency of mortality, morbidity signs, gross and microscopic lesions at 3 days post challenge with the P1, P2, and P3-H9N2, is concluded. An increase in certain morbidity signs and specific lesions was observed in P2- and P3-H9N2 challenged broilers compared to birds challenged with P1-H9N2. A conserved R-S-S-R amino acid sequence at the HA1 cleavage site was observed in the differently passaged H9N2, associated with a variability in the NA stalk-a.a sequences. The passaging of the low pathogenic H9N2 virus in broilers leads to a trend of increase in pathogenicity, manifested in higher frequency of morbidity signs, and of specific gross and microscopic lesions of the examined organs. This passaging was associated with a conserved a.a. sequence of the hemaglutinin cleavage site and a variability in the sequence of the neuraminidase stalk. A detailed study of the potential of the detected variability in the neuraminidase stalk of H9N2 in induction of a higher pathogenicity in broilers will be the subject of future investigations.展开更多

ATCC No. 50 041 in vitro. The minimum inhibitory concentration (MIC) and minimum bactericidal concentration (MBC) values of the 9-octadecanoic acid-hexadecanoic acid-tetrahydrofuran-3,4-diyl ester were determined by using the broth microdilution dilution (BMD) method at different concentrations ranging from 20 to 0.625 mg mL-1. Its time-inhibition curve against E. coli was also tested and showed that the MIC values for the bacterial strains S. aureus, E. coli and Salmonella sp. were 20, 5 and 10 mg mL-1, respectively. Its MBC values were 20, 20 and 10 mg mL-1, respectively. The antibacterial activity of 9-octadecanoic acid-hexadecanoic acid-tetrahydrofuran-3,4-diyl ester against three strain tested showed the relationship with time and concentration.展开更多

OBJECTIVE Propane-2-sulfonic acid octadec-9-enyl-amide(N15),an analogue of oleoylethanolamide(OEA),is a novel PPARα/γdual agonist.Our previous studies verified the positive effects of OEA on the acute and delayed st...OBJECTIVE Propane-2-sulfonic acid octadec-9-enyl-amide(N15),an analogue of oleoylethanolamide(OEA),is a novel PPARα/γdual agonist.Our previous studies verified the positive effects of OEA on the acute and delayed stages of cerebral ischaemia.However,it is not clear whether N15 is effective against ischaemic cerebral injury.In the present study,male Kunming mice were subjected to middle cerebral artery occlusion(MCAO).METHODS To evaluate its preventive effects,N15(50,100 or 200 mg·kg-1,ip)was administered for3 d before ischaemia.To evaluate its therapeutic effects,N15(200 mg·kg-1,ip)was administered 1 h before reperfusion or 0,1,2 or 4 h after reperfusion.Neurological deficit scores,infarct volume and the degree of brain oedema were determined at 24 h after reperfusion.Blood brain barrier(BBB)disruption was evaluated by Evans blue(EB)leakage at 6 h after reperfusion.The activation/inflammatory responses of microglia were detected using immunohistochemistry and Western blotting.RESULTS N15 pretreatment improved neurological dysfunction,reduced infarct volume and alleviated brain oedema in a dose-dependent manner;the most effective dose was 200 mg·kg-1.The therapeutic time window was within 2 h after reperfusion.Moreover,N15was more potent in the treatment of cerebral ischaemia injury than OEA.N15 treatment preserved the BBB integrity and suppressed the activation of microglia.N15 inhibited inflammatory cytokine expression not only in MCAO mice but also in lipopolysaccharide(LPS)-stimulated BV-2microglial cells.Moreover,N15 decreased the phosphorylation levels of NF-κBp65,STAT3,and ERK1/2 both in vivo and in vitro.CONCLUSION Our findings demonstrated that N15 exerts neuroprotective effects and was more potent than OEA.Additionally,the neuroprotective effects of N15 on cerebral ischaemia may be attributed to its anti-inflammatory properties,at least in part,by enhancing PPARα/γdual signalling and inhibiting the activation of the NF-κB,STAT3,and ERK1/2 signalling pathways.These findings suggest that N15 may be a potential therapeutic choice for the prevention and treatment of ischaemic stroke.展开更多

In this study, high temperature(HT) suppressed the expression of the jasmonic acid(JA) biosynthesis gene allene oxide cyclase 2(GhAOC2), reducing JA content and causing male sterility in the cotton HT-sensitive line H05. Anther sterility was reversed by exogenous application of methyl jasmonate(MeJA) to early buds. To elucidate the role of GhAOC2 in JA biosynthesis and identify its putative contribution to the anther response to HT, we created gene knockout cotton plants using the CRISPR/Cas9 system. Ghaoc2 mutant lines showed male-sterile flowers with reduced JA content in the anthers at the tetrad stage(TS), tapetum degradation stage(TDS), and anther dehiscence stage(ADS). Exogenous application of MeJA to early mutant buds(containing TS or TDS anthers) rescued the sterile pollen and indehiscent anther phenotypes, while ROS signals were reduced in ADS anthers. We propose that HT downregulates the expression of GhAOC2 in anthers, reducing JA biosynthesis and causing excessive ROS accumulation in anthers, leading to male sterility. These findings suggest exogenous JA application as a strategy for increasing male fertility in cotton under HT.展开更多

Glycine, glycylglycine, histidine, trigly...A highly sensitive HPLC method for the detection of amino acids and oligopeptides with 9-acridine formyl chloride by pre-column fluorescence derivatization has been developed. Glycine, glycylglycine, histidine, triglycine and glutathione were separated on a reversed-phase C-18 column with methanol-water-triethylamine eluent, derivatization and chromatographic conditions were optimized. The five derivatives were eluted in 28 min with a good reproducibility. Linear range of the calibration graph was 0.08-260 nmol/ml(-1). The relative standard deviations(n=6) are < 5%. Detection limits (signal-to-noise ratio=3) for the five derivatives are 20-40 fmol.展开更多

文摘OBJECTIVE Propane-2-sulfonic acid octadec-9-enyl-amide(N15),an analogue of oleoylethanolamide(OEA),is a novel PPARα/γdual agonist.Our previous studies verified the positive effects of OEA on the acute and delayed stages of cerebral ischaemia.However,it is not clear whether N15 is effective against ischaemic cerebral injury.In the present study,male Kunming mice were subjected to middle cerebral artery occlusion(MCAO).METHODS To evaluate its preventive effects,N15(50,100 or 200 mg·kg-1,ip)was administered for3 d before ischaemia.To evaluate its therapeutic effects,N15(200 mg·kg-1,ip)was administered 1 h before reperfusion or 0,1,2 or 4 h after reperfusion.Neurological deficit scores,infarct volume and the degree of brain oedema were determined at 24 h after reperfusion.Blood brain barrier(BBB)disruption was evaluated by Evans blue(EB)leakage at 6 h after reperfusion.The activation/inflammatory responses of microglia were detected using immunohistochemistry and Western blotting.RESULTS N15 pretreatment improved neurological dysfunction,reduced infarct volume and alleviated brain oedema in a dose-dependent manner;the most effective dose was 200 mg·kg-1.The therapeutic time window was within 2 h after reperfusion.Moreover,N15was more potent in the treatment of cerebral ischaemia injury than OEA.N15 treatment preserved the BBB integrity and suppressed the activation of microglia.N15 inhibited inflammatory cytokine expression not only in MCAO mice but also in lipopolysaccharide(LPS)-stimulated BV-2microglial cells.Moreover,N15 decreased the phosphorylation levels of NF-κBp65,STAT3,and ERK1/2 both in vivo and in vitro.CONCLUSION Our findings demonstrated that N15 exerts neuroprotective effects and was more potent than OEA.Additionally,the neuroprotective effects of N15 on cerebral ischaemia may be attributed to its anti-inflammatory properties,at least in part,by enhancing PPARα/γdual signalling and inhibiting the activation of the NF-κB,STAT3,and ERK1/2 signalling pathways.These findings suggest that N15 may be a potential therapeutic choice for the prevention and treatment of ischaemic stroke.

文摘A novel palladium(II) complex [(TMEDA)Pd(anca)2].2DMSO (1, TMEDA = N,N,N',N'-tetramethylethylethylenediamine, anca = anthracene-9-carboxylic acid) was synthesized and characterized by single-crystal X-ray diffraction, 1H NMR, UV-Vis and fluorescence spectroscopy. The cell belongs to orthorhombic Aba2 space group with a = 19.698(7), b = 21.045(7) c = 9.062(3) A, Z = 4, V= 3757(2) A3, C40H46N206PdS2, Mr = 821.31, D, = 1.452 g/cm3, F(000) = 1704, the final R = 0.0272, wR = 0.0667 and GOOF = 0.999. Complex 1 exhibits an interesting distorted clip-shaped molecular configuration which is stabilized by intramolecular C-H...O hydrogen bonds. The cells are packed into a 3D supramolecular structure based on the intermolecular C-H...zr interactions which further construct the tubular channels serving as guest molecular channels to include the DMSO solvent molecules inside. Furthermore, the spectroscopic properties of 1 were also investigated.

文摘Cotton(Gossypium spp.) yield is reduced by stress. In this study, high temperature(HT) suppressed the expression of the jasmonic acid(JA) biosynthesis gene allene oxide cyclase 2(GhAOC2), reducing JA content and causing male sterility in the cotton HT-sensitive line H05. Anther sterility was reversed by exogenous application of methyl jasmonate(MeJA) to early buds. To elucidate the role of GhAOC2 in JA biosynthesis and identify its putative contribution to the anther response to HT, we created gene knockout cotton plants using the CRISPR/Cas9 system. Ghaoc2 mutant lines showed male-sterile flowers with reduced JA content in the anthers at the tetrad stage(TS), tapetum degradation stage(TDS), and anther dehiscence stage(ADS). Exogenous application of MeJA to early mutant buds(containing TS or TDS anthers) rescued the sterile pollen and indehiscent anther phenotypes, while ROS signals were reduced in ADS anthers. We propose that HT downregulates the expression of GhAOC2 in anthers, reducing JA biosynthesis and causing excessive ROS accumulation in anthers, leading to male sterility. These findings suggest exogenous JA application as a strategy for increasing male fertility in cotton under HT.
文摘A highly sensitive HPLC method for the detection of amino acids and oligopeptides with 9-acridine formyl chloride by pre-column fluorescence derivatization has been developed. Glycine, glycylglycine, histidine, triglycine and glutathione were separated on a reversed-phase C-18 column with methanol-water-triethylamine eluent, derivatization and chromatographic conditions were optimized. The five derivatives were eluted in 28 min with a good reproducibility. Linear range of the calibration graph was 0.08-260 nmol/ml(-1). The relative standard deviations(n=6) are < 5%. Detection limits (signal-to-noise ratio=3) for the five derivatives are 20-40 fmol.
