Methods: The contents of nucleosides were determined by using high performance capillary electrophoresis (HPCE). Beckman P/ACE System 5010 apparatus equipped with a UV detector and a Beckman untreated fused-silica capillary (57 cm 75 mm, 50 cm effective length) was used. Before sample injection, the capillary was rinsed with 1 molL-1 sodium hydroxide solution and running buffer for 5 min, respectively. A voltage of 20 kV was applied for the separation. Pressure injection was 586 kPa for 6 seconds, and the wavelength of detector was 254 nm. The running time was 20 min at 20 oC. The effect of humidity and heat on the contents of nucleosides from natural Cordyceps sinensis and cultured Cordyceps mycelia was observed for 1, 3, 5 and 10 days at temperature 40 oC, and relative humidity 75%. Results: The content of nucleosides from natural Cordyceps sinensis was higher than that from cultured Cordyceps mycelia. But the contents of nucleosides from freshly collected natural Cordyceps sinensis were very low, even below the limit of quantitation. The contents of nucleosides from natural Cordyceps sinensis were significantly increased by humidity and heat, but this phenomenon was not observed in cultured Cordyceps mycelia. Conclusion: There are differences between the nucleosides from natural Cordyceps sinensis and cultured Cordyceps mycelia. The nucleosides in natural Cordyceps sinensis may be derived from the degradation of nucleic acids. This implies that adenosine being used for the quality control of natural Cordyceps sinensis may have to be reconsidered.展开更多

Methods: The SCI of Wistar rats was done by Allens weight dropping way by a 10 g×2.5 cm impact on the posterior of spinal cord T 8. NGF (3 g/L, 20 μl) or normal saline was injected through catheter into subarachnoid space 2, 4, 8, 12 and 24 h after SCI. The expression of N-methyl-D-asparate receptor 1 (NMDAR 1) and neuronal constitutive nitric oxide synthase (ncNOS) mRNA in rat spinal cord was detected by in situ hybridization. Results: Abnormal expression of NMDAR 1 and ncNOS mRNA appeared in spinal ventral horn motorneuron in injured rats, as compared with that in control group. The expression of NMDAR 1 and ncNOS mRNA in NGF group was significantly lower than that in saline group (P<0.01). Conclusion: NGF can protect spinal cord against injury in vivo. One of the mechanisms is that NGF can prohibit NMDAR 1 and nitric oxide (NO) production after spinal cord injury.展开更多

The amount of radioactivity excreted in urine, feces and bile within 24 h afterintravenous administration of ^(125)I recombinant E. col L-asparaginase to rats was 68.95% ,4.44%and 5.36% of the dose respectively. ^(125)I recombinant E. coli L-asparaginase in plasma samples wasdetermined. The levels of structural intact molecule in plasma samples were evaluated by SDS-PAGEand bio-imaging analyzer system. Pharmacokinetic parameters were assessed with a model-dependentmethod. The concentration-time curves of recombinant E. coli L-asparaginase after intravenousinjection at 1 250 IU·kg^(-1), 2 500, IU·kg^(-1), 5 000 IU·kg^(-1) to rats were consistent withthe two-compartment model. The first and terminal elimination t_(1/2) were 0.52 ~ 0.63 h and 2.39 ~2.76 h respectively. AUC was linearly related to the doses. The results of distribution in tissuesor organs and excretion in urine suggested that the metabolites of the enzyme were cleared bymechanisms of urinary excretion. Pharmacokinetics parameters of recombinant E. coli L- asparaginasein rats are warranted for the design of future clinical trials.展开更多

The aspartic acid(ASP),an environment-friendly corrosion inhibitor,was explored to intercalate into the ZnAl-layered double hydroxide(ZnAl-LDH)and MgAl-LDH coatings on AZ31 magnesium alloys by a facile one-step hydrot...The aspartic acid(ASP),an environment-friendly corrosion inhibitor,was explored to intercalate into the ZnAl-layered double hydroxide(ZnAl-LDH)and MgAl-LDH coatings on AZ31 magnesium alloys by a facile one-step hydrothermal method.The morphology,composition,structure and corrosion resistance of the prepared coatings were comparatively investigated.It is found that the uniform and dense layered nanosheet(NS)vertically grows on the substrate,and the MgAl−ASP-LDH films exhibit a three-dimensional(3D)rose-like sheet structure with high ratio of pore coverage.The MgAl−ASP-LDH coating exhibits better corrosion resistance and durability of longtime immersion than ZnAl−ASP-LDH coating.The corrosion current density of two coatings is two or three orders of magnitude lower than that of bare Mg alloy,indicating that ZnAl/MgAl-LDH films intercalated with organic ASP anions can significantly improve the corrosion resistance of Mg alloy.展开更多

Environmentally friendly flotation reagent,polyaspartic acid(PAPA),was tested as a potential selective depressant in the flotation separation of chalcopyrite and Cu-activated sphalerite.The depression mechanism of PAP...Environmentally friendly flotation reagent,polyaspartic acid(PAPA),was tested as a potential selective depressant in the flotation separation of chalcopyrite and Cu-activated sphalerite.The depression mechanism of PAPA was revealed by contact angle measurements,Zeta potential measurements,Fourier transform infrared spectroscopy(FT-IR)analysis and inductively coupled plasma(ICP)measurement.The micro-flotation tests with single minerals showed that PAPA selectively depressed Cu-activated sphalerite,while chalcopyrite remained floatable.Moreover,a concentrate containing 31.40%Cu with a recovery of 92.43%was obtained in flotation tests of artificially mixed minerals.Results of contact angle measurements,Zeta potential measurements and FT-IR spectrum revealed that PAPA exerted a much stronger adsorption on Cu-activated sphalerite surface than on chalcopyrite surface,preventing the further adsorption of sodium diethyl dithiocarbamate(DDTC)on its surface.ICP measurements indicated that PAPA had an excellent complexing ability with Cu^(2+)in flotation pulp,weakening the activation of Cu species on sphalerite surface and producing selective depression.展开更多

An important antiapoptosis factor, Bcl-2 protein, contributes in caspase pathway of apoptosis. Calcium, an important intracellular signal element in cells, is also observed to have changes during apoptosis, which maybe affected by Bcl-2 protein. We have previously reported that in Harringtonine (HT) induced apoptosis of HL-60 cells, there’s a change of intracellular calcium distribution, moving from cytoplast especially Golgi’s apparatus to nucleus and accumulating there with the highest concentration. We report here that caspase-3 becomes activated in HT-induced apoptosis of HL-60 cells, which can be inhibited by overexpression of Bcl-2 protein. No sign of apoptosis or intracellular calcium movement from Golgi’s apparatus to nucleus in HL-60 cells overexpressing Bcl-2 or treated with Ac-DEVD-CHO, a specific inhibitor of caspase-3. The results indicate that activated caspase-3 can promote the movement of intracellular calcium from Golgi’s apparatus to nucleus, and the process is inhibited by Ac-DEVD-CHO (inhibitor of caspase-3), and that Bcl-2 can inhibit the movement and accumulation of intracellular calcium in nucleus through its inhibition on caspase3. Calcium relocalization in apoptosis seems to be irreversible, which is different from the intracellular calcium changes caused by growth factor.展开更多

A variety of primer construction, primer concentration, dNTP concentration, and annealing temperature of primers were systematically examined. Pair primers specifi c to rtL180M and rtM204V were selected for YVDD detection. Primer specif ic to rtM204I with an additional 3’-penultimate base mismatched to both the mutant and wild-type sequence was selected for YIDD detection. We applied this assay to study YMDD mutants in 28 chronic hepatitis B patients before and after lamivudine treatment.RESULTS: We could detect as little as 0.001%-0.00001% of mutant viruses coexisting in 108-109 copies of wild-type HBV using this assay. YMDD mutants were detected in 8 of 12 HBeAg-positive patients and 8 of 16 HBeAg-negative patients before lamivudine treatment. After treatment, two more patients in HBeAg-positive patients and seven more patients in HBeAg-negative patients developed YMDD mutations. CONCLUSION: We developed a highly sensitive and specifi c assay for detecting YMDD mutants. This assay can be applied to monitor chronic hepatitis B patients before and during lamivudine treatment.展开更多

T. harzianum shared 53% identity of secreted aspartic proteinase gene with G. zeae, 37% with N. crassa and 36% with C. globosum. The full-length cDNA sequence of secreted aspartic proteinase gene from T. harzianum was cloned for the first time by using BD SMART RACE technique, which provides a foundation to obtain and validate functional genes of T. harzianum.展开更多
Using tropical cyclone (TC) observations over a 58-yr period (1949-2006) from the China Meteorological Administration, the 40-year ECMWF Reanalysis (ERA-40), NCEP-NCAR reanalysis, and the Hadley Centre sea ice a...Using tropical cyclone (TC) observations over a 58-yr period (1949-2006) from the China Meteorological Administration, the 40-year ECMWF Reanalysis (ERA-40), NCEP-NCAR reanalysis, and the Hadley Centre sea ice and sea surface temperature (HadISST) datasets, the authors have examined the behaviors of tropical cyclones (TCs) in the western north Pacific (WNP) in boreal winter (November-December-January-February). The results demonstrate that the occurrences of wintertime TCs, including super typhoons, have decreased over the 58 years. More TCs are found to move westward than northeastward, and the annual total number of parabolic-track-type TCs is found to be decreasing. It is shown that negative sea surface temperature anomalies (SSTAs) related to La Nifia events in the equatorial central Pacific facilitate more TC genesis in the WNP region. Large-scale anomalous cyclonic circulations in the tropical WNP in the lower troposphere are observed to be favorable for cyclogenesis in this area. On the contrary, the positive SSTAs and anomalous anticyclonic circulations that related to E1 Nifio events responsible for fewer TC genesis. Under the background of global warming, the western Pacific subtropical high tends to intensify and to expand more westward in the WNP, and the SSTAs display an increasing trend in the equatorial eastern-central Pacific. These climate trends of both atmospheric circulation and SSTAs affect wintertime TCs, inducing fewer TC occurrences and causing more TCs to move westward.展开更多

Cell proliferation, collagen content, and type 1 collagen expression were examined in activated HSC-T6 cells. Collagen was determined by estimating the hydroxyproline content. In rats with DMN-induced hepatic fi brosis, serum aspartate aminotransferase and alanine aminotransferase concentrations, liver hydroxyproline and lipid peroxides were determined. Pathologic changes were examined by hematoxylin & eosin staining.RESULTS: GT administration prevented the development of hepatic fibrosis in the rat model of DMN-induced liver fi brosis. These results were confi rmed both by liver histology and by quantitative measurement of hepatic hydroxyproline content, a marker of liver collagen deposition. Accordingly, inhibition of proliferation, reduced collagen deposition, and type 1 collagen expression were observed in activated HSC-T6 cells following GT treatment. These results imply that GT reduced the proliferation of activated HSC and down regulated the collagen content and expression of collagen type 1, thereby ameliorating hepatic fibrosis.CONCLUSION: This study demonstrates that greentea administration can effectively improve liver fibrosis caused by DMN, and may be used as a therapeutic option and preventive measure against hepatic f ibrosis.展开更多

[Results] The treatment of 5‰ humic acid + 3‰ PASP + compound fertilizer exhibited the plant height, stem diameter and yield increased by8.3%, 13.3% and 10.3%, respectively, and thus was the best; and the treatment showed a single curd weight of 478 g and a commodity rate of 89.4%, and had lower economic cost than treatment of 10‰ humic acid + 6‰ PASP + compound fertilizer. [Conclusion] In summary, 5‰ humic acid + 3‰ PASP + compound fertilizer is suitable for wide application in broccoli culture.展开更多

Methods LC3B expression was detected by immunohistochemistry in 53 human colorectal cancer tissues and 20 normal colon tissues.The protein levels of LC3B-Ⅱand active caspase-3 were also determined by Western blot analysis in 23 human colorectal cancer tissues and 10 normal colon tissues. Results LC3B was expressed both in cancer cells and normal epithelial cells.LC3B expression in the peripheral area of cancer tissues was correlated with several clinicopathological factors,including tumor differentiation(P=0.002),growth pattern of the tumor margin (P=0.028),pN(P=0.002),pStage(P=0.032),as well as vessel and nerve plexus invasion(P=0.002).The protein level of LC3B-Ⅱin cancer tissue was significantly higher than in normal tissue(P=0.038),but the expression of active forms of procaspase-3 in cancer tissue was lower(P=0.041).There was a statistically significant positive correlation between the expression levels of LC3B-Ⅱand the active forms of procaspase-3(r=0.537,P=0.008). Conclusions Autophagy has a prosurvival role in human colorectal cancer.Autophagy enhances the aggressiveness of colorectal cancer cells and their ability to adapt to apoptotic stimulus.展开更多

文摘[Objective]The study aimed to investigate the effects of the amino acids of aspartate family on the biosynthesis of CoQ10 in Rhodpseudomonas palustris J001.[Method]The impacts of amino acids of this family on the biosynthesis of CoQ10 in Rhodopseudomnas palustris J001 were investigated by feeding these amino acids at the end of the logarithmic phase during incubation,which aim was for the optimization of the fermentation medium and genetic improvement of the strain for CoQ10 production.[Result]The results showed that feeding proper amount of methione(125 mg/L)could increase CoQ10 production by 20.2%,but feeding of lysine(above 500 mg/L),threonine(above 400 mg/L)and/or isoleucine(above 400 mg/L)repressed the biosynthesis of CoQ10.The results indicated that the aspartate kinase is subject to feedback inhibition or repression by lysine,threonine and isoleucine in the strain,which was unfavorable to the formation of methioine and then caused the decrease of CoQ10 production.[Conclusion]Lysine,threonine and isoleucine auxotrophic mutants with resistance to analogues of lysine,threonine and isoleucine could increase the production of CoQ10.
文摘Objective:To explore the protective mechanisms of nerve growth factor (NGF) on spinal cord injury (SCI) and provide theoretical basis for its clinical application. Methods: The SCI of Wistar rats was done by Allens weight dropping way by a 10 g×2.5 cm impact on the posterior of spinal cord T 8. NGF (3 g/L, 20 μl) or normal saline was injected through catheter into subarachnoid space 2, 4, 8, 12 and 24 h after SCI. The expression of N-methyl-D-asparate receptor 1 (NMDAR 1) and neuronal constitutive nitric oxide synthase (ncNOS) mRNA in rat spinal cord was detected by in situ hybridization. Results: Abnormal expression of NMDAR 1 and ncNOS mRNA appeared in spinal ventral horn motorneuron in injured rats, as compared with that in control group. The expression of NMDAR 1 and ncNOS mRNA in NGF group was significantly lower than that in saline group (P<0.01). Conclusion: NGF can protect spinal cord against injury in vivo. One of the mechanisms is that NGF can prohibit NMDAR 1 and nitric oxide (NO) production after spinal cord injury.

Method Ugifour-component condensation reaction was used to synthesize peptide mimic CPP32 inhibitors; ResultsA key isocyanide component (aspartate-derived isocyanide 3) and one of the designed CPP32inhibitors 4 (as a template) were synthesized; Conclusion The CPP32 inhibitor 4 was synthesized bythe newly developed procedure, which is an Ugi four-component condensation reaction based onaspartate-derived isocyanide 3. This method can be used to build up the CPP32 inhibitor library.
文摘Aim To synthesize the tripepide Weinreb amide Boc Asp(OBzl) β Ala Asp(OBzl) N(OMe)Me (7) as a useful precursor of aspartyl peptide aldehyde derivatives; Methods DCC, IBCF method was used for preparation of Weinreb amide; N hydroxysuccinimide activated ester was used in peptide synthesis; and Boc as N protecting group of amino acid. Results Boc Asp(OBzl) N(OMe)Me (3), Boc β Ala Asp(OBzl) N(OMe)Me (5), and Boc Asp(OBzl) β Ala Asp(OBzl) N(OMe)Me (7) were synthesized successfully. Conclusion An useful precursor of tripeptide aspartyl aldehydes was synthesized.

文摘The distribution of ^(125)I recombinant E. coli L-asparaginase in tissues ororgans and the excretion in urine, feces and bile were studied with in vivo radioactive tracertechnique. The amount of radioactivity excreted in urine, feces and bile within 24 h afterintravenous administration of ^(125)I recombinant E. col L-asparaginase to rats was 68.95% ,4.44%and 5.36% of the dose respectively. ^(125)I recombinant E. coli L-asparaginase in plasma samples wasdetermined. The levels of structural intact molecule in plasma samples were evaluated by SDS-PAGEand bio-imaging analyzer system. Pharmacokinetic parameters were assessed with a model-dependentmethod. The concentration-time curves of recombinant E. coli L-asparaginase after intravenousinjection at 1 250 IU·kg^(-1), 2 500, IU·kg^(-1), 5 000 IU·kg^(-1) to rats were consistent withthe two-compartment model. The first and terminal elimination t_(1/2) were 0.52 ~ 0.63 h and 2.39 ~2.76 h respectively. AUC was linearly related to the doses. The results of distribution in tissuesor organs and excretion in urine suggested that the metabolites of the enzyme were cleared bymechanisms of urinary excretion. Pharmacokinetics parameters of recombinant E. coli L- asparaginasein rats are warranted for the design of future clinical trials.

文摘Polyaspartic acid(PASP)was used as a novel environmental-friendly depressant,and its inhibition effect on flotation performance of scheelite and calcite using sodium oleate(NaOL)as a collector was investigated by ways of flotation experiments,zeta potential measurement,contact angle measurement and infrared spectroscopic analysis(IR).The results show that PASP exhibits stronger inhibition capability and selectivity than acidified water glass,and the flotation separation of scheelite from calcite can be realized in the presence of 6 mg/L PASP and 20 mg/L NaOL at pH>6.In the presence of PASP,the zeta potential of calcite surface almost keeps unchanged after adding NaOL,and the contact angle decreases sharply.Therefore,it is indicated that PASP significantly decreases the adsorption of NaOL on calcite surface.By contrast,it has no distinct effect on the adsorption of NaOL on the scheelite surface,which is further confirmed by IR results.

文摘The aspartic acid(ASP),an environment-friendly corrosion inhibitor,was explored to intercalate into the ZnAl-layered double hydroxide(ZnAl-LDH)and MgAl-LDH coatings on AZ31 magnesium alloys by a facile one-step hydrothermal method.The morphology,composition,structure and corrosion resistance of the prepared coatings were comparatively investigated.It is found that the uniform and dense layered nanosheet(NS)vertically grows on the substrate,and the MgAl−ASP-LDH films exhibit a three-dimensional(3D)rose-like sheet structure with high ratio of pore coverage.The MgAl−ASP-LDH coating exhibits better corrosion resistance and durability of longtime immersion than ZnAl−ASP-LDH coating.The corrosion current density of two coatings is two or three orders of magnitude lower than that of bare Mg alloy,indicating that ZnAl/MgAl-LDH films intercalated with organic ASP anions can significantly improve the corrosion resistance of Mg alloy.

文摘Polyaspartic acid(PASP)is suitable for the inhibition of scale deposition from water.To enhance its in- hibition efficiency,PASP was modified by reacting aspartic acid(Asp)with glutamic acid(Glu)to provide Asp-Glu copolymer under microwave irradiation.The influence of reaction parameters on conversion,molecular weight and inhibition of CaCO3 precipitation was investigated Infra-red.(IR), 1H nuclear magnetic resonance( 1H NMR)and 13C nuclear magnetic resonance( 13C NMR)spectroscopies were used to characterize the copolymer.The results show that copolymerization of aspartic acid and glutamic acid is catalyzed by a small amount of phosphorous acid (H3PO4)in solvent,the product conversion is 98.05%under the following conditions:the molar ratio of glutamic acid to reactant[Glu/(Asp+Glu)]is 0.3 and that of catalyst(Cat)to reactant[Cat/(Glu+Asp)]is 0.05(0.65ml H3PO4),the volume of solvent dimethylformamide is 16ml,the microwave power used is 720W and the reaction for 3 min.The weight average molecular weight of copolymer synthesized under these conditions is 2709 and the inhi- bition rate for CaCO3 is 97.75%.

文摘Environmentally friendly flotation reagent,polyaspartic acid(PAPA),was tested as a potential selective depressant in the flotation separation of chalcopyrite and Cu-activated sphalerite.The depression mechanism of PAPA was revealed by contact angle measurements,Zeta potential measurements,Fourier transform infrared spectroscopy(FT-IR)analysis and inductively coupled plasma(ICP)measurement.The micro-flotation tests with single minerals showed that PAPA selectively depressed Cu-activated sphalerite,while chalcopyrite remained floatable.Moreover,a concentrate containing 31.40%Cu with a recovery of 92.43%was obtained in flotation tests of artificially mixed minerals.Results of contact angle measurements,Zeta potential measurements and FT-IR spectrum revealed that PAPA exerted a much stronger adsorption on Cu-activated sphalerite surface than on chalcopyrite surface,preventing the further adsorption of sodium diethyl dithiocarbamate(DDTC)on its surface.ICP measurements indicated that PAPA had an excellent complexing ability with Cu^(2+)in flotation pulp,weakening the activation of Cu species on sphalerite surface and producing selective depression.

文摘Apoptosis manifests in two major execution programs downstream of the death signal: the caspase pathway and organelle dysfunction. An important antiapoptosis factor, Bcl-2 protein, contributes in caspase pathway of apoptosis. Calcium, an important intracellular signal element in cells, is also observed to have changes during apoptosis, which maybe affected by Bcl-2 protein. We have previously reported that in Harringtonine (HT) induced apoptosis of HL-60 cells, there’s a change of intracellular calcium distribution, moving from cytoplast especially Golgi’s apparatus to nucleus and accumulating there with the highest concentration. We report here that caspase-3 becomes activated in HT-induced apoptosis of HL-60 cells, which can be inhibited by overexpression of Bcl-2 protein. No sign of apoptosis or intracellular calcium movement from Golgi’s apparatus to nucleus in HL-60 cells overexpressing Bcl-2 or treated with Ac-DEVD-CHO, a specific inhibitor of caspase-3. The results indicate that activated caspase-3 can promote the movement of intracellular calcium from Golgi’s apparatus to nucleus, and the process is inhibited by Ac-DEVD-CHO (inhibitor of caspase-3), and that Bcl-2 can inhibit the movement and accumulation of intracellular calcium in nucleus through its inhibition on caspase3. Calcium relocalization in apoptosis seems to be irreversible, which is different from the intracellular calcium changes caused by growth factor.
文摘AIM: To develop a PCR assay using mutant-specific primers to detect mutation of tyrosine-methionine-aspartate-aspartate (YMDD) motif of HBV to tyrosine-valine-aspartate-aspartate (YVDD) or tyrosine-isoleucine-aspartate-aspartate (YIDD).METHODS: Cloned wild-type and mutant HBV sequences were used as templates to test the sensitivity and specificity of the assay. A variety of primer construction, primer concentration, dNTP concentration, and annealing temperature of primers were systematically examined. Pair primers specifi c to rtL180M and rtM204V were selected for YVDD detection. Primer specif ic to rtM204I with an additional 3’-penultimate base mismatched to both the mutant and wild-type sequence was selected for YIDD detection. We applied this assay to study YMDD mutants in 28 chronic hepatitis B patients before and after lamivudine treatment.RESULTS: We could detect as little as 0.001%-0.00001% of mutant viruses coexisting in 108-109 copies of wild-type HBV using this assay. YMDD mutants were detected in 8 of 12 HBeAg-positive patients and 8 of 16 HBeAg-negative patients before lamivudine treatment. After treatment, two more patients in HBeAg-positive patients and seven more patients in HBeAg-negative patients developed YMDD mutations. CONCLUSION: We developed a highly sensitive and specifi c assay for detecting YMDD mutants. This assay can be applied to monitor chronic hepatitis B patients before and during lamivudine treatment.

文摘Total RNA was isolated from mycelium of T. harzianum by Total RNA extraction kit, and two clear bands of rRNA (28S and 18S) were observed in agarose electrophoresis. By joining the 3'end sequence with the known SA76 EST from cDNA library of T. harzianum, a full-length cDNA sequence of 2019bp was obtained, whose open reading frame contained 1593bp, a stop codon TAA, a 5'untranslated region (5'UTR) of 266bp, a 3'untranslated region (3'UTR) of 201bp, and poly (A) 29 encoded a protein of 530 amino acids, had a signal peptide. T. harzianum shared 53% identity of secreted aspartic proteinase gene with G. zeae, 37% with N. crassa and 36% with C. globosum. The full-length cDNA sequence of secreted aspartic proteinase gene from T. harzianum was cloned for the first time by using BD SMART RACE technique, which provides a foundation to obtain and validate functional genes of T. harzianum.

文摘Using tropical cyclone (TC) observations over a 58-yr period (1949-2006) from the China Meteorological Administration, the 40-year ECMWF Reanalysis (ERA-40), NCEP-NCAR reanalysis, and the Hadley Centre sea ice and sea surface temperature (HadISST) datasets, the authors have examined the behaviors of tropical cyclones (TCs) in the western north Pacific (WNP) in boreal winter (November-December-January-February). The results demonstrate that the occurrences of wintertime TCs, including super typhoons, have decreased over the 58 years. More TCs are found to move westward than northeastward, and the annual total number of parabolic-track-type TCs is found to be decreasing. It is shown that negative sea surface temperature anomalies (SSTAs) related to La Nifia events in the equatorial central Pacific facilitate more TC genesis in the WNP region. Large-scale anomalous cyclonic circulations in the tropical WNP in the lower troposphere are observed to be favorable for cyclogenesis in this area. On the contrary, the positive SSTAs and anomalous anticyclonic circulations that related to E1 Nifio events responsible for fewer TC genesis. Under the background of global warming, the western Pacific subtropical high tends to intensify and to expand more westward in the WNP, and the SSTAs display an increasing trend in the equatorial eastern-central Pacific. These climate trends of both atmospheric circulation and SSTAs affect wintertime TCs, inducing fewer TC occurrences and causing more TCs to move westward.

The crystal of this chiral complex belongs to monoclinic, space group P21 with a = 13.208(1), b = 10.741(1), c = 19.398(1) ? b = 104.39(1)o, V = 2665.8(4) 3, Z = 4, Dc = 1.435 g/cm3, m(MoKa) = 0.780 mm-1 and F(000) = 1198. The final R = 0.0504 and wR = 0.1267 for 9159 observed reflections (I > 2s(I)), and R = 0.065 and wR = 0.1299 for 11006 independent reflections. The complex assumes a distorted octa- hedral coordinate geometry formed by one aspartate dianion and three benzimidazole ligands. The aspartate chelates to the Ni atom as a tridentate ligand. The lattice water molecules are hydrogen- bonded to the Ni complex molecule.
文摘AIM: To examine the protective effect of green tea extract (GT) on hepatic fi brosis in vitro and in vivo in dimethylnitrosamine (DMN)-induced rats.METHODS: HSC-T6, a rat hepatic stellate cell line, was used as an in vitro assay system. Cell proliferation, collagen content, and type 1 collagen expression were examined in activated HSC-T6 cells. Collagen was determined by estimating the hydroxyproline content. In rats with DMN-induced hepatic fi brosis, serum aspartate aminotransferase and alanine aminotransferase concentrations, liver hydroxyproline and lipid peroxides were determined. Pathologic changes were examined by hematoxylin & eosin staining.RESULTS: GT administration prevented the development of hepatic fibrosis in the rat model of DMN-induced liver fi brosis. These results were confi rmed both by liver histology and by quantitative measurement of hepatic hydroxyproline content, a marker of liver collagen deposition. Accordingly, inhibition of proliferation, reduced collagen deposition, and type 1 collagen expression were observed in activated HSC-T6 cells following GT treatment. These results imply that GT reduced the proliferation of activated HSC and down regulated the collagen content and expression of collagen type 1, thereby ameliorating hepatic fibrosis.CONCLUSION: This study demonstrates that greentea administration can effectively improve liver fibrosis caused by DMN, and may be used as a therapeutic option and preventive measure against hepatic f ibrosis.

文摘[Objective] This study was conducted to analyze the effects of humic acid and PASP on the growth of broccoli to provide a basis for fertilizer application.[Method] A field experiment was carried out to investigate broccoli growth, yield and quality by using humic acids and PASP as fertilizer synergists in the mixture with compound fertilizer. [Results] The treatment of 5‰ humic acid + 3‰ PASP + compound fertilizer exhibited the plant height, stem diameter and yield increased by8.3%, 13.3% and 10.3%, respectively, and thus was the best; and the treatment showed a single curd weight of 478 g and a commodity rate of 89.4%, and had lower economic cost than treatment of 10‰ humic acid + 6‰ PASP + compound fertilizer. [Conclusion] In summary, 5‰ humic acid + 3‰ PASP + compound fertilizer is suitable for wide application in broccoli culture.

文摘Objective To investigate LC3B-Ⅱand active caspase-3 expression in human colorectal cancer to elucidate the role of autophagy and to explore the relationship of autophagy with apoptosis in human colorectal cancer. Methods LC3B expression was detected by immunohistochemistry in 53 human colorectal cancer tissues and 20 normal colon tissues.The protein levels of LC3B-Ⅱand active caspase-3 were also determined by Western blot analysis in 23 human colorectal cancer tissues and 10 normal colon tissues. Results LC3B was expressed both in cancer cells and normal epithelial cells.LC3B expression in the peripheral area of cancer tissues was correlated with several clinicopathological factors,including tumor differentiation(P=0.002),growth pattern of the tumor margin (P=0.028),pN(P=0.002),pStage(P=0.032),as well as vessel and nerve plexus invasion(P=0.002).The protein level of LC3B-Ⅱin cancer tissue was significantly higher than in normal tissue(P=0.038),but the expression of active forms of procaspase-3 in cancer tissue was lower(P=0.041).There was a statistically significant positive correlation between the expression levels of LC3B-Ⅱand the active forms of procaspase-3(r=0.537,P=0.008). Conclusions Autophagy has a prosurvival role in human colorectal cancer.Autophagy enhances the aggressiveness of colorectal cancer cells and their ability to adapt to apoptotic stimulus.
