[ Result] The optimum concentration of 20 μl reaction system was 10 × Buffer, 2.00 mmol/L Mg^2+ , 2 U/μl Taq DNA polymerase, 0.2 mmol/L dNTP, 0.2 μmol/L primer and 35 ng/μl DNA template. [ Conclusion] The optimum annealing temperature for cpSSR-PCR reaction system is 52 ℃, and the cpSSR reaction system is steady and reproducible. 展开更多

[Objective] This study aimed to investigate the variation dynamics of total nitrogen and nicotine of burtey tobacco at various growing stages under different ni- trogen nutrition levels to provide reference for optimizing fertilization technologies for burley tobacco. [Method] Fresh burley tobacco leaves were collected at various growing stages and dried to measure the content of total nitrogen, protein and nico- tine by using INTEGRAL automated chemical analyzer and analyze the correlation with nitrogen application level. [Result] Regardless of the nitrogen application level, the content of total nitrogen and protein showed a downward trend since root ex- tending stage and reached the minimum at mature stage; the content of nicotine showed an upward trend since early vigorous growing stage and increased to the maximum at mature stage; the content of total nitrogen, protein and nicotine all in- creased after air curing; the content of total nitrogen, protein and nicotine showed positive correlation with nitrogen application level, while total nitrogen/nicotine was negatively correlated. The results indicate that nitrogen level is closely related to the total nitrogen and nicotine of burley tobacco at growing period and post-air curing, rational application of nitrogen is an important measure to regulate the content of total nitrogen and nicotine of tobacco. [Conclusion] This study provides scientific ba- sis for rational fertilization of burley tobacco. 展开更多

With the help of FESEM, high resolution electron backscatter diffraction can investigate the grains/subgrains as small as a few tens of nanometers with a good angular resolution (~0.5°). Fast development of EBSD speed (up to 1100 patterns per second) contributes that the number of published articles related to EBSD has been increasing sharply year by year. This paper reviews the sample preparation, parameters optimization and analysis of EBSD technique, emphasizing on the investigation of ultrafine grained and nanostructured materials processed by severe plastic deformation (SPD). Detailed and practical parameters of the electropolishing, silica polishing and ion milling have been summarized. It is shown that ion milling is a real universal and promising polishing method for EBSD preparation of almost all materials. There exists a maximum value of indexed points as a function of step size. The optimum step size depends on the magnification and the board resolution/electronic step size. Grains/subgrains and texture, and grain boundary structure are readily obtained by EBSD. Strain and stored energy may be analyzed by EBSD. 展开更多

Liquid-phase oxidation of toluene with air has become the main technology for producing benzoic acid in a reactor at present. Based on the kinetic model of the toluene oxidation process obtained from laboratory and mass balance of key component, a novel model is established to simulate the industrial toluene oxidation process, in which the effects of benzaldehyde and benzyl alcohol are considered and the kinetic parameters are revised by industrial data. The simulation results show that the error of benzoic acid yield is within 3.5%. Based on the simulation model, to maximize the benzoic acid yield, an optimization model is proposed to optimize the operating parameters, including toluene feed-in mass flux and temperature. The optimization result indicates that on the allowable operating conditions the maximum benzoic acid yield obtained with the reaction temperature at 167.2 C an the mass flux at 104.1 t·h^- 1 is greater than the current one, which can be used to guide industrial reactor s operation. 展开更多

Both the theoretical analysis and numerical simulation are employed to study the bearing capacity of CBF and the relationship between loads and ground deformation. Furthermore, monopile, high-rise pile cap, tripod and CBF designs are compared to analyze the bearing capacity and ground deformation, with a 3-MW wind generator as an example. The resuits indicate that CBF can effectively bear horizontal load and large bending moment resulting from upper structures and environmental load. 展开更多

The supervised rules of intelligent concept are introduced into genetic algorithms (GAs) to solve the constraint optimization problem. One of the main characteristics of SGA is its searching space can be limited in feasible regions rather than infeasible regions. The superiority of SGA to other GAs lies in that some supervised search rules in which the information comes from the problems are incorporated into SGA. The simulation results show that SGA improves the ability of searching an optimum solution and accelerates the convergent process up to 20 times. 展开更多

In order to maximize the economic benefits of the ARGG unit a string of measures, including the modification of catalyst formulation, retention of high catalyst activity, application of mixed termina-tion agents to control the reaction temperature and once-through operation, and optimization of catalyst regeneration technique, were adopted to adapt the ARGG unit to processing of the heavy feedstock with its carbon residue equating to 7 % on an average. The cultivation of the first filial generation of monoploid gametophyte clones of different Laminaria species (hybrid Laminaria) is an effective way of utilizing heterozygous vigor (heterosis). A female gametophyte clone of L. japonica and a male gametophyte clone of L. longissima were hybridized, generating Dongfang No.2 hybrid Laminaria. The parentage of this hybrid Laminaria was determined using AFLP of total DNA, SNP of the ITS region of ribosomal RNA transcription unit and microsatellite DNA variation at two loci. In addition to 167 AFLP bands shared by Dongfang No.2, L. japonica and L. longissima, Dongfang No.2 hybrid Laminaria shared another 70 and 55 bands with L. japonica and L. longissima, respectively, which were obviously more than 11 bands shared by L. japonica and L. longissima. Dongfang No.2 held both ‘T’ and ‘C’ at position 847 of the ITS region, while ‘T’ at this position was specific for L. japonica and ‘C’ for L. longissima, respectively. Dongfang No.2 also held the microsatellite DNA alleles of two parents together at two microsatellite DNA marker loci. These observations clearly proved that Dongfang No.2 is a true hybrid of L. japonica and L. longissiuma. Unfortunately, the origin of the chloroplast of Dongfang No.2 was not determined based on the variation of RuBisCo spacer. More sequence variants of both ITS region and RuBisCo spacer were identified in Dongfang No.2 and most of them did not exist in either L. japonica or L. longissima. The unexpected variants may be due to the mutation of ga- metophyte clones occurring during their vegetative amplification. 展开更多

In this paper, a novel unmanned aerial vehicle(UAV)-enabled full duplex decode-and-forward(DF) technique is used in mobile relaying system. Compared with conventional static relaying, mobile relaying provides more degree of freedom for experiencing better channel conditions and further improving the system reliable performance. The source and relay transmit power as well as relay trajectory are jointly optimized for sum outage probability minimization in mobile relaying system. Due to the non-convex nature of the original problem, block coordinate decent optimization techniques are employed to decompose it into two sub-problems, which leads to an efficient iterative algorithm. Specifically, for the pre-determined relay trajectory, the optimal transmit power of source and relay are obtained by solving Karush-Kuhn-Tucker(KKT) conditions. For the given source/relay power allocation, the optimal UAV trajectory is obtained by solving dual problem. Based on the two steps, an iterative algorithm is proposed to jointly optimize source/relay power allocation and UAV trajectory alternately. Numerical results show the performance gain of our proposed scheme. 展开更多

The methylotrophic yeast Pichia pastoris is a highly successful system for production of a variety of heterologous proteins due to its unique features/abilities for effective protein expression, and tremendous efforts have been made to increase heterologous protein productivity by P. pastoris in recent years. When new engineered yeast strains are constructed and are ready to use tot industrial protein production, process control and optimization techniques should be applied to improve the fermentation performance in the following aspects: (1) increase recombinant cell concentrations in fermentor to high density during growth phase; (2) effectively induce heterologous proteins by enhancing/stabilizing titers or concentrations of the proteins during induction phase; (3) decrease operation costs by relieving the working loads of heat-exchange and oxygen supply. This article reviews and discusses the key and commonly used techniques in heterologous protein production by P. pastoris, with the focus on optimizations of fermentation media and basic operation conditions, development of optimal glycerol feeding strategies for achieving high density cultivation of P. pastoris and effective heterologous protein induction methods by regulating specific growth rate, methanol concentration, temperatures, mixture ratio of multi-carbon substrates, etc. Metabolic analysis for recombinant protein production by P. pastoris is also introduced to interpret the mechanism of sub-optimal heterologous protein production and to explore further optimal expression methods. 展开更多

Copy-Move Forgery(CMF) is one of the simple and effective operations to create forged digital images.Recently,techniques based on Scale Invariant Features Transform(SIFT) are widely used to detect CMF.Various approaches under the SIFT-based framework are the most acceptable ways to CMF detection due to their robust performance.However,for some CMF images,these approaches cannot produce satisfactory detection results.For instance,the number of the matched keypoints may be too less to prove an image to be a CMF image or to generate an accurate result.Sometimes these approaches may even produce error results.According to our observations,one of the reasons is that detection results produced by the SIFT-based framework depend highly on parameters whose values are often determined with experiences.These values are only applicable to a few images,which limits their application.To solve the problem,a novel approach named as CMF Detection with Particle Swarm Optimization(CMFDPSO) is proposed in this paper.CMFD-PSO integrates the Particle Swarm Optimization(PSO) algorithm into the SIFT-based framework.It utilizes the PSO algorithm to generate customized parameter values for images,which are used for CMF detection under the SIFT-based framework.Experimental results show that CMFD-PSO has good performance. 展开更多
