Mesenchymal stem cells(MSCs)are stromal multipotent stem cells that can differentiate into multiple cell types,including fibroblasts,osteoblasts,chondrocytes,adipocytes,and myoblasts,thus allowing them to contribute t...Mesenchymal stem cells(MSCs)are stromal multipotent stem cells that can differentiate into multiple cell types,including fibroblasts,osteoblasts,chondrocytes,adipocytes,and myoblasts,thus allowing them to contribute to the regeneration of various tissues,especially bone tissue.MSCs are now considered one of the most promising cell types in the field of tissue engineering.Traditional petri dish-based culture of MSCs generate heterogeneity,which leads to inconsistent efficacy of MSC applications.Biodegradable and biocompatible polymers,poly(3-hydroxyalkanoates)(PHAs),are actively used for the manufacture of scaffolds that serve as carriers for MSC growth.The growth and differentiation of MSCs grown on PHA scaffolds depend on the physicochemical properties of the polymers,the 3D and surface microstructure of the scaffolds,and the biological activity of PHAs,which was discovered in a series of investigations.The mechanisms of the biological activity of PHAs in relation to MSCs remain insufficiently studied.We suggest that this effect on MSCs could be associated with the natural properties of bacteria-derived PHAs,especially the most widespread representative poly(3-hydroxybutyrate)(PHB).This biopolymer is present in the bacteria of mammalian microbiota,whereas endogenous poly(3-hydroxybutyrate)is found in mammalian tissues.The possible association of PHA effects on MSCs with various biological functions of poly(3-hydroxybutyrate)in bacteria and eukaryotes,including in humans,is discussed in this paper.展开更多

Specimens of bending cylinder under gravity are used to avoid contact problem between testing device and sample, and a beam model is presented for evaluating the curvatures of gel beams with large elastic deformation. A self-adaptive algorithm is also proposed to search for the best estimation of gels' elastic moduli by comparing the experimental bending curvatures with those computed from the beam model with preestimated moduli. Application to the measurement of the property of polyacrylamide gels indi- cates that the material compliance varies with the concentrations of bis-acrylamide, and the gels become softer after being immersed in a culture medium for a period of time, no matter to what extent they are polymerized.展开更多

Maintainance of sulfomucin is a key end point in the treatment of diarrhea and inflammatory bowel disease(IBD).However,the mechanisms underlying the microbial sense to sulfomucin are poorly understood,and to date,ther...Maintainance of sulfomucin is a key end point in the treatment of diarrhea and inflammatory bowel disease(IBD).However,the mechanisms underlying the microbial sense to sulfomucin are poorly understood,and to date,there are no therapies targeting the secretion and maturation of sulfomucin in IBD.Herein,we biosynthesized poly-β-hydroxybutyrate(PHB)and found that PHB could alleviate inflammation caused by diarrhea and colitis by enhancing the differentiation of sulfomucin.Microbiota transplantation and clearance together demonstrate that PHB promoting sulfomucin is mediated by Lactobacillus johnsonii(L.johnsonii).Further studies revealed that PHB provides a favorable niche for L.johnsonii biofilm formation to resist disturbance and support its growth.L.johnsonii-biofilm alleviates colitis by regulating fucose residues to promote goblet cell differentiation and subsequent sulfomucin maturation.Importantly,PHB alleviates colitis by enhancing sulfomucin secretion and maturation in a L.johnsonii-dependent manner.PHB represents a class of guardians,acting as a safe probiotic-biofilm delivery system that significantly promotes probiotic proliferation.Altogether,this study adds weight to the possible role of probiotics and functional materials in the treatment of intestinal inflammation.The application of PHB and biofilm self-coating L.johnsonii carries high translational potential and may be of clinical relevance.展开更多
In this survey article,we present two applications of surface curvatures in theoretical physics.The first application arises from biophysics in the study of the shape of cell vesicles involving the minimization of a m...In this survey article,we present two applications of surface curvatures in theoretical physics.The first application arises from biophysics in the study of the shape of cell vesicles involving the minimization of a mean curvature type energy called the Helfrich bending energy.In this formalism,the equilibrium shape of a cell vesicle may present itself in a rich variety of geometric and topological characteristics.We first show that there is an obstruction,arising from the spontaneous curvature,to the existence of a minimizer of the Helfrich energy over the set of embedded ring tori.We then propose a scale-invariant anisotropic bending energy,which extends the Canham energy,and show that it possesses a unique toroidal energy minimizer,up to rescaling,in all parameter regime.Furthermore,we establish some genus-dependent topological lower and upper bounds,which are known to be lacking with the Helfrich energy,for the proposed energy.We also present the shape equation in our context,which extends the Helfrich shape equation.The second application arises from astrophysics in the search for a mechanism for matter accretion in the early universe in the context of cosmic strings.In this formalism,gravitation may simply be stored over a two-surface so that the Einstein tensor is given in terms of the Gauss curvature of the surface which relates itself directly to the Hamiltonian energy density of the matter sector.This setting provides a lucid exhibition of the interplay of the underlying geometry,matter energy,and topological characterization of the system.In both areas of applications,we encounter highly challenging nonlinear partial differential equation problems.We demonstrate that studies on these equations help us to gain understanding of the theoretical physics problems considered.展开更多
Objective:To determine the effect of steroidogenic acute regulatory protein(StAR) overexpression on the levels of adenosine triphosphate(ATP)-binding cassette transporter A1(ABCA1) and ATP-binding cassette transporter...Objective:To determine the effect of steroidogenic acute regulatory protein(StAR) overexpression on the levels of adenosine triphosphate(ATP)-binding cassette transporter A1(ABCA1) and ATP-binding cassette transporter G1(ABCG1) in an endothelial cell line(bEnd.3).Methods:The StAR gene was induced in bEnd.3 cells with adenovirus infection.The infection efficiency was detected by fluorescence activated cell sorter(FACS) and fluorescence microscopy.The expressions of StAR gene and protein levels were detected by real-time polymerase chain reaction(PCR) and Western blot.The gene and protein levels of ABCA1 and ABCG1 were detected by real-time PCR and Western blot after StAR overexpression.Results:The result shows that StAR was successfully overexpressed in bEnd.3 cells by adenovirus infection.The mRNA and protein expressions of ABCA1 and ABCG1 were greatly increased by StAR overexpression in bEnd.3 cells.Conclusion:Overexpression of StAR increases ABCA1 and ABCG1 expressions in endothelial cells.展开更多

文摘目的构建组织蛋白酶B(Cathepsin B,CTSB)小RNA(siRNA)慢病毒载体,并探讨其对小鼠脑微血管内皮细胞(bEND.3)的影响。方法设计并合成含有干扰Cathesin B基因的19 nt的双链寡DNA片段,将此片段克隆到携有绿色荧光蛋白(GFP)的慢病毒表达载体质粒pGCSIL-GFP上,经测序正确后,命名为pGCSIL-GFP-CTSB,将慢病毒表达载体pGCSIL-GFP-CTSB、慢病毒包装载体pHelper1.0和pHelper2.0 3质粒共同转染于293T细胞,获得携带Cathepsin B基因的RNAi慢病毒(Cathesin B-RNAi-Lentivirus,即CTSB-RNAi-LV),通过Real time-PCR和Western blotting方法观察CTSB-RNAi-LV对bEND.3的影响。结果 1.pGCSIL-GFP-CTSB中携带有正确的Cathesin B siRNA基因;2.目的基因Cathepsin B siRNA被RNAi慢病毒高效地转导入靶细胞bEND.3内,并达到稳定的表达;3.CTSB-RNAi-LV能有效降低bEND.3中Cathepsin B mRNA和蛋白表达。结论成功构建了RNAi慢病毒载体pGCSIL-GFP-CTSB;并成功包装了RNAi慢病毒CTSB-RNAi-LV;CTSB-RNAi-LV能有效地抑制bEND.3中Cathepsin B mRNA和蛋白表达水平下调。
