The microstructures and phase transformation of Ti-43Al-4Nb alloy in as-cast and heat-treated states were investigated by using optical microscopy, scanning and transmission electron microscopy as well as differential scanning calorimetry. The results show that a fine microstructure of the as-cast alloy can be obtained by solidifying through the β phase. γ grains can nucleate directly from the β phase. The coexistence of β phase and γ phase along primary α grain boundaries contributes to the decrease in the grain size of the as-cast alloy. The phase transformation sequence during solidification of the Ti-43Al-4Nb alloy is suggested as L→L+β→β→α+β→α+βr→α+γ+βr→lamellae(α2+γ)+γ+βr. The microstructure of the alloy after heat treatment at 1 250 ℃ for 16 h exhibits a certain coarsening compared with that of the as-cast state. The remnant β phase can be removed by the heat treatment process due to the diffusion of Nb and the non-equilibrium state of β phase. 展开更多

The effects of on-line solution, off-line solution and aging heat treatment on the microstructure and hardness of the die-cast AZ91D alloys were investigated. Brinell hardness of die-cast AZ91D alloy increases through on-line solution and off-line aging treatment but decreases after off-line solution treatment. By X-ray diffractometry, optical microscopy, differential thermal analysis, scanning electron microscopy and X-ray energy dispersive spectroscopy, it is found that the microstructures of the die-cast AZ91D magnesium alloy before and after on-line solution and off-line aging are similar, consisting of α-Mg and β-Al12Mg17. The precipitation of Al element is prevented by on-line solution so that the effect of solid solution strengthening is enhanced. The β-Al12Mg17 phases precipitate from supersaturated Mg solid solution after off-line aging treatment, and lead to microstructure refinement of AZ91D alloy, so the effect of precipitation hardening is enhanced. The β-Al12Mg17 phases dissolve in the substructure after off-line solution treatment, which leads to that the grain boundary strengthening phase is reduced significantly and the hardness of die cast AZ91D is reduced. 展开更多

Tetrandrine inhibits activation of rat hepatic stellate cells in vitro via transforming growth factor-β signaling 被引量:11

AIM: To investigate the effect of various concentrations of tetrandrine on activation of quiescent rat hepatic stellate cells (HSCs) and transforming growth factor-β (TGF-β) signaling in vitro.METHODS: HSCs were isolated from rats by in situperfusion of liver and 18% Nycodenz gradient centrifugation, and primarily cultured on uncoated plastic plates for 24 hwith DMEM containing 20% fetal bovine serum (FBS/DMEM) before the culture medium was substituted with 2% FBS/DMEM for another 24 h. Then, the HSCs were cultured in 2% FBS/DMEM with tetrandrine (0.25, 0.5, 1,2 mg/L, respectively). Cell morphological features were observed under an inverted microscope, smooth muscleα-actin (α-SMA) was detected by immunocytochemistry and image analysis system, laminin (LN) and type Ⅲprocollagen (PCⅢ) in supernatants were determined byradioimmunoassay. TGF-β1 mRNA, Smad 7 mRNA and Smad 7 protein were analyzed with RT-PCR and Western blotting, respectively.RESULTS: Tetrandrine at the concentrations of 0.25-2 mg/L prevented morphological transformation of HSC from the quiescent state to the activated one, while α-SMA, LN and PCⅢ expressions were inhibited. As estimated by gray values, the expression of α-SMA in tetrandrine groups (0.25, 0.5, 1, 2 mg/L) was reduced from 21.3% to 42.2%(control: 0.67, tetrandrine groups: 0.82, 0.85, 0.96, or 0.96, respectively, which were statistically different from the control, P<0.01), and the difference was more significant in tetrandrine at 1 and 2 mg/L. The content of LN in supernatants was significantly decreased in tetrandrine groups to 58.5%, 69.1%, 65.8% or 60.0% that of the control respectively, and that of PCⅢ to 84.6%, 81.5%,75.7% or 80.7% respectively (P<0.05 vs control), with no significant difference among tetrandrine groups. RTPCR showed that TGF-β1 mRNA expression was reduced by tetrandrine treatments from 56.56% to 87.90% in comparison with the control, while Smad 7 mRNA was increased 1.4-4.8 times. The TGF-β1 mRNA and Smad 7 mRNA expression was in a significant negative correlation (r= -0.755, P<0.01), and both were significantly correlated with α-SMA protein expression (r = -0.938, P<0.01;r = 0.938, P<0.01, respectively). The up-regulation of Smad 7 protein by tetrandrine (1 mg/L)was confirmed by Western blotting as well.CONCLUSION: Tetrandrine has a direct inhibiting effect on the activation of rat HSCs in culture. It up-regulates the expression of Smad 7 which in turn blocks TGF-β1 expression and signaling. 展开更多

In recent years,certain foundry processes have made it possible to obtain products with very thin parts,below the4mmthreshold of the permanent mold casting technology.The safety margins of these castings have been reduced,so the T6heattreatment conditions adopted for the Al?7Si?Mg alloys need to be investigated to identify the best combination of strength andductility.Furthermore,the cost and the production time associated with T6heat treatment have to be optimized.In the present work,an experimental study was carried out to optimize the solution treatment and artificial aging conditions in gravity cast thin bars ofB356aluminum alloy modified with Sr.Two solution temperatures were selected,530°C and550°C,respectively,with solutiontime ranging from2to8h,followed by water quenching and artificial aging at165°C with aging time from2to32h.The results ofhardness and tensile tests were correlated with differential scanning calorimetry(DSC)analysis.The best combination of mechanicalproperties and heat treatment duration was obtained with2h solutionizing at550°C and8h aging at165°C.DSC analysis showedthat the alloy’s mechanical properties reach the maximum value when theβ''phase is completely developed during the artificialaging. 展开更多

The microstructural evolution and tensile properties of a forged Ti−42Al−5Mn alloy subjected to different heat treatments were studied.The results showed that,when the forged alloy was aged at 800℃ for 24 h,the interlamellar spacing(λ)andγgrain size at colony boundaries are generally coarsened.Whereas,when the alloy was first annealed at 1300℃ and then aged at 800℃ for 24 h,this coarsening of related microstructures appears less pronounced.The suggested annealing temperatures for the forged Ti−42Al−5Mn alloy are in the range of 1250−1300℃.It was found that,on the condition of the same annealing system,both the strength and ductility were improved as the aging temperature changed from 1000 to 800℃.The secondary precipitatedβo(β_(o,sec))at colony boundaries could be responsible for improving the strength,and theγphase at colony boundaries with the grain size about 6μm might be one of the main reasons for the better ductility. 展开更多

The cast bars were hot swaged at 700 °C and then heat treated by two different regimes which resulted in fine and coarse lamella structures, respectively. The grain size of the as-cast structure was estimated to be 660 μm and the swaged samples obtained a very fine grain size in the range of 50 μm. The overall best combination of hardness, tensile properties, and wear resistance of theα+βtitanium alloy was achieved by heat treating the samples at 1050 °C for getting fine lamellar structure. The maximum compression strength was reported for the heat treated samples at 800 °C with coarse lamella structure. The minimum wear rate was reported for the heat treated samples with fine lamellar structure and the maximum wear rate was obtained for as-cast samples due to its coarse and heterogeneity microstructure. 展开更多

The goal of the present work was to study the effects of acid treatment on the foaming properties of a soybean protein isolate (SPI) and its fractions, glycinin (11S) and β-conglycinin (7S). The structural characteristics, interfacial properties, foaming capacity and stability of the treated proteins were studied. Results from surface hydrophobicity and differential scanning calorimetry (DSC) showed that the acid treatment caused the complete denaturation of 11S and a partial denaturation of 7S. This protein unfolding affected their interracial properties, which led to an improvement in the foaming properties of both protein fractions and isolate. Treated 7S showed the best behavior in the rearrangement process, probably due to its smaller size and its modified structural characteristics. All treated proteins showed stronger interracial films. The foams of treated proteins were destabilized mostly due to gravitational drainage rather than Ostwald ripening. 展开更多

The effects of sub-transus(α+β)annealing treatment(ST),followed by single aging(SA)or duplex aging(DA)on the microstructural evolution and mechanical properties of near-βTi-4Al-1Sn-2Zr-5Mo-8V-2.5Cr(mass fraction,%)alloy were investigated using optical microscopy,scanning electron microscopy,and transmission electron microscopy.The results show that the finer secondaryαphase precipitates in the alloy after DA than SA(e.g.,149 nm for SA and 69 nm for DA,both after ST at 720℃).The main reason is that the pre-aging step(300℃)in the DA process leads to the formation of intermediateωphase nanoparticles,which assist in the nucleation of the acicular secondaryαphase precipitates.In addition,the strength of the alloy after DA is higher than that of SA at the specific ST temperature.A good combination is achieved in the alloy subjected to ST at 750℃,followed by DA(UTS:1450 MPa,EL:3.87%),which is due to the precipitation of nanoscale secondaryαphase by DA.In conclusion,DA is a feasible process for this new near-βtitanium alloy. 展开更多

To study the relationship between the microstructure and tensile properties of the novel metastable β titanium alloy Ti-5.5Cr-5Al-4Mo-3Nb-2Zr,a heat treatment process of ABFCA(solid solution in α+βregion with subsequent furnace cooling followed by aging treatment finally)was designed,by which α phases of different sizes can be precipitated in the β matrix.The results show that the microstructure obtained by this heat treatment process is composed of primary α(α_(p))phase,submicro rod-like α(α_(r))phase and secondary α(α_(s))phase.The alloy with multi-scale α phase has an excellent balance between strength and ductility.The elongation is about 18.3% at the ultimate tensile strength of 1125.4 MPa.The relationship between the strength of the alloy and the α phase was established.The strength of the alloy is proportional to the power of‒1/2 of the average spacing and width of α phase.The α_(s) phase with a smaller size and phase spacing can greatly improve the strength of the alloy by hindering dislocation slip.The transmission electron microscope analysis shows that there is a large amount of dislocation accumulation at the α/β interfaces,and many deformation twins are found in the α_(p) phase after tensile deformation.When the dislocation slip is hindered,twins occur at the stress concentration location,and twins can initiate some dislocations that are difficult to slip.Meanwhile,the plastic strain is distributed uniformly among the α_(p),α_(r),α_(s) phases and β matrix,thereby enhancing the ductility of the alloy. 展开更多
