Smooth muscle progenitor cells (SPCs), especially from peripheral blood, may offer an alternative cell source for tissue engineering involving a less invasive harvesting technique. Methods: SPCs were isolated from 5-ml fresh rat peripheral blood by density-gradient centrifugation and cultured for 3 weeks in endothelial growth medium-2-MV (EGM-2-MV) medium containing platelet-derived growth factoroBB (PDGF BB). Before seeded on the synthesized scaffold, SPC-derived smooth muscle outgrowth cell (SOC) phenotypes were assessed by immuno-fluorescent staining, Western blot analysis, and reverse transcription polymerase chain reaction (RT-PCR). The cells were seeded onto the silk fibroin-modified poly(3-hydroxybutyrate-co-3-hydroxyhexanoate) (SF-PHBHHx) scaflblds by 6× 10^4 cells/cm^2 and cultured under the static condition for 3 weeks. The growth and proliferation of the seeded cells on the scaffold were analyzed by 3-(4,5-dimethylthiazol-2-yl)-diphenyltetrazolium bromide (MTT) assay, scanning electron microscope (SEM), and 4,6-diamidino-2-phenylindole (DAPI) staining. Results: SOCs displayed specific "hill and valley" morphology, expressed the specific markers of the SMC lineage: smooth muscle (SM) a-actin, calponin and smooth muscle myosin heavy chain (SM MHC) at protein and messenger ribonucleic acid (mRNA) levels. RT-PCR results demonstrate that SOCs also expressed smooth muscle protein 22a (SM22a, a contractile protein, and extracellular matrix components elastin and matrix Gla protein (MGP), as well as vascular endothelial growth factor (VEGF). After seeded on the SF-PHBHHx scaffold, the cells showed excellent metabolic activity and proliferation. Conclusion: SPCs isolated from peripheral blood can be differentiated the SMCs in vitro and have an impressive growth potential in the biodegradable synthesized scaffold. Thus, SPCs may be a promising cell sointo urce for constructing TEBVs.展开更多

Five fibre feedstuffs--palm kernel...This study was conducted to investigate the effect of feeding different fibre feedstuffs on the performance, nutrient utilization and economics of production of weaning/growing pigs. Five fibre feedstuffs--palm kernel cake (PKC), wheat offal (WO), corn bran (CB), rice bran (RB) and brewers' dried grain (BDG) were used at 25% in each of the five experimental diets that were formulated in this study. Twenty weaner pigs (12.75 ± 0.6 kg) were used for the growth study while 15 pigs (14.95 ± 0.57 kg) were used for the digestibility study. The design of the experiment was completely randomized. The results of the study showed that the performance of the experimental animals were significantly influenced (P 〈 0.05) by the dietary treatments. The apparent digestibility of the crude protein, crude fiber and ether extract showed significant differences (P 〈 0.05) while there was no significant difference (P 〉 0.05) in the apparent digestibility of their dry matter, ash and nitrogen free extract. The diet that contained WO had the best growth response and apparent nutrient digestibility. Results of economics of production also showed that diet with WO gave the best (P 〈 0.05) value in feed cost/'kg gain and net profit/animal. It can be concluded from this study that although all the animals fed the different fibre feedstuffs performed satisfactorily on all the parameters monitored, but for optimum growth, apparent nutrients digestibility and economics of production. WO should be used as a fibre feedstuff in the diets of weaning/growing pigs in the tropics.展开更多

Philosophy of science has arisen as alternative to epistemology, because scientific development wanted another kind of explanation than traditional epistemological one. The latter kind of explanation is theoretically ...Philosophy of science has arisen as alternative to epistemology, because scientific development wanted another kind of explanation than traditional epistemological one. The latter kind of explanation is theoretically loaded and based on latent ontological assumptions. Epistemology offers science a "road map" for researcher's thinking. Thus epistemology knows what science should be, and philosophy of science take science for granted--existing as an empirical fact. Philosophy of science had always been a reflective and critical discipline that didn't want any a-priory knowledge. The main distinction between these fields of knowledge is as follows: epistemology is theoretical and projective kind of knowledge whereas philosophy of science is a non-theoretical, or better, a "post-theoretical" discipline. All facts and statements arising from philosophy of science lie within the heterogeneous space, where there is continuous transit of pre-theoretical, theoretical and post-theoretical versions of knowledge. The inevitability of conservation and utilization of human thought products is recognized, which is supplemented by the need to study the impact of theoretical thinking upon social practices.展开更多
