To reduce the error in transfer alignment caused by reference information delay,a time delay estimation method is developed based on least-squares curve fitting of the angular rate integration.First,the gyro sensor me...To reduce the error in transfer alignment caused by reference information delay,a time delay estimation method is developed based on least-squares curve fitting of the angular rate integration.First,the gyro sensor measurements of the main strapdown inertial navigation system(M-SINS) and the slave strapdown inertial navigation system(S-SINS) are recorded for a few seconds and the integration of the data is calculated.Then,the possible maximum range of the delay value is defined and the points of the curve at different intervals are moved.The square of the differences between the corresponding points are calculated.Finally,the delay estimation can be acquired by the least-squares curve fitting of the M-SINS and the S-SINS.A delay compensation method by local data shifting is also presented.The simulation results demonstrate the effectiveness of delay estimation and indicate that the estimation accuracy is independent of the delay value.And the local data shifting compensation method can effectively reduce the errors of the transfer alignment caused by the reference information delay.展开更多

[ Method ] The bending properties such as elastic modulus, bending strength, flexural rigidity, moment of inertia, density and water content of the second base internodes of Zhengmai 9023 and Yumai 25 were determined. [ Result] The results show that during filling stage, there are significant differences in the elastic modulus, moment of inertia, flexural rigidity and density among wheat varieties, while there are no significant differences in the bending strength and water content among wheat varieties. The moment of inertia, flexural strength and flexural rigidity have positive relationship to density but negative relationship to water content. [ Conclusion] The study results provide some references for the research on high yield cultivation and lodging resistance of wheat.展开更多

Then we propose the control mesh representation of the generalized helicoid using the AHT Bézier basis. A significance-based optimization method of the parameters was proposed based on the finite element (FE) simulation, and the significance analysis of the processing parameters on the forming quality in terms of the maximum wall thinning ratio and the maximum cross section distortion degree was implemented using the fractional factorial design. The optimum value of the significant parameter, the clearance between the tube and the wiper die, was obtained, and the values of the other parameters, including the friction coefficients and the clearances between the tube and the dies, the mandrel extension length and the boost velocity were estimated. The results are applied to aluminum alloy tube NC bending d50 mm×1 mm×75 mm and d70 mm×1.5 mm×105 mm (initial tube outside diameter D0 × initial tube wall thickness t0 × bending radius R), and qualified tubes are produced.展开更多

The proposed method can capture the buckling and post buckling behaviors of a thin plate in different geometrical and load boundary conditions. The analysis confirms that there occur snap and bifurcation behaviors in the post buckling stage of the plate. AIM:To investigate recurrent variceal hemorrhage and long-term survival rates of patients treated with partial proximal splenorenal venous shunt.METHODS:Patients with variceal hemorrhage who were treated with small-di...AIM:To investigate recurrent variceal hemorrhage and long-term survival rates of patients treated with partial proximal splenorenal venous shunt.METHODS:Patients with variceal hemorrhage who were treated with small-diameter proximal splenorenal venous shunt in Ruijin Hospital between 1996 and 2009 were included in this study.Shunt diameter was determined before operation using Duplex Doppler ultrasonography.Peri-operative and long-term results in term of rehemorrhage,encephalopathy and mortality were followed up.RESULTS:Ninety-eight patients with Child A and B variceal hemorrhage received small-diameter proximal splenorenal venous shunt with a diameter of 7-10 mm.After operation,the patients’mean free portal pressure (P<0.01)and the flow rate of main portal vein(P< 0.01)decreased significantly compared with that before operation.The rates of rebleeding and mortality were 6.12%(6 cases)and 2.04%(2 cases),respectively.Ninety-one patients were followed up for 7 mo-14 years (median,48.57 mo).Long-term rates of rehemorrhage and encephalopathy were 4.40%(4 cases)and 3.30% (3 cases),respectively.Thirteen patients(14.29%)died mainly due to progressive hepatic dysfunction.Fiveand ten-year survival rates were 82.12%and 71.24%,respectively.CONCLUSION:Small-diameter proximal splenorenal venous shunt affords protection against variceal rehemorrhage with a low occurrence of encephalopathy in patients with normal liver function.展开更多
AIM: TO investigate the role of Lactobacillus crispatus (L. crispatus) strain China Center for Type Culture Col- lection (CCTCC) M206119 in intestinal inflammation.METHODS: Forty 8-wk-old Balb/c mice (20± ...AIM: TO investigate the role of Lactobacillus crispatus (L. crispatus) strain China Center for Type Culture Col- lection (CCTCC) M206119 in intestinal inflammation.METHODS: Forty 8-wk-old Balb/c mice (20± 2 g) were divided into four groups of 10 mice each. Three groups that had received dextran sulfate sodium (DSS) were administered normal saline, sulfasalazine or CCTCC M206119 strain, and the fourth group received none of these. We assessed the severity of colitis using a disease activity index, measured the colon length and weight, collected stools and mesenteric lymph nodes for bacterial microflora analysis. One centimeter of the proximal colon, middle colon and distal colon were collected and fixed in 10% buffered formalin, dehydrated in ethanol, and embedded in paraffin. Interleukin (IL)- 1β, IL-6 and tumor necrosis factor (TNF)-α expression was detected using reverse transcription polymerase chain reaction. Protective factors zonula occludens (ZO)-1 and β-defensin 2 were detected by immunoblot-ting. The features of CCTCC M206119 strain were identified based on morphology, biochemical profile, and 16S RNA sequencing.RESULTS: DSS-colitis animals treated with CCTCC M206119 had markedly more severe disease, with greater weight loss, diarrhea, fecal bleeding, and shortened colon length. In addition, the CCTCC-M206119- treated group had comparatively higher histologi- cal scores and more neutrophil infiltration than the controls. Expression of protective factors ZO-1 and β-defensin 2 was downregulated due to destruction of the mucosal barrier after CCTCC M206119 strain treatment. An in vitro assay demonstrated that CCTCC M206119 strain increased the nuclear translocation of nuclear factor-κB in epithelial cells. Intestinal proinflam- matory or anti-inflammatory cytokine responses were evaluated. Proinflammatory colonic cytokine (IL-Iβ, IL-6 and TNF-α) levels were clearly increased in CCTCC- M206119-treated animals, whereas anti-inflammatory colonic cytokine (IL-10) level was lowered compared with saline or 5-aminosalicylic-acid-treated DSS-colitis mice. Next, CCTCC M206119 strain was characterized as 1. crispatus by microscopic morphology, biochemical tests and 16S rRNA gene level.CONCLUSION: Not all lactobacilli are beneficial for in- testinal inflammation, and L. crispatus CCTCC M206119 strain is involved in exacerbation of intestinal inflamma- tion in DSS-colitis mice.展开更多
Five multiparameter empirical criteria were exclusively evaluated by comparing them with the strength data covering various stress conditions to find out which failure criterion best fits the test data and describes t...Five multiparameter empirical criteria were exclusively evaluated by comparing them with the strength data covering various stress conditions to find out which failure criterion best fits the test data and describes the mechanical behavior of the salt rock sequence (halite,bedded composite specimens and anhydrite interlayers).Full-scale comparison of all criteria for the three rock types was conducted based on five standard statistics calculated from least squares curve-fitting,which measures both the goodness of fitting and the quality of future prediction.The results indicate that all five nonlinear criteria with a basic power form are efficient in predicting the strength trend in the low tension area as well as in the high compression area of the soft rocks.The parameters obtained for the bedded rock salt are somewhat in the ones for the "pure" rocks and are even closer to those obtained for the halite.The generalized Hoek-Brown criterion is proven to perform best to two rock strength data followed by one for the Bieniawski empirical criterion,thus is the best candidate for the analysis of the salt rock.The Sheorey empirical criterion consistently achieves an intermediate performance for all the three rocks.It seems that the superiority of the poly-axial criteria (the Mogi 1967 criterion and the N-type criterion) over the former three triaxial criteria no longer exists when applied to the conventional triaxial strength data.Besides,the method of tension cut-off was proposed to solve the ambiguity problem of the two poly-axial criteria in the tension field in the plane of the major (σ1) andminor principal stress (σ3).展开更多

The moving least square method is introduced, for the first time, to fit a stratum surface. The results show that, using the same-degree base function, compared with a traditional least square method, the moving least square method can produce lower fitting errors, the fitting surface can describe the morphological characteristics of stratum surfaces more accurately and the principal curvature values vary within a wide range and may be more suitable for the prediction of the distribution of structural fractures. The moving least square method could be useful in curved surface fitting and stratum curvature analysis.展开更多

The Horton equation is one of the popular models used in the characterization of field infiltration. In this study, the least square curve firing technique was employed to estimate the model parameters from fifteen field measured data and gave resultant mean regression coefficients (R2) value of 0.811. Furthermore, plotting the measured against the calculated infiltration rate for the first six (6) measurement points yielded R2 values close to unity in the regression curve indicating a marked relationship between the two. This indicates that the Horton infiltration model can be applied to estimate infiltration characteristics of soils in Samaru, Northern Guinea Savanna of Nigeria.展开更多

METHODS:Ten healthy volunteers were enrolled in this study.The subjects were divided into two groups:a placebo group given low-dose aspirin plus placebo and a rebamipide group given low-dose aspirin plus rebamipide for a period of 14 d.Capsule endoscopy and contrast-enhanced ultrasonography were performed before and after administration of drugs.Areas under the curves and peak value of time-intensity curve were calculated. RESULTS:Absolute differences in areas under the curves were-1102.5(95%CI:-1980.3 to-224.7,P=0.0194) in the placebo group and-152.7(95%CI:-1604.2 to 641.6,P=0.8172) in the rebamipide group. Peak values of time intensity curves were-148.0(95% CI:-269.4 to-26.2,P=0.0225) in the placebo group and 28.3(95%CI:-269.0 to 325.6,P=0.8343) in the rebamipide group.Capsule endoscopy showed mucosal breaks only in the placebo group. CONCLUSION:Short-term administration of low-dose aspirin is associated with small bowel injuries and blood flow.展开更多
Colonoscopy sometimes causes pain during insertion,especially in difficult cases.Over-insufflation of air causes elongation or acute angulations of the colon,making passage of the scope difficult and causing pain.We p...Colonoscopy sometimes causes pain during insertion,especially in difficult cases.Over-insufflation of air causes elongation or acute angulations of the colon,making passage of the scope difficult and causing pain.We previously reported a sedative-risk-free colonoscopy insertion technique,namely,"Water Navigation Colonoscopy".Complete air suction after water infusion not only improves the vision,but also makes water flow down to the descending colon,while the sigmoid colon collapses and shortens.While non-sedative colonoscopy can be carried out without pain in most cases,some patients do complain of pain.Most of these patients have abnormal colon morphology,and the pain is caused while negotiating the "hairpin" bends of the colon.The "hairpin" bends of the colon should be negotiated by gently pushing the full-angled colonoscope.The proximal 10-20 cm from the angulated part of the conventional colonoscope is stiff,with a wide turning radius,therefore,a conventional colonoscope cannot be negotiated through the "hairpin" bends of the colon without stretching them and causing pain.The "passive-bending colonoscope" has a flexible tip with a narrow turning radius,so that the scope can be negotiated through the "hairpin" bends of the colon with a minimum turning radius and minimal discomfort.Therefore,the intubation and pain-reducing performance of the "passive-bending colonoscope" was assessed in difficult cases.展开更多

An innovative approach for the identification of cracks from the dynamic responses of girder bridges was proposed.One of the key steps of the approach was to transform the dynamical responses into the equivalent stati...An innovative approach for the identification of cracks from the dynamic responses of girder bridges was proposed.One of the key steps of the approach was to transform the dynamical responses into the equivalent static quantities by integrating the excitation and response signals over time.A sliding-window least-squares curve fitting technique was then utilized to fit a cubic curve for a short segment of the girder.The moment coefficient of the cubic curve can be used to detect the locations of multiple cracks along a girder bridge.To validate the proposed method,prismatic girder bridges with multiple cracks of various depths were analyzed.Sensitivity analysis was conducted on various effects of crack depth,moving window width,noise level,bridge discretization,and load condition.Numerical results demonstrate that the proposed method can accurately detect cracks in a simply-supported or continuous girder bridges,the five-point equally weighted algorithm is recommended for practical applications,the spacing of two discernable cracks is equal to the window length,and the identified results are insensitive to noise due to integration of the initial data.展开更多
Ten seed borne fungi (Alternaria sp., Aspergillus sp., Aureobasidium sp., Cladosporium sp., Dreschslera sp., Penicillium sp., Rhizoctonia sp., Stemphylium sp., Mueor sp. and Rhizopus sp.) were isolated and identifie...Ten seed borne fungi (Alternaria sp., Aspergillus sp., Aureobasidium sp., Cladosporium sp., Dreschslera sp., Penicillium sp., Rhizoctonia sp., Stemphylium sp., Mueor sp. and Rhizopus sp.) were isolated and identified from two wheat varieties, the highest frequency of seed borne fungi was observed on wheat cultivar site Moll4 Alternaria sp.. Their mean and standard deviation was (5.5 ~ 1.69) while the lowest frequency fungal isolated was Dreschslera sp. and Rhizopus sp.. Their mean and standard deviation was (0.1 ~ 0.64). The aflatoxin-producing isolates appeared as gray or black colonies in the UV photographs, whereas nonproducing isolates appeared as white colonies, the plate five colony four (P5CO4) showed the positive results which means the presence of aflatoxin as compaired to the control which showed the ngative results. Ammonium Hydroxide Vapor-Induced Color Change method used which the dish was inverted and 1 or 2 drops of concentrated ammonium hydroxide solution are placed on the inside of the lid. The undersides of aflatoxin-producing colonies quickly turn plum-red after the bottom of the Petri dish has been inverted over the lid containing the ammonium hydroxide aspositive result in (P5CO4) and (P7CO4) observed. Essentially no color change occurs on the undersides of colonies that are not producing aflatoxinsthis indicate to the negative results (control). The main objective of this study is to isolation, identification and rapid detection of aflatoxin from wheat seed borne fungi.展开更多

To interfere with the potentially effective target, plasmid named p-mTNFRlshRNA complimentary to the sequence responsible for mTNFR1 was also constructed and further confirmed by sequence analysis. To investigate the effect of mTNFRlshRNA plasmid on mTNFR1 expression in vivo and the disease progress in MHV-3 induced fulminant hepatitis mice model. By hydrodynamic injection of mTNFRlshRNA plasmid, the survival rate of mice, hepatic pathological change were examined and compared between mice with/without mTNFRlshRNA plasmid intervention. The expression of mTNFR1 was detected by Real-time PCR, immunohistochemistry staining. The mTNFRlshRNA plasmid significantly reduced mTNFR1 expression in vivo, markedly ameliorates inflammatory infiltration, prolonged the survival time period and elevated the survival rate from 0 up to 13.3% in Balb/cJ mice with MHV-3 induced fulminant hepatitis. This study was designed to explore the opportunity of RNA interference technique in inhibiting TNFR1 expression, which has been reported to be involved in the development of a variety of diseases including fulminant viral hepatitis and severe chronic hepatitis B.展开更多

The proposed method can capture the buckling and post buckling behaviors of a thin plate in different geometrical and load boundary conditions. The analysis confirms that there occur snap and bifurcation behaviors in the post buckling stage of the plate. And these results show the validity of the variational method for solving buckling problems of thin plate.
文摘AIM:To investigate recurrent variceal hemorrhage and long-term survival rates of patients treated with partial proximal splenorenal venous shunt.METHODS:Patients with variceal hemorrhage who were treated with small-diameter proximal splenorenal venous shunt in Ruijin Hospital between 1996 and 2009 were included in this study.Shunt diameter was determined before operation using Duplex Doppler ultrasonography.Peri-operative and long-term results in term of rehemorrhage,encephalopathy and mortality were followed up.RESULTS:Ninety-eight patients with Child A and B variceal hemorrhage received small-diameter proximal splenorenal venous shunt with a diameter of 7-10 mm.After operation,the patients’mean free portal pressure (P<0.01)and the flow rate of main portal vein(P< 0.01)decreased significantly compared with that before operation.The rates of rebleeding and mortality were 6.12%(6 cases)and 2.04%(2 cases),respectively.Ninety-one patients were followed up for 7 mo-14 years (median,48.57 mo).Long-term rates of rehemorrhage and encephalopathy were 4.40%(4 cases)and 3.30% (3 cases),respectively.Thirteen patients(14.29%)died mainly due to progressive hepatic dysfunction.Fiveand ten-year survival rates were 82.12%and 71.24%,respectively.CONCLUSION:Small-diameter proximal splenorenal venous shunt affords protection against variceal rehemorrhage with a low occurrence of encephalopathy in patients with normal liver function.
文摘AIM: TO investigate the role of Lactobacillus crispatus (L. crispatus) strain China Center for Type Culture Col- lection (CCTCC) M206119 in intestinal inflammation.METHODS: Forty 8-wk-old Balb/c mice (20± 2 g) were divided into four groups of 10 mice each. Three groups that had received dextran sulfate sodium (DSS) were administered normal saline, sulfasalazine or CCTCC M206119 strain, and the fourth group received none of these. We assessed the severity of colitis using a disease activity index, measured the colon length and weight, collected stools and mesenteric lymph nodes for bacterial microflora analysis. One centimeter of the proximal colon, middle colon and distal colon were collected and fixed in 10% buffered formalin, dehydrated in ethanol, and embedded in paraffin. Interleukin (IL)- 1β, IL-6 and tumor necrosis factor (TNF)-α expression was detected using reverse transcription polymerase chain reaction. Protective factors zonula occludens (ZO)-1 and β-defensin 2 were detected by immunoblot-ting. The features of CCTCC M206119 strain were identified based on morphology, biochemical profile, and 16S RNA sequencing.RESULTS: DSS-colitis animals treated with CCTCC M206119 had markedly more severe disease, with greater weight loss, diarrhea, fecal bleeding, and shortened colon length. In addition, the CCTCC-M206119- treated group had comparatively higher histologi- cal scores and more neutrophil infiltration than the controls. Expression of protective factors ZO-1 and β-defensin 2 was downregulated due to destruction of the mucosal barrier after CCTCC M206119 strain treatment. An in vitro assay demonstrated that CCTCC M206119 strain increased the nuclear translocation of nuclear factor-κB in epithelial cells. Intestinal proinflam- matory or anti-inflammatory cytokine responses were evaluated. Proinflammatory colonic cytokine (IL-Iβ, IL-6 and TNF-α) levels were clearly increased in CCTCC- M206119-treated animals, whereas anti-inflammatory colonic cytokine (IL-10) level was lowered compared with saline or 5-aminosalicylic-acid-treated DSS-colitis mice. Next, CCTCC M206119 strain was characterized as 1. crispatus by microscopic morphology, biochemical tests and 16S rRNA gene level.CONCLUSION: Not all lactobacilli are beneficial for in- testinal inflammation, and L. crispatus CCTCC M206119 strain is involved in exacerbation of intestinal inflamma- tion in DSS-colitis mice.

文摘Five multiparameter empirical criteria were exclusively evaluated by comparing them with the strength data covering various stress conditions to find out which failure criterion best fits the test data and describes the mechanical behavior of the salt rock sequence (halite,bedded composite specimens and anhydrite interlayers).Full-scale comparison of all criteria for the three rock types was conducted based on five standard statistics calculated from least squares curve-fitting,which measures both the goodness of fitting and the quality of future prediction.The results indicate that all five nonlinear criteria with a basic power form are efficient in predicting the strength trend in the low tension area as well as in the high compression area of the soft rocks.The parameters obtained for the bedded rock salt are somewhat in the ones for the "pure" rocks and are even closer to those obtained for the halite.The generalized Hoek-Brown criterion is proven to perform best to two rock strength data followed by one for the Bieniawski empirical criterion,thus is the best candidate for the analysis of the salt rock.The Sheorey empirical criterion consistently achieves an intermediate performance for all the three rocks.It seems that the superiority of the poly-axial criteria (the Mogi 1967 criterion and the N-type criterion) over the former three triaxial criteria no longer exists when applied to the conventional triaxial strength data.Besides,the method of tension cut-off was proposed to solve the ambiguity problem of the two poly-axial criteria in the tension field in the plane of the major (σ1) andminor principal stress (σ3).

文摘With the east section of the Changji sag Zhunger Basin as a case study, both a principal curvature method and a moving least square method are elaborated. The moving least square method is introduced, for the first time, to fit a stratum surface. The results show that, using the same-degree base function, compared with a traditional least square method, the moving least square method can produce lower fitting errors, the fitting surface can describe the morphological characteristics of stratum surfaces more accurately and the principal curvature values vary within a wide range and may be more suitable for the prediction of the distribution of structural fractures. The moving least square method could be useful in curved surface fitting and stratum curvature analysis.
文摘Field infiltration measurement is often a tedious task thus can be easily estimated from proposed infiltration models. The Horton equation is one of the popular models used in the characterization of field infiltration. In this study, the least square curve firing technique was employed to estimate the model parameters from fifteen field measured data and gave resultant mean regression coefficients (R2) value of 0.811. Furthermore, plotting the measured against the calculated infiltration rate for the first six (6) measurement points yielded R2 values close to unity in the regression curve indicating a marked relationship between the two. This indicates that the Horton infiltration model can be applied to estimate infiltration characteristics of soils in Samaru, Northern Guinea Savanna of Nigeria.

文摘AIM:To investigate the relationship between low-dose aspirin-induced small bowel mucosal damage and blood flow,and the effect of rebamipide. METHODS:Ten healthy volunteers were enrolled in this study.The subjects were divided into two groups:a placebo group given low-dose aspirin plus placebo and a rebamipide group given low-dose aspirin plus rebamipide for a period of 14 d.Capsule endoscopy and contrast-enhanced ultrasonography were performed before and after administration of drugs.Areas under the curves and peak value of time-intensity curve were calculated. RESULTS:Absolute differences in areas under the curves were-1102.5(95%CI:-1980.3 to-224.7,P=0.0194) in the placebo group and-152.7(95%CI:-1604.2 to 641.6,P=0.8172) in the rebamipide group. Peak values of time intensity curves were-148.0(95% CI:-269.4 to-26.2,P=0.0225) in the placebo group and 28.3(95%CI:-269.0 to 325.6,P=0.8343) in the rebamipide group.Capsule endoscopy showed mucosal breaks only in the placebo group. CONCLUSION:Short-term administration of low-dose aspirin is associated with small bowel injuries and blood flow.
文摘Colonoscopy sometimes causes pain during insertion,especially in difficult cases.Over-insufflation of air causes elongation or acute angulations of the colon,making passage of the scope difficult and causing pain.We previously reported a sedative-risk-free colonoscopy insertion technique,namely,"Water Navigation Colonoscopy".Complete air suction after water infusion not only improves the vision,but also makes water flow down to the descending colon,while the sigmoid colon collapses and shortens.While non-sedative colonoscopy can be carried out without pain in most cases,some patients do complain of pain.Most of these patients have abnormal colon morphology,and the pain is caused while negotiating the "hairpin" bends of the colon.The "hairpin" bends of the colon should be negotiated by gently pushing the full-angled colonoscope.The proximal 10-20 cm from the angulated part of the conventional colonoscope is stiff,with a wide turning radius,therefore,a conventional colonoscope cannot be negotiated through the "hairpin" bends of the colon without stretching them and causing pain.The "passive-bending colonoscope" has a flexible tip with a narrow turning radius,so that the scope can be negotiated through the "hairpin" bends of the colon with a minimum turning radius and minimal discomfort.Therefore,the intubation and pain-reducing performance of the "passive-bending colonoscope" was assessed in difficult cases.

文摘An innovative approach for the identification of cracks from the dynamic responses of girder bridges was proposed.One of the key steps of the approach was to transform the dynamical responses into the equivalent static quantities by integrating the excitation and response signals over time.A sliding-window least-squares curve fitting technique was then utilized to fit a cubic curve for a short segment of the girder.The moment coefficient of the cubic curve can be used to detect the locations of multiple cracks along a girder bridge.To validate the proposed method,prismatic girder bridges with multiple cracks of various depths were analyzed.Sensitivity analysis was conducted on various effects of crack depth,moving window width,noise level,bridge discretization,and load condition.Numerical results demonstrate that the proposed method can accurately detect cracks in a simply-supported or continuous girder bridges,the five-point equally weighted algorithm is recommended for practical applications,the spacing of two discernable cracks is equal to the window length,and the identified results are insensitive to noise due to integration of the initial data.
文摘Ten seed borne fungi (Alternaria sp., Aspergillus sp., Aureobasidium sp., Cladosporium sp., Dreschslera sp., Penicillium sp., Rhizoctonia sp., Stemphylium sp., Mueor sp. and Rhizopus sp.) were isolated and identified from two wheat varieties, the highest frequency of seed borne fungi was observed on wheat cultivar site Moll4 Alternaria sp.. Their mean and standard deviation was (5.5 ~ 1.69) while the lowest frequency fungal isolated was Dreschslera sp. and Rhizopus sp.. Their mean and standard deviation was (0.1 ~ 0.64). The aflatoxin-producing isolates appeared as gray or black colonies in the UV photographs, whereas nonproducing isolates appeared as white colonies, the plate five colony four (P5CO4) showed the positive results which means the presence of aflatoxin as compaired to the control which showed the ngative results. Ammonium Hydroxide Vapor-Induced Color Change method used which the dish was inverted and 1 or 2 drops of concentrated ammonium hydroxide solution are placed on the inside of the lid. The undersides of aflatoxin-producing colonies quickly turn plum-red after the bottom of the Petri dish has been inverted over the lid containing the ammonium hydroxide aspositive result in (P5CO4) and (P7CO4) observed. Essentially no color change occurs on the undersides of colonies that are not producing aflatoxinsthis indicate to the negative results (control). The main objective of this study is to isolation, identification and rapid detection of aflatoxin from wheat seed borne fungi.

文摘Previous study on TNFRl-mediated hepatocyte apoptosis has been implicated in the development of fulminant viral hepatitis. To interfere with the potentially effective target, plasmid named p-mTNFRlshRNA complimentary to the sequence responsible for mTNFR1 was also constructed and further confirmed by sequence analysis. To investigate the effect of mTNFRlshRNA plasmid on mTNFR1 expression in vivo and the disease progress in MHV-3 induced fulminant hepatitis mice model. By hydrodynamic injection of mTNFRlshRNA plasmid, the survival rate of mice, hepatic pathological change were examined and compared between mice with/without mTNFRlshRNA plasmid intervention. The expression of mTNFR1 was detected by Real-time PCR, immunohistochemistry staining. The mTNFRlshRNA plasmid significantly reduced mTNFR1 expression in vivo, markedly ameliorates inflammatory infiltration, prolonged the survival time period and elevated the survival rate from 0 up to 13.3% in Balb/cJ mice with MHV-3 induced fulminant hepatitis. This study was designed to explore the opportunity of RNA interference technique in inhibiting TNFR1 expression, which has been reported to be involved in the development of a variety of diseases including fulminant viral hepatitis and severe chronic hepatitis B.
