The relationships between 17α,20β dihydroxy 4 pregnene 3 one (17α,20β DP) and plasma IgM and total protein levels were investigated in rainbow trout, Oncorhynchus mykiss .IgM and total protein levels decreased in both sexes of mature rainbow trout when 17α,20β DP levels increased during the spawning season,while the elevated 17α,20β DP suppressed IgM levels and reversibly enhanced total protein concentrations in immature trout.This represents the first report of the effect of 17α,20β DP on fish immunity.These data suggest that the increase of 17α,20β DP in spawning season may be related to infectious skin diseases. 展开更多

Complex 1 exhibits a two-dimensional(2 D) network, which was stabilized through O–H···O hydrogen bonding interactions. 4', 6-Dihydroxy-4-methoxyisoaurone inhibits TNF-α-induced NF-KB activation and expressions of NF-KB-regulated target gene products

The nuclear factor-KB (NF-KB) transcription factors control many physiological processes including in- flammation, immunity, apoptosis, and angiogenesis. In our search for NF-KB inhibitors from natural resources, we identified 4',6-dihydroxy-4-methoxyisoaurone (ISOA) as an inhibitor of NF-KB activation from the seeds of Tricho- santhes kirilowii. However, the mechanism by which ISOA inhibits NF-KB activation is not fully understood. In the present study, we demonstrated the effect of ISOA on NF-KB activation in TNF-α-stimulated HeLa cells. This com- pound suppressed NF-KB activation through the inhibition of IKB kinase (IKK) activation. ISOA also has an influ- ence on upstream signaling of IKK through the inhibition of expression of adaptor proteins, TNF receptor-associated factor 2 (TRAF2) and receptor interacting protein 1 (RIP1). Consequently, ISOA blocked the phosphorylation and degradation of the inhibitor of NF-KB alpha (IKBα) , and subsequent phosphorylation and nuclear translocation of p65. The suppression of NF-KB activation by ISOA led to the down-regulation of target genes involved in inflam- mation, proliferation, angiogenesis and invasion, as well as potentiation of TNF-α-induced apoptosis at least in part through activation of caspase-8. Taken together, this study extends our understanding on the mechanisms underly- ing the anti-inflammatory and anti-cancer activities of ISOA. Our findings provide new insight into the molecular mechanisms and a potential application of ISOA for inflammatory diseases as well as certain cancers associated with abnormal NF-KB activation. 展开更多

Excited-state double proton transfer(ESDPT)is a controversial issue which has long been plagued with theoretical and experimental communities.Herein,we took 1,8-dihydroxy-2-naphthaldehyde(DHNA)as a prototype and used combined complete active space selfconsistent field(CASSCF)and multi-state complete active-space second-order perturbation(MS-CASPT2)methods to investigate ESDPT and excited-state deactivation pathways of DHNA.Three different tautomer minima of S1-ENOL,S1-KETO-1,and S1-KETO-2 and two crucial conical intersections of S1 S0-KETO-1 and S1 S0-KETO-2 in and between the S0 and S1 states were obtained.S1-KETO-1 and S1-KETO-2 should take responsibility for experimentally observing dual-emission bands.In addition,two-dimensional potential energy surfaces(2 D-PESs)and linear interpolated internal coordinate paths connecting relevant structures were calculated at the MS-CASPT2//CASSCF level and confirmed a stepwise ESDPT mechanism.Specifically,the first proton transfer from S1-ENOL to S1-KETO-1 is barrierless,whereas the second one from S1-KETO-1 to S1-KETO-2 demands a barrier of ca.6.0 kcal/mol.The linear interpolated internal coordinate path connecting S1-KETO-1(S1-KETO-2)and S_(1) S0-KETO-1(S1 S0-KETO-2)is uphill with a barrier of ca.12.0 kcal/mol,which will trap DHNA in the S_(1) state while therefore enabling dual-emission bands.On the other hand,the S1/S0 conical intersections would also prompt the S_(1) system to decay to the S_(0) state,which could be to certain extent suppressed by locking the rotation of the C5-C8-C9-O10 dihedral angle.These mechanistic insights are not only helpful for understanding ESDPT but also useful for designing novel molecular materials with excellent photoluminescent performances. 展开更多

Use of free air as oxidant and copper(II)chloride as catalyst,3,3'-di-tert-butylbi-phenyl-2,5,2',5'-diquinone(BBDQ)was prepared via catalytic oxidation of 2,2'-dihydroxy-3,3'-di-tert-butyl-5,5'-dimethoxy-biphenyl(di-BHA).The yield of reaction attained 95% and the selectivity for BBDQ was 100%.The single-crystal X-ray diffraction analysis reveals that the dihedral angle between two rings(benzene rings for di-BHA and quinone rings for BBDQ)changes from 89.8 to 45.3o,indicating the steric hindrance effects of methyl disappear in the oxidation process.The crystal structures of di-BHA and BBDQ are further confirmed by their spectral characterizations.The probable mechanism for this oxidation process is also discussed. 展开更多

Novel segmented thermoplastic polyurethane (TPU) copolymers were synthesized using two-step solventless bulk polymerization. 4,4’-methylenediphenyl diisocyanate (MDI) and 1,4-Butanediol (BDO) were used to form hard segment of TPU and α,ω-dihydroxy-[poly(propyleneoxide)-poly (dimethylsiloxane)-poly(propyleneoxide)] (α,ω-dihydroxy-(PPO-PDMS-PPO)) was used to form soft segment of TPU, where the molar ratio of the –N=C=O/OH was 1.02 and the hard segment weight percentage was 30%. A series of TPUs were characterized by fourier transform infrared spectroscopy (FT-IR). The investigation of triblock oligomer’s PPO molecular weight impact on the derived TPU’s mechanical properties, thermal performance, surface water repellency and morphology performance was carried by Instron material tester, thermogravimetric analysis (TGA), differential scanning calorimetry (DSC), water contact angles (WCA), scanning electron microscope and energy dispersive X-ray spectroscopy (SEM-EDX) and wide angle X-ray diffraction (WAXD), respectively. FT-IR confirmed α,ω-dihydroxy-(PPO-PDMS-PPO) well cooperating into urethane structure and analyzed hydrogen bonding between N-H group with hard segment C=O group and N-H group with soft segment C-O-C group. DSC and WAXD results showed α,ω-dihydroxy-(PPO-PDMS-PPO) segments crystallization. SEM-EDX results showed that the presence of a spherulitic morphology, which arose from the crystallization of the PPO segments. The thermal properties measured by TGA and DSC were slightly affected by molecular weight of PPO and microphase separation. The weight loss of TPUs started between 294°C and 300°C, and Tg was in the range of -70°C to -107°C. TPU copolymers’ surface hydrophobicity property was excellent with WCA range of 95°?to 112°. TPU-3 with 1000 molecular weight PPO has the optimized mechanical properties with tensile strength 16.4 MPa and the modulus at 100% elongation 6.2 MPa and elongation 398%. 展开更多

Novel segmented urethane-siloxane copolymers were synthesized by a two-step bulk polymerization procedure using 4,4'-methylenediphenyl diisocyanate( MDI) and 1,4-butanediol( BDO)as the hard segment components and novel polypropyleneoxide polyether( PPO) blocked polydimethylsilxoane( PDMS) α,ω-dihydroxy-( PPO-PDMS-PPO) as the soft segment component,where the hard segment content was 30% by weight,and the molar ratio of NCO / OH was 1. 02. A series of urethane-siloxane copolymers were prepared from different molecular weights of α,ω-dihydroxy-( PPO-PDMS-PPO). The soft segments were different in the molecular of PDMS block segment. Chemical structure,morphology,mechanical properties,surface properties and thermal properties of thermoplastic polyurethane( TPU) were investigated by Fourier transform infrared spectroscopy( FT-IR),scanning electron microscopy and energy dispersive X-ray spectroscopy( SEM-EDX),mechanical properties( MP) test,water contact angle( WCA) test,differential scanning calorimetry( DSC),thermogravimetric analysis( TGA), respectively. According to the obtained results, the hydrophobicity of silicone modified TPU promoted,surface WCA increased from 73° to 120°. Low temperature resistant performance was improved,and the glass transition temperature decreased from- 44. 2℃ to- 120. 8℃. Thermal stability was also improved,the beginning temperature of thermal degradation increased from 281 ℃to 299 ℃. 展开更多

目的研究串钱柳中间苯三酚类化学成分及其抗耐甲氧西林金黄色葡萄球菌(MRSA)活性,为综合开发串钱柳提供科学依据。方法采用常规柱层析技术对串钱柳间苯三酚类化合物进行纯化,通过理化常数和NMR等方法鉴定化合物的结构;并通过刃天青法、... 目的研究串钱柳中间苯三酚类化学成分及其抗耐甲氧西林金黄色葡萄球菌(MRSA)活性,为综合开发串钱柳提供科学依据。方法采用常规柱层析技术对串钱柳间苯三酚类化合物进行纯化,通过理化常数和NMR等方法鉴定化合物的结构;并通过刃天青法、细胞膜去极化、SYTOX Green和扫描电镜等实验对所获化合物进行抗MRSA活性研究。结果从串钱柳乙酸乙酯萃取部位中分离纯化得到2个间苯三酚类化合物:2,6-dihydroxy-4-methoxyisobutyrophenone(1)、2,6-dihydroxy-4-methoxyisovalerophenone(2);化合物1和2具有显著的抗MRSA活性,抑菌机制为破坏细菌的细胞膜。结论串钱柳间苯三酚衍生物具有良好的抗MRSA活性,化合物1和2为开发新型抗MRSA药物提供物质基础与科学依据。 展开更多

Cultivation of an endophytic fungus Aspergillus niger EN-13 that was isolated from the inner tissue of the marine brown alga Colpomenia sinuosa resulted in the characterization of a new naphthoquinoneimine derivative, namely, 5,7-dihydroxy-2-[1-(4- methoxy-6-oxo-6H-pyran-2-yl)-2-phenylethylamino]-[ 1,4]naphthoquinone. The structure of the new compound was established on the basis of various NMR spectroscopic analyses including 2D NMR techniques, El-MS, and HR-ESI-MS.
