背景:松脂醇二葡萄糖苷可以促进骨形成与骨基质的合成,加速骨组织修复,但其对成骨细胞的影响和作用机制仍需进一步探索。目的:基于Wnt/β-catenin信号通路探讨松脂醇二葡萄糖苷对地塞米松干预成骨细胞的影响和作用机制。方法:设置不同... 背景:松脂醇二葡萄糖苷可以促进骨形成与骨基质的合成,加速骨组织修复,但其对成骨细胞的影响和作用机制仍需进一步探索。目的:基于Wnt/β-catenin信号通路探讨松脂醇二葡萄糖苷对地塞米松干预成骨细胞的影响和作用机制。方法:设置不同浓度的地塞米松组和松脂醇二葡萄糖苷组对成骨细胞干预24 h,筛选最佳干预浓度;使用地塞米松、松脂醇二葡萄糖苷和Wnt/β-catenin抑制剂XAV-939对成骨细胞进行干预,设置对照组、地塞米松组、抑制剂组、松脂醇二葡萄糖苷组、松脂醇二葡萄糖苷+抑制剂组。CCK-8法检测细胞活性,检测各组细胞碱性磷酸酶和半胱氨酸天冬氨酸蛋白酶3/7酶活性,Annexin V/PI染色与EdU法检测细胞凋亡与增殖情况,Real-Time qPCR检测Wnt3a、β-catenin、c-myc、骨钙素、Ⅰ型胶原蛋白的mRNA表达水平,Western Blot检测Wnt3a、β-catenin、c-myc、骨钙素、Ⅰ型胶原蛋白的蛋白表达水平。结果与结论:①地塞米松和松脂醇二葡萄糖苷干预成骨细胞24 h后,发现浓度为10μmol/L的地塞米松细胞抑制率为实验最佳干预浓度;松脂醇二葡萄糖苷浓度在100μmol/L时,细胞增殖最为明显;②与地塞米松组比较,松脂醇二葡萄糖苷组的碱性磷酸酶活性显著增强(P<0.05),半胱氨酸天冬氨酸蛋白酶3/7酶活性显著降低(P<0.05);Annexin V/PI染色和EdU法细胞增殖检测结果表明,松脂醇二葡萄糖苷可抑制地塞米松干预后成骨细胞的凋亡,促进成骨细胞增殖;③与地塞米松组和抑制剂组比较,松脂醇二葡萄糖苷组和松脂醇二葡萄糖苷+抑制剂组中Wnt3a、β-catenin、c-myc、骨钙素、Ⅰ型胶原蛋白mRNA和蛋白表达水平均显著升高(P<0.05);④结果表明,松脂醇二葡萄糖苷可以抑制地塞米松干预后的成骨细胞凋亡,通过激活Wnt/β-catenin信号通路来保护成骨细胞活性,促进成骨细胞增殖,可能发挥延缓激素性股骨头坏死的作用。 展开更多

Expressions of E-cadherin, p120ctn, β-catenin and NF-κB in Ulcerative Colitis 被引量:4

This study was aimed to investigate the expressions of E-cadherin, p120 ctn, β-catenin and NF-κB in ulcerative colitis(UC) tissues and the implications of their expressions in the pathogenesis of UC. The expressions of E-cadherin, p120 ctn, β-catenin and NF-κB were detected by immunohistochemistry, and those of p120 ctn and NF-κB by Western blotting in 23 cases of UC and 17 cases of normal colonic tissues. The relationship between the expression of E-cadherin or NF-κB and that of p120 ctn was analyzed by Spearman rank correlation analysis. The results showed that in UC and normal colonic groups, the abnormal expression rate of E-cadherin, p120 ctn, β-catenin, and NF-κB was 52.2% vs. 0(P〈0.05), 73.9% vs. 23.5%(P〈0.05), 65.2% vs. 17.6%(P〈0.05) and 78.4% vs. 23.5%(P〈0.05), respectively. p120 ctn expression was positively correlated with E-cadherin expression(r=0.404, P〈0.05), but negatively with nuclear NF-κB expression(r= – 0.347, P〈0.05). Western blotting showed that as compared with the normal controls, the p120 ctn protein level was significantly decreased(P〈0.05), whereas the NF-κB protein level was increased(P〈0.05) in UC tissues. It was concluded that in the colonic tissues of UC patients, the expressions of E-cadherin, p120 ctn and β-catenin are decreased, suggesting the mucosal barrier is impaired in UC. Moreover, NF-κB is increased and activated in the UC tissues, resulting in the inflammation in UC. p120 ctn may influence the UC development through modulating intercellular adhesion and inflammatory response. 展开更多

Objective: To investigate the expressions and correlations of Pin1, β-catenin and cyclin D1 in elderly lung carcinomas. Methods: The expressions of Pin1, β-catenin and cyclin D1 were examined in the specimens of 92 elderly lung carcinomas and 10 normal lung tissues by immunohistochemistry and explored the relationship between the expression levels and clinicopathological factors. Results: (1) The overexpression of Pin1 and cyclin D1 in lung carcinomas was 46 (50%) cases and 60 (65.22%) cases respectively and 56 (60.82%) cases showed positive immunoreactivity for 13-catenin in the nuclear and (or) cytoplasmic fraction in tumor tissues. In normal tissue, the expressions of Pin1 and cyclin D1 were negative, the expression of β-catenin was lied in cell membrane. (2) In lung carcinomas the expressions of Pin1, β-catenin and cyclin D1 correlated with tumor differentiation (P 〈 0.05). The pesitive expression rate and intensity of Pin1 correlated with tumor stage (P = 0.032) and lymph node positive disease (P = 0.041). The expression of β-catenin correlated with lymph node positive disease (P = 0.012). (3) High expression levels of Pin1 correlated with aberrant I]-catenin expression (P = 0.000) but did not show a correlation with cyclin D1 (P = 0.157). Conclusion: In elderly lung carcinomas, the positive expression of Pin1 causes abnormal accumulation of β-catenin and actives its target gene, however, this target gene was not cyclin DI. The detection of Pin1 expression had some clinical significance in estimating prognosis of elderly patient with lung carcinomas. 展开更多

Lengthening periods of hospitalization, increasing numbers of patients with age-related complications, and a shortage of nursing staff have been of great concern in medical psychiatry in Japan. Under these circumstances, countries such as Japan that face a super-aging society and a decline in the working-age population, have been recommended for use of advanced information and communications technology (ICT) to improve the efficiency of medical treatment and care. This study aims to develop the “Monitoring Application for Inappropriate Expressions in Nursing Records” for using PsyNACS<sup>©</sup>, which will enable psychiatric nursing plans to harness the advantages of ICT. The functions considered necessary are as follows: 1) identification of users who enter information;2) a necessary database for lists of inappropriate expressions;3) development of a matching function to recommend proper writing input and a warning function;and 4) a management function for an inappropriate expression list database. A demonstration experiment for developing the application in this study was conducted at a specialized psychiatric hospital. To introduce them to the application, nurses and nurse managers were informed about the system developed in this study, and a survey regarding their opinions on the functions of the application was conducted with ten nurse managers. The results were evaluated in terms of usefulness for nursing care, documentation, and the education of nurses from an ethical perspective. This study suggests that matching their input with an inappropriate expression database will allow nurses to record more appropriate expressions. From an ethical perspective, the ability to use appropriate expressions makes records more likely to withstand disclosure requests from patients and their families. 展开更多

Objective:To compare the expression of Epstein-Barr virus encoded LMP1 and E-cadherin/β-catenin in primary and metastatic nasopharyngeal carcinoma(NPC)for the purpose of understanding their relationship.Methods:Twenty-two pairs of biopsies taken from the nasopharynx and cervical lymph node(s)of the same patient with nasopharyngeal carcinoma were collected.The expression of LMP1,E-cadherin andβ-catenin was observed on immunostained slides using LSAB method.Results:The expression rate of LMP1 in the 22 metastatic tumors(86.36%,19/22)was significantly higher than that in the 22 primary growths(68.18%,15/22),P<0.05.The mean expression percentages of E-cadherin andβ-catenin in metastatic tumors(50.11%±22.53%and 66.36±21.05%,respectively)were significantly lower than those in primary growths(71.52±24.34%and 79.40%±15.05%,respectively),P<0.05.There was a positive correlation between the expressions of E-cadherin andβ-catenin either in primary growths or metastatic tumors.Conclusion:The LMP1 is more likely to be expressed in metastatic neoplastic cells of NPC than in primary carcinoma cells,and on the contrary the expression of E-cadherin/β-catenin in metastatic cells was decreased.Accordingly,the LMP1 might have the ability to downregulate the expression of E-cadherin/β-catenin,resulting in enhancement of the invasive capacity of metastatic NPC cells. 展开更多

Neuronal activity,synaptic transmission,and molecular changes in the basolateral amygdala play critical roles in fear memory.Cylindromatosis(CYLD)is a deubiquitinase that negatively regulates the nuclear factor kappa-B pathway.CYLD is well studied in non-neuronal cells,yet underinvestigated in the brain,where it is highly expressed.Emerging studies have shown involvement of CYLD in the remodeling of glutamatergic synapses,neuroinflammation,fear memory,and anxiety-and autism-like behaviors.However,the precise role of CYLD in glutamatergic neurons is largely unknown.Here,we first proposed involvement of CYLD in cued fear expression.We next constructed transgenic model mice with specific deletion of Cyld from glutamatergic neurons.Our results show that glutamatergic CYLD deficiency exaggerated the expression of cued fear in only male mice.Further,loss of CYLD in glutamatergic neurons resulted in enhanced neuronal activation,impaired excitatory synaptic transmission,and altered levels of glutamate receptors accompanied by over-activation of microglia in the basolateral amygdala of male mice.Altogether,our study suggests a critical role of glutamatergic CYLD in maintaining normal neuronal,synaptic,and microglial activation.This may contribute,at least in part,to cued fear expression. 展开更多

[Objective] This study was carried out to determine the induction effect of jasmonic acid(JA)on powdery mildew resistance in wheat,the activation effect on the expressions of plant disease resistance related genes,and to investigate the relationship between the induced resistance and the gene expression patterns.[Method] Three powdery mildew susceptible cultivars of "Chinese Spring","Pumai 9" and "Zhoumai 18" typically representing different phenotypes in the field were employed.The powdery mildew was assessed by detached leaf assay,and real time quantitative RT-PCR was used to determine the expression patterns of 9 disease resistance related genes of PR1(PR1.1),PR2(β,1-3 glucanase),PR3(chitinase),PR4(wheatwin1),PR5(thaumatin-like protein),PR9(TaPERO,peroxidase),PR10,TaGLP2a(germin-like)and Ta-JA2(jasmonate-induced protein)in leaf of the three cultivars.[Result] MeJA application enhanced the powdery mildew resistances of "Chinese Spring","Pumai 9" and "Zhoumai 18".The induced powdery mildew resistance could be detected from 12 h to 96 h after MeJA treatment,and the peak value was at 24 h.Though there were differences between the three cultivars,MeJA significantly effect on the expressions of the 8 disease resistance related genes except TaGLP2a,and the peak values were at 12 h,24 h or 48 h after treatments.The strongest activation of MeJA was on PR9 and PR1 that their expressions could reach more than 100 times of the untreated samples.MeJA strongly activated PR2、PR4、PR5、PR3、PR10 and Ta-JA2,their expression could reach 10 to 70 times,and there was almost no activation effect on TaGLP2a.The induced powdery mildew resistance positively correlated with the induced expressions of the 8 disease related genes.[Conclusion] The induced powdery mildew resistance positively correlated with the induced expressions of the disease related genes.Jasmonate signalling plays a role in defence against Blumeria graminis f.sp.tritici.and future manipulation of this pathway may improve powdery mildew resistance in wheat. 展开更多

Lectin and leghemoglobin in legumes play the important roles, respectively, in recognition of host plants to their rhizobial bacteria, and lowering the oxygen partial pressure around bacteroids and protecting nitrogenase from oxygen in symbiotic nitrogen-fixing nodules. In order to extend the host range of the rhizobial bacteria and to make them fix nitrogen in non-legumes, pea lectin gene (pl) and Parasponia hemoglobin gene ( phl,) have been constructed into a plant expression vector (pCBHUL) and the vector pCBHUL was introduced into rice calli from immature young embryos by particle bombardment. After the calli were regenerated into plantlets on the resistant-selecting media containing hygromycin, they were identified by PCR and Southern blot hybridization. It was indicated that the pi and phb genes were integrated into nucleic genome of the transformed rice plants. GUS activity and the product of the pi gene were determined by GUS staining, Western blot and in situ hybridization at translational level. Eighteen out of 40 plants resistant to hygromycin were positively identified by PCR analysis with the rate of 45%. The pi gene was expressed in 3 out of 18 plants with 17% and 7.5% in 40 plants. The results may provide a clue for exploring whether Rhizobium leguminosarum by. viceae could extend its host range and make the transgenic rice plants have the possibility of being symbiotic, or associative to nitrogen fixation. 展开更多

[Objective] The research aimed to discuss the effects of rosiglitazone and serum on the expressions of PPARα and PPARγ genes in the induced differentiation process of pig preadipocyte.[Method] The pig preadipocyte was separated by using the collagenase digestion method.Three kinds of different differentiation culture solutions were used to induce the differentiation of pig preadipocyte.The oil red O staining extraction method was used to contrast the influences of different differentiation culture solutions on the variation of cellular fat content in the differentiation process.Moreover,the variation trends of PPARα and PPARγ expressions in the cellular differentiation process in the different differentiation culture solutions were detected by the real-time quantification PCR.[Result] The cellular fat accumulation was the fastest in MII which contained rosiglitazone and was the slowest in MI which didn't contain rosiglitazone.Rosiglitazone could significantly increase the expression of PPARγ gene(P0.01),but had the certain inhibition effect on the expression of PPARα gene,which wasn't significant.The serum had the extremely significant up-regulation effect on the expression of PPARγ gene(P0.01),but had the extremely significant down-regulation effect on the expression of PPARα gene(P0.01).[Conclusion] Rosiglitazone could greatly promote the expression of PPARγ gene,which increased the cellular fat deposition.Maybe the activator of PPARγ gene existed in the serum,and the inhibitor of PPARα gene existed simultaneously. 展开更多

目的:探讨补中益气汤含药血清协同Siβ-catenin调控Wnt/β-catenin信号通路干预A549/DDP细胞顺铂耐药性的分子机制研究。方法:采用siRNA干扰沉默β-catenin的表达,Western blot检测正常血清组、顺铂组、siCTNNB1-235组、siCTNNB1-510组... 目的:探讨补中益气汤含药血清协同Siβ-catenin调控Wnt/β-catenin信号通路干预A549/DDP细胞顺铂耐药性的分子机制研究。方法:采用siRNA干扰沉默β-catenin的表达,Western blot检测正常血清组、顺铂组、siCTNNB1-235组、siCTNNB1-510组、siCTNNB1-547组A549/DDP细胞的β-catenin蛋白的相对表达量;干扰β-catenin后,Western blot检测各组A549/DDP细胞β-catenin和Survivin的蛋白表达量,MTT法检测各组A549/DDP细胞对顺铂的IC50,Annexin V/PI染色法检测各组A549/DDP细胞顺铂诱导的凋亡率。结果:相较于正常血清组(1.00),siCTNNB1-235组(0.323±0.021)对β-catenin表达抑制率达67%(P=0.000)。RNAi技术沉默β-catenin后,与正常血清组(1.00)相比,补中益气汤联合顺铂组的Survivin(0.247±0.015)和β-catenin(0.257±0.015)蛋白的表达下调更为明显(P=0.000,P=0.000)。IC50和正常血清组(28.330±1.029)µmol/L相比,单独补中益气汤含药血清(20.350±1.155)µmol/L或单独瞬时转染siCTNNB1-235组(15.577±1.535)µmol/L均可降低A549/DDP细胞顺铂的IC50(P=0.000,P=0.000),2者联合(10.453±0.999)µmol/L使用可进一步降低顺铂的IC50(P=0.000)。与正常血清组(9.130±0.384)%相比,瞬时转染siCTNNB1-235联合顺铂组(64.393±0.244)%和补中益气汤联合顺铂组(70.120±0.400)%的总凋亡率均升高(P=0.000,P=0.000)。结论:补中益气汤含药血清和瞬时转染siβ-catenin在抑制Wnt/β-catenin信号通路改善肺腺癌顺铂耐药性方面具有协同效应,且补中益气汤具有siβ-catenin瞬时转染样效应。 展开更多

From viewpoint of chemical element and microstructure of wood, this paper makes a discussion on thermal diffusivity of wood and two theoretical expressions of thermal diffusivity for the choral and radial directions were derived. The thermal diffusivities of the choral and radial directions for about 20 species of trees were calculated with the derived theoretical expressions and compared with the experimental values. The average error of the theoretical values of thermal diffusivity was 7.5% for choral direction and 6.2% for radial direction. 展开更多

The expression of c-kit was elevated both in positive rate and positive index by TGF-01 in both time- and dose-dependent manners. Ing/ml rhTGF-β1 simultaneously enhanced the expression of c-fms and PDGF-R which is not detected in 50 ng / ml GM-CSF treatment. Endoglin was down-regulated after TGF-β treatment and up-regulated in J6-2 cells after GM-CSF treatment, c-kit Expression was elevated by TGF-β in J6-1 cells while decreased by both in J6-2 cells. 展开更多

创面愈合是由于外伤、糖尿病溃疡、压疮、感染和术后创面不愈合等多种因素导致的如表皮、真皮、肌肉、筋膜等局部组织的损伤,其愈合过程是一个复杂且动态的程序,涉及到多种细胞反应和周围微环境的相互配合。目前,创面发病率呈逐年增长... 创面愈合是由于外伤、糖尿病溃疡、压疮、感染和术后创面不愈合等多种因素导致的如表皮、真皮、肌肉、筋膜等局部组织的损伤,其愈合过程是一个复杂且动态的程序,涉及到多种细胞反应和周围微环境的相互配合。目前,创面发病率呈逐年增长的趋势,造成了全球范围内巨大的社会和经济负担,老龄人口中发生的慢性损伤随着年龄的增长而逐年攀升。当前,修复创面的药物种类及方法多样,一定程度上减轻了患者的生理、心理和经济社会压力。中药单体及复方制剂修复创面具有独特的优势和力量,引起社会各界广泛的关注及应用,Wnt/β-catenin是调节细胞增殖、分化及凋亡,高度参与创面修复,是皮肤损伤修复过程中最重要的经典信号通路之一。Wnt/β-catenin信号通路通过中药单体及复方制剂降低机体创面中蛋白激酶、炎症反应,促进血管新生和表皮干细胞等表达,对创面愈合的治疗具有重要的参考价值。本文复习国内外相关文献,对Wnt/β-catenin信号通路与创面的关系及中药单体和中药复方通过调控该信号通路影响创面愈合的机制进行归纳总结,为中药治疗创面损伤提供新的思路和方向。 展开更多

Lectins and leghemoglobins in legumes play the important roles, respectively, in recognition of host plants to their own rhizobia, and lowering the oxygen partial pressure surround the bacteroids and protecting nitrogenase from oxygen in symbiotic nitrogen_fixing nodules.In order to investigate the non_leguminous recognition of rhizobial bacteria relating to nitrogen fixation, plant expression vectors containing pea lectin gene ( pl ) and Parasponia hemoglobin gene ( phb ) have been, respectively, constructed in a plasmid and the plasmid has been introduced into tobacco ( Nicotiana tabacum L.) using Agrobacterium tumefaciens (Smith et Townsend) Conn as a vehicle for transformation. PCR and Southern blot demonstrated that the two genes were integrated into the genome of the tobacco plants. Histochemical staining for GUS activity, Western blotting,and in situ hybridization of pea lectin showed that they were expressed at translational level in the plants. These results may provide a clue for exploring whether Rhizobium leguminosarum bv. viciae could extend its host range and make the transgenic tobacco plants have the possibility of being symbiotic, or associative to nitrogen fixation. 展开更多
