AIM:To characterize the differentially expressed gene profiles in livers from biliary atresia (BA) patients including,ascertain genes,functional categories and pathways that play a central role in the pathogenesis of ...AIM:To characterize the differentially expressed gene profiles in livers from biliary atresia (BA) patients including,ascertain genes,functional categories and pathways that play a central role in the pathogenesis of BA,and identify the novel prognostic markers for BA.METHODS:Liver tissue samples from control patients,neonatal cholestasis patients,and BA patients at the age of < 60 d,60-90 d,and > 90 d were pooled for DNA microarray analysis.Bioinformatics analysis was performed using,series test cluster of gene ontology,and Pathway-Finder software.Reverse-transcription polymerase chain reaction was performed to confirm changes in selected genes.Relation between RRAS gene expression and prognosis of 40 BA patients was analyzed in a 2-year follow-up study.RESULTS:The 4 identified significant gene expression profiles could confidently separate BA liver tissue from normal and other diseased liver tissues.The included genes were mainly involved in inflammation response and reconstruction of cellular matrix.The significant pathways associated with BA were primarily involved in autoimmune response,activation of T lymphocytes and its related cytokines.The RRAS,POMC,SLC26A6 and STX3 genes were important regulatory modules in pathogenesis of BA.The expression of RRAS was negatively correlated with the elimination rate of jaundice and positively correlated with the occurrence rate of cholangitis.CONCLUSION:Autoimmune response mediated by T lymphocytes may play a vital role in the pathogenesis of BA.The RRAS gene is an important regulatory module in the pathogenesis of BA,which may serve as a novel prognostic marker for BA.展开更多

Methods: Retrospective study of 7 patients with hepatic angiosarcoma. Results: Symptoms and signs such as abdominal pain were extremely nonspecific. The levels of the tumor markers were within the normal range. Four cases were positive for hepatitis B viruses. Among the 5 patients who received chemotherapy, improvement in survival was seen in 3 cases which also received surgery. Conclusion: Hepatic angiosarcoma is a rare and diffficult-to-diagnose malignant liver tumor. Surgery is the definitive treatment and adjuvant chemotherapy after surgery may improve survival.展开更多

Methods: Artificially synthesize biotin labeled peptide ZS-9, anchored ZS-9 in the enzyme label plate coupled by avidin, used ZS-9 as probe to screen cDNA library of human lung cancer, after screening, obtained bacteriophage clone specifically binding with anchored polypeptide ZS-9. Extracted plasmid of bacteriophage and performed sequencing after amplified by PCR. Results: It was demonstrated by bioinformatic analysis on the sequence of ligand binded by lung cancer specific peptide ZS-9 that the ligand was the cytoskeletal protein periplakin on the surface of lung cancer cells, suggesting that periplakin might be a new marker for non-small-cell lung cancer in lung cancer. Conclusion: Use specific lung cancer binding peptide to screen new tumor marker periplakin in lung cancer and further studies on its biologic functions in genesis and development of lung cancer are still needed.展开更多
To determine geochemical indicators for depositional environment favored by terrestrial petroleum source rocks, we selected 40 source rock samples from the Late Cretaceous Qingshankou Formation (Kzqn) and the first ...To determine geochemical indicators for depositional environment favored by terrestrial petroleum source rocks, we selected 40 source rock samples from the Late Cretaceous Qingshankou Formation (Kzqn) and the first member of Nenjiang Formation (K2n1) in the Songliao Basin to qualify saturate fraction and aromatic fraction using GC-HRT (gas chromatography high resolution time-of-flight mass spectrometry) and quantify important biomarkers using GC-MS. The results reveal that source rocks from the 1st member of Qingshankou Formation (K2qn1) are characterized by not only high contents of terpanes, regular steranes and 4-methylsteranes but also high contents of dinosteranes, C31 steranes and aryl isoprenoids. Presence of specific biomarkers like elementary sulfur and lanostanes indicates a depositional environment of lagoon characterized by water stratification and high salinity. In the 2nd-3rd members of Qingshankou Formation (K2qn2+3), source rocks contain lower contents of biomarkers, indicating a depositional environment of shallow fresh-water lake delta. Source rocks in the K2n1 contain high contents of terpanes, regular steranes and 4-methylsteranes but lower contents of dinosteranes, C31 steranes and aryl isoprenoids, indicating a depositional environment of fresh-brackish open lake characterized by low salinity and poor water stratification, where organic matter is seriously altered by bacteria. Overall analysis shows that primary geochemical indicators for terrestrial petroleum source rocks are as follows: 1) C30 hopanes 〉 1500 ppm; 2) gammacerane 〉190 ppm; 3) C27 steranes 〉200 ppm; 4) 4-methylsteranes 〉 100 ppm; 5) aryl isoprenoids 〉 3 ppm; 6) dehydroxyl-vitamin E 〉 10 ppm.展开更多
Induced pluripotent stem cells (iPSCs) are an attractive cell source for regenerative medicine through cell therapy or drug screening. But application of iPSCs in regenerative medicine requires rapid and accurate ch...Induced pluripotent stem cells (iPSCs) are an attractive cell source for regenerative medicine through cell therapy or drug screening. But application of iPSCs in regenerative medicine requires rapid and accurate charac- terization of iPSCs. Here, we demonstrate the detection of multiple antigens present in iPSC lysate using rapid, label- free surface plasmon resonance imaging (SPRi) assay. Validation of pluripotency is an important aspect of iPSC research. In this study, we fabricated antibody array against pluripotency biomarkers and found that our array suc- cessfully detect corresponding antigens in stem cell lysate. Each antibody recognized its specific antigens presented in iPSC lysates and a certain degree of variability was observed in comparison with other cell lysates. The results suggested that SPRi is a versatile technology feasible for the detection of multiple antigens presented in iPSC lysate. Further extension of this method may be applied in the characterization and high-throughput biomarker profiling of iPSCs.展开更多
